The Chlamydia species are obligate intracellular bacteria that proliferate only within the infected cell. Since the extracellular bacteria are metabolically inert and there are no cell-free systems for characterizingChlamydia metabolism, we studied metabolic changes related to ATP synthesis and glycolysis in HeLa cells infected withChlamydia psittaci during the course of the 2-day infection cycle using noninvasive 31P and 13C NMR methods. We find that the infection stimulates ATP synthesis in the infected cell, with a peak of ATP levels occurring midway through the infection cycle, when most of the metabolically active bacteria are proliferating. As the most common cause of sexually transmitted disease in women, chlamydial infections can lead to pelvic inflammatory disease, infertility, and ectopic pregnancy. To better understand the role played by sex hormones in modulating the immune response of the genital tract to microbial infections, we have developed a rat model to study Chlamydia trachomatis infection. Inbred female Lewis rats were primed with progesterone and inoculated by intrauterine instillation of C. trachomatis (mouse pneumonitis strain MoPn) into each uterine horn. Results on the subcellular localization of the mineralocorticoid receptor (MR) have been controversial. To determine the subcellular distribution and trafficking of the MR in living cells after binding of agonists and antagonists, we expressed a MR-green fluorescent protein (GFP) chimera in mammalian cells lacking endogenous MR. The GFP-tagged MR (GFP-MR) remained transcriptionally active, as determined in cotransfection experiments with the MR-responsive reporter, TAT3-LUC. The subcellular localization of GFP-MR was monitored by fluorescence time-lapse microscopy. In the absence of hormone, MR was present both in the cytoplasm and nucleus. Configural theories of hippocampal function predict that hippocampal dysfunction should impair acquisition of the transverse patterning task, which involves the concurrent solution of three discrimination problems: A+ versus B-; B+ versus C-; and C+ versus A-. The present study tested this prediction in rats using computer-graphic stimuli presented on a touchscreen. Experiment 1 assessed the effects of fornix lesions when the three problems were introduced sequentially (phase 1: A+ vs B-; phase 2: A+ vs B-, B+ vs C-; phase 3: A+ vs B-, B+ vs C-, C+ vs A-). In this study, adult volunteers were fed tcpA and mshA deletion mutants of V. cholerae O139 strain CVD 112 to determine the role of toxin-coregulated pili (TCP) and mannose-sensitive hemagglutinin (MSHA) in intestinal colonization. Eight of 10 volunteers who received CVD 112 or CVD 112 ΔmshA shed the vaccine strains in their stools; the geometric mean peak excretion for both groups was 1.4 × 105 CFU/g of stool. In contrast, only one of nine recipients of CVD 112 ΔtcpA shed vibrios in his stool (P < 0.01); during the first 24 h after inoculation, 3 × 102 CFU/g was recovered from this volunteer. Homotypic vacuole fusion in yeast requires Sec18p (N-ethylmaleimide-sensitive fusion protein [NSF]), Sec17p (soluble NSF attachment protein [alpha-SNAP]), and typical vesicle (v) and target membrane (t) SNAP receptors (SNAREs). We now report that vacuolar v- and t-SNAREs are mainly found with Sec17p as v-t-SNARE complexes in vivo and on purified vacuoles rather than only transiently forming such complexes during docking, and disrupting them upon fusion. In the priming reaction, Sec18p and ATP dissociate this v-t-SNARE complex, accompanied by the release of Sec17p. Sporulation is a developmental process in which temporally distinct sets of genes regulate the processes of meiosis and ascospore formation. Sporulation-specific genes are classified into early, middle, and late genes based on their peak time of expression during sporulation. Regulation of two late genes, SPR1 and SPR2, was studied. The function of SPR2 is unknown. Its regulatory region contains two Mid Sporulation Elements (MSE) repeated in tandem that are sufficient to confer sporulation-specific regulation to a CYCl-lacZ reporter. Physiologists say that ions and neutral solutes can cross biological membranes via “transporters” and “channels.” We tend to think about the difference between transporters and channels in terms of gating mechanisms. Ion channels exhibit a wide range of selectivity properties and permeation rates, but their gating at the most basic level can be thought of in terms of a single barrier or gate acting as a switch. When the gate is closed, ions can’t permeate; when the gate is opened, a permeation pathway for ions allows flux, often at very high rates (up to 108/sec). Transporters and ion …

The rapid re-uptake of extracellular glutamate mediated by a family of high-affinity glutamate transporter proteins is essential to continued glutamatergic signaling and neuronal viability, but the contributions of individual transporter subtypes toward cellular physiology are poorly understood. Because the physiology of glutamate transport in the salamander retina has been well described, we have examined the expression and function of glutamate transporter subtypes in this preparation. cDNAs encoding five distinct salamander excitatory amino acid transporter (sEAAT) subtypes were isolated, and their molecular properties and distributions of expression were compared. Glutamate transporters remove this neurotransmitter from the synaptic cleft by a two-stage electrogenic process, in which glutamate is first cotransported with three sodium ions and a proton. Subsequently, the cycle is completed by translocation of a potassium ion in the opposite direction. Recently, we have identified an amino acid residue of the glutamate transporter GLT-1 (Glu-404) that influences potassium coupling. We have now analyzed the effect of seven other amino acid residues in the highly conserved region surrounding this site. Glutamate transporters remove this transmitter from the extracellular space by cotransport with three sodium ions and a proton. The cycle is completed by translocation of a potassium ion in the opposite direction. Recently we have identified two adjacent amino acid residues of the glutamate transporter GLT-1 that influence potassium coupling. Using the scanning cysteine accessibility method we have now explored the highly conserved region surrounding them. Replacement of each of the five consecutive residues 396–400 by cysteine abolished transport activity but at several other positions the substitution is tolerated. Signal transductions by the dual-function CXCR4 and CCR5 chemokine receptors/HIV type 1 (HIV-1) coreceptors were electrophysiologically monitored in Xenopus laevis oocytes that also coexpressed the viral receptor CD4 and a G protein-coupled inward-rectifying K+ channel (Kir 3.1). Large Kir 3.1-dependent currents generated in response to the corresponding chemokines (SDF-1α for CXCR4 and MIP-1α; MIP-1β and RANTES for CCR5) were blocked by pertussis toxin, suggesting involvement of inhibitory guanine nucleotide-binding proteins. Prolonged exposures to chemokines caused substantial but incomplete desensitization of responses with time constants of 5–7 min and recovery time constants of 12–19 min. The excitatory amino acid transporter EAAT4 is expressed predominantly in Purkinje neurons in the rat cerebellum (1-3), and it participates in postsynaptic reuptake of glutamate released at the climbing fiber synapse (4). Transporter-mediated currents in Purkinje neurons are increased more than 3-fold by arachidonic acid, a second messenger that is liberated following depolarization-induced Ca2+ activation of phospholipase A2 (5). In this study we demonstrate that application of arachidonic acid to oocytes expressing rat EAAT4 increased glutamate-induced currents to a similar extent. The behavior of a Cl channel associated with a glutamate transporter was studied using intracellular and patch recording techniques in Xenopus oocytes injected with human EAAT1 cRNA. Channels could be activated by application of glutamate to either face of excised membrane patches. The channel exhibited strong selectivity for amphipathic anions and had a minimum pore diameter of ∼5Å. Glutamate flux exhibited a much greater temperature dependence than Cl flux. This study was undertaken to examine the laminar distribution of spinotelencephalic tract neurons in the rat, by means of retrograde fluorescent labelling and to estimate the role of NADPH-diaphorase-reactive spinal cells as sources of spino-supraspinal pathways in the rat. The localization, size and shape of some neurons were similar in both labelling methods. Cells were mostly multipolar in the medial part of nucl. proprius, and fusiform and spindle-shaped in lamina I and X. Dexfenfluramine is a variation of fenfluramine which is used for weight reduction. The aim of the study is to determine the hemodynamic and structural effects of this drug on pulmonary circulation. An experimintal study was carried out with New Zealand Rabbits. A control group (n=30) and an experimintal group (n=20) was constituted. A four-week normal diet for control group and dexfenfluramine plus normala diet for experimental group were applied. Body weights and pulmonary artery pressures of groups were recorded and lungs were collected for pathologic examination. An experimental study was designed to investigate the effect of diclofenac sodium (DS) on both renal parenchyma during complete unilateral ureteral obstruction. In 40 adult rats, left ureteropelvic junction were completely obstructed. Rats were divided into 4 equal groups and treated with cefoperazone (Group 2), DS (Group 3), cefoperazone and DS (Group 4) for one week postoperatively. No drug was given to Group 1 (control). Then, both kidneys of all rats were removed and examined histopathologically. The parenchymal thickness of left kid-neys were less than those of rights in all groups. This study was carried out to assess the role of tumor necrosis factor (TNF-\alpha) and interleukin-6 (IL-6 ) in plasma and peritoneal fluid in higher rates of adhesion formation following standard bowel injury. Forty-five albino rabbits were divided into three equal groups. Blood was obtained from all the rabbits preoperatively. All the rabbits were subjected to a laparotomy. This study is an evaluation of serum levels in the tumor markers CA 19-9 and CA 72-4 in gastric cancer patients in preoperative, postoperative and follow-up periods. These tumor markers were measured in 35 patients with primary gastric carcinoma (28 advanced stage, 7 early stage), 27 with benign gastric disease and 20 healthy control subjects. Serum CA 72-4 levels were found to have increased (>6.7kU/l) in 16 (14 with advanced stage, 2 with early stage) patients with primary gastric carcinoma. ESWL is an effective and noninvasive method of treatment for patients with urinary stones. However, especially in the treatment of larger stones impact of the small stone particles in the lower ureter (steinstrasse) is a serious complication that affects the result of the procedure. If the renal unit has not been catheterized before the ESWL procedure, removal of these particles by open surgical or endoscopic intervention within a very short time is mandatory. Objective: Our purpose was to determine whether unexplained elevations in maternal serum human chorionic gonadotropin (hCG)in the second trimester is associated with an increased risk of preeclampsia. The effect of zinc salts is defined by several studies. The aim of this study is to investigate how the short period toothbrushing with equal volumes of a toothpaste containing 0.75% zinccitrate effects the salivary zinc levels over a period of 7 hours. In our study a significant increase in zinc measurements was observed 5 minutes after toothbrushing. Although 7 th hour measurements were higher than 0 hour measurements; these values were found approximately similar to intial control measurements. Cartilage grafts are widely used in maxillo-facial reconstruction operations. There are some difficulties of conventional suture techniques in these operations such as long operation time, mobilization, shifting, and implantation or fixation of small pieces of cartilage. Therefore the advantages of a tissue adhesive over conventional suture techniques became evident in these operations. Butyl 2-Cyanoacrylate (B2-CA) is one of the least histotoxic cyanoacrylate derivatives and is used as a tissue adhesive. The effect of different techniques and materials on peripheral shaping of the complete denture impressions were investigated by stereophotogrammetric method. Six impressions were taken from each of the 20 edentulous patients by using two different techniques and three impression materials. A non-metric camera was used to produce stereo paired photographs of each model. The measurements were made at the buccal vestibule regions, labial vestibule regions and labial frenilum region on 120 casts. When buccal vestibule regions were evaluated no statistically significant difference was found between double phase and mono phase impression materials.
