A silica gel-petroleum (SGP) medium was developed which is reliable, approximately the same cost as that of the corresponding agar-based petroleum medium, and can be made sufficiently firm to use in streaking or the spread-plate technique. Advantages of the medium include: the incorporation of all of the major ions of seawater in accordance with Dittmar's Law (the salinity of which can easily be varied from 0 to 35 parts per thousand); a wide workable pH range (5.5 to 11.6); negligable pH drift(~ 0.2 pH units after 166 days); the elimination of all but negligable amounts of syneresis, the ability to …

Methodologies for the improved recovery of selected pathogenic bacteria from estuarine sediments were investigated. Laboratory and field studies were conducted to document the sensitivity of the techniques for isolating Salmonella species, Pseudomonas aeruginosa, and Vibrio parahaemolyticus. Vibrio parahaemolyticus-like organisms were enriched in a seawater yeast extract broth and isolated from Thiosulfate Citrate Bile Salts Sucrose plated media. Enrichment procedures with and without elevated temperatures and subsequent transfers to selective and differential plated media have been examined for the isolation of Salmonella species and P. aeruginosa. This investigation evaluated the gingival condition of areas with minimal (less than or equal to 1.0 millimeter) and appreciable (greater than or equal to 2.0 millimeters) widths of keratinized gingiva.

Sixteen dental, dental hygiene, and dental assisting students and dental faculty who had contralateral or unilateral areas of minimal and appreciable widths of keratinized gingiva on the mid-buccal surface of mandibular bicuspids were examined. Gingival exudate, plaque, gingival inflammation based on color change and/or swelling and bleeding on probing were evaluated. The results showed that all areas examined were plaque free. The bacterial flora of acute biliary tract disease has not been completely elucidated. The organisms identified most often were coliforms. Anaerobic culture technics were not precise enough to provide proper identification. In a 12-month study of 250 patients operated upon for inflammatory gallbladder disease, culture specimens were collected in a most strict and rigorous fashion. It was hoped that a more exact understanding of the relationship between bacteria and inflammatory biliary tract disease might be appreciated. Important technics were developed for gathering specimens, delivery to the laboratory and culturing. More meaningful results are to be expected In the future.

This report describes investigations into the physiology of microorganisms that are commonly involved in the oxidation of inorganic sulfur compounds. The metabolic activities of these bacteria play a potent role in several fields of economic importance such as strip mining operations, water pollution, corrosion, metallurgy, petroleum technology and soil fertility processes.

The oxidation of inorganic sulfur compounds was studied in the chemolithotrophs Thiobacillus denitrificans, Thiobacillus A2, Thiobacillus neapolitanus, and a photolithotroph Rhodopseudomonas palustris. Cell suspensions from all of the thiobacilli catalyzed completely the oxidation of sulfide, thiosulfate, and sulfite to sulfate. The oxidation of thiosulfate in R. palustris …

An in vitro study using polyethylene tubes filled with Grossman's sealer, Rickert's sealer, and N2 permanent was conducted over a six month period in an effort to determine their lasting antimicrobial properties against eight different microorganisms.

From the study it was observed that N2 permanent possesses a long-lasting antimicrobial effect, but this effect appears to diminish greatly with time. Further investigation is needed to determine whether or not the effect is actually permanent in nature. The N2 permanent was also shown to have broader antimicrobial effectiveness against the eight microorganisms tested. N2 permanent produced zones of inhibition against five of …

The results of experimentation approximately twenty years ago with variuos bacteriophages by Ralston and Krueger16, Anderson and Felix1, Luria and Human12, and by Bertani and Weigle2 have shown that the host range of a given phage depends directly on the bacterial strain on which the phage has last propagated. This event came to be known as host-induced modification or host-controlled variation. Proteolytic activity of Streptococcus cremoris HP, P2 and AM2; Streptococcus lactis-C2, ML8 and a commercial mixed culture was determined in skimmilk and whey by measuring the increase in Absorbency at 274.5 nm of a pH 4.6 soluble filtrate, following incubation in a 2.5% casein solution. Proteolytic activity, rate of acid production and growth rate decreased in order of C2, HP, mixed culture, AM2, ML8 and P2 at a regulated pH (6.1 - 6.2) and 32 C. Cytochrome transport particles from Nitrobacter catalyzed nitrite, ascorbate as well as NADH oxidation with concomitant phosphate esterification yielding P/0 ratios of 1.0, 0.6 and 2.0 respectively. Phosphorylation coupled to nitrite oxidation was not effected by rotenone, amytal or antimycin while 50 and 70% inhibition of the NADH-linked phosphorylation was observed in the presence of HOQNO and rotenone respectively. Cell-free extracts from Nitrobacter also catalyzed an energy-dependent reduction of NAD+ by nitrite. The reduction of cytochrome c by NŌ2 was energy-dependent which involved the reversal of electrons from cytochrome a1. The subsequent energy-linked reduction of the flavoproteins …

The Role Of D-Gluconic Acid In The Regulation Of The Synthesis Of The Enzymes Of The Entner-Doudoroff Pathway In Pseudomonas Fluorescens, Steven C. Quay

Induction of Entner-Duodoroff pathway enzymes in Pseudomonas fluorescens was investigated to study the role of gluconate as a possible inducer. Glucose oxidase-deficient mutants were isolated and characterized. One of these mutants, gox-7, was deficient in particulate glucose oxidase; another mutant, gox-17, was deficient in particulate glucose and gluconate oxidase activities. Gluconate, but not glucose, induced synthesis of gluconokinase and 6-phosphogluconate dehydratase in both mutants. High constitute levels of 2-keto-3-deoxy-6-phosphogluconate aldolase were found when both mutants were grown on glucose. Growth of parent and both mutant strains on glycerol also resulted in high levels of Entner-Doudoroff pathway enzymes. It was concluded …

In order to determine the duration of the effect of glucose, fructose, sucrose, honey, starch, and orange juice on neutrophilic phagocytosis, 10 subjects underwent a complete series of carbohydrate tolerance tests. Blood samples were drawn at specific time periods following ingestion of 100 grams of each carbohydrate. Blood was then analyzed for glucose and phagocytic index (the mean number of bacteria engulfed per leukocyte); a red blood cell and white blood cell count and differential white cell count were done also on about 1/2 of the samples. Two group A phages, P9c/2 and P4/2, were studied. Their density, sedimentation velocity, and biochemical properties were compared. Their densities were determined by density gradient centrifugation in 38.3% cesium chloride. Sedimentation velocity was used to determine their molecular weights. To accomplish this, the DNA of P9c/2 and P4/2 was labeled with tritium and T2 DNA, labeled with 32P, was used as a standard. The DNA of P9c/2 and P4/2 was separately mixed with T2 DNA and centrifuged in a 0-15% sucrose gradient. The molecular weights were calculated using the formula D2/D1= (M2 …

The chemical composition of algae grown in batch culture depends mainly on environmental conditions, nutrient availability, presence of predators, cell age, and species. The effects of nutrient availability and cell age on the composition of three unialgal cultures (algae + bacteria) and one hetergeneous culture (algae + bacteria + microscopic animals) were evaluated. The cultures were grown in batch culture under both nutrient-abundant and nutrient deficient conditions and the changes in compositions were observed. The purpose of this investigation was to determine the sites of overwintering of Erwinia amylovora (Burrill) Winslow et al. outside of living tissue in Utah. Bacterial isolates were taken from the surfaces of blighted twigs and mummified fruits remaining on the tree; also from internal dead tissue of blighted twigs and mummies, and from soil beneath blighted trees. Each isolate was tested for agglutination with antiserum specific for Erwina amylovora, susceptibility to Erwinia amylovora bacteriophage, production of symptoms in pear seedlings and development of bacterial ooze on green pears.

Both virulent and avirulent isolates were taken from all sites tested. …

The purpose of this study was to find out the immediate and residual antibacterial effects of the two preoperative skin preparation procedures most commonly used in the operating room of this selected hospital.

This study was conducted on one hundred selected surgical patients from the general medical and surgical units of this hospital.
