Genetically fat and lean chickens reared identically in the same environment were used to investigate the effect of the apoVLDL-II gene (intron I) on total cholesterol, triglyceride, and lipoprotein concentrations. The overall allele frequency in the 2 populations was 0.59 for allele A and 0.41 for allele B. The allele frequency in the Rugao breed was 0.52 for allele A and 0.48 for allele B; however, in the Anka breed it was 0.65 for allele A and 0.35 for allele B. Phenotypic Characterisation Of Enterococcus Spp. From Femoral Head Necrosis Lesions Of Chickens, Nevi̇n Oktay, Seran Temelli̇, K. Tayfun Çarli

Two types of Enterococcus species were isolated from femoral head necrosis lesions of chickens. A total of 150 femoral articular tissues, of which 121 were from commercial broilers, 18 from broiler breeders, and 11 from layer breeders, were sampled. Out of 48 Enterococcus isolates of these clinical samples, 37 and 11 isolates were identified as Enterococcus faecalis and Enterococcus faecium, respectively. Additionally, 42 (28%) Escherichia coli and 33 (22%) Staphylococcus aureus isolates were obtained either alone or together with 1 of the 2 Enterococcus species from the same clinical samples, while no bacteria were detected from the remaining 56 samples. …

In this study Mentofin^® was used in commercial broiler chickens naturally exposed to infectious bursal disease virus (IBDV) and administered live Newcastle disease virus (NDV) vaccine to evaluate the ability of Mentofin^® to reduce Escherichia coli-related respiratory lesions, immunomodulate Newcastle disease (ND) vaccine response, change pharyngeal aerobic bacterial counts, and have certain impacts on specific production parameters. Presence of the chicken infectious anemia virus (CIAV) infection in chicken flocks in some provinces of Turkey was investigated by ELISA and PCR in this study. From 38 flocks (16 commercial layer, 10 commercial broilers, 4 breeder layers and 8 breeder broilers), 922 serum samples were tested for CIAV-specific antibodies using a commercially available competitive ELISA. CIAV antibodies were positive in 609 (66.0%) sera from 34 flocks (89.5%). A total of 95 thymus samples from 25 flocks, 57 samples from 15 commercial layers, and 38 samples from 10 commercial broiler flocks were tested by PCR. In 53 (55.8%) thymus samples …

Alleles of physiological candidate genes for reproductive traits, insulin-like growth factor-1 (IGF-1), and growth hormone receptor (GHR) were assessed to determine their associations with total egg production (NE), average days of continual egg-laying (ADCE), and number of double yolk eggs (DYE) in Wenchang chickens (Chinese indigenous breed). PCR-RFLP was used for genotype identification. The frequency of restriction enzyme C1/C2 alleles in the population was 0.53 (C1) and 0.47 (C2) for IGF-1. The objective of this experimental study was to examine the pathological effects of the intramuscular administration of gentamicin to growing chickens. Two experiments of 35 days´ duration were conducted simultaneously. In both experiments White Leghorn cockerels of 6 weeks of age were randomly divided into 4 groups and gentamicin was administered intramuscularly to the birds on days 1 and 3 at a dose of 10, 50, 100, and 0 mg/kg body weight, respectively. Immunomodulatory And Protective Effects Of Sugar Cane Juice In Chickens Against Eimeria Infection, Masood Akhtar, M. Abdul Hafeez, Faqir Muhammad, Ahsan Ul Haq, M. Irfan Anwar

Present paper reports the effects of sugar cane juice (SCJ) on immune responses and its protection against Eimeria (mixed species) infections in broiler chickens. Experiment 1 was conducted to test the dose efficacy of SCJ and its effects on chickens against coccidiosis. Maximum weight gain per day was recorded in chickens given SCJ @ 400 mg/kg body weight. Significantly (P < 0.01) low mortality and increased body weight gain per day were recorded in chickens administered SCJ orally and challenged with mixed species of genus Eimeria. Furthermore, mild hemorrhages, lower lesion scores (+1), and significantly lower number of oocysts shed in faeces were recorded in immunized chickens compared to control. Experiment 2 was carried out to evaluate the immunomodulatory effects of SCJ on cellular and humoral responses. Highest antibody titres (1:1024-1:16; GMT 367.28) were recorded in chickens administered SCJ @ 400 mg/kg body weight in comparison with control (1:16-1:2; GMT 2.82). To demonstrate the cell mediated immunity (CMI), amplitude of toe-web swelling 72 h post avian tuberculin injection was recorded in the experimental and control groups. Maximum swelling (8.13 ± 0.21 mm) was recorded in the experimental group as compared to control (1.04 ± 0.10); indicating higher cell mediated immune response. From these results it was concluded that SCJ elicit a significantly higher cellular and humoral responses compared to control, which may provide protection against Eimeria infections (mixed species) in chickens.

The aim of this study was to isolate Escherichia coli from chickens and to determine the presence of the eaeA gene, a virulence factor detected in E. coli, in the isolates by polymerase chain reaction (PCR). Different chicken organs were inoculated onto blood agar and biochemical tests were performed on the suspicious isolates. E. coli was isolated from 48% (48/100) of the samples. This study was undertaken to investigate the relation between abdominal fat and serum biochemical indices in chicken. In the study, 120 chickens at 12 weeks of age from Anka and Rugao breeds were used. They were reared under the same environment and management. Blood samples were taken, the serum was harvested by centrifugation, and then the total cholesterol (TCH), triglycerides (TG), and high-density lipoprotein (HDL) were assayed using an enzymatic kit. The objective of this study was to isolate, identify, and genotype Campylobacter jejuni from various sources in the province of Kayseri, Turkey. A total of 6667 samples consisting of 5167 human fecal swabs, 600 dog rectal swabs, 600 cattle gallbladders, and 300 chicken carcasses were examined. The samples were plated onto mCCDA (cefoperazone charcoal desoxycholate agar) agar. In order to identify C. jejuni, phenotypic tests and PCR (polymerase chain reaction) were performed. The aim of this study was to demonstrate the presence of estrogen and progesterone receptors (ERs and PRs, respectively) in oviduct adenocarcinomas in laying hens. In all, 32 oviduct adenocarcinomas (18 primary tumors and 14 intestinal metastases) from Babcock B-380 laying hens 16-18 months old were stained with commercially available monoclonal antibodies against mouse-anti-human ERs and mouse-anti-human PRs. Glutathione reductase (GR) is important in maintaining high levels of reduced glutathione in cells. The enzyme is inhibited by some drugs used in antimicrobial chemotherapy. The effects of some antibiotics on GR activity were investigated in this study. At first, the enzyme, having a specific activity of 88.28 EU/mg proteins, was purified 1636-fold with a yield of 35%. Purity of the enzyme was checked by means of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The histochemical and ultrastructural effects of water Cu on the anterior latissimus dorsi (ALD) muscle of 120 newly hatched broilers were investigated in a 45-day experiment. Two doses of water Cu (250 mg Cu/kg water and 500 mg Cu/kg water) were given to the poults. Two distinct categories (IIIA and IIIB) of slow tonic fibers in the control groups were detected by enzyme reactions. This study was carried out to isolate Clostridium perfringens from chickens and to detect the gene encoding the alpha toxin produced by all types of C. perfringens by polymerase chain reaction (PCR). Intestinal contents of 160 slaughtered chickens from 8 different farms in Elazığ province were analyzed. C. perfringens was isolated from 8 (5%) of the samples. The development and extent of quinolone-resistance in broiler isolates of campylobacters over a 15-year period was investigated. Minimal inhibitory concentrations of enrofloxacin, ciprofloxacin and nalidixic acid for 567 Campylobacter jejuni and 233 Campylobacter coli strains were determined by the agar dilution method. Of the strains, 180, 420 and 200 were isolated during 1987, 1992 and 2000, respectively. Enrofloxacin or ciprofloxacin resistance was not found in isolates from 1987. The first fluoroquinolone resistance (1.4% to enrofloxacin and 1.2% to ciprofloxacin) appeared in strains isolated in 1992, approximately 2 years after the licensing of enrofloxacin in Turkey. A total of 426 serum samples were examined from 9 broiler and 7 broiler breeder flocks showing clinical signs of swollen head syndrome (SHS). Turkey rhinotracheitis virus antibodies in 8 broiler flocks were between 31.8 and 75.9% and in broiler breeder flocks between 5.4 and 48.6%, with a commercial ELISA. Typical clinical symptoms were observed for SHS in broilers and broiler breeders. During the study, turkey flocks showing clinical signs were not detected. Detection Of Listeria Monocytogenes In Faeces From Chickens, Sheep And Cattle In Elazığ Province, Hakan Kalender

Faecal samples collected from 206 chickens, 170 sheep and 130 cattle were investigated for the presence of Listeria monocytogenes in animals slaughtered at local abattoirs in Elazığ. The prevalance of L. monocytogenes was 4.36% in chickens, 0.58% in sheep and 1.53% in cattle. Of the 12 L. monocytogenes isolates, seven (58.33%) and five (41.66%) belonged to serotypes 1 and 4, respectively. Other species isolated were L. innocua, L. welshimeri, L. seeligeri, L. murrayi and L. grayi. These results indicate that animal faeces could be a source of contamination of carcasses with L. monocytogenes and may lead to foodborne listeriosis.

In this study, organ and tissue samples were collected for routine histopathological examinations and also for the detection and definitive diagnosis of Marek's disease (MD) viral antigens by the Labeled StreptAvidin-Biotin Peroxidase (LSAB) method during post-mortem examinations of dead or sacrificed hens. This study aimed at an examination of the effects of different housing systems on the body weight, serum calcium levels, digit lengths and various parameters of the humerus and tibiotarsus. For this purpose 40 Tetra SL chickens were kept in caged, aviary, perchery and litter housing systems with 10 animals in each. After 72 weeks, body weight, digit lengths and serum calcium levels as well as the weight, volume, cortical thickness and bone ash contents of the humerus and tibiotarsus were measured. A change in the vaccination strategy used in the Central Anatolia region of Turkey to combat the effects of infections with endemic virulent Newcastle disease virus (NDV) in maternally immune birds is described in this retrospective work. A novel vaccination schedule was applied to approximately 2,230,000 chickens from different breeds. The alterations were based on the use of an inactivated NDV + infectious bursal disease virus (IBDV) and the application of aerosol vaccines with different timings compared to traditional programmes. Respiratory diseases due to infectious agents are an important problem in the broiler chicken industry. One of the most important infectious agents of chickens is Ornithobacterium rhinotracheale, which has recently been isolated in Turkey and around the world. In this report, O. rhinotracheale infection was detected in broiler chickens submitted to the poultry disease laboratory with respiratory problems, increased mortality and retarded growth.

The treatment of chicken caecal coccidiosis with sulfamonomethoxine, at a concentration of 0.05 % in drinking water for 3 days, was performed on different days post oocyst inoculation.162 chickens were divided into 6 groups, 3 cages per group with 9 chicken each group. Chickens in groups 1-5 obtained 7x104 oocysts of Eimeria tenella, via a stomach tube. Chickens in group 1, 2 and 3 were treated on days 1-3, 2-4 and 3-5 post inoculation respectively. In this study we examined factors affecting the colonization of campylobacters in chick intestines, colonization times and elicited immune responses. In chicks, infected orally by minimal infective dose _{100} (MID_{100}), colonization in the intestines was observed to reach 100% during the 3-week period following C. jejuni and C. coli inoculation. When the SDS-PAGE protein profiles of the inoculated and isolated strains were examined similarities of 98.3% among C. jejuni strains and 98.5% among C. coli strains were determined. The purpose of this study was to isolate Campylobacter species from the intestines and livers of chicken and to identify Campylobacter coli and Campylobacter jejuni by both conventional methods and Polymerase Chain Reaction (PCR). Four specific primers derived from the ceuE gene present in the genomes of C. coli and C. jejuni were used for PCR identification. In the examination of 150 intestine and liver samples by culture and PCR, 25 (16.6%) and 32 (21.3%) were identified as C. coli and C. jejuni, respectively. In this study, two subjects, 1) whether Visceral Larva Migrans (VLM) takes place when T. canis larvae-infected chick livers are eaten and 2) whether any differences are present between larval distributions, clinical signs and pathological disorders of the infections caused by eggs and infected liver, were investigated in the chick-mouse model. A total of 251 tracheal swabs and a total of 102 tracheae, lungs, airsacs, livers and spleens were collected from commercial layer, broiler and broiler parent stock flocks with respiratory infection. From turkey flocks, 96 tracheal swabs and 5 tissue samples from the trachea, lung, airsac, liver and spleen were also taken. Cultivations on Blood Agar supplemented with 5-10%sheep blood, on Chocolate Agar and on MacConkey Agar were incubated at 37ºC for 24-72 hours in the presence of 10%CO_2.
