Isolation And Preparation Of Monoclonal Antibody To Ovineimmunoglobulin M, Hüseyi̇n Yilmaz, Aysan Tantaş, Ati̇lla Ilgaz, Kenton L. Morgan
Turkish Journal of Veterinary & Animal Sciences
Ovine immunoglobulin M was isolated from sheep serum using gel filtration and affinity chromatography. Polyclonal and monoclonal antibodies against this isotype were prepared. The specificity of the monoclonal antibody was investigated using SDS-Page, immunoblotting, an enzyme-linked immunosorbent assay (ELISA) and affinity chromatography. On immunoblotting, monoclonal anti-IgM reacted with a 70 kD protein which corresponded to ovine IgM.

The objectives of this study were to investigate the effectiveness of ultrasonography in monitoring ovarian activity by examining follicular growth and regression and studying the correlation between corpus luteum (CL) measurements and plasma progesterone (P4) concentrations, and to compare high-and low-production genetic lines of dairy cows regarding endogenous bovine somatotropin (bST) and milk production effects on follicular wave patterns, size and number of follicles, CL growth and plasma P4 concentrations during the estrous cycle monitored by ultrasound. In animal breeding, when there is a relationship between the dependent (Y) and independent (X) variables, regression analysis is applied. But when one of the variables has one or more missing observations regression analysis cannot be applied. The location of extrinsic afferent and efferent neurons projecting to the rat gastrointestinal tract were mapped using the retrograde tracer, True Blue (TB). Injection of retrograde tracer, TB into stomach, proximal colon and distal colon resulted in labelled cells in the dorsal root ganglia. Cells were also labelled in the nodose ganglia following injection of tracer into stomach and proximal colon. TB-labelled cells were also seen in the prevertebral ganglia (coeliacomesenteric complex and inferior mesenteric ganglion), after injection of TB into the stomach, proximal and distal colon. In this study, 6 six-week-old puppies born of a stray bitch were used. Four of them were 5 kg, one of them 7 kg and the last 8 kg. Before egg inoculation, fecal examination by flotation technique showed that the animals had no helminth infection. Each puppy was infected orally with 150 embriyonated Toxocara canis eggs. The present work was undertaken to show the histological structure and histometrically features of thymus and bursa of Fabricius of native geese. The general histologic structure of bursa of Fabricius was similar to the structure of many other birds stated in literature. By histometrical measurements, it was determined that the mean thickness of cortex and medulla of the lymphoid follicles in plicae were 89.06±3.23 ??and 196.8±3.08 ??respectively. The mean diameters of lympocytes in cortex were 4.54±0.08 ??while those of plasma cells were 6.36±0.33 ?. In this study, 30 Budgerigars ( Melopsittacus undulatus) with Knemidocoptes pilae infection were treated with a spot-on application of moxidectin (Cydectin® Injection For Cattle) at a dose of 1 mg (0.1 ml Cydectin) per bird. This application was given once to 10 birds and twice at 10 days intervals to 20 birds. Clinical signs in all birds of both groups disappeared 30-40 days after treatment. No side effects were seen.

Performance data on fertility of ewes, pre-weaning mortilaty of lambs and productivity were collected during two years from 24 village flocks in Cnetral Anatolia. The data included flocks with sheep from the Akkaraman and Merino breeds and their crosses. The overall means for lambing rate, weaning rate, twining rate and pre-weaning lamb mortality were 92.1%, 89.8%, 4.8% and 6.9%, respectively. The fertility traits were significant effected by genetic group, area were the flock was located and age of ewe and the lamb mortality by the first two effects. In the present study, the growth of the Mediterranean mussel Mytilus galloprovincialis Lam., 1819 in the Black Sea on ropes has been studied. The result of a 18 month-trial showed that the mussels grew 72.84±0.74 mm in lenght. Temperature of seawater has been measured (minimum of 7ú°C in the month of February and maximum of 27.7°C in August). Salinity varied from 17.2 to 19.3.

This study aimed to investigate relationships between steroidogenic characteristics and abilities, and some macroscopic characteristics of bovine ovarian follicles and to relate this criterion to the physiological status of the follicles. Follicular fluid of 5 to 8 mm diameter follicles from bovine ovaries (n=46) were analysed and steroidogenic activities of granulosa cells were studied. For the macroscopic classification of the follicles (n=232), appearence of the follicles, thecal vascularization, the colour of theca interna and the presence of follicular debri were determined. In this study; macroscopic and microscopic changes induced by Avian Encepholomyelitis Virus (AEV) in turkey embryos were investigated. For purpose 0,1 ml. AEV-VR (Van Roekel Strain) at Embriyo Infective Dose 50 of 10 -3 diluted in steril saline at 1/10 ratio was inoculated into 7-day-old emriyonated turkey eggs via yolk sac. It was reportked a case of naturally occurring zinc deficiency in a calf with it's clinical, histopathological, haematological and biochemical findings in province of Elazığ, Turkey. Zinc oxide was administered per orally, in the dose of 100 mg/day for 8 days, and then 40 mg/day for 7 days, and 40 mg/week for 4 weeks. A four-month study of the distribution of morphologic lesions relating to encephalitozoonosis and the staining proper-ties of Encephalitozoon cuniculi was carried out on 40 rabbits from a breeding colony of 100 White New Zealand rabbits. Prominent and constant gross lesions, distinguished by subcapsular, small depressions, were identified in the kidneys of 37 rabbits (92.5%). These lesions were bilateral, and exhibited focal and multifocal distribution. Microscopic examinations of the kidneys revealed that the lesions were characterized by non-purulent granulomatous interstitial nephritis (55%) and chronic interstitial nephritis (45%). This paper reports an investigation of the effects of acute and chronic carbon tetrachloride intoxications on the histologic structure of the liver and hematological values and electrocardiograms in rabbits. The rabbits were divided into 3 groups of 7. The first group was the control group, the second group was acutely intoxicated and the third group was chronically intoxicated. In the acutely intoxicated group, caryorexsis, along with necrosis was observed in the hepatocyte of the centrolobular area of the liver. Infectious bursal disease (IBD) field viruses were isolated from the bursa Fabricius (BF) materials collected from the IBD infected or suspected flocks in different geographical regions of Turkey during 1992-1994. The field viruses were adapted to embryonated spesific-pathogen-free (SPF) eggs and chicken embryo fibroblast (CEF) cell cultures by serial blind passages and characterized by virus neutralization test (VN). The investigation was performed to develope sire lines for lamb production at Acipayam state Farm. For this purpose, German Black Face (A) x Acipayam (AC) and Ramliç (R) x Acipayam (AC) cross-bred were used. According to the results; Fecundities are 109%, 72.0%, 85.0%, 95.0% and 85.0% for ACxAC, AxAC, RxAC, AxAC and RxRAC cross-bred groups respectively. Weaning weights are also 22.45 kg, 22.23 kg, 24.38 kg, 21.48 kg and 23.06kg for AC, AAC, AAAC, RAC and RAAC genotype respectively.

This study was carried out on cows, sheep and goats in Salkaya Village, Kovancılar, Baskıl and Sivrice towns and in Elazığ during 1994 and 1995. Fifty two myiasis cases were detected throughout the study. Forty seven (90.38%) and 5 (9.62%) of the cases were determined in sheep and cows respectively, no myiasis case was encountered in goats. Twenty two (42.32%) of the cases were seen in June during 1994 and 1995, 10 (19.23%) in May, 9 (17.31%) in July, 8 (15.38%) in September and 3 (5.77%) in August. Caecal coccidiosis was determined in 12 white Leghorn chickens aged 18 weeks. Macroscopically, slight thickness, con-gestion and petechie were observed in the caecum. The lumen of caecum was full of blood and, necrotic areas were seen on the mucosal surface due to the damagae of villi epithelisl cells. Microscopically, it was observed that erythrocyte, epithelial cells and forms of parasites were present in the lumen of caecum due to the damage of villi epithelial cells. Lymphocyte, plasma cell and heterophil cell infiltrations were found in the propria mucosa as well as gangrenous typhilitis containing focal necrosis.

Postoperative clinical, histopathological and radiological findings were evaluated for disc fenestration, open discolysis (chy-mopapain) and percutan discolysis which had been performed on three groups of dogs, each group containing 6 healthy mongrels of different ages, sexes and breeds. The degrees of intervertebral space narrowing were classified as mild, medium and severe according to postoperative radiological examinations. Sections which were prepared from intervertebral areas which had been operated on, were investigated histopathologically. This study was made on 79 dogs and 24 cats with dermatomycosis were came to the clinic of Internal medicine of Veterinary Faculty, İstanbul University with complaint of a skin problem to detect the incidence dermatomycosis. Mycological cul-tures were made from the skin samples of infected animals in the laboratory of Microbiology Department. 65% of Microsporum canis, 20% M. nanumand %15 of Candida spp.was diagnosed in all cases. The aim of this study was investigated the bones of pelvic limb of two porcupines (Hystrix cristata). Incisura acetabuli was narrow and when examining at ventral direction the surface areas of the ilium bones were parallel while the wing (ala ossis ilii) and the body (corpus ossis ilii) were very wide and narrow, respectively. A haematological study was carried out with rainbow trouts experimentally infected with Yersinia ruckeri. One hundred rainbow trouts ( Oncorhynchus mykiss) at 192±0.24 g were examined. The present study was conducted to determine of incidence and serotype distribution of salmonellae in ground beef on markets in Ankara. Between February 1994 and January 1995, a total of 120 samples were obtained from different butcheries and supermarkets of different regions in Ankara. Salmonellae were found in 3.3% (4/100) of ground beef samples. Three different serotypes were identified among the cultures isolated namely S. anatum (2), S. typhimurium (1) and S. telaviv (1). The arrhythmias occurring and caused to death during the anaesthesia is generated endogenous release of epinephrine or norepinephrine. Therefore, the effects of yohimbine (alpa-2 blocker), prazosin (alpha-1 blocker), metoprolol (beta-1 blocker) on the arrhythogenic dose of epinephrine in 10 dogs anesthetized with xylazine and kethamine combination, were researched in this study. The infusion of epinephrine, 1 mg/ml/min; yohimbine, 0.125 mg/kg; prazosin, 0.1 mg/kg; metoprolol 0.5 mg/kg in dose were made intravenously. In this study, the changes (IN) the iodine number (IN), the peroxide value (POV), and the refractive index (RI) of whiting ( Merlangius merlangus euxinus Nord., 1840) liver oil stored in a refrigerator and the relationships between them (4±1°C) were investigated. In a nine-week period, the (IN), the POV and the RI of whiting liver oil stored in the refrigerator were determined to be 169.39- 112.06, 3.56-15.25 meqg O 2 /kg and 1.4818-1.4777 respectively.
