The Effects Of Bsa And Fcs On In Vitro Maturation Of Bitch Oocytes, Mi̇that Evecen, Alper Baran, Serhat Alkan, Ö. Banu Özdaş, Çağatay Tek
Turkish Journal of Veterinary & Animal Sciences
The effects of protein sources added to the medium for maturation of bitch oocytes were investigated. Ovaries from spayed bitches were transported to the laboratory in PBS at 38 ºC within an hour. Recovered oocytes were divided into three groups and left for 72 h maturation at 38.5 ºC under an atmosphere of 5% CO_2 and high humidity. Group 1 (Control): TCM 199 medium, Group 2 (BSA): TCM 199 medium supplemented with 0.3% BSA, Group 3 (FCS): TCM 199 medium supplemented with 5% FCS. At the end of the maturation period, all oocytes were denuded from cumulus cells by pipetting …

Detection Of Cattle Infected With Theileria Annulata In Fields By Nested Pcr, Ifat And Microscopic Examination Of Blood Smears, Zati̇ Vatansever, Serpi̇l Nalbantoğlu
Turkish Journal of Veterinary & Animal Sciences
The aim of this study was to diagnose Theileria annulata infected cattle by microscopy, IFAT and nested PCR and to compare the sensitivity and specificity of these diagnostic methods. A total of 147 blood and sera samples were collected from healthy cattle in four localities of Polatlı district, Ankara, where tropical theileriosis is prevalent. Examination results revealed positivity rates of 31.3%, 44.9% and 61.2% for microscopy, IFAT and nested PCR, respectively. It was shown that nested PCR is more sensitive and specific when compared to microscopical examination and serological findings. It was also shown that nested PCR is more sensitive …

Turkish Journal of Veterinary & Animal Sciences
The liver damage and obstructions of the biliary tract were evaluated ultrasonographically in experimentally bile duct ligated dogs. Seven healthy dogs of different breeds were used. The common bile duct was double ligated at the duodenal region after clinical and ultrasonographic examinations. After that, clinical examinations, biochemical analyses and examinations of the liver, gallbladder and biliary tract were done ultrasonographically over 6 weeks. All dogs were necropsied and macroscopic and histologic alterations were determined at the end of the study. It was determined that the levels of serum ALT, ALP, GGT and T. Bilirubin were (p < 0.001-0.05) statistically significantly increased after the operation in all dogs. As a result of ultrasonographic examinations, it was determined that the bile ducts and gallbladder were distended, the neck of the gallbladder was larger and more tortuous than normal, hypoechoic sludge was prominent in the gallbladder, and perivascular, multifocal hyperechoic areas appeared in the liver parenchyma toward the end of the study. These alterations were confirmed macroscopically and microscopically at the end of the autopsy in all dogs. We consider that as well as blood serum parameters, ultrasonographic examinations are useful in the early diagnosis of biliary diseases and in differentiating them from hepatic diseases.

The objective of the present study was to transfer sheep embryos produced in vitro to recipient ewes. Ovaries were taken from slaughtered Kıvırcık ewes and transferred to the laboratory in phosphate buffered saline (PBS) at 30-35 ºC. The cumulus-oocyte complexes were obtained by slicing and washing 1-6 mm diameter follicles and matured for 24 h in medium 199 supplemented with sodium pyruvate, follicle stimulating hormone (FSH), luteinizing hormone (LH) and 10% fetal calf serum (FCS) at 38.5 ºC under 5% CO_2 in humidified atmosphere. Fresh semen was collected from three Kıvırcık rams, pooled and prepared for in vitro fertilization by …

Comparatively higher values of dry matter, crude ash and lipid were determined for the claw meat of males in winter. In the same season the crude protein and lipid composition of claws and crude ash of body meat were higher in females than other meat. The …

In this study, pastırma samples were produced using potassium nitrate and commercial starter culture (Staphylococcus carnosus + Lactobacillus pentosus). The meat for pastırma was dry cured with NaCl, glucose and saccarose together with KNO_3. The use of starter culture in pastırma production had a significant effect on counts of total aerobic mesophilic bacteria, Micrococcus/Staphylococcus and lactic acid bacteria. The maximum counts of microorganisms were found in the 2^{nd} stage of the drying process. There was no significant difference between yeast mould counts in control and starter added samples. Nitrite and nitrate residues were lower in commercial starter added samples than …

Lysozyme (EC 3.2.1.17) is an enzyme that catalyzes the hydrolysis of \beta(1-->4) glycosidic bonds between N-acetylglucosamine (NAG) and N-acetylmuramic acid (NAM), which are present in peptidoglycan heteropolymers of the prokaryotic cell wall. The purpose of this study was the cloning and expression of the bacteriophage T4 lysozyme gene (gene e) via PCR amplification in Escherichia coli using the pUC18 and pRS416 vectors. PCR amplification is carried out by using the primers having Bam HI recognition sites at their 5' ends. After digesting the vectors and PCR product with Bam HI endonuclease, two recombinant plasmids were constructed by ligation reaction …

In this research, the effects of some environmental factors on the reproductive characters of Awassi and Redkaraman ewes reared at the Research and Application Farm of Agricultural College at Atatürk University were investigated. Year, age of ewes, breed and the linear effect of body weight of ewes at the beginning of mating were studied. The year had no significant effect on any of the reproductive characters. Age of ewes, breed and body weight of ewes at the start of mating caused highly significant (P
