Cholesterol Has An Inflammatory Influence In The Effects Of Fats On Metabolism In Endotoxemic Rats, Hali̇t Tanju Besler, Robert Francis Grimble

Turkish Journal of Medical Sciences

We showed that butter suppresses metabolic responses to endotoxin in Wistar rats. Responses included increases in plasma caeruloplasmin and liver zinc and protein concentrations, and decreases in plasma zinc and albumin concentration. However, butter lost its suppressive effect at dietary concentrations above 90g/kg diet. The present study examined whether this phenomenon was due to the total dietary cholesterol intake rather than the intrinsic fatty acid composition of butter. Rats received diets, for four weeks, containing either 100g/kg corn oil, 90 g/kg butter, 190g/kg butter, or 90 g/kg butter supplemented with cholesterol to render the latter two diets identical in cholesterol …

Turkish Journal of Medical Sciences

It has been hypothesized that treating hypercholesterolemic patients with statins will lead not only to a reduction in cholesterol, but also to inhibited synthesis of other compounds that derive from the synthetic pathway of cholesterol. One important by-product is ubiquinone (CQ), which has a pivotal role in mitochondrial electron transport and antioxidant activity. We therefore investigated the effect of 2 months of simvastatin treatment (20 mg/day) on blood ATP concentration, plasma total antioxidant capacity and ubiquinone levels in 17 hypercholesterolemic patients (age range: 40 to 65). To evaluate the possible muscle-related side effects, plasma CK activity and myoglobin concentrations were …

High-Resolution Physical Map For Chromosome 16q12.1-Q13, The Blau Syndrome Locus, Xiaoju Wang, Helena Kuivaniemi, Gina Bonavita, Charlene J. Williams, Gerard Tromp

Wayne State University Associated BioMed Central Scholarship

Abstract

Background

The Blau syndrome (MIM 186580), an autosomal dominant granulomatous disease, was previously mapped to chromosome 16p12-q21. However, inconsistent physical maps of the region and consequently an unknown order of microsatellite markers, hampered us from further refining the genetic locus for the Blau syndrome. To address this problem, we constructed our own high-resolution physical map for the Blau susceptibility region.

Results

We generated a high-resolution physical map that provides more than 90% coverage of a refined Blau susceptibility region. The map consists of four contigs of sequence tagged site-based bacterial artificial chromosomes with a total of 124 bacterial artificial …

Positive And Negative Roles Of Cd72 In B Cell Function., Hsin-Jung Wu, Subbarao Bondada Jan 2002

Positive And Negative Roles Of Cd72 In B Cell Function., Hsin-Jung Wu, Subbarao Bondada

Microbiology, Immunology, and Molecular Genetics Faculty Publications

Regulation of B cell activation depends on integration of signals transmitted by the B cell receptor (BCR) and a variety of co-receptors. CD72 is a B cell co-receptor that is expressed in all stages of B cell development except plasma cells. Ligation of CD72 enhances B cell growth and differentiation. Recently, the class IV semaphoring, CD100, has been identified as the natural ligand for CD72. Cytoplasmic domain of CD72 has been shown to be associated with SHP-1 leading to the proposal that the positive effects of CD72 on B cell response may result from sequestration of negative signals from BCR. …

Increased Production Of Pro-Inflammatory Cytokines And Enhanced T Cell Responses After Activation Of Human Dendritic Cells With Il-1 And Cd40 Ligand, Amy Wesa, Anne Galy

Wayne State University Associated BioMed Central Scholarship

Abstract

Background

Various microbial, inflammatory and immune signals regulate the activation of dendritic cells (DC), determining their ability to interact with naïve T cells and to produce cytokines that direct T cell development. In particular, CD40L and IL-1 cooperatively activate DC to secrete high levels of IL-12. The immuno-stimulatory capacity of such DC is otherwise not well-defined prompting further characterization of the effects of IL-1 and family members on DC activation in comparison with other pro-inflammatory stimuli.

Results

Human DC co-activated in vitro by CD40L and IL-1β expressed numerous cytokine genes including IL-12β, IL-23 p19, IL-1β, IL-1α, IL-1Ra, IL-10, IL-6, …

Wayne State University Associated BioMed Central Scholarship

Abstract

Background

Cerebrovascular disease is the third leading cause of death in the United States, and about one-fourth of cerebrovascular deaths are attributed to ruptured intracranial aneurysms (IA). Epidemiological evidence suggests that IAs cluster in families, and are therefore probably genetic. Identification of individuals at risk for developing IAs by genetic tests will allow concentration of diagnostic imaging on high-risk individuals. We used model-free linkage analysis based on allele sharing with a two-stage design for a genome-wide scan to identify chromosomal regions that may harbor IA loci.

Methods

We previously estimated sibling relative risk in the Finnish population at between …

Turkish Journal of Medical Sciences

We investigated whether the activity of double-stranded RNA activated kinase (p68/PKR) is necessary for induction of IDO (indolamine 2,3 -dioxygenase) gene by IFN-\gamma. For this purpose, we planned to abrogate the function of cellular wt-PKR gene. Clones of ME180 cells expressing the dominant negative form of PKR were selected in the presence of neomycin. Then these cells were treated with IFN-\gamma and accumulation of IDO protein was determined by Western blot analysis. We found that clones expressing the dominant negative form of the PKR gene were not only resistant to IFN-\gamma-mediated killing, but were also incapable of inducing accumulation of …

Comparison Of Antibiotic Resistance Patterns Of Uropathogenic Escherichia Coli In Outpatient And Inpatient Populations From 1990 To 1996 In Bemidji, Mn, Gail R. Conway, Patrick Guilfoile

Journal of the Minnesota Academy of Science

A retrospective study of antibiotic susceptibility was conducted on data from all uropathogenic Escherichia coli submitted to the MeritCare Clinic-Bemidji, MN and the North Country Regional Hospital-Bemidji, MN during 1990, 1993, and 1996. The proportion of antibiotic resistant strains was generally equal or higher in E. coli isolates from community-acquired infections than with E. coli isolates from nosocomial infections. Comparison with antibiotic prescriptions during 1990, 1993, and 1996 provided no clear relation between antibiotics prescribed for outpatient urinary tract infections and the proportion of E. coli strains resistant to those antibiotics. This suggests that factors other than selection for resistance …

Turkish Journal of Medical Sciences

Breast cancer is the most prevalent cancer in women and an important cause of both morbidity and mortality. Although long-term disease-free survival is offered to women with early stage disease by current treatment modality, some, 30-40% of patients will have metastatic disease. Treatment of patients with metastatic disease is a great challenge for oncologists. Therefore, new drugs and therapeutic options for women with metastatic breast cancer are urgently required. Arsenical compounds have cytotoxic effects on several cancer cell lines including promyelocytic leukemia, esophageal carcinoma, megakaryocytic leukemia, and malignant lymphocytic cell lines. In addition, recent studies showed that arsenic trioxide (As_2O_3) …
