Near-Field Electrospinning And Characterization Of Biodegradable Small Diameter Vascular Grafts, William E. King Iii

The ideal “off the shelf” tissue engineering, small-diameter (< 6 mm inner diameter (ID)) vascular graft hinges on designing a template that facilitates transmural ingrowth of capillaries to regenerate an endothelized neointimal surface. Previous traditionally electrospun (TES) approaches to create bioresorbable vascular grafts lack the pore sizes required to facilitate transmural capillary ingrowth required for successful in situ neovascular regeneration. Therefore, the ability to create scaffolds with program-specific architectures independent of fiber diameter via the relatively recent sub-technique of near-field electrospinning (NFES) represents a promising solution to create tissue engineering vascular grafts. These programmed large pore sizes are anticipated to promote in situ regeneration and improve the outcomes as well as the quality of life of patients with arterial disease.
In this dissertation, we manufactured via NFES as well as characterized biodegradable polydioxanone (PDO) small-diameter vascular grafts. Chapter 1 introduces the need for off-the-shelf, small-diameter vascular grafts to facilitate in situ regeneration, the process and pore size limitations of TES vascular grafts, and the promising use of NFES to develop precisely tailored PDO vascular grafts. Chapter 2 describes the process of NFES and details the current progress in NFES of biomedical polymers as well as the major limitations that exist in the field. The Japanese health practice of Reiki attempts to maximize the latent ability of the human system to heal itself. The Reiki system, established over a century ago, combines multiple Asian health traditions, experimenting with practices that maximize the natural processes of the body to perform its own repairs. Reiki encourages healthy behaviors that balance the mind and body, return the human system to a lowered stress level, and allow for an optimal recovery state for the patient. It is widely known that patients suffer diminished long-term outcomes in the years following secondary infections, infectious diseases that occur during or after treatment of a primary infection such as influenza or COVID-19. One common secondary infection is pneumonia, which has become the most fatal hospital-acquired infection. The bacterium responsible for pneumonia is known as Pseudomonas aeruginosa. Ionic liquids (ILs) are emerging as an eco-friendlier alternative to traditional organic compounds. The hope with ILs in this project is to incorporate them with resins used in 3D printing to act as plasticizers which will in turn help prolong the life of the printed object. A common issue with plasticizers like phthalate in particular is they can leach out of plastics due to their vapor pressure which leads to decreased performance and can also pose a health risk being released into its surroundings or atmosphere. Due to rapid advances in sequencing technology, it is becoming increasingly easier to assemble unknown genomes from millions of short sequencing reads of nucleotides taken from the full genomic sequence (Hernandez, 2008). In this study, we used various computational programs to align reads from unsequenced strains of the bacteria Staphylococcus epidermidis and Staphylococcus hominis into a hitherto undefined, single contiguous genomic sequence. Endotracheal intubation is an essential component within the skill set of anesthesia providers. Rapid arterial desaturation remains a primary risk factor during laryngoscopy and intubation. The risk for hypoxemia exists because during endotracheal intubation, patients are apneic or hypo-ventilating (Wong et al., 2017). Apneic oxygenation (AO) has been advocated to attenuate hypoxemia during endotracheal intubation (Pavlov et al., 2017).

Anatomic changes occurring mid-treatment for patients undergoing radiation therapy for locally advanced lung cancer can degrade the quality of the intended treatment plan. These changes include tumor regression, geometric misalignment, and lung density changes (atelectasis and pleural effusion), and are visible on the daily 3D cone beam CT (CBCT). To maintain the intended treatment quality, adaptive radiotherapy (ART) can be employed to modify the treatment plan to account for these anatomic changes. Health-related Quality of Life (QoL) is generally poorer in individuals with opioid use disorder (OUD) in comparison with those suffering from other chronic diseases. Little is known about the difference each buprenorphine/naloxone (BUP/NX) or methadone maintenance therapy (MMT) may make in the QoL for the HIV-infected people with OUD who are new to HIV care. Direct-to-consumer (DTC) testing provides consumers access to genetic testing without involvement or advice from a genetics specialist. While the expansion of DTC testing has increased consumer access to genetic interpretations, these reports can often provide a false sense of security and rarely cover enough genetic information to make significant interpretations about disease risk. Patient-initiated testing involves consumers seeking out and ordering more comprehensive genetic testing on a cash pay basis. In the development of a bioink for cartilage 3D printing, Hyaluronic acid (HA), Alginate (Alg) were modified into amine-hyaluronic acid (HA-NH2) and aldehyde alginate (Alg-CHO). Magnetic Nuclear Resonance (NMR) and Fourier Transform Infrared Spectroscopy (FTIR) confirmed the successful modification of HA-NH2 and Alg-CHO. The hydrogel HA-Alg was crosslinked as a result of Schiff's base reaction, between primary amines and aldehydes to form imine bonds. By varying volume ratios, the optimal hydrogel HA-Alg (5:5) exhibited its printability, biocompatibility, and functionality as a bioink for cartilage tissue. Our body’s functioning depends on the ability of cells to sense and react to their local mechanical environment; this process is known as mechanotransduction. Despite the importance of understanding how cells interact with mechanical stimuli, the specific mechanisms governing such processes have yet to be elucidated. Using microscopy to detect the early responses of living cells to mechanical loads and forces would be a critical step towards further understanding cellular mechanotransduction. Dynamic and high-frequency cyclical loads are relevant to human physiology and disease. Proton therapy is a radiotherapy modality that can offer a better physical dose distribution when compared to photon radiotherapy by taking advantage of the Bragg peak, a narrow region of rapid energy loss. Proton therapy is also known to offer an enhanced relative biological effectiveness (RBE) compared to photons. In the current clinical standard, RBE is fixed at 1.1 at all points along the proton beam, meaning protons are assumed to require 10% less dose than photons to achieve target coverage and organ at risk (OAR) sparing. Colorectal Cancer (CRC) is the third-most common malignancy for men or women, with chronic inflammation considered as a primary risk factor. Obesity is also considered a chronic inflammatory disease and is associated with increased CRC incidence. Further, obesity and CRC occur in men and women differently with the highest incidence of either disease found in men, suggesting that female sex hormones may play a protective role in inflammatory diseases. Macrophages can promote inflammation and are a driving force in obesity-associated metabolic dysfunction. Familial hypercholesterolemia (FH) is a common genetic disease and has been studied with the aim of finding a curative measure for decades. FH is caused by mutations in the low-density lipoprotein receptor (LDLR) resulting in defects in LDL-cholesterol (LDL-C)-receptor mediated endocytosis and development of premature cardiovascular disease (CVD). Here I describe the use of a corrected and non-corrected LDLR FH cell model to investigate receptor-mediated endocytosis and statin effects. Replication of clinical research is requisite for forming effective clinical decisions and guidelines. While rerunning a clinical trial may be unethical and prohibitively expensive, the adoption of EHRs and the infrastructure for distributed research networks provide access to clinical data for observational and retrospective studies. Purpose: This work investigated the delivery accuracy of high-dose lung and spine stereotactic treatments delivered with the Elekta Infinity and Versa HD platforms. The accuracy of these platforms will be used for consideration in implementing a spine stereotactic radiosurgery (SSRS) program at Mary Bird Perkins Cancer Center.
Methods: A geometric phantom was used to perform Winston-Lutz type tests that assessed the relevant degrees of freedom (gantry, collimator, and couch) of the delivery system. While effective, it can result in progressive and debilitating cognitive impairment that can diminish quality of life. Mossy cells are a major population of excitatory neurons located within the dentate hilus and highly involved in hippocampal circuitry. Purpose: Nursing students are prone to stress-related diseases, such as depression and anxiety, which are associated with chronically elevated cortisol levels due to the excessive firing of the Hypothalamic Pituitary Adrenal (HPA) axis. This study assessed three speeds of yoga and how they acutely affected salivary cortisol levels within female nursing students. Radioembolization of liver tumors using yttrium-90 (Y-90) labeled microspheres is a proven method of treatment. The primary decay mode of Y-90 is beta-minus decay. The electrons produced during decay deliver a cytotoxic radiation dose in a localized volume. The radiation dose is sufficient to kill tumor cells yet spare the normal liver. The Y-90 microspheres are delivered via a catheter that has been placed in an artery that supplies blood to the tumor. Microspheres are injected and the blood distributes the microspheres. Purpose: The delivery of post-mastectomy radiotherapy (PMRT) with bolus electron conformal therapy (BECT) for patients with left-sided breast cancer can reduce second cancer complication probability (SCCP) compared to modern rotational intensity modulated x-ray (IMXT) techniques such as volumetric modulated arc therapy (VMAT) or Tomotherapy. However, rotational IMXT yields superior levels of dose homogeneity compared to BECT.
