Microfluidics has enabled numerous applications in, for example, analytical chemistry, medical diagnostics, microelectronics, and soft robotics. In most of these applications, the geometries of the microchannels are of fixed dimensions that (ideally) remain invariant during operation. In soft robotics, however, the geometries of the microchannels contained in soft actuation systems are inherently dynamic, and the specific dimensions are expected to change during operation, and, by extension, the fluid transport properties of the system are variable. It is known that an isolated single-molecule magnet tends to become super- paramagnetic even at an ultralow temperature of a few Kelvin due to the low spin switching barrier. Herein, single-molecule ferroelectrics/multiferroics is proposed, as the ultimate size limit of memory, such that every molecule can store 1 bit data. The primary strategy is to identify polar molecules that possess bistable states, moderate switching barriers, and polarizations fixed along the vertical direction for high-density perpendicular recording. First- principles computation shows that several selected magnetic metal porphyrin molecules possess buckled structures with switchable vertical polarizations that are robust at ambient conditions. …

Utilizing screen-printed carbon electrodes (SPCEs), a fast, simple, and sensitive approach toward the detection, identification, and quasi-quantitation of fentanyl was achieved both in an electrochemical cell and as a drop on the electrode surface. Electro-oxidation of fentanyl at the electrode was demonstrated using adsorptive stripping square-wave voltammetry between -0.5 V and +1.6 V with 100 mM Tris-HCl buffer at pH 8.5 as supporting electrolyte. Parameter optimization was conducted during method development to include supporting electrolyte and pH, electrochemical technique, pre-treatment and equilibration time, and various surface modifications. In this work we demonstrate the power of compound-specific isotope analysis (CSIA) to analyze proteinaceous biological materials in three distinct forensic applications, including: 1) linking necrophagous blow flies in different life stages to their primary carrion diet; 2) identifying the harvesting area of oysters for food authentication purposes; and 3) the ability to predict biometric traits about humans from their hair.

Novel psychoactive substances (NPSs) are synthetically derived compounds designed to mimic the effects of other illicit drugs. An endless cycle of NPSs continually reach the drug market due to limitations in drug legislation creating two problems: (1) non-availability of standards and (2) multiple isobars indistinguishable even to high resolution mass spectrometry. Gas chromatography-mass spectrometry (GCMS) is the most common instrument used for compound identification in non-targeted seized drug analysis. Human exposure to biomass smoke is a health concern worldwide. Although many studies have measured particulate matter in wood smoke as a health concern, exposure to mutagenic and carcinogenic volatile and semivolatile compounds remains understudied. This research introduces a novel method of quantitative measurement of exposure to these compounds using silicone wristbands. The study developed a method to extract analytes of interest from the wristbands and quantify a few volatile organic compounds and polycyclic aromatic hydrocarbons with known ill health effects, and then performed linear regressions between extracted levels and exposure to those analytes using controlled exposure studies. Male Hymenochirus sp. frogs are known to release pheromone that attracts females of the same species. Four methods for collecting secretions containing pheromone in Hymenochirus sp. were tested: norepinephrine injection, gonadotropin-releasing hormone injection, homogenization of gland tissue, and electrostimulation of the skin over the breeding gland area. The samples collected were analyzed using high-performance liquid chromatography (HPLC) and mass spectrometry. The HPLC chromatograph for the male norepinephrine sample contained a peak at 6.4 min that was not in the female norepinephrine sample HPLC chromatograph. Antibody drug conjugates, which take advantage of both antibodies and cytotoxic agents, showed high selectivity towards cancer cells. Laser-induced breakdown spectroscopy (LIBS) was investigated to determine magnesium (Mg) content in infant formula powder. To predict Mg content in the range established by the Codex Alimentarius, a partial least squares regression (PLSR) model was developed using a calibration data set (n = 30) based on full cross-validation and validated using an independent validation data set (n = 21). Choosing a stationary phase is the first step in developing a liquid chromatography (LC) separation method, where the selectivity is governed by the differential interactions of the analytes with the stationary and mobile phases. This work presents a reliable, cost-effective, rapid and in-field voltammetric method for the detection of barium. The optimized method consists of an ultrathin mercury film deposited in situ on a glassy carbon electrode in dilute potassium chloride without deoxygenation, using differential pulse anodic stripping voltammetry (DP-ASV). Application of the method allowed for the quantitative determination of barium concentration in a variety of waters and brake pad dust samples. Comparative analysis of sample results from DP-ASV with inductively coupled plasma mass spectroscopy (ICP-MS) showed a mean percent difference of 1.8%.

Organic aerosol affects human health and climate. These effects are largely determined by the composition of the organic aerosol, which is a complex mixture of species. Understanding the complexity of organic aerosol is critical to determining its effect on human health and climate. In this study, long range transported organic aerosol collected at the Pico Mountain Observatory was analyzed using ultrahigh resolution mass spectrometry. Organic aerosol transported in the free troposphere had an overall lower extent of oxidation than aerosol transported in the boundary layer. By analyzing Electrochemical Impedance Spectroscopy (EIS) in applications of dynamic proton exchange membrane fuel cell (PEMFC), bottlenecks which restrict EIS tool development have been pointed out in this paper. Though the high-frequency resistance in EIS is largely accepted as cell inner-resistance, this can only be applied for cell with low current. The low-frequency resistance is difficult to be realized due to its relation with mass transfer. In this work, ferricyanide-doped polyaniline (PANI) is electropolymerized by constant current and cyclic voltammetry (CV) measurements with simultaneous monitoring of mass changes using electrochemical quartz crystal microbalance (eQCM). Raman spectroscopy, Fourier transform infrared spectroscopy and CV measurements confirm the incorporation of the ferricyanide into the film and indicate modified electrical conductivity. UV-Vis absorbance spectroscopy and Raman spectroscopy confirm the conducting ‘emeraldine-salt’ state and confirm the presence of polarons, i.e. charge carriers.

In this thesis, we demonstrate an electrochemical approach for monitoring the electric-field induced melting of surface-bound double-stranded DNA. The electrochemical routine involves repeated chronoamperometry pulses to melt the duplex and square wave voltammetry to monitor the extent of melting. We utilize a scanning potential and constant potential technique to generate melting curves and access the stability and kinetics of the DNA duplexes. This thesis investigates the combustion potential of 2-phenylethanol and the superalkali properties of small lithium substituted silicon compounds. All combustion experiments were performed at the Advanced Light Source of Lawrence Berkeley National Laboratory at the Chemical Dynamics Beamline 9.0.2. The chlorine initiated oxidation of 2PE was investigated at 298 and 550 K using a multiplex photoionization mass spectrometer, coupled with the tunable vacuum ultraviolet radiation. Reaction products were identified using kinetic time traces and photoionization spectra.

Additionally, the stability of small superalkali silicon-lithium compounds has also been investigated. All structures and energetics were calculated using the CBS-QB3 composite method.

The vitreous humor (VH) is located in between the lens and the retina. It is composed of 98% water, hyaluronan (HA), collagen, proteins, phospholipids (PLs), and other metabolites. With aging, the VH undergoes liquefaction, a process that causes the gel-like structure of the VH to turn into a liquid and can lead to serious ocular diseases, including retinal detachment, vitreal detachment, and macular hole formation. The liquefaction process is expedited in diabetic VHs. This project focuses on understanding the molecular changes that lead to liquefaction and the reasons for which this process is accelerated with diabetes. Relative Permittivity As A Function Of Co-Solvent And Impedance Spectroscopy For Quantifying Anions In Solution, Yahya Sami Alseiha

This work measured relative permittivity from 15.0 to 55.0 °C of aqueous solutions containing acetone or tetrahydrofuran using a BI 870 dielectric meter. A cubic polynomial fit resulted in coefficients for calculating permittivity as a function of temperature and mole fraction. Literature values for other co-solvent system resulted in similar polynomial coefficients. Using BI 870, permittivity measurements of ionic solutions were not possible because of high conductivity. A simple method using impedance spectroscopy measured these solutions. Impedance decreases with increasing temperature and decreases with increasing ionic strength. Due to the temperature dependence of impedance, all measurements were at 25.0 °C. …

Zebra mussels (Dreissena polymorpha) are an exotic and invasive mollusk that have spread extensively through various rivers and lakes in the United States. These mussels are notorious for their “biofouling” capabilities costing billions of dollars in their removal from industrial, public, and power plant water supply lines. While having been present in the Tennessee River drainage with occasional small reproducing populations since the early 1990s, zebra mussels were never abundant or widespread. The drug overdose deaths in the USA increased by ~20% from 2015 to 2016 to the total >63,600 drug overdose deaths in 2016. Kentucky is among the highest rate of drug overdose death states in the USA. The current estimates of the prevalence of substance abuse are based on the self-reported surveys, overdose/toxicological reports, and drug-related crime statistics. Survey-based conventional approaches are not only cost and time-intensive but also underestimate the actual consumption of drugs. The development and awareness of Machine Learning and “big data” has led to a growing interest in applying these methods to bioanalytical research. Methods such as Mass Spectrometry (MS), and Nuclear Magnetic Resonance (NMR) can now obtain tens of thousands to millions of data points from a single sample, due to fundamental instrumental advances and ever-increasing resolution. This work was done in order to deepen the understanding of interactions occurring during sorption of anthropogenic organic compounds and dissolved organic matter (DOM) onto soil. As well as the transformation DOM undergoes during the sorption process and the overall transformation OM undergoes in the environment. This was achieved through the use of a “bottom-up” synthetic investigation, “top-down” natural system study, and a combination of these approaches.

The “bottom-up” approach, discussed in Chapter 2, involved the synthesis of a modular multi-tiered engineered soil surrogate (ESS) designed to mimic OM and general trends identified in the literature that impacts sorption behavior. …
