Morphine and Δ9-THC are drugs that produce analgesia and rewarding effects. However, chronic treatment with morphine and Δ9-THC produces problematic side-effects including tolerance and physical dependence. The cellular mechanisms underlying opioid and cannabinoid antinociceptive tolerance have been studied for years. Research has demonstrated that the expression of morphine and Δ9-THC antinociceptive tolerance may be mediated through intracellular signaling pathways, such as the adenylyl cyclase /Protein Kinase A (PKA) cascade. The present study investigated the role of PKA in the expression of morphine and Δ9-THC antinociceptive tolerance. Semen cryopreservation has an important role in assisted reproductive technology however, the cooling, freezing and thawing processes often result in a significant loss of sperm motility, viability and nuclear integrity. Many links exist between cocaine abuse and stress. The literature and laboratory studies in rats suggest that this could be because stress increases the reinforcing efficacy of cocaine. Using male Long-Evans rats, experiments in this thesis tested effects of footshock on the reinforcing efficacy of cocaine using a progressive ratio schedule of reinforcement. They also examined effects of footshock on the reinforcing efficacy of a half-maximal dose of cocaine. Finally, they tested the effects of footshock on cocaine self-administration in rats initially resistant to acquisition of cocaine self-administration. Despite distinct dissimilarities, diverse cancers express several common pro-tumorigenic traits. We present here evidence that the pro-apoptotic protein Par-4 utilizes one such common tumorigenic trait to become selectively activated and induce apoptosis in cancer cells. Elevated PKA activity noted in cancer cells activated the apoptotic function of ectopic Par-4 or its SAC domain, which induces apoptosis selectively in cancer cells and not in normal or immortalized cells. PKA preferentially phosphorylated Par-4 at the T155 residue within the SAC domain in cancer cells. Moreover, pharmacological-, peptide- or siRNA-mediated inhibition of PKA activity in cancer cells resulted in abrogation of both T155 …

Colon cancer is the third leading cause of cancer-related death in this country.5 The majority of medical societies recommend some form of colon cancer screening for asymptomatic adult patients over age 50. The evaluation of rectal bleeding is different from screening because the risk of serious disease is higher and it is unclear whether early diagnosis and treatment of serious disease results in improved mortality once gross bleeding has occurred. The optimal evaluation strategy for rectal bleeding is unknown. BACKGROUND: The Structural Maintenance of Chromosome 3 protein (SMC3) plays an essential role during the sister chromatid separation, is involved in DNA repair and recombination and participates in microtubule-mediated intracellular transport. SMC3 is frequently elevated in human colon carcinoma and overexpression of the protein transforms murine NIH3T3 fibroblasts. In order to gain insight into the mechanism of SMC3-mediated tumorigenesis a gene expression profiling was performed on human 293 cells line stably overexpressing SMC3. As a spirochete, the genus Treponema is one of the few major bacterial groups whose natural phylogenic relationships are evident at the level of gross phenotypic characteristics such as their morphology. Treponema spp. are highly invasive due to their unique motility in dense media, and their ability to penetrate cell layers. This feature is associated with the helical cell body and the presence of flagellar filaments in the periplasm. Studies of transgenic mice with accelerated accumulation of mtDNA mutations specifically in the heart lead us to propose that apoptotic signaling and cell death is central to the pathogenesis of mtDNA mutations in aging. It is the cellular response to that apoptotic signaling and the organ?s compensatory response to a loss of cells that specify the phenotype of an accumulation of mtDNA mutations. In the heart, cardiomyocytes induce a vigorous anti-apoptotic, pro-survival response to counteract mitochondrial apoptotic signaling. BACKGROUND: Two-photon-excitation fluorescence lifetime imaging (2P-FLIM) was used to investigate the association of protein kinase C alpha (PKCalpha) with caveolin in CHO cells. PKCalpha is found widely in the cytoplasm and nucleus in most cells. Upon activation, as a result of increased intracellular Ca2+ and production of DAG, through G-protein coupled-phospholipase C signalling, PKC translocates to a variety of regions in the cell where it phosphorylates and interacts with many signalling pathways. Due to its wide distribution, discerning a particular interaction from others within the cell is extremely difficult. Dysfunctional trophoblasts, impaired development of spiral arteries, imbalance in systems controlling the dilation and contraction of spiral arteries, placental fibrin clots and intervillous thrombosis are all possible factors that can result in an insufficient placental circulation. Bioluminescent imaging (BLI) is particularly well suited for imaging small animals, and it can be readily used by scientists who are already routinely using the luciferase gene as a reporter in cell-based assays. This strategy relies on ATP and an O_2 dependent photochemical reaction between luciferin and luciferase, resulting in the release of photons from only live cells. Glutathione (GSH) exerts a variety of cytoprotecive effects, but is readily depleted from cells under a variety of stressful stimuli. The impact of GSH depletion on receptor-mediated activity in rat hepatocytes has been studied with regard to glycogenolysis stimulated with a-adrenoceptor agonists or glucagon, which exert their actions through different signalling pathways. Depletion of GSH content (70%) by diethyl maleate had no effect on the redox status of the cells or on basal or glucagon-stimulated glycogenolysis, but significantly reduced the response to the a-adrenoceptor agonists adrenalin and phenylephrine. The aim of this work was to evaluate changes in lymphocyte subpopulations, especially helper and cytotoxic T cells, in acute brucellosis patients undergoing treatment. Forty-three acute brucellosis patients were included in the study. Twenty healthy subjects served as controls. Total lymphocytes and the CD3^+, CD4^+, CD8^+, CD19^+ and CD (16+56)^+ subpopulations were counted by two-color flow cytometric analysis. The CD4+ counts in patients before and after treatment were not statistically different (p = 0.7), but healthy subjects had significantly more of these cells (p = 0.001 and p = 0.001 compared to pre- and post-treatment patients, respectively). The imbalance between coagulation and fibrinolysis leads to vascular complications in diabetics. In our study, we have investigated the effects of the protein C-thrombomodulin system as a cause of diabetic vascular complications. Whereas PC-Ag levels were lower, thrombomodulin (TM) levels of diabetics were higher than that of the controls (P < 0.001, P < 0.001 respectively). Furthermore, patients were divided into three groups according to their urinary albumin excretion (UAE). Group 1, 2 and 3 consisted of 27 patients with UAC £ 30 µg/ml, 16 patients with UAC in range 30-140 µg/ml, 3 patients with UAC >140 µg/ml, respectively. Plasma PC-Ag levels were significantly decreased in group 1 and 2 with respect to controls (P < 0.01, P < 0.01). Plasma TM concentrations were significantly increased in group 1, 2 and 3 compared with the control group (P < 0.001, P < 0.01, P < 0.01). Patients were also divided into two groups according to the duration of their illness; patients whose diabetic age < 10 years (Group A) and whose diabetic age > 10 years (Group B). In both groups, PC-Ag levels were lower and TM levels higher than that of the control group (P < 0.01, P < 0.01; P < 0.001, P < 0.001 respectively). There was no significant difference between group A and B in PC-Ag levels (P > 0.50) but a significant difference in TM levels (P < 0.001).

Methicillin-resistant Staphylococcus aureus (MRSA) is an important nosocomial pathogen. The prevalence of MRSA in many countries is increasing and, in some hospitals, more than half of all S. aureus disease isolates are MRSA. MRSA strains are becoming increasingly multiresistant, and have recently developed resistance to vancomycin, which has been used successfully to treat MRSA for more than 30 years. In-vitro determination of resistance patterns of S. aureus is critical in terms of administering suitable antimicrobial treatments. The objective of this study is to evaluate effects of four dentin bonding agents PromptL-Pop, Syntac, Pekabond, Scotchbond-1 on cell proliferation in human gingival fibroblast cultures. Under aseptic conditions, test specimens were placed in the centre of 24-well tissue culture trays. Each well was covered at a concentration of 104 cells/cm2. The cultures were incubated at 37 ºC and cell proliferation was determined by the MTT method 24 h and 72 h after exposure. Statistical analysis was performed applying the Student t test. Having previously shown that CCL20/macrophage inflammatory protein 3 and tumor necrosis factor alpha (TNF-alpha) are released by polarized primary rat uterine epithelial cells (UEC) in response to Escherichia coli but not to Lactobacillus rhamnosus , we sought to determine if epithelial cells are responsive to pathogen-associated molecular patterns (PAMP), including lipopolysaccharide (LPS), lipoteichoic acid (LTA), and Pam 3 Cys, a bacterial lipoprotein analog. Epithelial cells were grown to confluence on Nunc cell culture inserts prior to apical treatment with PAMPs. We investigated the role of interleukin-17 (IL-17)/IL-17 receptor (IL-17R)-mediated signaling in the protective immunity against Toxoplasma gondii. IL-17R−/− mice developed a normal adaptive immunity against the parasite. However, increased mortality in the knockout animals can be attributed to a defect in the migration of polymorphonuclear leukocytes to infected sites during early infection.

Background: Acyl-CoA thioesterases are enzymes that hydrolyze acyl-CoAs to the free fatty acid and coenzyme A (CoASH). These enzymes have been identified in several cellular compartments and are thought to regulate intracellular levels of acyl-CoAs, free fatty acids and CoASH. However, to date no patients deficient in acyl-CoA thioesterases have been identified. Design: Acyl-CoA thioesterase activity was measured in human skin fibroblasts. Western blot analysis was used to determine Type-II acyl-CoA thioesterase protein levels in patients.
