This thesis aimed to elucidate the demographic characteristics associated with elevated or reduced rates of pneumococcal conjugate 13 (PCV13) vaccination.
A retrospective cohort study was performed using the Truven Health MarketScan® Database. Three cohorts were created corresponding to populations for which the CDC recommends PCV13 vaccination. Cohort 1: children < 36 months of age. Cohort 2: adults 19-64 years of age with high infection risk. Cohort 3: adults > 65 years of age. Odds of having a PCV13 claim were calculated for each cohort.
For Cohort 1, 78% out of a total of 353,214 subjects had a sufficient number of PCV13 doses to meet CDC recommendations. For Cohort 2, 3.7% out of a total of 673,157 subjects had a PCV13 …

Aspirin and other nonsteroidal anti-inflammatory drugs (NSAIDs) are currently widely used as fever and pain relief in patients with arthritis and other inflammatory symptoms. NSAIDs effect by inhibiting cyclooxygenase-1 (COX-1) and/or cyclooxygenase-2 (COX-2). COX isozymes (COXs) are key enzymes in the biosynthesis of prostaglandin H2 (PGH2) from arachidonic acid (AA). It is now clear that prostaglandin E2 (PGE2), one of the downstream products of PGH2, is the main mediator in both chronic and acute inflammation. Microsomal prostaglandin E synthase (mPGES-1) is the terminal enzyme of COX-2 in the PGE2 biosynthesis pathway. …

Current smoking cessation guidelines recommend nicotine replacement therapy (NRT) to assist pregnant smokers to quit, but this is without strong evidence for effectiveness and safety. Nicotine, the main addictive component of tobacco, is known to exert physiological effects by binding to its receptor, the nicotinic acetylcholine receptor (nAChR). Recent studies have identified the presence of nAChRs in non-neuronal cells, and in macrophages, functional alteration upon stimulation with nicotine has been documented.

More than 80% of lung cancer patients die from drug-resistant, metastatic disease. Our focus is to identify new drug targets and alternative therapeutic strategies to improve outcomes for this majority of lung cancer patients. We aimed to satisfy the need for new treatment approaches by leveraging the information gained from the development of two multigene biomarker predictors of Epidermal Growth Factor Receptor Inhibitors (EGFRI) response in Non-Small Cell Lung Cancer (NSCLC). Aging is the main risk factor for cognitive decline. The hippocampus, a brain region critical for learning and memory formation, is especially vulnerable to normal and pathological age-related cognitive decline. Dysregulation of both insulin and intracellular Ca2+ signaling appear to coexist and their compromised actions may synergistically contribute to neuronal dysfunction with aging. This dissertation focused on the interaction between insulin, Ca2+ dysregulation, and cognition in hippocampal neurons by examining the contributions of insulin to Ca2+ signaling events that influence memory formation. Dehydroepiandrosterone supplementation is used to treat a variety of conditions. Rapid-dissolving tablets are a relatively novel choice for compounded dehydroepiandrosterone dosage forms. While rapid-dissolving tablets offer ease of administration, there are uncertainties about the physical and chemical stability of the drug and dosage form during preparation and over long-term storage. This study was designed to evaluate the stability of dehydroepiandrosterone rapid-dissolving tablets just after preparation and over six months of storage. The Professional Compounding Centers of America rapid-dissolving tablet mold and base formula were used to prepare 10-mg strength dehydroepiandrosterone rapid-dissolving tablets. Background: Pharmacy education in India has expanded to include the Doctorate of Pharmacy degree (Pharm.D.). With clinical practice in early development, job opportunities in India are limited. Graduates often consider pharmacy opportunities abroad.
Aims: This study compares Indian students’ career aspirations related to the Pharm.D. degree before and after beginning their programmes.
Methods: A 5-point Likert scale paper survey with open ended questions was distributed to all Pharm.D. students (Year 1- 6) at a medical university in India.

Introduction: Cannabis is listed as a Schedule I substance under the Controlled Substances Act of 1970, meaning the US federal government defines it as an illegal drug that has high potential for abuse and no established medical use; however, half of the states in the nation have enacted “medical marijuana” (MM) laws. Clinicians must be aware of the evidence for and against the use of MM in their patients who may consider using this substance.

Background: Simulation-based counselling using standardised patients (SPs) provide pharmacy students an authentic approach to training; limited data exists regarding student performance using immediate feedback approaches.
Aims: To compare grades of students receiving immediate feedback verses (vs.) delayed feedback.
Methods: A pre-trial assessment of student perceptions and an unblinded randomised trial comparing immediate and delayed feedback. Third year pharmacy students (n=153) counselled SPs in four clinical “experiences”; student grades were the primary outcome. Student t-test and repeated measures were used to compare grades between groups and grades over time.

"Both the clinical benefits and the adverse effects of nonsteroidal anti-inflammatory drugs (NSAIDs) are mediated by the inhibition of cyclooxygenase (COX) 1 and 2.[1] COX-1 is expressed constitutively in tissues, whereas COX-2 is produced under periods of stress and inflammation. The physiologic function of COX-1 and COX-2 is to convert arachidonic acid to prostaglandins, thromboxane A2 (TXA2), and prostacyclins. These products influence multiple systems such as cardiac, immune, gastrointestinal (GI), renal, vascular, and pulmonary systems.[12] COX enzymes are present in various locations throughout the body including the blood vessels where they produce vasodilatory prostacyclins and TXA2, which causes platelet aggregation.[3] …

The implementation of Molecularly Imprinted Polymers (MIPs) into sensing systems has been demonstrated abundantly over the past few decades. In this article, a novel application for an MIP-based thermal sensing platform is introduced by using the sensor to characterize the drug release kinetics of a nanoporous silver-organic framework. This Ag nanoporous matrix was loaded with acetylsalicylic acid (aspirin) which was used as a model drug compound in this study. Objective. To determine if Grit-S scores correlate with academic success in a doctor of pharmacy (PharmD) program, as well as the pursuit and attainment of pharmacy postgraduate (residency or fellowship) training.
Methods. A 28-item survey was administered to third- and fourth-year (P3 and P4) pharmacy students. Variables queried included Grit-S score, demographics, pharmacy experience prior to the PharmD program, and factors that may affect academic performance during didactic coursework. Didactic coursework GPA was used as a surrogate for academic success. Information about pursuit and attainment of a postgraduate training position was also documented and used in the analyses. …

AIM: Over 7 million traumatic brain injuries (TBI) are reported each year in the United States. However, treatments and neuroprotection following TBI are limited because secondary injury cascades are poorly understood. Lipopolysaccharide (LPS) administration before controlled cortical impact can contribute to neuroprotection. However, the underlying mechanisms and whether LPS preconditioning confers neuroprotection against closed-head injuries remains unclear.
METHODS: The authors hypothesized that preconditioning with a low dose of LPS (0.2 mg/kg) would regulate glial reactivity and protect against diffuse axonal injury induced by weight drop. LPS was administered 7 days prior to TBI. LPS administration reduced locomotion, which recovered completely …

Functionalized piperidines, azepanes, azamacrocycles, morpholines, and thiomorpholines are common structural motifs found in a wide range of pharmaceuticals such as carmegliptine, levofloxacin, thioridazine, claviciptic acid, and azithomycin. As a result, there is a strong desire to construct highly functionalized nitrogen-bearing ring scaffolds in order to construct a wide range of drug possibilities. The aim of this study was to develop cream containing mangosteen pericarp extract (MPE) encapsulated in solid lipid nanoparticles (SLNs) in order to achieve enhanced photoprotection and to be an alternative for synthetic sunscreens in the market. The MPE was prepared by maceration method and evaluated for SPF value by UV-Vis spectrophotometer. SLNs were prepared by ultrasonication method. วัตถุประสงค์ : เพื่อศึกษาอิทธิพลร่วมกันของภาวะพหุสัณฐาน POR*28 และ CYP3A5 ต่อสัดส่วนของระดับความเข้มข้นต่ำสุดต่อขนาดยา tacrolimus (C0/D) ในผู้ปลูกถ่ายไตชาวไทย วิธีการดำเนินการวิจัย : รูปแบบการศึกษาวิเคราะห์แบบย้อนหลัง, ศึกษาในผู้ปลูกถ่ายไตจำนวน 230 คน โดยผู้ปลูกถ่ายไตทุกคนจะได้รับยากดภูมิคุ้มกัน 3 รายการ ประกอบด้วย tacrolimus, mycophenolate mofetil หรือ sodium, และ steroids ข้อมูลพื้นฐานและข้อมูลทางห้องปฏิบัติการเก็บรวบรวม ณ วันที่ 3, 7, 90, และ 180 หลังการปลูกถ่ายไต ใช้สถิติ two-way ANCOVA ในการวิเคราะห์อิทธิพลร่วมกันของภาวะพหุสัณฐาน POR*28 และ CYP3A5 ต่อ C0/D ของยา tacrolimus โดยควบคุมปัจจัยอายุ, ฮีโมโกลบิน, อัลบูมิน, และขนาดยา steroids ผลการศึกษา : ไม่พบอิทธิพลร่วมกันของภาวะพหุสัณฐาน POR*28 และ CYP3A5 ต่อ Log C0/D ของยาtacrolimus (ณ วันที่ 3, 7, 90, และ 180 หลังการปลูกถ่ายไต, P>0.05) แต่อย่างไรก็ตามพบว่า ภาวะพหุสัณฐาน CYP3A5 มีอิทธิพลต่อ Log C0/D ของยา tacrolimus ทุกช่วงเวลาที่ทำการศึกษา (P<0.001) โดยผู้ที่มีภาวะพหุสัณฐาน CYP3A5 expressers จะมีค่าเฉลี่ยของ Log C0/D ของยา tacrolimus ต่ำกว่า CYP3A5 non-expressers สำหรับภาวะพหุสัณฐาน POR*28 พบว่ามีอิทธิพลต่อ Log C0/D ของยา tacrolimus เฉพาะวันที่ 7 หลังการปลูกถ่ายไต (P=0.047) และพบว่าผู้ที่มีภาวะพหุสัณฐาน POR*28 T carriers จะมีค่าเฉลี่ย Log C0/D ของยา tacrolimus ต่ำกว่า POR*28 CC อีกทั้งยังพบว่าอายุ, ฮีโมโกลบิน, และขนาดยา steroids มีอิทธิพลต่อ Log C0/D ของยา tacrolimus สรุปผลการศึกษา : ณ วันที่ 3, 7, 90, และ 180 หลังการปลูกถ่ายไต ใม่พบอิทธิพลร่วมกันระหว่างภาวะพหุสัณฐาน POR*28 และ CYP3A5 ต่อสัดส่วนของ Log C0/D ของยา tacrolimus และพบว่าภาวะพหุสัณฐาน CYP3A5 เป็นปัจจัยที่มีอิทธิพลต่อเภสัชจลนศาสตร์ของยา tacrolimus มากที่สุด

The aim of this study was to prepare optimized nanoparticles and hot melt cast films of quercetin and to investigate the release and permeability profile across corneal membranes. The results demonstrate that all formulations can be successfully employed for delivery of quercetin into the eye through the topical route of administration, with films showing significantly better transcorneal permeability.

Investigators measured students' needs for healthcare services during their transition from pediatric to adult healthcare at The University of Mississippi. Also, investigators examined chronically ill students' adherence to their conditions needs and measured a baseline of college freshman's knowledge about healthcare. Data was collected using a self-administered cross-sectional survey online through Qualtrics. Introduction Cardiovascular diseases are the leading cause of morbidity and mortality among individuals with diabetes. Despite the beneficial effects of antidiabetic drugs (ADDs) in terms of lowering haemoglobin A1c, several ADDs have been shown to increase the risk of cardiovascular events. Given the high prevalence of cardiovascular disease among individuals with diabetes, it is important to weigh the benefits of ADDs against their cardiovascular safety. Polyphenols content and antioxidant activity in Artemisia sieversiana Willd. were investigated in relation to the altitude of the plant growth in the Western Pamir region. Antioxidant activity generally correlated with the polyphenols content. However, no significant association was found between an altitude of the sample collection and polyphenols content or antioxidant activity.

Oral vaccines present an attractive alternative to injectable vaccines for enteric diseases due to ease of delivery and the induction of intestinal immunity at the site of infection. However, susceptibility to gastrointestinal proteolysis, limited transepithelial uptake and a lack of clinically acceptable adjuvants present significant challenges.
