Objective: To describe the clinical features of a novel fused in sarcoma (FUS) mutation in a young adult female amyotrophic lateral sclerosis (ALS) patient with rapid progression of weakness and to experimentally validate the consequences of the P525R mutation in cellular neuronal models.

Methods: We conducted sequencing of genomic DNA from the index patient and her family members. Immunocytochemistry was performed in various cellular models to determine whether the newly identified P525R mutant FUS protein accumulated in cytoplasmic inclusions. OBJECTIVE: The brain blood vessels of patients with type 2 diabetes and dementia have deposition of amylin, an amyloidogenic hormone cosecreted with insulin. It is not known whether vascular amylin deposition is a consequence or a trigger of vascular injury. We tested the hypothesis that the vascular amylin deposits cause endothelial dysfunction and microvascular injury and are modulated by amylin transport in the brain via plasma apolipoproteins.

The purpose of this study was to provide a comprehensive strength-based physiological profile of women’s collegiate basketball and gymnastic athletes; and to make sport-specific comparisons for various isokinetic and maximal and rapid isometric strength characteristics of the knee flexor and extensor muscles. A focus on antagonist muscle balance (hamstrings to quadriceps ratios, H:Q) will help elucidate particular vulnerabilities in these at-risk female athletes. Very limited experimental techniques are available to properly estimate inhaled doses and distribution of the drug inside the mouse lungs. Poxviruses have evolved unique proteins and mechanisms to counteract the nuclear factor κB (NF-κB) signaling pathway, which is an essential regulatory pathway of host innate immune responses. Here, we describe a NF-κB inhibitory virion protein of orf virus (ORFV), ORFV073, which functions very early in infected cells. Infection with ORFV073 gene deletion virus (OV-IA82Δ073) led to increased accumulation of NF-κB essential modulator (NEMO), marked phosphorylation of IκB kinase (IKK) subunits IKKα and IKKβ, IκBα and NF-κB subunit p65 (NF-κB-p65), and to early nuclear translocation of NF-κB-p65 in virusinfected cells (30 min post infection). UBQLN proteins have 3 functional domains: N-terminal ubiquitin-like domain (UBL), C-terminal ubiquitin-associated domain (UBA) and STI chaperone-like regions in the middle. Alterations in UBQLN1 gene have been detected in a variety of disorders including Alzheimer’s disease, Amyotropic Lateral Sclerosis and lung cancer. Activation of TLR4 signaling in neutrophils is beneficial for effective clearance of LPS-bearing Gram-negative bacteria, but may also drive aberrant inflammation if not stringently regulated. Using the biochemically tractable Xenopus oocyte model system, we have previously characterized a novel metabolic regulation of cell death. We found that glucose-6-phosphate (G6P) via the pentose phosphate pathway leads to increased nicotinamide adenine dinucleotide phosphate (NADPH) levels, a subsequent increase in cytosolic acetyl-coenzyme A and activation of Ca2+/calmodulin-dependent protein kinase II (CaMKII). We recently identified coenzyme A (CoA), derived from the breakdown of acetyl-CoA, as the key metabolic signal that mediates a novel mechanism of calmodulindependent activation of CaMKII. Although limited, the literature suggests alterations in activation of cognitive control regions in adults and adolescents with a history of childhood abuse. The current study examined whether such alterations are increased in the face of emotionally-distracting as compared to emotionally neutral information, and whether such alterations occur in brain regions that exert cognitive control in a more top-down sustained manner or a more bottom-up transient manner.

Once the disease has progressed far enough, dialysis treatments are mandatory to sustain life. To further understand how older adults receiving dialysis treatments cope with their disease, psychosocial factors were evaluated to investigate possible correlations with life satisfaction using a Life Satisfaction Index Z (LSI-Z). Expected correlations with life satisfaction were sociability, financial satisfaction, importance of spirituality, and activities of daily living scores. TB dapat meningkatkan progresivitas HIV dan meningkatkan risiko kematian bagi penderita HIV. Tujuan penelitian ini adalah untuk mengetahui prediktor yang berhubungan dengan kejadian TB pada ODHA di salah satu RS tahun 2014-2016. Desain studi yang digunakan adalah cross sectional dengan menggunakan data register ART dan Rekam Medis. Sampel berjumlah 817 pasien HIV. Kematian bayi didefinisikan sebagai kematian yang terjadi pada tahun pertama kehidupan. Angka kematian bayi di Indonesia dan Kamboja sendiri masih berada diatas AKB Asia Tenggara, sedangkan Filipina sudah sama dengan AKB Asia Tenggara. Jarak kelahiran merupakan salah satu faktor yang memegang peran penting pada kematian bayi. Tujuan penelitian ini adalah untuk mengetahui pengaruh jarak kelahiran terhadap kematian bayi di Indonesia, Filipina dan Kamboja. Penelitian menggunakan data dari Demographic Health Survey (DHS). Desain penelitian adalah cross sectional dan sampel pada masing-masing negara berjumlah 10.162, 4.741 dan 4.330 bayi. Hasil penelitian memperlihatkan, jarak kelahiran < 18 bulan memiliki risiko paling besar terhadap kematian bayi di Indonesia (OR = 2,43: 95% CI 1,26 - 4,70), Kamboja (OR = 4,39: 95% CI 1,76 - 10,94) dibandingkan jarak kelahiran 18 - 23 bulan, 24 - 35 bulan dan > 36 bulan. Stigma terhadap ODHA menjadi salah satu hambatan paling besar dalam pencegahan, perawatan, pengobatan, dan dukungan HIV/AIDS. Pengetahuan tentang HIV/AIDS mempengaruhi terjadinya stigma terhadap ODHA. Tujuan penelitian ini untuk mengetahui hubungan pengetahuan tentang HIV/AIDS dengan stigma terhadap ODHA di kalangan remaja usia 15-19 tahun di Indonesia. Penelitian ini menggunakan data Survei Demografi dan Kesehatan Indonesia (SDKI) Tahun 2012 dengan disain cross-sectional. Sampel penelitian sebanyak 8.316 orang. Traumatic brain injury (TBI) produces profound and lasting neuroinflammation that has both beneficial and detrimental effects. Recent evidence has implicated microRNAs (miRNAs) in the regulation of inflammation both in the periphery and the CNS. We examined the expression of inflammation associated miRNAs in the context of TBI using a mouse controlled cortical impact (CCI) model and found increased levels of miR-21, miR-223 and miR-155 in the hippocampus after CCI. The expression of miR-155 was elevated 9-fold after CCI, an increase confirmed by in situ hybridization (ISH). OBJECTIVES: In this analysis of 2-year outcomes in the ADAPT-DES (Assessment of Dual AntiPlatelet Therapy with Drug-Eluting Stents) study, the authors sought to examine the independent associations between platelet reactivity to both aspirin and clopidogrel and subsequent outcomes.

BACKGROUND: The relationship between platelet reactivity and long-term adverse events following implantation of drug-eluting stents (DES) has been incompletely characterized.

METHODS: The ADAPT-DES study was a multicenter registry of patients undergoing routine platelet function testing following percutaneous coronary intervention with DES.
