Ion Mobility-Resolved Collision-Induced Dissociation And Electron Transfer Dissociation Of N-Glycopeptides: Gathering Orthogonal Connectivity Information From A Single Mass- Selected Precursor Ion Population, Venkata Kolli, Katherine N. Schumacher, Eric Dodds
Department of Chemistry: Faculty Publications
Glycopeptide-level mass spectrometry (MS) and tandem mass spectrometry (MS/MS) analyses are commonly performed to establish site-specific protein glycosylation profiles that are of central importance to gaining structure-function insights on glycoproteins. Confoundingly, the complete characterization of glycopeptide connectivity usually requires the acquisition of multiple MS/MS fragmentation spectra. Complementary ion fragmentation techniques such as collision-induced dissociation (CID) and electron transfer dissociation (ETD) are often applied in concert to address this need. While structurally informative, the requirement for acquisition of two MS/MS spectra per analyte places considerable limitations upon the breadth and depth of large-scale glycoproteomic inquiry. Here, a previously developed method of …
Relationship Of The Fractal And Chemical Characteristics Of Self-Assembled Natural Organic Matter, Dennis K. Gibson
Relationship Of The Fractal And Chemical Characteristics Of Self-Assembled Natural Organic Matter, Dennis K. Gibson
Electronic Theses and Dissertations
Natural Organic Matter (NOM) is a heterogeneous mixture formed by the degradation of organic substances during early diagenesis in surficial environments. It has been shown that the interaction of the components that comprise this mixture has a significant impact on its microbial mineralization to CO2. The extent of NOM selfassembly is emerging as an important factor in understanding its role in the global geochemical carbon cycle, and it is beginning to appear that it may be more important than the chemical composition of a sample. The overall goal of this research is to establish NOM’s self-assembled “architecture” and …
Investigating Stress Response Across Integrated Biological Systems, Alice A. Han
Investigating Stress Response Across Integrated Biological Systems, Alice A. Han
Graduate Theses, Dissertations, and Problem Reports (ETD)
The impact that a stressor has on a living system, whether it is on a cellular, organ, or even a whole organism level, can affect the overall health of the system. Monitoring the biochemical response resulting from a stressful experience offers insight into the numerous potential outcomes ranging on the spectrum of survival and death. Accessing this information not only provides a heightened understanding of the biochemical adaptions that occur, but also allows for the development of prediction models (to assess prospective influences of the stressor) or potential therapeutic treatments (to alleviate adverse effects) using measurable, quantifiable, and comparable metrics. …
Energetic Effects Of Hole Transporting Materials On The Performance Of Organometal Halide Perovskite Photovoltaic Cells, So Min Park
Theses and Dissertations--Chemistry
Efficient, inexpensive, lightweight and flexible solar cells are desired to help meet the world’s growing energy needs. Organometal halide perovskite (OMHP) photovoltaic (PV) cells have shown dramatic increases in solar cell efficiencies increase over the last 5 years. OMHP PV cells have attracted significant attention due to their broad absorption spectra, high electron and hole mobility, and low production cost. The interface between hole transporting layer (HTL) and perovskite thin films have a significant influence on charge transfer and overall solar cell performance. 2,2’,7,7’-tetrakis(N,N-di-p-methoxyphenylamine)9,9’-spirobifluorene (Spiro-OMeTAD) is a small molecule largely used as HTL in perovskite solar cells. However, this material …
Mechanisms Of Heterogeneous Oxidations At Model Aerosol Interfaces By Ozone And Hydroxyl Radicals, Elizabeth A. Pillar-Little
Mechanisms Of Heterogeneous Oxidations At Model Aerosol Interfaces By Ozone And Hydroxyl Radicals, Elizabeth A. Pillar-Little
Theses and Dissertations--Chemistry
Atmospheric aerosols play an important role in climate by scattering and absorbing radiation and by serving as cloud condensation nuclei. An aerosol’s optical or nucleation properties are driven by its chemical composition. Chemical aging of aerosols by atmospheric oxidants, such as ozone, alters the physiochemical properties of aerosol to become more hygroscopic, light absorbing, and viscous during transport. However the mechanism of these transformations is poorly understood. While ozone is a protective and beneficial atmospheric gas in the stratosphere, it is a potent greenhouse gas in the troposphere that traps heat near the Earth’s surface. It also impacts human heath …
Heterogeneous Base Metal Catalyzed Oxidative Depolymerization Of Lignin And Lignin Model Compounds, John Adam Jennings
Heterogeneous Base Metal Catalyzed Oxidative Depolymerization Of Lignin And Lignin Model Compounds, John Adam Jennings
Theses and Dissertations--Chemistry
With the dwindling availability of petroleum, focus has shifted to renewable energy sources such as lignocellulosic biomass. Lignocellulosic biomass is composed of three main constituents, lignin, cellulose and hemicellulose. Due to the low value of cellulosic ethanol, utilization of the lignin component is necessary for the realization of an economically sustainable biorefinery model. Once depolymerized, lignin has the potential to replace petroleum-derived molecules used as bulk and specialty aromatic chemicals. Numerous lignin depolymerization strategies focus on cleavage of β-aryl ether linkages, usually at high temperatures and under reductive conditions.
Alternatively, selective benzylic oxidation strategies have recently been explored for …
Semiconductor Photocatalysis: Mechanisms, Photocatalytic Performances And Lifetime Of Redox Carriers, Ruixin Zhou
Semiconductor Photocatalysis: Mechanisms, Photocatalytic Performances And Lifetime Of Redox Carriers, Ruixin Zhou
Theses and Dissertations--Chemistry
Photocatalytic reactions mediated by semiconductors such as ZnS, TiO2, ZnO, etc. can harvest solar energy into chemical bonds, a process with important prebiotic and environmental chemistry applications. The recycling of CO2 into organic molecules (e.g., formate, methane, and methanol) facilitated by irradiated semiconductors such as colloidal ZnS nanoparticles has been demonstrated. ZnS can also drive prebiotic reactions from the reductive tricarboxylic acid (rTCA) cycle such as the reduction of fumarate to succinate. However, the mechanism of photoreduction by ZnS of the previous reaction has not been understood. Thus, this thesis reports the mechanisms for heterogeneous photocatalytic reductions …
Characterization And Application Of Hybrid Nanostructures For Enhanced Biological Imaging Using Fluorescence Microscopy Techniques, William E. Martin
Characterization And Application Of Hybrid Nanostructures For Enhanced Biological Imaging Using Fluorescence Microscopy Techniques, William E. Martin
Theses and Dissertations--Chemistry
Fluorescence microscopy is a powerful tool for interpreting the structure and function of biomolecules, and their interactions with one another. Understanding fundamental biological mechanisms is important to the development of improved treatments for a variety of diseases. Fluorescent tags are used to track the motion and longevity of such events, with the ability to monitor several biomolecules at once. Currently, many of these studies are conducted using bulk measurements, or several biological events at the same time, because the added light from several emitters can more easily overcome the high fluorescence background inherent to biological systems. Although important in their …

Epigenetic Modifications To Cytosine And Alzheimer’S Disease: A Quantitative Analysis Of Post-Mortem Tissue, Elizabeth M. Ellison
Theses and Dissertations--Chemistry
Alzheimer’s disease (AD) is the most common form of dementia and the sixth leading cause of death in the United States, with no therapeutic option to slow or halt disease progression. Development of two characteristic pathologic lesions, amyloid beta plaques and neurofibrillary tangles, in the brain are associated with synaptic dysfunction and neuron loss leading to memory impairment and cognitive decline. Although mutations in genes involved in amyloid beta processing are linked to increased plaque formation in the inherited familial form of AD, the more common idiopathic form, termed sporadic AD, develops in the absence of gene mutations. In contrast, …
Application Of High-Resolution Accurate Mass (Hram) Mass Spectrometry For Analysis Of Lignin Model Compounds And The Post-Pretreatment Products, Fan Huang
Theses and Dissertations--Chemistry
Lignin, one of main components in the woody cell walls, is a complex heterogeneous biopolymer, which provides structural support and transportation of water in plants. It is highly recalcitrant to degradation (both chemically and environmentally) and protects cellulose from being degraded/hydrolyzed. Due to the structural complexity of native lignin, complete characterization and elucidation of lignin’s structure remains very challenging. The overarching goal of this work is to develop mass spectrometry based analytical methods to contribute to a better understanding of lignin structures.
This dissertation will focus on the development and application of High-Resolution Accurate-Mass (HRAM) Mass Spectrometry (MS) as main …

Theses and Dissertations--Chemistry
Gene therapy holds promise for the treatment a wide range of diseases ranging from cystic fibrosis to cardiovascular disease to cancer. The need for safe and efficient gene delivery methods remains the primary barrier to human gene therapy. Non-viral vector materials, including polymers, can be designed to be biocompatible and non-immunogenic, but lack the efficiency to be clinically relevant. Gene therapy awaits the development of new materials that are both safe and efficient. Here, we have synthesized a series of modified zwitterionic polymers based on the common transfecting agent polyethylenimine (PEI). Using a variety of biochemical and biophysical methods we …

Bioavailable Iron In Equatorial Pacific Ocean Aerosol Samples, Hsiang T Teng
Bioavailable Iron In Equatorial Pacific Ocean Aerosol Samples, Hsiang T Teng
All Master's Theses
Oceanic iron (Fe) fertilization experiments performed in remote regions have established that Fe additions draw carbon into the ocean, at least over the months-long time frame of the experiments. However, the mechanisms that control Fe speciation in atmospheric aerosol particles before and after deposition into the surface ocean remain largely unknown. This is in part due to the analytical challenge of quantifying Fe at environmentally significant sub-nano molar levels. The flow injection analysis method combined with the luminol chemiluminescence analytical system allows us to explore the near-real time determination of pico-molar levels of both Fe(II) and H2O2 …

The method development process in liquid chromatography (LC) involves optimization of a variety of method parameters including stationary phase chemistry, column temperature, initial and final mobile phase compositions, and gradient time when gradient mobile phases are used. Here, a general simulation program to predict the results (i.e., retention time, peak width and peak shape) of LC separations, with the ability to study various complex chromatographic conditions is described. The simulation program is based on the Craig distribution model where the column is divided into discrete distance (Δz) and time (Δt) segments in a grid and is …
Separation Of Transition And Heavy Metals Using Stationary Phase Gradients And Chelation Thin Layer Chromatography / Evaluation Of The Effectiveness Of Pogil-Pcl Workshops, Stacy L. Stegall
Theses and Dissertations
Gradient surfaces exhibit a variation in functionality along the length of the surface. One method for preparing gradients is controlled-rate infusion (CRI). In Part 1 of this work, CRI was used to prepare gradients for the purpose of separating transition and heavy metals. Initial work on this project was focused on controlling the retention of the metal ions by varying the number of amine groups, aminoalkoxysilane concentration, and the infusion time. The retention factors of four metal ions varied predictably with increasing number of amine groups, increasing aminoalkoxysilane concentration, and increasing infusion time, producing small but useful changes in the …

Theses and Dissertations
Surface chemical gradients are materials that exhibit continuous, gradually varying chemical or physical properties along and across the length of a substrate. As a result, each point on the gradient surface can represent an individual sample. They are broadly classified as chemical and physical gradients depending upon the properties that the gradient exhibits. A physical gradient involves a continuous variation of physical properties such as surface roughness and film porosity on the micrometer scale. Chemical gradients involve a gradual variation of chemical properties such as polarity, acidity and basicity, etc. Such gradients have found various applications in cell adhesion, nanoparticle …
Natural Organic Matter Characterization And Lead Speciation As Indicators Of Potential Lead Bioavailability In Aquatic Environments, Giselle Cimprich
Natural Organic Matter Characterization And Lead Speciation As Indicators Of Potential Lead Bioavailability In Aquatic Environments, Giselle Cimprich
Theses and Dissertations (Comprehensive)
Trace metals and natural organic matter (NOM) are ubiquitous in aquatic environments. Some trace metals are essential for aquatic life, while other non-essential metals (like lead) can be toxic if present in great enough concentrations. Natural waters contain a combination of inorganic, and organic ligands capable of binding metals. While the chemistry of inorganic Pb species is well understood, and National Institute of Standards and Technology (NIST) certified K values are readily available, the interactions between metals and organic matter is not so clearly defined. It is important to understand how NOM interacts with metals in the environment, as the …

Identification Of “Kratom” (Mitragyna Speciosa) Alkaloids In Commercially Available Products, Julia Grzymkowski, Michelle R. Peace, Justin Poklis
Undergraduate Research Posters
“Kratom” is the common name for the botanical mitragyna speciosa. It is a tree native to Southeast Asia in which leaves contain the psychoactive alkaloids mitragynine and 7-hydroxymitragynine. Kratom is often ingested as teas, chewed, or smoked. It acts as a stimulant in small doses and as an opioid in large doses. Overdoses can result in vomiting, seizures, and death. Recently the Drug Enforcement Agency (DEA) placed Kratom on Schedule 1, but, due to public outcry, it was almost immediately removed.
Eleven kratom based products were obtained from various tobacco shops, “headshops” and via the internet including: Choice brand …

An Stm Study On The Structure Of Pt(100)/Ionic Liquid Omipf6 Interface, Jin Peng, Mian-Gang Li, Li-Qiang Xie, Jia-Wei Yan, Bing-Wei Mao
Journal of Electrochemistry
Potential-dependent structures of Pt(100)/ionic liquid 1-methyl-3-octylimidazolium hexafluorophosphate (OMIPF6) interface have been studied by electrochemical scanning tunneling microscopy (ECSTM). The cation OMI+ forms ordered structure on Pt(100) surface, which exists in a potential region of about 1.2 V. When the potential is more negative than -0.6 V, it can be seen that the ordered structure transforms to disordered structure. When the potential shifts positively to +0.6 V, the desorption of cations OMI+ occurs, which indicates that strong electrostatic repulsion is needed to overcome chemical interaction between OMIPF6 and Pt(100) surface, leading to the desorption. The above results …

Journal of Electrochemistry
An electrocatalyst for oxygen reduction reaction (ORR) is an important component for fuel cells. An investigation at interfacial electrochemical reactions toward ORR at a molecular scale benefits mechanistic understanding as well as rational design of catalysts. Scanning tunneling microscopy (STM) has been proven to be a powerful tool to monitor chemical reactions and to provide in-situ information about the interfacial electrochemical reactions at a molecular level. This review summarizes the recent STM studies in monitoring the interface processes such as morphological changes, molecular changes, reaction intermediates, and oxidation products. The prospects of future development in this field are outlined.

A grand unified model (GUM) is developed to achieve fundamental understanding of rich structures of all 71 liganded gold clusters reported to date. Inspired by the quark model by which composite particles (for example, protons and neutrons) are formed by combining three quarks (or flavours), here gold atoms are assigned three ‘flavours’ (namely, bottom, middle and top) to represent three possible valence states. The ‘composite particles’ in GUM are categorized into two groups: variants of triangular elementary block Au3(2e) and tetrahedral elementary block Au4(2e), all satisfying the duet rule (2e) of the valence shell, akin to …

Environmental Emission Of Pharmaceuticals From Wastewater Treatment Plants In The Usa, Bikram Subedi, Bommanna G. Loganathan
Environmental Emission Of Pharmaceuticals From Wastewater Treatment Plants In The Usa, Bikram Subedi, Bommanna G. Loganathan
Faculty & Staff Research and Creative Activity
The residual drugs, drug bioconjugates, and their metabolites, mostly from human and veterinary usage, are routinely flushed down the drain, and enter wastewater treatment plants (WWTP). Increasing population, excessive use of allopathic medicine, continual introduction of novel drugs, and existing inefficient wastewater treatment processes result in the discharge of large volumes of pharmaceuticals and their metabolites from the WWTPs into the environment. The effluent from the WWTPs globally contaminate ~25% of rivers and the lakes. Pharmaceuticals in the environment, as contaminants of emerging concerns, behave as pseudo-persistent despite their relatively short environmental half-lives in the environment. Therefore, residual levels of …
