HSV-1 acquires its final envelope by budding into cytoplasmic vesicles thought to be derived from Trans-Golgi Network (TGN) membranes. This process is facilitated by interactions among the carboxyl termini of viral glycoproteins and tegument proteins. To investigate the relative importance of different viral glycoproteins in cytoplasmic virion morphogenesis, a set of recombinant viruses were constructed silencing expression of the glycoprotein E (ΔgE), the carboxyl terminus of glycoprotein D (gDΔcp), and the membrane protein UL20 (ΔUL20). In addition, recombinant viruses were constructed having the ΔgE+gDΔcp, and the ΔgE+ΔgM (glycoprotein M) deletions. A conditional silencing system using anti-gB siRNAs was devised to investigate the structure and function of the KSHV gB. Transient co-transfection of plasmids constitutively expressing gB and anti-gB siRNAs in 293 cells substantially inhibited gB mRNA levels and protein production. Similarly, transient expression of siRNAs into the basal cavity-based lymphoma cells (BCBL-1) caused substantial reduction of gB transcription and protein synthesis. TaqMan real-time PCR and infectivity assays showed that gB was essential for virion egress and infectivity. Transfection of a codon-optimized gB not recognized by the anti-gB siRNAs, efficiently rescued virion egress and infectivity in BCBL-1 cells in the presence …

Oncolytic herpes simplex virus type-1 (HSV-1) has shown great potential as an effective agent for cancer therapy ¨C oncolytic virotherapy. To enhance oncolytic capabilities of previously reported agents, new oncolytic and fusogenic HSV-1 OncSyn and OncdSyn viruses were constructed based on wild type HSV-1 (F) strain. To provide for safety and tumor selectivity, the viruses carried a large deletion including one of the two ¦Ã134.5 genes. Although the biochemical characterization of E. ictaluri, the subsequent disease progression of enteric septicemia of catfish (ESC), and the associated pathologic lesions are well characterized, the mechanism of invasion of E. ictaluri into a susceptible host is poorly understood. Identification and confirmation of virulence factors and associated genes of E. ictaluri is crucial to elucidating the pathogenesis of this important disease. A signature tagged mutagenesis (STM) study conducted by Thune et al. (2006) identified 50 E. ictaluri clones with transposon insertions in genes potentially involved in pathogenesis. The Lyme disease spirochete Borrelia burgdorferi has a very unusual genome composed of one linear chromosome and up to 21 linear and circular plasmids. Several plasmids are known to be important either for mammalian infection or tick colonization. A single spirochete harbors up to 7 different cp32 plasmids; however, nothing is known about their role in mammalian infection. The plasmids in this family are well maintained during in vitro cultivation, making it difficult to study their functions. Microglia/macrophage activation has been associated with the pathogenesis of various neurological diseases including human immunodeficiency virus encephalitis, transmissible spongiform encephalitis, and Alzheimer's disease (AD). In vitro studies have indicated a role for TNFα in activating these cells which leads to their migration, proliferation, and secretion of proinflammatory cytokines and chemokines that may potentially damage brain tissue. In the current study, we analyzed the phenotype of microglia and macrophages enriched from wild type and TNFα deficient mice infected with a neurovirulent murine retrovirus. Viral infections of the central nervous system (CNS) in infants are rare; however, they are associated with high morbidity and mortality rates. These virus infections often induce strong innate immune responses in the brain including: the production of cytokines and chemokines, the activation of astrocytes and microglia and the recruitment of macrophages. Innate immune responses are often initiated by toll-like receptors (TLR). Herpes simplex virus type-1 (HSV-1) morphogenesis occurs in multiple stages within infected cells. Initially, the virion capsid assembles within the nucleus and buds through the nuclear membrane into the cytoplasm. Within the cytoplasm, additional tegument proteins attach to the capsid and the fully tegumented capsids bud into trans-Golgi network (TGN) derived vesicles. Enveloped virions are ultimately secreted to extracellular spaces. The process by which the cytoplasmic capsids bud into TGN-derived vesicles is not well understood. Brucella melitensis is a facultative intracellular bacterial pathogen that causes abortions in goats and sheep and Malta fever in humans. The zoonotic disease brucellosis causes severe economic losses in the Mediterranean region and parts of Africa, Asia, and Latin America. With the completion of the genomic sequences of B. abortus 2308 and B. melitensis 16M, no classical virulence factors were found; and the chromosomes were virtually identical. However, in B. melitensis, a putative hemagglutinin gene was identified which is absent in B. abortus. West Nile virus (WNV), a member of the genus Flavivirus transmitted by mosquitoes, first appeared in the New York in 1999. Within five years WNV was detected throughout the contiguous 48 states causing disease in reservoirs and accidental hosts alike. In Louisiana, WNV was first detected in 2001 with one human case, ten equine cases, and six dead birds reported. The introduction of WNV into Louisiana presented an unique opportunity to observe an emerging disease unfold, so a study was launched to gain insight into the epidemiology of WNV in Louisiana. Brugia infections occur via the bite of an infected mosquito. Third stage infective larvae (L3) deposited on the skin during feeding migrate into the bite wound, through skin and into the lymphatic system. It is hypothesized that L3 excretory/secretory products (ES) are important in this initial phase of the infection. A model for these early migrations has been established by inoculating L3s into the dermis (ID) of the permissive gerbil host. In this model, most L3s injected ID in the louer hind limb travel to the popliteal lymph node by 3 days post infection. Adult parasites are located primarily in …

Extracts of adult Haemonchus contortus were purified and used as a vaccine against the blood feeding parasite in goats as previously studied in sheep. The proteins used were H11 and H-gal-GP, hidden gut antigens from the microvillar membrane of the gut of the worm and combined with Quil A as adjuvant, then administered to a group of goats kept on concrete then artificially infected with H. contortus. The drive to exercise voluntarily likely results from complex interactions between genes in many organ systems and various psychological parameters, such as motivation and the perception of fatigue. Reproducible variations in exercise intensity and duration are well established in laboratory rodents, but the genes responsible remain largely unknown. Also, to date, studies addressing the adaptive changes to exercise that might prevent dietary-induced obesity have focused primarily on energy intake and nutrient oxidation/partitioning, as opposed to genetics. Summer pasture-associated obstructive pulmonary disease (SPAOPD), a naturally occurring airway disease of horses, is characterized by clinical exacerbation associated with exposure to pasture environment during the summer. Aeroallergens are believed to trigger exacerbation of SPAOPD, cytokines are likely associated with the anamnestic response to aeroallergens, and endothelin (ET)-1 is a potential mediator of airway obstruction. The SARS-Coronavirus (SARS-CoV) is the etiological agent of the severe acute respiratory syndrome (SARS). The SARS-CoV spike (S) glycoprotein mediates membrane fusion events during virus entry and virus-induced cell-to-cell fusion. Investigations, described herein, have focused on the genetic manipulation of the SARS-CoV S glycoprotein in order to delineate functional domains within the protein. This was accomplished by incorporating single point mutations, cluster-to-lysine and cluster-to-alanine mutations, as well as carboxyl terminal truncations into the protein and investigating these mutants in transient expression experiments. An Edwardsiella ictaluri strain with disruption of ureG was identified through the use of signature tagged insertion mutagenesis as being attenuated for virulence in the channel catfish host. Sequencing of the flanking regions surrounding the insert showed that the gene was part of a urease gene complex that included ureE, ureF, ureG, ureD, ureI, and an ammonium transporter homologue. The ureG gene encodes a GTP-binding accessory protein which is thought to function in energy-dependent urease assembly. During the course of their disease, leprosy patients may experience two types of inflammatory reactions- erythema nodosum leprosum (ENL) or reversal reaction (RR). Thalidomide is effective treatment for ENL, but not for RR. Using concentrations of thalidomide similar to that achieved in the treatment of ENL, we investigated thalidomide’s effect on reactions, viability of M. leprae, and integrity of plasma membranes. Brucellosis and pseudorabies lead to abortion in pregnant sows and are perpetuated by feral swine reservoirs. A multivalent oral vaccine for these diseases would improve vaccination and eradication programs worldwide. Previous studies have shown that the rough attenuated Brucella strains RB51 and VTRS1, when administered subcutaneously to swine, stimulate host immune responses, transiently colonize tissues, and provide partial protection against virulent B. suis infection in pregnant sows. Studies conducted to evaluate DNA vaccination against CCV compared encoded gene, dose, multiple DNA vaccines, and immune response to vaccination. Validation of diagnostic assay performance is hampered where no gold standard exists. Bayesian estimates of the sensitivity and specificity were applied to studies of two persistent bovine infections: bovine immunodeficiency virus (BIV), and bovine viral diarrhea virus (BVDV). BVDV pathogenesis is well described. Losses of $20 million in 1992 alone led the Danish government to mandate eradication. Two enzyme-linked immunosorbent assay (ELISA) tests were developed; one antibody-based to detect exposure, one antigen-based to detect persistent infection. The host-parasite interaction during early filarial nematode migration is poorly understood. The objective of this study was to develop a model of early cutaneous filarid migration using Brugia pahangi in the jird (gerbil) host and measure the histologic and cytokine responses during this period. Male gerbils were intradermally inoculated in the left hindlimb with 100 B. pahangi L3 then necropsied at 3 hours, 24 hours, 3 days, 7 days, and 28 days post-infection. Larvae were recovered and tissues collected for histology and cytokine measurement. At 3 hours, most larvae (96.3%) were recovered from tissues associated with the infection site. Migration …

The Herpes Simplex Virus life cycle contains a number of membrane fusion events that must function properly to ensure a productive infection, including: virus attachment and entry into susceptible cells, de-envelopment at the outer nuclear lamellae, and virus-induced cell-to-cell fusion. The first event in the infection process of Ostertagia ostertagi in cattle is the process of exsheathment. Before trichostrongylid nematodes can transition from a free-living infective stage larva (L3) on pasture to a parasitic existence within the ruminant host, it must first undergo exsheathment. Exsheathment is the process whereby the L2 cuticle retained from the previous molt is cast from the L3. Exsheathment enables the developmental transition from a free-living stage on pasture to a parasitic existence in the bovine host.
