Over a decade, proteasome inhibitors (PIs), bortezomib, carfilzomib (Cfz) and ixazomib, have contributed to a significant improvement in the overall survival for multiple myeloma (MM) patients. However, the response rate of PI was fairly low, leaving a huge gap in MM patient care. Given this, mechanistic understanding of PI resistance is crucial towards developing new therapeutic strategies for refractory/relapsed MM patients.

In this dissertation work, we found H727 human bronchial carcinoid cells are inherently resistant to Cfz, yet susceptible to other PIs and inhibitors targeting upstream components of the ubiquitin-proteasome system (UPS). It indicated H727 cells may serve as a …

Background: Infections caused by carbapenem-resistant Enterobacteriaceae (CRE) such as Escherichia coli and Klebsiella pneumoniae are among the most urgent threats of the infectious disease realm. The incidence of these infections has been increasing over the years and due to very limited treatment options, mortality is estimated at about 50%. By 2050, mortality from antimicrobial resistant infections is expected to surpass cancer at 10 million deaths annually.

Methods: We evaluated 18 contemporary antimicrobials against 122 carbapenem-resistant Enterobacteriaceae using a variety of antimicrobial susceptibility testing methods according to Clinical Laboratory Standards Institute guidelines. Attention Deficit Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder characterized by difficulty paying attention, impulsivity, and hyperactivity. Diagnosis of ADHD rose 42% from 2003–2004 to 2011–2012. In 2011, 3.5 million children were treated with drugs. Optimizing therapy can take a year, and may not be completely effective.

A clinical trial is currently being conducted of a device/drug combination using the computer game Minecraft, to determine how certain activities affect executive function, working memory, and restraint in patients diagnosed with ADHD. Overdose and addiction are two medical complications of cocaine abuse. To date, there is no FDA approved pharmacotherapy specific for cocaine abuse. Cocaine hydrolases (CocHs) have been extensively investigated for its potential in anti-cocaine therapy. Previous studies have demonstrated that CocHs efficiently hydrolyze cocaine to generate biologically inactive metabolites both in vivo and in vitro. However, it has not been studied whether there is gender difference in the therapy using CocHs. In addition, the effectiveness of CocHs is unknown for treating cocaine toxicity when alcohol is co-administered.

Nanoparticulate drug delivery systems (NDDS) such as nanocrystals, nanosuspensions, solid-lipid nanoparticles often formulated for the bioavailability enhancement of poorly soluble drug candidates are stabilized by a mixture of excipients including surfactants and polymers. Most literature studies have focused on the interaction of excipients with the NDDS surfaces while ignoring the interaction of excipients in solution and the extent to which the solution-state interactions influence the affinity and capacity of adsorption. Mechanisms by which excipients stabilize NDDS and how this information can be utilized by formulators a priori to make a rational selection of excipients is not known.

Introduction: Genetic rearrangements in Ewing sarcoma, prostate, and leukemia cells result in activation of oncogenic ETS transcription factor fusions. Mithramycin (MTM) has been identified as an inhibitor of EWS-FLI1 transcription factor, a gene fusion product responsible for oncogenesis in Ewing sarcoma. Despite preclinical success, a phase I/II clinical trial testing MTM therapy in refractory Ewing sarcoma was terminated. Liver and blood toxicities resulted in dose de-escalation and sub-therapeutic exposures. However, the promise of selectively targeting oncogenic ETS transcription factors like EWS-FLI1 prompted us to undertake the discovery of more selective, less toxic analogues of MTM. Many proteins are unstable in solution and must be formulated in the solid state. This has led to an increase in the use of lyophilized dosage forms. Lyophilization is a complicated processing method consisting of three major steps: freezing, primary drying, and secondary drying. Induced by cardiomyocyte death, MI initiates a prolonged and uncontrolled inflammatory response which impairs the healing process. Immune cells, such as macrophages, play a central role in organizing the early post-MI inflammatory response and the subsequent repair phase. Introduction: Low blood-brain barrier (BBB) penetration and hematopoietic side effects limit the therapeutic development of erythropoietin (EPO) for Alzheimer's disease (AD). A fusion protein of EPO and a chimeric monoclonal antibody targeting the mouse transferrin receptor (cTfRMAb) has been engineered. The latter drives EPO into the brain via receptor-mediated transcytosis across the BBB and increases its peripheral clearance to reduce hematopoietic side effects of EPO. Objectives: The aim of this study is to unravel the variabilities posed by alteration of the extraction technique employed on the contents and composition of essential oils derived from the same plant species Methods: Volatile oils of four different Ocimum species (Ocimum basilicum L., O. africanum Lour., O. americanum L. and O. minimum L. family Lamiaceae) were individually extracted from their fresh aerial parts using green microwave assisted extraction (MAE) method and conventional hydrodistillation (HD) and steam distillation (SD) methods. Proton- and sodium-dependent monocarboxylate transporters (MCTs (SLC16A) and SMCTs (SLC5A)) transport monocarboxylates such as ketone bodies, lactate and pyruvate, as well as drugs such as gamma-hydroxybutyric acid. CD147 acts as an ancillary protein for MCT1 and MCT4, and is involved in membrane trafficking. Previously, it has been shown that MCT expression changes under different sex hormone conditions in skeletal muscle and Sertoli cells. However, it is unknown if MCTs, SMCTs or CD147 demonstrate sex differences in tissues where they play an important role in drug disposition. Photoelectron Photoion Coincidence (PEPICO) spectroscopy is a robust tool for elucidating complex unimolecular dissociation mechanisms and for determining thermochemical and kinetic data of gas-phase ion dissociations with high accuracy. In this work, the dissociative photoionization of two sets of isomeric systems were analyzed with PEPICO: 1) C7H7+ ions of toluene (Tol) and 1,3,5-cycloheptatriene (CHT), and 2) two butyl alcohol isomers, 1-butanol and isobutanol. Threshold dissociative photoionization data on these four molecules of interest were collected on the imaging PEPICO apparatus at the VUV beamline of the Swiss Light Source. Vital autonomic and cardiovascular functions are susceptible to dysfunction after spinal cord injury (SCI), with cardiovascular dysregulation contributing to morbidity and mortality in the SCI population. Autonomic dysreflexia (AD) is a condition that develops after injury to the sixth thoracic spinal segment or higher and is characterized by potentially dangerous and volatile surges in arterial pressure often accompanied with irregular heart rate, headache, sweating, flushing of the skin, and nasal congestion. Introduction: Outpatient parenteral antimicrobial therapy (OPAT) has gained popularity since its first description in 1974. At Community Health Network (CHN), the OPAT clinic was developed three years ago by an ambulatory care clinical pharmacist and includes pharmacist driven monitoring for all patients receiving OPAT. The pharmacist ensures labs are drawn weekly to assess for renal or hepatic function changes, medication levels are drawn and adjusted as needed, and repeat labs and levels are ordered to continue assessing therapy. Mixture Of Medium Chain Fatty Acids For Protein Precipitation In Purification Process Of Equine Anti-Rabies Immunoglobulin As F(Ab′)2, Sirisak Sathorn

Besides the widely used caprylic acid (C8) as protein precipitating agent, capric acid (C10) and lauric acid (C12) were investigated as alternative medium chain fatty acids (MCFAs) for their capabilities on protein purification. The optimization process conducted with normal bovine plasma as a model antibody demonstrated the essential effect of temperature during filtration to significantly reduce the content of albumin, a contaminant marker, compared with filtration at room temperature (p<0.05). From SDS-PAGE analysis, the optimal concentrations of 3% C8 and 5% C10 increased IgG purity from 30.2% to 94.6% and 80.4%, respectively. Interestingly, precipitation by C10 provided the lowest turbidity of filtrates. Purification by C12 showed unsatisfied results that albumin content was minutely decreased when increasing C12 concentration. C8 and C10 were then variously combined into 2 levels of 3% and 5% binary mixture for plasma purification. The synergist effect of C8 and C10 mixtures was clearly demonstrated in albumin elimination that the purity of fractionates were increased to 95%. Moreover, binary mixtures with higher concentration of C10 improved the filtration velocity in plasma model. Selected C8:C10 mixtures were then applied for both ERIG-IgG and ERIG-F(ab′)2 purification. SDS-PAGE analysis confirmed the absence of albumin protein in all purified samples. For ERIG-IgG purification, C8:C10 mixture at ratio 1.5:3.5 demonstrated the best in homogeneity with high recovery rate of bioactive antibody, while the mixtures at 2.25:0.75 showed the best purification capabilities to obtain highly purified ERIG-F(ab′)2. Furthermore, the purification by C8:C10 mixtures displayed superior process characteristic than the commercially non-chromatographic technique for ERIGF(ab′)2 purification in terms of antibody recovery, purity and lower in aggregate formation. The novelty on beneficial use of MCFA mixture as protein precipitating agent for antibody purification has been demonstrated.

The objective of this study was to determine student motivations and perceptions of the benefits of participation in Dance Marathon. This was a quantitative and qualitative cross-sectional study administered using an online survey software. Undergraduate students from three different Indiana programs were invited to participate. Pharmacists are capable and interested in providing a targeted weight management intervention addressing the high rates, and associated risk, of obesity. A series of in-depth phone interviews with Mississippi community pharmacists were used to gain their perspective on a systematic approach for delivering a weight and obesity management service to patients. Using qualitative content analysis, three themes were identified and analyzed. These themes included: pharmacist’s perceptions of patient’s needs, program structural components, and pharmacist’s implementational needs. Lung cancer leads in mortality among all types of cancer in the US and Non-small cell lung cancer (NSCLC) is the major type of lung cancer. Immuno-compromised mice bearing xenografts of human lung cancer cells represent the most common animal models for studying lung cancer biology and for evaluating potential anticancer agents. Medication reconciliation, also known as “med recs”, are an important part of a patient’s care during their hospitalization. A med rec is when hospital personnel generate a list of the medications the patient takes at home. Med recs are vital in the hospital admission process because home medications need to be restarted at appropriate times to fully care for the patient. Vancomycin is an antibiotic used regularly in hospitals across the world. The most concerning adverse effect of vancomycin is its documented effect on kidney function. This study examines the impact of two dosing strategies of vancomycin on patients’ kidney function at the University of Kentucky Albert B. Chandler Medical Center. It specifically focuses on patients on the orthopedic service because these patients traditionally receive higher doses of vancomycin for longer periods of time due to the severity of their infections, which puts them at an increased risk of vancomycin-induced nephrotoxicity compared to other patients.

Abstract: Mass spectrometry is a fundamental tool for modern proteomics. The increasing availability of mass spectrometry data paired with the increasing sensitivity and fidelity of the instruments necessitates new and more potent analytical methods. To that end, we have created and present XFlow, a feature detection algorithm for extracting ion chromatograms from MS1 LC-MS data. XFlow is a parameter-free procedurally agnostic feature detection algorithm that utilizes the latent properties of ion chromatograms to resolve them from the surrounding noise present in MS1 data.
