In the present study, 178 blood samples from buffaloes were tested against indirect enzyme linked immunosorbent assay (I-ELISA), rose bengal plate test (RBPT), microagglutination test (MAT), modified microagglutination test (mMAT), and polymerase chain reaction (PCR) to select the most suitable test for efficient and effective diagnosis of bovine brucellosis. Win Episcope 2 software was used to determine the agreement between tests (kappa values at 95% confidence interval). I-ELISA was pair compared with all the other tests. The efficacy of blood and buffy coat smear examination, commercially available dot-enzyme-linked immunosorbent assay (ELISA) kit, and nested polymerase chain reaction (PCR) was evaluated in the diagnosis of canine ehrlichiosis in 40 dogs exhibiting symptoms suggestive of the disease. The intracytoplasmic rickettsial organisms were seen in the smears of only 1 dog (2.5%) with an extremely low level of parasitemia. Nested PCR with 16S rRNA revealed characteristic 378 bp bands indicating positive reaction in 8 (20.0%) dogs and sera samples of 21 (52.5%) dogs revealed anti-Ehrlichia antibodies. The objective of this study is to establish a polymerase chain reaction (PCR) laboratory for detection of multidrug resistance1 (MDR1) mutation in Thailand. Blood samples were collected from 67 dogs, namely 16 Collies, 9 Shetland Sheepdogs, 5 Border Collies, 6 Shih tzus, 13 German Shepherds and 18 mixed breeds. Genomic DNA was extracted and PCR was performed to amplify 341 and 577 bp fragments to detect homozygous wild-type (+/+) and homozygous mutant (-/-), respectively. This study aimed to determine the prevalence of Escherichia coli O157 in retail red meat and meat products with the Vitek Immunodiagnostic Assay System, including H7 Escherichia coli phage technology (VIDAS ECPT), and a real-time polymerase chain reaction system (LightCycler PCR; LCPCR). A total of 106 red meat and meat product samples were analyzed with VIDAS ECPT and LCPCR. Clostridium perfringens is a gram-positive, anaerobic bacterium that causes a wide range of diseases in humans and animals. The purposes of this study were to determine, using the enzyme-linked immunosorbent assay (ELISA), the clostridial toxins in intestinal samples of lambs with suspected enterotoxemia; to conduct molecular typing of C. perfringens isolates; and to investigate the presence of the genes of 4 major toxins (\alpha, \beta, \epsilon, and \iota) in the isolates by polymerase chain reaction (PCR). This study was to investigate the virulence and pathogenicity of Thai isolated Mycoplasma gallisepticum (MG)inoculated into embryonated eggs and the chance of virulence study in embryonated eggs instead of experimentalchickens. Fasciolosis caused by Fasciola spp. (Platyhelminthes: Trematoda: Digenea) is considered the most important helminth infection of ruminants in tropical countries, causing considerable socioeconomic problems. Samples identified morphologically as Fasciola sp. in Equus caballus host species from the city of Tunis (north of Tunisia) were genetically characterized, for the first time, by sequences of the 1st (ITS-1), the 5.8S and 2nd (ITS-2) Internal Transcribed Spacers (ITS) of nuclear ribosomal DNA (rDNA). A Maedi-Visna virus (MVV) infection was diagnosed in a 10-month-old lamb from a Turkish flock. The animal suddenly showed nervous signs and died within 2 days. At necropsy, different pulmonary and nervous lesions were observed. A Coenurus cerebralis cyst was also present in the right-brain hemisphere. Severe nonpurulent meningoencephalitis and lymphoproliferative pneumonia were detected by histopathology. MVV proviral DNA was demonstrated by polymerase chain reaction (PCR) in formalin-fixed and paraffin-embedded samples collected from the brain and the lungs. Tuberculosis caused by Mycobacterium tuberculosis is an important zoonotic disease that concerns the survival of endangered species and public health aspects. A 17-year-old, female Malayan tapir from a zoo in Thailand died with a sign of panting after anesthesia for the treatment of her oral problems. Grossly, multiple caseous white nodules were scattered throughout the parenchyma and bulged from the capsular surface in affected organs such as lung, lymph node and liver. The Laguna Madre in Mexico is a nursery, growth, and refuge area for shrimp and other wildlife species. These organisms support an important fishery for local industry (third place in Mexico), which may suffer diseases that are few studied. The aim of this study was to investigate the antimicrobial susceptibility and to determinate the resistance genes by Polymerase Chain Reaction (PCR) among enterococci isolated from healthy cats and dogs in İzmir, Turkey. From a total of 114 rectal swabs, 91 (31 cats and 60 dogs) stains were isolated as enterococci. Twenty eight isolates were identified as Enterococcus faecalis and 63 as Enterococcus faecium with the API 20 IDStrep system. Antimicrobial susceptibilities were determined by disc diffusion. This study was to conduct a surveillance of Mycoplasma synoviae (MS) infection in mixed Thai native chickens. Samples were submitted from 30 mixed Thai native chicken flocks, 15 birds per flock, aged between 1-4.5 months in the area of Nakornpathom province in the September 2005-October 2006 period. Twenty-two sick koi with cumulative mortality showed clinical signs of depression, brachial hemorrhage and necrosis, focal hemorrhage, pale patches and ulcers on the skin. Clinical examination showed the heavy infection by opportunistic organisms such as Dactylogyrus sp., Gyrodactylus sp. and Saprolegnia sp. of the skin, gill and fins. Histopathologically, the gill showed severe acute diffuse necrosis of the branchial epithelial cells and severe diffuse lymphocytic-monocytic interstitial nephritis with necrosis of the tubular epithelial cells were predominantly found in most sick fish. The aim of this study was to identify the presence and prevalence of bovine tuberculosis in the eastern Anatolian border province of Van and its villages and to collect the preliminary data required for programs for the prevention and control of this zoonotic disease. Lung, trachea, and serum samples obtained from turkeys showing clinical signs of respiratory disease were examined for the presence of Bordetella avium infection (bordetellosis) using culture, enzyme-linked immunosorbent assay (ELISA), and polymerase chain reaction (PCR). Presence of the chicken infectious anemia virus (CIAV) infection in chicken flocks in some provinces of Turkey was investigated by ELISA and PCR in this study. From 38 flocks (16 commercial layer, 10 commercial broilers, 4 breeder layers and 8 breeder broilers), 922 serum samples were tested for CIAV-specific antibodies using a commercially available competitive ELISA. CIAV antibodies were positive in 609 (66.0%) sera from 34 flocks (89.5%). This study is aimed to detect Coxiella burnetii in cattle by Polymerase Chain Reaction (PCR). A total of 138 cattle blood samples were collected from 8 farms and then examined in laboratory conditions. The examination of Coxiella burnetii was carried out by PCR with specific primers. In this study, a total of 6 (4.3%) cattle serum samples were found PCR positive for Coxiella burnetii. This result proves that cattle are an important reservoir of Coxiella burnetii infection.

The objective of this study was to develop a reliable Taqman® 5' Nuclease Assay for genotyping sheep for scrapie susceptibility. The sheep prion gene contains 2 single nucleotide polymorphisms (SNPs) that may mediate resistance to classical scrapie, one at codon 136, alanine (A) or valine (V), and another at codon 171, arginine (R) or glutamine (Q). The origin of horse, dog, cat, bovine, sheep, porcine, and goat meat was determined by the polymerase chain reaction (PCR) technique, using species-specific primers. Test mixtures of meat were prepared by adding 5%, 2.5%, 1%, 0.5%, and 0.1% levels of pork, horse, cat, or dog meat to beef, sheep, and goat meat. Samples taken from those combinations were analyzed by PCR for species determination. This experiment was designed to study the results of laboratory diagnosis of Mycoplasma gallisepticum (MG) infection in layer chicken receiving MG vaccines and MG organisms. One hundred and twenty 1-day-old-malelayer chickens were raised in separate isolation rooms and equally allocated into 4 separated groups. When 4 weeks old, groups 1, 2, 3 and 4 served as a negative control, a MG commercially inactivated vaccine group, a MG commercially live vaccine group and a MG infected group, respectively. The aim of this study was to evaluate the role of stuffed mussel as a source of Listeria monocytogenes. Polymerase chain reaction is a rapid procedure with both sensitivity and specificity for quick detection and identification of L. monocytogenes. A total of 50 mussel samples were investigated for L. monocytogenes. L. monocytogenes was not identified by the conventional method. However, PCR amplification products demonstrated that 5 out of 50 samples showed positive reactions with L. monocytogenes. The PCR positive samples showed specific amplification at approximately 343 bp for L. monocytogenes.

Dichelobacter nodosus is an essential causative agent of footrot in ruminants, particularly in sheep, goats and cattle. In this study, more than 100 footrot samples were collected from 4 different farming regions in New Zealand (NZ). Selective media were chosen and isolation and routine growth conditions were optimized for NZ D. nodosus serotypes. Identification Of Vibrio Anguillarum By Pcr (Rpon Gene) Associated With Vibriosis In Marine Fish In Turkey, Di̇dem Demi̇rcan, Akin Candan

The main causative agent of vibriosis is Vibrio anguillarum. In this study, 33 V. anguillarum strains were isolated from the internal organs of marine fish showing typical clinical signs of vibriosis, and these strains were identified by biochemical tests. For comparison of these results, API 20E and BIONOR Mono-Va agglutination kits were used. Finally strains were amplified by PCR using the rpoN gene. Although the phenotypic properties of V. anguillarum strains varied, the rpoN gene from chromosomal DNA was amplified in all strains. In conclusion, this gene could be used in the diagnosis of V. anguillarum strains isolated in Turkey.

The detection of Listeria monocytogenes in Helix pomatia, both raw and cooked is presented here. Polymerase chain reaction (PCR) is a rapid procedure with both sensitivity and specificity for quick detection and identification of Listeria monocytogenes from raw and cooked Helix pomatia. A total of 30 bags (10 g each) of H. pomatia samples were investigated for the Listeria monocytogenes inlB gene with the PCR method. PCR amplification products demonstrated that 18 of the 30 samples showed positive reactions to Listeria monocytogenes in the PCR. All PCR positive samples showed specific amplification of the 343 bp fragment for Listeria monocytogenes.

In this study, the presence of Arcanobacterium spp., which causes a variety of purulent infections involving the skin, joints and visceral organs, was investigated in abscessed kidney samples of cattle. A total of 500 cattle were examined at postmortem and 100 samples with abscess were collected. Arcanobacterium pyogenes was isolated in 40 (40%) of the samples examined. DNA extracted from the isolates were amplified by Polymerase Chain Reaction (PCR) using specific primers derived from plo gene of A. pyogenes and all the isolates were determined to be positive by PCR. This study was carried out to isolate Clostridium perfringens from chickens and to detect the gene encoding the alpha toxin produced by all types of C. perfringens by polymerase chain reaction (PCR). The purpose of this study was to isolate Campylobacter species from the intestines and livers of chicken and to identify Campylobacter coli and Campylobacter jejuni by both conventional methods and Polymerase Chain Reaction (PCR). Four specific primers derived from the ceuE gene present in the genomes of C. coli and C. jejuni were used for PCR identification. The aim of this study was to diagnose Theileria annulata infected cattle by microscopy, IFAT and nested PCR and to compare the sensitivity and specificity of these diagnostic methods. A total of 147 blood and sera samples were collected from healthy cattle in four localities of Polatlı district, Ankara, where tropical theileriosis is prevalent. Examination results revealed positivity rates of 31.3%, 44.9% and 61.2% for microscopy, IFAT and nested PCR, respectively.
