The latency-associated transcript (LAT) is the only abundant herpes simplex virus type 1 (HSV-1) transcript expressed during latency. In the rabbit eye model, LAT null mutants do not reactivate efficiently from latency. We recently demonstrated that the LAT null mutant dLAT2903 induces increased levels of apoptosis in trigeminal ganglia of infected rabbits compared to LAT+ strains (G.-C. Perng, C. Jones, J. The Use Of Fresh, Salt-Stored And Frozen, Immature And Mature Oocytes, To Assess The Penetration Ability Of Porcine Spermatozoa, Adisorn Adirekthaworn, Mongkol Techakumphu, Sudson Sirivaidyapong, Saroch Gnamkum

The objective of this study was to compare the penetration rate of sperm, collected from two Landrace Boars A and B, into fresh, salt-stored and frozen, immature and matured oocytes. The first experiment, performed on immature oocytes, revealed that the penetration rates for the three groups of oocytes was from Boar A 59.6%(59/99), 78.1%(75/96) and 77.8%(77/99) (p<0.05) and the number of sperm penetration each oocyte was 2.79±0.42, 2.97±0.29 and 2.29±0.26 respectively (p>0.05). While for Boar B, the percentages were 65.3%(62/95), 76.8%(73/95) and 77.8%(77/99) with the number of sperm per oocyte being 2.25±0.28, 3.63±0.42 and 2.57±0.36. The highest values were found in the salt-stored group (p<0.05). The second experiment involved sperm penetration into in vitro mature oocytes. No differences were found for the sperm penetration rate and the number of sperm per oocyte using semen from Boar A which were 85.1%(74/87), 86.1%(56/65) and 89.1%(79/88) and 13.87±1.45, 17.69±2.61 and 14.45±1.75. While for Boar B, the lowest penetration rate was found when using fresh oocyte, lower than the two other groups; 52.6%(50/95), 67.3%(64/95) and 69.1%(67/ 97)(p<0.05). The number of sperm entering each oocyte was also inferior in the fresh oocytes than in salt-stored and frozen ones; 1.55±0.31, 2.80±0.35 and 2.87±0.40 (p<0.05) respectively. The conclusion that can be drawn from the experiment is that a) there are significant differences in the sperm penetration ability of different the boars used and b) the type of oocytes; immature vs. mature and fresh vs. salt-stored vs. frozen influenced the results that were obtained. It maybe possible to apply such an in vitro penetration assay to determine boar semen fertility under field conditions.

The effect of methylparathion on cholinesterase activity in foot muscle tissues of cockle were evaluated. Ninety - five healthy cockle were randomly divided into control and experimental groups. The experimental group was exposed to 10 ppb of methylparathion for 90 hours. Measurements of cholinesterase activity revealed increasing enzyme activity in foot muscle (p < 0.05) in the exposed animals while clinical observations showed no differences between the groups. The results indicate that methylparathion affects the observed activity of cholinesterase in cockle. However, it should be noted that the data do not correlate with those of previous studies reported in other aquatic animal species.

Formalin fixed tissues from an aborted foal which had been diagnosed as having Equine Herpesvirus (EHV) infection, on histopathological criteria, were examined for EHV antigen using immunohistochemistry. EHV antigen was found in the focal necrotic area of the liver, especially in hepatocytes and Kupffer cells. Intranuclear and intracytoplasmic, immuno-positive labelings were also prominent in bronchiolar epithelial cells. The method allowed the visualization of the antigenic site distribution and their subsequent correlation with histopathological changes.

Ornithobacterium rhinotracheale is a recently discovered bacterium of the rRNA superfamily V, first named in 1994. The first occurence was reported in wild birds and later shown to be widespread worldwide in commercial poultry. It has not been reported in Thailand. The bacterium causes important ecomonic losses in the poultry industry. O. rhinotracheale can affect almost all avian species and is associated with respiratory diseases. Airsacculitis and pneumonia are the most common features of infection. These clinical signs are aggravated by other factors, especially respiratory viruses and bacterial infections. A Comparison Of Resazurin Test Scores On Milk, After 1 And 3 Hours And Comparing It With Total Bacteria, Coliform And Somatic Cell Counts, Sophat Chawalkul, Witaya Suriyasathaporn

The present study was conducted to compare results of a resazurin test on milk, scoring it after 1 hour and 3 hours and relating it to total bacterial count, coliform count and somatic cell counts. Milk samples from the bulk tanks of 337 dairy farms in Amphur Muang and Kumpangsan, Nakhon Pathom Province, were collected during October to December, 1998. Results from multiple regression analyses showed that the resazurin test scored 1 hour later was associated only with the total bacterial count (p < 0.05), but after 3 hours associations with total bacteria, coliform and somatic cell counts were seen (p < 0.05). We conclude that a resazurin test scored after 3 hours gave a better result than the test scored after 1 hour when judging total bacteria, coliform and somatic cell counts

The clinical course and laboratory valuation of 21 patients coinfected with human immunodeficiency virus (HIV) and Ehrlichia chaffeensis or Ehrlichia ewingii are reviewed and summarized, including 13 cases of ehrlichiosis caused by E. chaffeensis, 4 caused by E. ewingii, and 4 caused by either E. chaffeensis or E. ewingii. Twenty patients were male, and the median CD4+ T lymphocyte count was 137 cells/μL. Exposures to infecting ticks were linked to recreational pursuits, occupations, and peridomestic activities. For 8 patients, a diagnosis of ehrlichiosis was not considered until ≥4 days after presentation. Severe manifestations occurred more frequently …

A prospective study established the etiology of febrile illnesses in residents of Slovenia that occurred within 6 weeks after a tick bite. A combination of laboratory and clinical criteria identified 64 (49.2%) of 130 patients as having confirmed, probable, or possible cases of tickborne disease during 1995 and 1996. Of the 130 patients, 36 (27.7%) had laboratory evidence of tickborne encephalitis, all of whom had clinically confirmed disease. Evidence of infection with Borreliab urgdorfersie sensu lato was identified in 26 patients; 10 (7.7%) had confirmed Lyme borreliosis. Epidemiologic and clinical characteristics of fatal and nonfatal cases of Rocky Mountain spotted fever (RMSF) were compared to identify risk factors for death caused by this disease. Confirmed and probable RMSF cases reported through US national surveillance for 1981- 1998 were analyzed. Among 6388 RMSF patients, 213 died (annual case-fatality rate, 3M; range, 4.9 % in 1982 to 1.1 % in 1996). Use of tetracycline-class antibiotics for treatment of RMSF increased significantly in the 1990s, compared with use in the 1980s. Older patients, patients treated with chloramphenicol only, patients for whom tetracycline antibiotics were not the primary therapy, and patients …

Scrapie is a naturally occurring prion (PrP) disease causing a fatal neurodegenerative disorder in sheep and goats. Previous studies suggest that scrapie is transmitted naturally through exposure to the scrapie agent in wasted placentas of infected ewes. This study determined the distribution and biochemical properties of PrP cellular (PrP-C) and the distribution of PrP scrapie (PrP-Sc) in reproductive, placental, and selected fetal tissues and fetal fluids in sheep. Glycosylated, N-terminally truncated, proteinase K-sensitive PrP-C with apparent molecular masses of 23–37 kDa was present in reproductive, placental, and fetal tissues and fetal fluids. Following diagnosis of scrapie in a clinically suspect Suffolk sheep, 7 clinically normal flockmates were purchased by the Pennsylvania Department of Agriculture to determine their scrapie status using an immunohistochemical procedure. Two of the 7 euthanized healthy sheep had positive immunohistochemical staining of the prion protein of scrapie (PrP-Sc) in their brains, nictitating membranes, and tonsils. The PrP-Sc was localized in the areas of the brain where, histopathologically, there was neurodegeneration and astrocytosis. The PrP-Sc occurred within germinal centers of the affected nictitating membranes and tonsils and was located in the cytoplasm of the dendrite-like cells, lymphoid cells, and macrophages. …

To determine the transmissibility of chronic wasting disease (CWD) to cattle and to provide information about clinical course, lesions, and suitability of currently used diagnostic procedures for detection of CWD in cattle, 13 calves were inoculated intracerebrally with brain suspension from mule deer naturally affected with CWD. Between 24 and 27 months postinoculation, 3 animals became recumbent and were euthanized. Gross necropsies revealed emaciation in 2 animals and a large pulmonary abscess in the third. Brains were examined for protease-resistant prion protein (PrPres) by immunohistochemistry and Western blotting and for scrapie-associated fibrils (SAFs) by negative-stain electron microscopy. Buggy Creek (BCR) virus is an arthropod-borne alphavirus that is naturally transmitted to its vertebrate host the cliff swallow (Petrochelidon pyrrhonota) by an invertebrate vector, namely the cimicid swallow bug (Oeciacus vicarius). We examined how the prevalence of the virus varied with the group size of both its vector and host. The study was conducted in southwestern Nebraska where cliff swallows breed in colonies ranging from one to 3700 nests and the bug populations at a site vary directly with the cliff swallow colony size. Twenty Departmental students completed their Bachelor of Science in Agricultural Science degrees in 2001, nineteen in the Veterinary Science Major and one in the Veterinary Technologist Major. Twenty-five Nebraska residents were admitted into the College of Veterinary Medicine at Kansas State University, twelve of them having completed all or part of their pre-veterinary studies at UNL. Seven students completed their Master of Science degree in Veterinary Science. During a survey of carnivores and omnivores for bovine tuberculosis conducted in Michigan (USA) since 1996, Mycobacterium bovis was cultured from lymph nodes pooled from six coyotes (Canis latrans) (four adult female, two adult male), two adult male raccoons (Procyon lotor), one adult male red fox (Vulpes vulpes), and one 1.5-yr-old male black bear (Ursus americanus). One adult, male bobcat (Felis rufus) with histologic lesions suggestive of tuberculosis was negative on culture but positive for organisms belonging to the Mycobacterium tuberculosis complex when tested by polymerase chain reaction. The summer drenching program has provided highly effective sheep worm control in Western Australia for many years - but recent research challenges its long term sustainability.
Trial results suggest that in large parts of the State, summer drenching is the main factor leading to the development of drench resistant worms. Alternative programs less likely to lead to drench resistance will require greater monitoring of worm burdens and panning pasture moves.

Bacillus subtilis RSKK246 was found to produce approximately a 65-kDa \alpha-amylase enzyme. A gene was isolated encoding \alpha-amylase activity that corresponded to this size and was inserted into pUC18 plasmid which was transferred to Escherichia coli. An approximately 1.7kbp fragment, which contains a whole \alpha-amylase gene, was excised and inserted into pUB110 and then transferred into the different B. subtilis strains including RSKK246, RSKK243, RSKK244, YB886 and ORBAM. The a-amylase gene was cloned into the plasmids and expressed with its own promoter, and this promoter sequence seemed to function in the E. coli and in all B. subtilis strains. Specific …

Clenbuterol is a β2 agonist/antagonist bronchodilator, and its identification in post-race samples may lead to sanctions. The objective of this study was to develop a specific and highly sensitive serum quantitation method for clenbuterol that would allow effective regulatory control of this agent in horses. This study was carried out to determine the prevalence of Cryptosporidium spp. oocysts in diarrhoeic calves in Kars province. The study was conducted between February 1999 and June 1999, involving 140 diarrhoeic calves from 8 different localities (Kars-Central district, Karakaş, Bulanık, Karakale, Hacıhalil, Boğazköy, Susuz-Central district, and Arpaçay). These localities were visited once during the study period, and fresh faecal samples were taken from the rectums of the calves up to 3 months old. This study was carried out in Malatya and vicinity to determine the seroprevalance of Babesia ovis in sheep. A total of 220 sheep sera were tested against B. ovis using the Enzyme-Linked Immunosorbent Assay (ELISA). In addition, peripheral blood smears from these sheep were prepared and examined for B. ovis. The ELISA test showed that 55.9% of sheep were seropositive for B. ovis infection. The infection rate was 49.2% in sheep which were 1-12 months old and 59% in sheep which were older than one year. In the microscopic examination of blood smears, four out of 220 sheep were found …

Some characteristics related with the fattening performance of Tushin lambs reared in the Research and Application Farm of Ataturk University and subjected to intensive fattening were determined. The inital fattening weight, final fattening weight, and total and daily weight gain during the fattening period were 19.71 kg, 38.07 kg, 18.35 kg and 203.48 g respectively. An 8-year-old cow admitted to the Veterinary Faculty, Internal Medicine Clinic (University of Fırat, Elazığ/TURKEY), was diagnosed with intussusception, and the diagnosis was confirmed by experimental laparotomy. Intussusception was corrected by enterectomy and intestinal anastomosis. Defecation on the day following the operation indicated normal intestinal flow. The animal was kept under observation for ten days after the operation, and had a full recovery.

This study was conducted in two stages to determine the ensiled qualities of corn, alfalfa and sugar beet pulp, and the effects on food intake and nutrient digestibility of corn and alfalfa silages used alone, together and with sugar beet pulp silage in lambs. In the first stage, corn, alfalfa and sugar beet pulp were ensiled and silos were opened after 45 days. Silage samples were taken for determination of silage quality and the samples were analysed. The sera collected from 152 sheep older than one year old sheep in various parts of Yozgat Province were tested for Toxoplasma gondii antibodies using the Sabin- Feldman dye test. Out of the 152 sera examined, 69 (45.4%) were seropositive at different dilutions (34 samples at 1/16, 21 at 1/64, 11 at 1/256 and 3 at 1/1024).

A Study On Direct And Indirect Effects Of Some Factors On Weaning Weight Of A Awassi Lambs, Mehmet Topal, Nuri̇ni̇sa Esenbuğa

The correlation coefficients divided into direct and indirect effects among ewe weight in the mating period and lambing, birth weight and weaning age of lamb, and weaning weight of Awassi lambs raised in Erzurum conditions were determined in this study. In path analysis, the direct and indirect impact of birth weight and weaning age of lamb on weaning weight were found to be highly significant. Partial correlation and standardized partial regression coefficients among weaning weight, birth weight and weaning age of lamb were also observed to be highly significant.
