Mouse Mammary Tumor Virus (Mmtv) Genomic Rna (Grna) Packaging: Selective Grna Encapsidation Is Facilitated By Long-Range Interactions (Lris) That Facilitate Specific Gag Binding, Suresha G. Prabhu

Thesis/ Dissertation Defenses

Mouse mammary tumor virus (MMTV) is a betaretrovirus that induces breast cancer and sporadically, T-cell lymphomas in mice. MMTV serves as a pertinent model for elucidating the mechanism(s) of oncogenesis and the genetics associated with the development of mammary tumors. Being a rodent retrovirus, MMTV is phylogenetically distant from human and other primate retroviruses; thus, positioning it ideally as a potential vector of choice for human gene therapy. This may circumvent safety concerns such as recombination with other related endogenous human retroviruses. In addition, MMTV is distinct from most other retroviruses, but similar to lentiviruses, in that it has the …

Using A Rabbit Model To Understand The Impact Of Oral Versus Intravenous Transmission Of Epstein-Barr Virus, Meera Mohammed Al Ameri Nov 2024

Using A Rabbit Model To Understand The Impact Of Oral Versus Intravenous Transmission Of Epstein-Barr Virus, Meera Mohammed Al Ameri

Thesis/ Dissertation Defenses

The isolation of Epstein-Barr Virus (EBV) in 1964 from a case of Burkitt lymphoma prompted a surge in research on the biology of this oncogenic virus and its role in the pathogenesis of associated malignancies. Over half a century later, a number of pertinent questions on the dynamics of EBV and the cellular targets of primary infection remain poorly defined. One major obstacle in EBV research has been the lack of a suitable small animal model. EBV is a highly human tropic virus. We have recently established a novel rabbit model of EBV infection that appears to mimic natural infection …

Clonality And Epidemiological Studies Of E. Coli Strain Collected From Abu Dhabi Emirate Using Ngs And Bioinformatic Tools, Ahmed Abdelrahman Mohamed

Theses

In the current landscape, where antibiotic resistance in bacteria is steadily rising, the role of bacterial genomics has assumed increasing significance. Therefore, it is necessary for microbiologists to acquire proficiency in the utilization of these methods and tools to harness their potential in gaining valuable insights into bacterial genomes. This can facilitate the development of innovative strategies for managing and treating bacterial infections. Here we present the power of the whole-genome sequencing technology in characterization of multidrug resistant Escherichia coli isolates. Fourteen Escherichia coli isolates were selected for this study. Following the standard microbiological techniques to confirm the sample identification, …

Exploring The Dynamics Of Primary Epsteinbarr Virus Infection Using A Novel Rabbit, Narendran Reguraman Oct 2022

Exploring The Dynamics Of Primary Epsteinbarr Virus Infection Using A Novel Rabbit, Narendran Reguraman

Dissertations

Epstein-Barr virus (EBV) is a common herpesvirus asymptomatically carried by over 90% of the world population. However, in certain circumstances, the virus can lead to life-threatening diseases. Although the virus was isolated over 55 years ago, several fundamental aspects of the biology of EBV and the mechanism by which this virus induces pathology remain unknown. One major obstacle has been the lack of a suitable small animal model for EBV infection. We have recently shown that healthy rabbits are susceptible to EBV infection, and the virus establishes long-term latent infection typically seen in humans. The main objective of this dissertation …

Associated Risk Of Blastocystis Infection In Colorectal Cancer, Lena Abdulbaset Labania Oct 2022

Associated Risk Of Blastocystis Infection In Colorectal Cancer, Lena Abdulbaset Labania

Theses

Blastocystis species is an anaerobic intestinal protozoan seen in humans and a wide range of animals. Only nine Blastocystis subtypes are seen in humans. The pathogenicity of Blastocystis spp. has long been controversial. Recently, a subtype-dependent association between Blastocystis spp. and colorectal cancer (CRC) is being debated. Thus, this thesis aims to assess the possible association between Blastocystis spp. infection and CRC condition compared to cancer outside the gastrointestinal tract (COGT) and a cancer-free control (CF) group. This study is the first of its kind in the UAE. Participants were divided into two groups; cancer patients and CF participants. The …

Investigating The Role Of Natural Killer (Nk) Cells In Ozanimod-Mediated Remission In Experimental Autoimmune Encephalomyelitis (Eae) In C57bl/6 Mice, Doua Youssef Kamyan Oct 2022

Investigating The Role Of Natural Killer (Nk) Cells In Ozanimod-Mediated Remission In Experimental Autoimmune Encephalomyelitis (Eae) In C57bl/6 Mice, Doua Youssef Kamyan

Theses

Experimental autoimmune encephalomyelitis (EAE) is an important model of multiple sclerosis (MS) in rodents. The disease is characterized by axonal damages mediated by activated autoreactive T lymphocytes, microglia, and macrophages. Ozanimod (RPC1063) is an immunomodulator that has been recently approved by the FDA for the treatment of relapsing-remitting multiple sclerosis (2020) and ulcerative colitis (2021). It is a selective agonist of the sphingosine1-phophate receptors (1 and 5) involved in lymphocyte trafficking and expressed on naïve and central memory T and B cells as well as NK cells. Ozanimod acts by inducing internalization and degradation of the S1PR which in turn …

Attenuated Bacteria Potentiate The Outcome Of Immunotherapy With Pd-L1 Blockade In A Pre-Clinical Model Of Colorectal Cancer, Besan Haytham Mohammad Al-Saafeen Nov 2021

Attenuated Bacteria Potentiate The Outcome Of Immunotherapy With Pd-L1 Blockade In A Pre-Clinical Model Of Colorectal Cancer, Besan Haytham Mohammad Al-Saafeen

Theses

After more than a century of investigation, it has become evident that cancer development and progression occur at the expense of a dysfunctional immune system. This realization has ushered cancer immunotherapy as a new modality of treatment. In the last decade, the use of immune checkpoint inhibitors to treat cancer patients resulted in unprecedented and durable clinical benefits. However, the response rate in many cancers remains rather modest, with colorectal cancer being at the lower end of the spectrum. Previous work from our laboratory demonstrated the efficacy of using attenuated bacteria as immunomodulatory anti-cancer agents. The current study investigated the …

Insights Into The Packaging Of Mouse Mammary Tumor Virus (Mmtv) Genomic Rna By Identifying Pr77ᴳᴬᴳ Binding Sites Involved During Its Selective Encapsidation, Akhil Chameettachal Jun 2021

Insights Into The Packaging Of Mouse Mammary Tumor Virus (Mmtv) Genomic Rna By Identifying Pr77ᴳᴬᴳ Binding Sites Involved During Its Selective Encapsidation, Akhil Chameettachal

Dissertations

Selective encapsidation and/or packaging of retroviral genomic RNA (gRNA) by Gag during retrovirus assembly is a crucial step for generating infectious virus particles. Despite having been studied extensively, the mechanism by which the retroviral Gag precursor selects and packages the retroviral genome remains largely unclear. Therefore, to understand the molecular mechanism(s) of mouse mammary tumor virus (MMTV) gRNA packaging, as a first step, expression of full-length recombinant Pr77Gag -His6-tag fusion protein in bacteria was done. The recombinant Gag protein was then purified from the soluble fractions of bacterial cultures using immobilized metal affinity chromatography (IMAC) and size exclusion chromatography …

Electrical Identification Of Innate Immune Cells, Rasha Ayman Nasser Dec 2020

Electrical Identification Of Innate Immune Cells, Rasha Ayman Nasser

Theses

This thesis is concerned with the electrical characterization of the key players of the innate immune cells. Innate immunity is basically the nonspecific immune response that is triggered by any foreign body that attacks the human system. The key players of the innate immune system are mainly dendritic cells and macrophages. Accurately classifying these cell types helps us understand the mechanism of the immune system thereby enabling the development of models to improve new prospects for therapeutics and diagnostics. The characterization described in this thesis is based on extracting the capacitance for each biological cell using I-V curves. The main …

Emergence Of Carbapenem Resistant Enterobacteriaceae In Abu Dhabi, 2009-2015, Aqdas Bilal Butt May 2018

Emergence Of Carbapenem Resistant Enterobacteriaceae In Abu Dhabi, 2009-2015, Aqdas Bilal Butt

Theses

The rapid emergence of carbapenem resistant Enterobacteriaceae (CRE) is a global phenomenon that has not spared the United Arab Emirates. However, unlike in most Western countries with similarly developed health care system, very little data have been available locally on the rate and dynamics of antimicrobial resistance in general, and that of CRE, in particular. Our laboratory has been collecting CRE isolates from Abu Dhabi hospitals since 2009. In the current study we subjected 394 such independent strains, representing 34.0% of the total CRE encountered, to a detailed analysis that included the determination of their antibiotic susceptibility profile, of the …
