Current testing methodologies within equine forensic toxicology focus on arbitrary thresholds and zero-tolerance policy. Modern analytical chemistry’s limits of detection are low enough that oftentimes femtogram-per-milliliter amounts of a substance can readily be identified in both blood and urine of a horse. For most pharmacologically relevant compounds, these concentrations have no relevance to pharmacological effect. It is therefore crucial that testing methodologies to determine appropriate thresholds and cut-offs be developed that are driven by biological activity rather than arbitrary limits of detection. Development Of Mucoadhesive Polymeric Nanoparticles For Intranasal Delivery Of Favipiravir In Treating Sars-Cov-2 Infection, Khent Alcantara

Nanoparticles (NPs) have been used as an effective carrier for transmucosal delivery of drugs through their prolonged residence time and deposition, or the enhanced permeation through the mucosa. In this study, we aimed to design a nanocarrier using the combined mucoadhesive (M) properties of chitosan (CS) and alginate (ALG) polymers for the intranasal delivery of favipiravir (FVR) in treating SARS-CoV-2 infection. The optimized FVR-loaded mucoadhesive CS-coated ALG-NPs (FVR-MCS-ALG-NPs) rendered suitable size (233.5 nm), zeta potential (-21.6 mV), loading capacity (26.0%), and encapsulation efficiency (84.6%) for transmucosal delivery. Treatment of central nervous system (CNS) diseases, such as Alzheimer’s disease, Parkinson’s diseases, and brain tumors, is still a challenge due to the existence of the blood-brain barrier (BBB). The BBB as a membrane barrier that separates systemic blood circulation from the CNS is structurally formed by endothelial cells, astrocyte, pericyte and neurons and is one of the crucial protection mechanisms for the CNS. The BBB protects the brain by preventing foreign compounds (non-endogenous compounds) from entering the brain and helps maintain brain homeostasis. In addition, the presence of drug efflux pumps (i.e., P-glycoproteins, multidrug-resistance proteins) in the BBB helps …

Regeneration of damaged periodontal tissues becomes one of the biggest challenges that is hard to overcome, even without the presence of any pathogens. Tissue engineering was proposed as a possible technique for the lost tissues’ restoration. In this study, methacrylated hyaluronic acid (MeHA) hydrogel was developed with different concentrations of low molecular weight (short-chain) hyaluronic acid to achieve the hydrogels with suitable physical properties and biological properties for dental tissue regeneration. Cancer stem cells (CSCs) drive aggressiveness and metastasis by utilizing stem cell-related signals. In this study, 5-O-(N-Boc-L-alanine)-renieramycin T (OBA-RT) was demonstrated to suppress CSC signals and induce apoptosis. OBA-RT exerted cytotoxic effects with a half-maximal inhibitory concentration of approximately 7 µM and mediated apoptosis as detected by annexin V/PI and nuclear staining assays. Mechanistically, OBA-RT exerted dual roles, activating p53-dependent apoptosis and concomitantly suppressing CSC signals. A p53-dependent pathway was indicated by the induction of p53 and the depletion of anti-apoptotic Mcl-1 and Bcl-2 proteins. Cleaved PARP was detected in OBA-RT-treated cells. Quinuclidine derivatives (s)-T1, (s)-T2, and (s)-T6 are the ligands of α3β4 nicotinic acetylcholine receptor (nAChR). α3β4 nAChRs are highly expressed in medial habenula (MHb). The MHb connects to the brain rewarding pathway and mediates nicotine induce rewarding effect. Various α3β4 nicotinic acetylcholine antagonists have been investigated for their anti-addictive effect in animal models of nicotine addiction. The present study was aimed to evaluate the anti-addictive effects and mechanisms of quinuclidine derivatives, (s)-T1, (s)-T2, and (s)-T6 in nicotine-induced conditioned place preference (CPP) in mice. While e-cigarette vaporizers are primarily used for the delivery of nicotine, they can also be used to administer a wide range of other substances of abuse including stimulants such as d-amphetamine and methamphetamine. Methamphetamine and prescription amphetamines are widely abused throughout the United States and across the world leading us to believe there may be abuse potential with vaping methamphetamine as vaping becomes increasingly popular. Electronic cigarettes (e-cigarettes) are one of the major sources of exposure to nicotine, an addictive chemical. Since 2016, the Food and Drug Administration (FDA) has started regulating these products under the Tobacco Control Act yet, specifications about the nicotine content in these products have not been established. In e-cigarettes, nicotine concentration ranges from 0 to >50mg/mL. Additionally, several e-cigarette related parameters have been identified which can affect nicotine delivery to users. PURPOSE: Albumin has been shown to be safe and effective in clinical practice for a wide variety of indications. The purpose of this medication use evaluation is to quantify the use of albumin in the community hospital setting based on indication and prescribing department.
METHODS: This study is a retrospective, single-center, chart review over a 6-month period of 186 patients aged 18 and older who were treated with IV human albumin 5% or 25% at a single 202-bed community hospital setting from February 1, 2020, to August 1, 2020. Purpose: This work describes the late-stage product portfolios of the biotechnology companies that completed initial public offerings (IPOs) from 1997 to 2016. We asked whether these emerging companies continue to develop innovative, biologic products and produce the innovation promised by the early biotechnology industry.
Methods: We identified therapeutic products that reached Phase III development from 1997 to 2016, the characteristics of the products, the dates of the initiation of Phase III and product approval, proxy indicators of the innovativeness of each product, and the contribution of each biotechnology company. การศึกษานี้เป็นการศึกษาเชิงสังเกตเพื่อหาปัจจัยที่สัมพันธ์กับการเกิดพิษต่อไตจากยาเคมีบำบัดกลุ่มแพลตินัมในผู้ป่วยมะเร็งชาวไทย เก็บข้อมูลจากเวชระเบียนผู้ป่วยที่อายุตั้งแต่ 18 ปีขึ้นไป ที่ได้รับสูตรยาเคมีบำบัดกลุ่มแพลตินัม ณ โรงพยาบาลจุฬาลงกรณ์ ระหว่างวันที่ 1 มกราคม พ.ศ. 2561 ถึง 31 ธันวาคม พ.ศ. 2561 โดยการประเมินพิษต่อไตประเมินจากระดับซีรัมครีเอทินีนที่เพิ่มขึ้นจากค่าพื้นฐานอย่างน้อยร้อยละ 25 ในการศึกษามีผู้ป่วย 240 คน อายุเฉลี่ย 55.6 ± 12.6 ปี เป็นเพศหญิงร้อยละ 55 ส่วนใหญ่เป็นมะเร็งระบบทางเดินอาหาร ร้อยละ 36.3 เป็นมะเร็งระยะแพร่กระจายร้อยละ 60 โดยส่วนใหญ่จะได้รับยาเคมีบำบัดกลุ่มแพลตินัมร่วมกับยาเคมีบำบัดชนิดอื่นในการรักษาร้อยละ 70.4 และได้รับรังสีรักษาร่วมด้วยร้อยละ 32.9 พบอุบัติการณ์การเกิดพิษต่อไต 63 คน (ร้อยละ 26.3) ผลการวิเคราะห์ถดถอยพหุคูณพบว่ามีปัจจัยที่สัมพันธ์กับการเกิดพิษต่อไตจากยาเคมีบำบัดกลุ่มแพลตินัมทั้งหมด 5 ปัจจัย โดยเป็นปัจจัยเสี่ยง 4 ปัจจัย ได้แก่ มีคะแนนสมรรถภาพร่างกาย ECOG PS 2 (ORadj = 11.447, 95%CI 1.84-71.40, p = 0.009) น้ำหนักตัวตั้งแต่ 60 กิโลกรัมขึ้นไป (ORadj = 2.200, 95%CI 1.05-4.63, p = 0.038) มีโรคประจำตัวตั้งแต่ 2 โรคขึ้นไป (ORadj = 2.884, 95%CI 1.30-6.38, p = 0.009) และโรคมะเร็งในระยะลุกลาม (ORadj = 4.595, 95%CI 1.30-16.28, p = 0.018) และปัจจัยป้องกัน 1 ปัจจัย คือ การได้รับคาร์โบพลาตินหรือออกซาลิพลาติน (ORadj = …

Ninety strains of lactic acid bacteria (LAB) isolated from fermented foods and bark of Tamarindus indica were screening cholesterol-lowering effects, selected and identified based on the phenotypic, chemotaxonomic characteristics and 16S rRNA gene sequence analysis. Flavonoids are a group of polyphenolic compounds, with established antioxidant properties, that are present ubiquitously in plants, fruits, and vegetables. Their universal presence in nature is evident by the fact that more than 4000 types of flavonoids have been identified till date. Additionally, they have also become part of the daily diet through consumption of fruits such as berries, grapes, pomegranate, and cherries, and vegetables such as onions, kale, and eggplant. Purpose
Pharmaceutical manufacturers execute quality control operations and Good Manufacturing Practices (GMP) to provide safe drugs. The Federal Drug and Food Administration (FDA) is tasked with ensuring manufacturers are performing such procedures. Faced with limited resources the FDA has developed novel tools to aid pharmaceutical manufacturing oversight. This paper briefly reviews these tools.
Methods
Current inspection approaches employed by the FDA are identified by searching the FDA’s guidances, the Code of Federal Regulations, public reports and other online resources.

Gram-negative unicellular myxobacteria, along with their multicellular lifestyle andbiologically active specialized metabolites, are known for the predatory interactions with Gram-negative/ Gram-positive bacteria and fungi. Although myxobacterial predation range have been exploited extensively, little is known about the prey associated molecules contributing to myxobacterial predator-prey dynamics. By employing transcriptomics and untargeted metabolomics approaches, we demonstrate two structurally distinct classes of signaling molecules from Gram-negative bacterial prey elicit significant omics responses from myxobacteria, Myxococcus xanthus and Cystobacter ferrugineus. In the rhizosphere, plants curate and maintain distinct rhizobiome, some of which serve as secondary defense mechanisms against pathogenic microorganisms. Additionally, in response to biotic stress, plants generate phytohormones which regulate signaling pathways to activate systemic resistance. These phytohormones contribute to the chemical space within the rhizosphere in addition to other low-molecular-weight metabolites used for cell-to-cell communication within species and across kingdoms. However, little is known about the influences of stress-related phytohormones on beneficial bacteria even though biocontrol strategies abundantly explore plant-beneficial microorganisms to manage pathogens in agricultural systems. Chronic hyperglycemia is one of the main characteristics of type 2 diabetes mellitus (T2DM). With the increased prevalence of T2DM, the risk of cardiovascular complications, including heart attack or stroke, is increased. Complications linked to the vasculature are increased vascular calcification (VC) or mineral deposition within the medial layer of conducting arteries. Under calcification conditions, vascular smooth muscle cells (VSMCs) undergo an osteogenic switch to an osteoblast-like cell and begin producing bone proteins. Chronic hyperglycemia can also result in increased extracellular matrix (ECM) synthesis, accumulation, and crosslinking. The antimalarial drug primaquine (PQ), which has minimal activity, has to be metabolized through the CYP2D6 enzyme for its most effective antimalarial activity. The stable form 5,6-orthoquinone-primaquine (5,6-PQ) was used as a representative marker for PQ active metabolite quantification. However, the relationship between CYP2D6 polymorphisms and the pharmacokinetic (PK) parameters of 5,6-PQ has not been reported. We assessed the relationship between CYP2D6 polymorphisms and PKs of both PQ and 5,6-PQ in the Thai army population on the Thailand-Cambodia border area. We found that 56% of volunteers were CYP2D6 normal metabolizers (CYP2D6 NMs) and 44% were CYP2D6 intermediate metabolizers (CYP2D6 IMs). …
