Production Of Recombinant Protein Ltb-Ctla4 By Transient Plant-Based Expression Using Geminiviral Vector, Sutita Yiemchavee
Chulalongkorn University Theses and Dissertations (Chula ETD)
Cancer is one of the leading global causes of death. In 2018, WHO estimated that 9.6 million patients died from cancer, with 18.1 million new cases emerged. In recent years, immunotherapy is considered as a treatment improving cancer-related immune function by manipulating the immune checkpoints in the immune system. Cytotoxic-T-lymphocyte antigen 4 (CTLA-4) is a immune checkpoint transmembrane protein. The expression of CTLA-4 on the surface of activated T-cell could downregulate the production and proliferation of cytotoxic T-lymphocyte cells (CTLs). CTLs are the host-defended immune cells that could retard the growth of cancer cells by recognizing the tumor-associated antigens expressed …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Cepharanthine (CEP), a natural compound isolated from Stephania cepharantha Hayata, has been shown to possess anti-cancer activity against several cancers including colorectal cancer (CRC) cells. Previously, it was found that cytotoxicity of CEP was associated with ROS generation and p21Waf1/Cip1 expression. However, information regarding to the molecular mechanisms underlying the apoptosis-inducing effects of CEP in CRC cells is still limited. Therefore, the present study aimed to evaluate the roles of ROS and p21Waf1/Cip1 in the apoptosis-inducing effect of CEP in two human colorectal cancer cells lines, p53 wild-type HCT116 cells and p53 mutant HT-29 cells. The results demonstrated that CEP …

Chulalongkorn University Theses and Dissertations (Chula ETD)
The shifting of autophagy, a catabolic process for cell survival, toward death could be a potential mode of anti-cancer drug action. In addition, autophagic cell death (ACD) is an alternative death mechanism in resistant malignant cancer cells. This study, we demonstrated how PE5, a stilbene compound, exhibits potent ACD-promoting activity and apoptosis in lung cancer cells that may offer an opportunity for novel cancer treatment. It was found that PE5 exerted a higher toxic effect toward cancer cells compared with its effect on normal cells. Interestingly, the cell death caused by PE5 was found to be concomitant with dramatic autophagy …

Lung cancer is one of leading cause of deaths worldwide due to cancer metastasis. The metastatic process that initiated by dissociation of cancer cells from the primary tumor and colonization of new tumor at distant organ. Cell migration and invasion play as the key early steps and required actin reorganization and extracellular matrix (ECM) degradation to achieve cell metastasis. The inhibition of these behaviors is an intriguing approach to minimize cancer metastasis. Erianthridin, a phenolic compound isolated from the Thai orchid Dendrobium formosum exhibits several pharmacological activities; however, the effect of erianthridin on lung cancer cell metastasis has not been …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Bacteria, exposed to stress from the environment or competition, release toxins called bacteriocins. Bacteriocins are classified by their mode of cytotoxicity such as pore former, endonuclease, etc. Colicins are the member of bacteriocins produced by Escherichia coli (E.coli). Among several types of colicins cytotoxicity, Pore-forming colicins has been reported in both bacteria and various type of cancer cells. Colicin N is also a pore-forming colicin and the anticancer activity of colicin N has not been extensively reported. Here we examined the cytotoxic effects of colicin N and its domain on cancer cells. The expression and purification of recombinant full-length and …
