An ongoing advancement in forensic trace evidence has driven the development of new and objective methods for comparing various materials. While many standard guides have been published for use in trace laboratories, different areas require a more comprehensive understanding of error rates and an urgent need for harmonizing methods of examination and interpretation. Two critical areas are the forensic examination of physical fits and the comparison of spectral data, which depend highly on the examiner’s judgment.

Coronavirus disease-2019 (COVID-19), which is caused by a novel coronavirus named severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), has ravaged the world for the past 3 years. Even today, there still exists a need for rapid, accurate, economical and non-invasive diagnostic testing platforms that yield high specificity and sensitivity towards the constantly mutating SARS-CoV-2. This attention is attributed to their low cost and excellent optoelectronic properties, including large absorption coefficients, tunable bandgaps, long charge-carrier diffusion lengths, and low densities of deep trap states. Phosphorus (P) is a fundamental element necessary for all life forms and a key component in the fertilizer industry. Meanwhile, the excessive load of P to water bodies due to human activities has the potential to promote eutrophication. Wastewater treatment plants remove P either biologically or chemically and produce P rich sludge which could be a potential renewable source for P. At present, commercial technologies exist for P recovery from biological wastewater sludge. Using a multiplatform and multiomics approach, we identified metabolites, lipids, proteins, and metabolic pathways that were altered in the liver after chronic ethanol administration. A functional enrichment analysis of the multiomics dataset revealed that rats treated with ethanol experienced an increase in hepatic fatty acyl content, which is consistent with an initial development of steatosis. The nuclear magnetic resonance spectroscopy (NMR) and liquid chromatography–mass spectrometry (LC-MS) metabolomics data revealed that the chronic ethanol exposure selectively modified toxic substances such as an increase in glucuronidation tyramine and benzoyl; and a depletion in cholesterol-conjugated glucuronides. Total ammonia nitrogen (TAN) is a pollutant agent found in water and generated from the metabolisms of living organisms and the decomposition of organic matter. This research aims to develop an efficient, low-cost, and environmentally friendly TAN analysis method. The principle of this method is based on the reaction of TAN injected in the donor stream with NaOH to form alkaline gaseous ammonia. The gaseous ammonia diffuses through a hydrophobic membrane into an acceptor stream containing Ruellia tuberosa extract. The growing demand of society for gas sensors for energy-efficient environmental sensing stimulates studies of new electronic materials. Here, we investigated quasi-one-dimensional titanium trisulfide (TiS3) crystals for possible applications in chemiresistors and on-chip multisensor arrays. TiS3 nanoribbons were placed as a mat over a multielectrode chip to form an array of chemiresistive gas sensors. The severity of pancreatic ductal adenocarcinoma (PDAC) is largely attributed to a failure to detect the disease before metastatic spread has occurred. CA19-9, a carbohydrate biomarker, is used clinically to surveille disease progression, but due to specificity challenges is not suitable for early discovery. As CA19-9 and other prospective markers are glycan epitopes, there is great clinical interest in understanding the glycobiology of pancreatic cancer. The widespread pollution of mercury motivates research into its atmospheric chemistry and transport. Gaseous elemental mercury (Hg(0)) dominates mercury emission to the atmosphere, but the rate of its oxidation to mercury compound (Hg(II)) plays a significant role in controlling where and when mercury deposits to ecosystems. The major focus of this thesis was to explore methods for the detection, identification, and characterization of organic gunshot residues (OGSRs) for forensic purposes using various spectroscopic techniques. There are four major projects detailed in this thesis. The first project details the development of a novel-two step method for the detection and identification of organic gunshot residues using fluorescence spectroscopy for detection and Raman spectroscopy as a confirmatory step. Non-muscle Invasive Bladder Cancer (NMIBC) accounts for 80% of all bladder cancers. Although it is mostly low-grade tumors, its high recurrence rate necessitates three-times-monthly follow-ups and cystoscopy examinations to detect and prevent its progression. A rapid liquid biopsy-based assay is needed to improve detection and reduce complications from invasive cystoscopy. Here, we present a rapid spectroscopic method to detect the recurrence of NMIBC in urine. Urine samples from previously-diagnosed NMIBC patients (n = 62) were collected during their follow-up visits before cystoscopy examination. The degradation of petroleum-derived organic pollutants in aquatic systems occurs through lengthened exposure to UV and visible light. In this thesis, organic pollutants include crude oil, refined fuels, and plastic particles or films. The photo-oxidation of these pollutants are monitored over natural waters. To characterize the molecular signatures of photo-oxidized petroleum, bench-scale spills of refined fuels and crude oil were irradiated over Alaskan waters. A 4-component fluorescence PARAFAC model revealed a unique feature associated with photo-oxidized refined fuel unlike traditional “microbial”- or “terrestrial-like” components. In contrast, crude oil photolytically decomposed into humic-like components and oxidized aliphatics. FT-ICR MS data corroborated …

The role of acids in the formation of emulsions from sunlight exposed petroleum is not well understood. To elucidate the impact of sunlight on the formation of emulsions from petroleum, Macondo, its surrogate and NIST 1619b and 2717a were separately exposed to simulated sunlight for up to 48 hours as a film dispersed into a quartz dish, or a film spread on water. Emulsion stability was scored from 0-5. All exposures demonstrated a correlation between emulsion stability and sunlight exposure with no observable emulsification in absence of photoproduction due to sunlight exposure. Ultramicroelectrodes (UMEs) are useful as probes for evaluating electroactive species in confined spaces (e.g., inside living cells) and for measuring fast electrochemical reactions. However, UME applications often require modification of the electrode surface to improve selectivity and sensitivity towards target analytes. Previous research in our group demonstrated that a simple soft nitriding method introduces surface nitrogen (N)-containing groups on carbon fiber (CF), leading to improved electroreduction of hydrogen peroxide (H2O2) on CF-UMEs. The peroxidation of unsaturated fatty acids is a widely recognized metabolic process that creates a complex mixture of volatile organic compounds including aldehydes. Elevated levels of reactive oxygen species in cancer cells promote random lipid peroxidation, which leads to an increase in a variety of aldehydes. Many of these volatile aldehydes are exhaled and are of interest as potential markers of disease. Chapter 1 presents a review of reported aldehydes in the exhaled breath of lung cancer patients. alpha,beta-Unsaturated aldehydes, detected primarily when derivatized during exhaled breath preconcentration, are underreported in the reviewed articles. Overall, the objective of this dissertation was to investigate the degree of sorption for the alkali cations on rutile to ascertain the impact of different cation properties, such as ion size and charge density, on sorption mechanics as well as probe how the ion may alter the surface – aqueous interface. Initial molecular dynamic simulations and batch experiments showed minimal surface sorption for any alkali cation at relatively low concentrations while simultaneously suggesting the enthalpy of deprotonation shifts slightly in the presence of the alkali cations at different ionic strengths. We have successfully designed a uranyl ion (U(VI)-specific peptide and used it in the fabrication of an electrochemical sensor. The 12-amino acid peptide sequence, (n) DKDGDGYIpTAAE (c), originates from calmodulin, a Ca(II)-binding protein, and contains a phosphothreonine that enhances the sequence’s affinity for U(VI) over Ca(II). The sensing mechanism of this U(VI) sensor is similar to other electrochemical peptide-based sensors, which relies on the change in the flexibility of the peptide probe upon interacting with the target. Anthropogenic impacts on marine environments can impact metal fluxes and concentrations available to marine species. Monitoring these impacts is necessary to better understand the interactions between the biotic and abiotic components of these ecosystems and mitigate the risk posed by harmful toxins introduced by human activities. Biomoniters, like macroscopic algae, are useful indicators that illuminate the bioaccumulation of toxins commonly introduced from anthropogenic activity. Pyrrole-imidazole polyamides (PIPs) are nanomolecular compounds designed to fit in the tight space of DNA minor grooves. As analogs of Netropsin and Distamycin A, PIPs are specifically designed to recognize the base pairs of DNA sequences. PIPs have many biological applications, such as regulating gene expression, biochemistry pathway regulations, and suppressing the development of cancer cells. SETMAR gene is the chimeric fusion of the SET domain with the mariner transposase. Its protein (Metnase) has functions involving DNA repairs in the NHEJ pathway, regulating gene expression, DNA decatenation, etc. Despite not having an active transposable element, SETMAR still has an unknown …

Ethylene is an important chemical feedstock for production of polymers and high-value organic chemicals, and yet its conventional purification process is plagued with high consumption of energy. Metal−organic frameworks (MOFs) provide a suitable adsorption platform for selective ethane/ ethylene separation thanks to their structural diversity, tunable pore characteristics, designable pore sizes, and high pore volumes. Although there are empirical design rules like avoiding open metal sites and creating nonpolar pore surfaces for development of adsorptive MOFs, it is still challenging to design robust MOFs that can realize direct ethane-selective separation. Determining the amount of inorganic ions in aqueous media is important in the food industry and environmentally. In the environment high levels of ions like nitrate (NO3-), nitrite (NO2-), and phosphate (PO43-) can lead to unhealthy conditions for aquatic species. Similarly, high levels of nitrate and nitrite can cause health issues in humans. Fourier transform infrared spectroscopy-attenuated total reflectance (FTIR–ATR) was used to analyze pure and adulterated honey samples. The FTIR spectra was analyzed using principal component analysis (PCA) and partial least squares (PLS) regression analysis to determine if these methods could differentiate between pure, commercial, and sucrose-adulterated honey samples. PCA showed a clear distinction between pure and adulterated honey samples. Commercial honey samples showed clustering around the unadulterated samples. PLS regression analysis correctly identified 81.8% of the standards and samples used in the PCA analysis. The five commercial samples were tested and shown to have a concentration of less than 3% adulterant, …

Pancreatic cancer remains one of the deadliest forms of cancer with a 5-year survival rate of only 11%. Difficult diagnosis and limited treatment options are the major causes of the poor outcome for pancreatic cancer. The human protein DNAJA1 has been proposed as a potential therapeutic target for pancreatic cancer, but its cellular and biological functions remain unclear. Previous studies have suggested that DNAJA10s cellular activity may be dependent upon its protein binding partners. Louise Nevelson’s large-scale, matte black assemblage sculpture Dawn’s Image, Night, 1969, is owned by and currently on display at SUNY Buffalo State College. The paint layers exhibited significant amounts of dust, some damages, vandalism, and unsightly fingerprints. Extensive scientific analysis and archival research were utilized to design an appropriate treatment and long-term preservation plan. Methods of analysis include: Fourier transform infrared spectroscopy, pyrolysis-gas chromatography-mass spectrometry, microchemical testing, and xray fluorescence spectroscopy. The project also addresses the ethical considerations surrounding the prior removal of original components for safety considerations, repainting as a possible treatment, and the overall obstacles of treating a …

Sensitized Photooxidation Of Prenylated Compounds: Mechanisms Of Downstream Dark Effects And Phototoxicity Priming, Shakeela Jabeen

This thesis consists of four chapters as detailed below.
Chapter 1 discusses a singlet oxygen priming mechanism. Airborne singlet oxygen derived from photosensitization of triplet dioxygen is shown to react with an alkene surfactant (8-methylnon-7-ene-1 sulfonate) leading to ‘ene’ hydroperoxides that in the dark inactivate planktonic E. coli. The ‘ene’ hydroperoxide photoproducts are not toxic on their own, but they become toxic after the bacteria are pretreated with singlet oxygen. The total quenching rate constant (kT) of singlet oxygen of the alkene surfactant was measured to be 1.1 × 106 M−1 s− …
