The Thai Journal of Veterinary Medicine

Tuberculosis caused by Mycobacterium tuberculosis is an important zoonotic disease that concerns the survival of endangered species and public health aspects. A 17-year-old, female Malayan tapir from a zoo in Thailand died with a sign of panting after anesthesia for the treatment of her oral problems. Grossly, multiple caseous white nodules were scattered throughout the parenchyma and bulged from the capsular surface in affected organs such as lung, lymph node and liver. Histologic examination revealed multifocal to coalescing granulomas characterized by caseous necrotic center, loosely organized inflammatory zone with low numbers of Langhans’s giant cells and thin fibrotic zone. The aim of the present study is to determine the distribution of immune cells within the endosalpinx related to the ovarian function in gilts culled due to repeat breeding and anoestrus. A total of 30 reproductive organs from culling crossbred Landrace x Yorkshire gilts were collected from commercial swine herds in Thailand and categorized into 3 groups, i.e. anoestrus, repeat breeding and non-involved in reproductive disorder gilts (control group). The ovaries and oviducts were separated from the reproductive tracts and macroscopically investigated. A study was conducted to determine the effects of dietary graded levels of melamine, urea-formaldehyde (UF) and their mixture on growth performance, carcass quality, melamine residues and microscopic changes in broiler tissues from d-1 to d-42 followed by a 7-d feeding of withdrawal diets. One thousand and forty 1-d-old equal mixed-sex Arber Acres broiler were assigned to 13 dietary treatments with 4 replicate pens of 20 chicks assigned to each treatment. The objective of this chronic toxicity study was to investigate the safety of ethanolic extract of chronic Kaempferia parviflora Wall ex Bak in Wistar rats. Salbutamol Residues In Swine Tissues And Body Fluids After Feeding, Jen Hung Ming, Hsiang Huang Han, Ling Chen Chao, Jen Wu Yu, Katie Marie Dixon, Liang Mao Chi

The Thai Journal of Veterinary Medicine

Salbutamol residues in swine tissues and fluids were investigated. Tissue, serum and urine samples were collected from 33 pigs after 14 days of oral administration of 3 ppm salbutamol. The concentrations of salbutamol were determined by enzyme-linked immunosorbent assay and further confirmed by capillary electrophoresis. The highest urinary concentration of salbutamol was 145.12 ng/ml at 18 hour and remained detectable for 30 days after the last feeding. Salbutamol was undetectable in serum samples. Forced impregnation process is normally performed under vacuum at freezing temperature which requires expensive plastination apparatus. To reduce the cost in the plastination process, forced impregnation of porcine hearts under two different environments, which are at the freezing temperature (-20oC) and at room temperature (22-25oC), was compared. Twelve porcine hearts were collected from a slaughter house, and divided into the groups, six in each, and subjected to be plastinated by S10 standard technique at the two temperatures. An Outbreak Of Avibacterium Paragallinarum Serovar B In A Thai Layer Farm, Kridda Chukiatsiri, Suwit Chotinun, Niwat Chansiripornchai

Avibacterium paragallinarum serovar B was diagnosed and revealed in a layer farm. The infected chickens showed mucous nasal discharge, facial edema and conjunctivitis. Egg production also decreased dramatically. Necropsy findings showed lesions of conjunctivitis and edematous tissue around the eyes. The infraorbital sinuses were filled with a caseopurulent mass and tracheitis. Bacterial isolation and identification was performed and confirmed by PCR technic. A hemagglutination inhibition test using specific antibodies against serovar B revealed the Avibacterium paragallinarum serovar B infection. Eleven microsatellite loci were evaluated for genetic profiles of Thai domestic elephants (Elephas maximus indicus) and their suitability as genetic markers for molecular testing. A total of 66 animals were tested. 10 out of 11 loci could be amplified and they demonstrated polymorphism with allelic numbers per locus ranging from 7 (LaT06) to 39 (LaT18). Values of expected heterozygosity (He) and Polymorphic Information Content (PIC) were between 0.6449 (LaT17) – 0.9593 (LaT05) and 0.5934 (LaT17) – 0.9578 (LaT05), respectively. In order to develop the most practical plastination specimens used for anatomical study, the aim of the present study was to compare between using acetone and absolute alcohol for dehydration in plastination procedure. Ten pig heart specimens were used in the present study and plastination procedure was processed by using acetone (group 1) or absolute alcohol (group 2) for dehydration. The results showed that dehydration by acetone resulted in more natural color with no shrinkage of the heart specimens while dehydration by alcohol caused darker color and more shrinkage. The antimicrobial activities of organic releasing chlorine disinfectant (Virusnip™) was evaluated and compared by semi-quantitative suspension-neutralization method under clean condition in vitro. The test organisms were clinical bacterial isolates of hemolytic Escherichia coli, Salmonella Typhimurium, Salmonella Enteritidis, Salmonella Hadar, Salmonella Infantis, and Salmonella Virchow. Fungal isolates were Candida albicans and Trichophyton mentagophytes. Four sets of Virusnip™ concentrations were prepared at 1:100, 1:200, 1:400 and 1:1000 for bacteria and 3 sets of 1:100, 1:200 and 1:400 for fungi. Prevalence Of The Cellular And Molecular Antimicrobial Resistance Against E. Coli Isolated From Thai Broilers, Sarawoot Mooljuntee, Piyarat Chansiripornchai, Niwat Chansiripornchai

The antimicrobial resistant characteristics both of the cellular and molecular levels of Escherichia coli in broilers were performed by disk diffusion test and polymerase chain reaction technique. Specific primers for the aadA, aac(3)-IV, cmlA, cat1, tetA, sul1, SHV, CITM, ereA and dhfrV gene were used to detect resistance to gentamicin, chloramphenicol, tetracycline, cephalothin, ampicillin, erythromycin, sulfonamide+trimetroprim. The 30 samples of E. coli were found to be 100% antimicrobial disk resistant to tetracycline, ampicillin and erythromycin and revealed a 90%, 93.3% and 73.3% resistant gene to tetA, CITM and ereA, respectively. The study was conducted to examine the effect of different parthenogenetic activation (PA) protocols on developmental competence and quality of embryos, and to establish the parthenogenetic embryonic stem (ptES)-like cells in pigs. Compacted cumulus-oocyte complexes (COCs) were collected from porcine ovaries and then cultured in M199-medium. An isolate of Actinobacillus (A.) pleuropneumoniae from porcine tonsil samples was identified as serotype 15-like strain, which was previously isolated in Australia, North America and Japan. It was characterized by biochemical testing performed for Actinobacillus sp. and confirmed as A. pleuropneumoniae by PCR and nucleotide sequencing of 16s rDNA. The isolate contains apxIBD, apxIICA, apxIIIBD, apxIIICA and apxIVA gene. The result of PCR-REA of apxIVA method was identical as A. pleuropneumoniae serotype 15 strain HS143 reference strain. The purpose of this study is to identify the relationship of calcium concentration in seminal plasma on elephant semen quality including of volume, concentration, pH and percentage of progressive motility, dead sperm and abnormal morphology, respectively. Semen collection and evaluation were done in 9 elephants of Thai Elephant Conservation Centre, National Elephant Institute, Forest Industry Organization at Lampang. Calcium in seminal plasma was measured by using colorimetric method. Data were analyzed by using Linear Regression. The results revealed that amount of calcium in seminal plasma was negative correlated with only percentage of progressive motility (p<0.05). Analysis On The Polymorphism And The Genetic Effects On Some Economic Traits Of Mx Gene S631n Mutation Site In Chicken, Luan De Qin, Bin Chang Guo, Wei Sheng Zhong, Liu Yan, Hong Chen Guo

PCR-RFLP technique was performed to detect the distribution differences of the resistant and sensitive alleles of S631N site of Mx gene in 8 chicken populations. The results showed the average frequencies of resistant allele A and sensitive allele G were 0.4995 and 0.5005, respectively, and the average value of observed heterozygosis and Shannon’s index were 0.6163 and 0.5232. Three populations were in Hardy-Weinberg equilibrium at this site (p>0.05). The Ewens-Watterson test indicated that this site was neutral in all the other populations except for Rugao. The characteristic of erythrocytes, leukocytes and thrombocytes of a captive Irrawaddy river dolphin (Orcaella brevirostris) was described using blood sample collected from caudal vein. By morphological and ultrastructural studies, erythrocytes showed similar size as in humans and contained cytoplasmic electron dense hemoglobin without nucleus. Multilobulated granulocytes composed of cytoplasmic round-to-elongated granulecontaining neutrophils and round-to-oval shaped granule-bearing eosinophils while basophil was very scanty. Monocytes displayed azulophilic granules and vacuolated cytoplasm with pseudopodia. Lymphocytes showed equal amounts of heterochromatin and euchromatin with numerous mitochondria in their cytoplasm. Chronic Mastitis In Small Dairy Cattle Herds In Muang Khon Kaen, Jaruwan Kampa, Varaporn Sukolapong, Anantachai Chaiyotwittakun, Sarinya Rerk-U-Suke, Arunee Polpakdee

Monthly records of bulk tank milk somatic cell count (BMSCC) of all 194 dairy herds in Muang, Khon Kaen were used as an indicator of chronic mastitis problems; i.e. BMSCC > 500,000 cells/ml at least 3 consecutive records. Twelve chronic mastitis herds in Muang, Khon Kaen were data collected on 1) general herd information, 2) milking equipment, 3) milking procedure and 4) awareness and management of farmers to the mastitis problem. Blood pressure, urinary protein creatinine (UPC) ratio and oxidative stress (plasma melondialdehyde, urinary melondialdehyde creatinine ratio; UMDA/Cr, the activities of red blood cell catalase; RBC-CAT and red blood cell glutathione) were studied in 42 dogs with urolithiasis and 14 control healthy dogs. The types and location of calculi were defined. The results showed that calculi preferentially found in male and female dogs were calcium oxalate and struvite, respectively. In female, calculi found mostly at urinary bladder while in male, they could be found in either urinary bladder and/or urethra. Dogs with urolithiasis were normotensive.
