PCR-based tests for various microorganisms or target DNA sequences are generally acknowledged to be highly "sensitive" yet the concept of sensitivity is ill-defined in the literature on these tests. We propose that sensitivity should be expressed as a function of the number of target DNA molecules in the sample (or specificity when the target number is 0). However, estimating this "sensitivity curve" is problematic since it is difficult to construct samples with a fixed number of targets. Nonetheless, using serially diluted replicate aliquots of a known concentration of the target DNA sequence, we show that it is possible to disentangle …

Latent class regression models are useful tools for assessing associations between covariates and latent variables. However, evaluation of key model assumptions cannot be performed using methods from standard regression models due to the unobserved nature of latent outcome variables. This paper presents graphical diagnostic tools to evaluate whether or not latent class regression models adhere to standard assumptions of the model: conditional independence and non-differential measurement. An integral part of these methods is the use of a Markov Chain Monte Carlo estimation procedure. We revisit the question of exact multiplicity of positive solutions for a class of Dirichlet problems for cubic-like nonlinearities, which we studied in 161. Instead of computing the direction of bifurcation as we did in [6], we use an indirect approach, and study the evolution of turning points. We give conditions under which the critical (turning) points continue on smooth curves, which allows us to reduce the problem to the easier case of f (0) = 0. We show that the smallest root of f (u) does not have to be restricted.

In this paper we study stopping time and impulse control problems for stochastic Navier-Stokes equation. Exploiting a local monotonicity property of the nonlinearity, we establish existence and uniqueness of strong solutions in two dimensions which gives a Markov-Feller process. The variational inequality associated with the stopping time problem and the quasi-variational inequality associated with the impulse control problem are resolved in a weak sense, using semigroup approach with a convergence uniform over path.

We give a detailed algebraic characterization of when a graph G can be imbedded in the projective plane. The characterization is in terms of the existence of a dual graph G∗ on the same edge set as G which satisfies algebraic conditions inspired by homology groups and intersection products in homology groups.

In non-randomized treatment studies a significant problem for statisticians is determining how best to adjust for confounders. Marginal structural models (MSMs) and inverse probability of treatment weighted (IPTW) estimators are useful in analyzing the causal effect of treatment in observational studies. Given an IPTW estimator a doubly robust augmented IPTW (AIPTW) estimator orthogonalizes it resulting in a more e±cient estimator than the IPTW estimator. One purpose of this paper is to make a practical comparison between the IPTW estimator and the doubly robust AIPTW estimator via a series of Monte- Carlo simulations. In many applications, it is often of interest to estimate a bivariate distribution of two survival random variables. Complete observation of such random variables is often incomplete. If one only observes whether or not each of the individual survival times exceeds a common observed monitoring time C, then the data structure is referred to as bivariate current status data (Wang and Ding, 2000). Let G be a connected graph that is 2-cell embedded in a surface S, and let G* be its topological dual graph. We will define and discuss several matroids whose element set is E(G), for S homeomorphic to the plane, projective plane, or torus. We will also state and prove old and new results of the type that the dual matroid of G is the matroid of the topological dual G*.

Laser-beam-induced-current (LBIC) imaging is a nondestructive technique used for the characterization of the electrical structure within a semiconductor. In this paper a model is formulated for this technique using the standard drift-diffusion model, and, subsequently, an approximate version and its dual are derived for the study of the inverse problem. The formulation is then applied to a cross-sectional model for n-on-p devices of finite depth to study in detail the relation between the LBIC images and the device parameters. Kaufman & Rousseeuw (1990) proposed a clustering algorithm Partitioning Around Medoids (PAM) which maps a distance matrix into a specified number of clusters. A particularly nice property is that PAM allows clustering with respect to any specified distance metric. In addition, the medoids are robust representations of the cluster centers, which is particularly important in the common context that many elements do not belong well to any cluster. Based on our experience in clustering gene expression data, we have noticed that PAM does have problems recognizing relatively small clusters in situations where good partitions around medoids clearly exist. The pool-adjacent violator-algorithm (Ayer, et al., 1955) has long been known to give the maximum likelihood estimator of a series of ordered binomial parameters, based on an independent observation from each distribution (see Barlow et al., 1972). This result has immediate application to estimation of a survival distribution based on current survival status at a set of monitoring times. This paper considers an extended problem of maximum likelihood estimation of a series of ‘ordered’ multinomial parameters. We use martingale methods to obtain an explicit formula for the expected wet period of the finite dam of capacity V, where the amounts of inputs are i.i.d exponential random variables and the output rate is one, when the reservoir is not empty. As a consequence, we obtain an explicit formula for the expected hitting time of either 0 or V and a new expression for the distribution of the number of overflows during the wet period, both without the use of complex analysis.

The objective of this paper is to study the stochastic version of a previous paper of the authors, in which hybrid control for deterministic systems was considered. The modelling is quite similar to the deterministic case. We have a system whose state is composed of a continuous part and a discrete part. They are affected by a continuous type control and an impulse control. The dynamics is moreover perturbed by noise, also a continuous and a discrete noise process. An initial-boundary value problem for nonlinear parabolic equations modelling surfactant diffusions is investigated. The boundary conditions are of nonlinear adsorptive types, and the initial value has a single point jump. We study the well-posedness of the problem, the convergence of a numerical scheme, and the regularity as well as quantitative behaviour of solutions.

A base β of a space X is called an OIF base when every element of B is a subset of only a finite number of other elements of β. We will explore the fundamental properties of spaces having such bases. In particular, we will show that in T2 spaces, strong OIF bases are the same as uniform bases, and that in T3 spaces where all subspaces have OIF bases, compactness, countable compactness, or local compactness will give metrizability.

The k-means algorithm and the principal curve algorithm are special cases of a self-consistency algorithm. A general self-consistency algorithm is described and results are provided describing the behavior of the algorithm for theoretical distributions, in particular elliptical distributions. The results are used to contrast the behavior of the algorithms when applied to a theoretical model and when applied to finite datasets from the model. The algorithm is also used to determine principal loops for the bivariate normal distribution.

Pertussis (whooping cough) incidence in the United States has oscillated with a period of about four years since data was first collected in 1922. An infection with pertussis confers immunity for several years, but then the immunity wanes, so that reinfection is possible. A pertussis reinfection is mild after partial loss of immunity, but the reinfection can be severe after complete loss of immunity. Three pertussis transmission models with waning of immunity are examined for periodic solutions. Equilibria and their stability are determined. Image resolution is the primary parameter for performance characterization of any imaging system. In this work, we present an axiomatic approach for quantification of image resolution, and demonstrate that a good image resolution measure should be proportional to the standard deviation of the point spread function of an imaging system.
