The intercistronic gene junctions of vesicular stomatitis virus (VSV) contain conserved sequence elements that are important for polyadenylation and transcription termination of upstream transcript as well as reinitiation of transcription of downstream transcript. To examine the role of the putative polyadenylation signal 3' AUACU75' at the gene junctions in polyadenylation and transcription termination, we constructed plasmids encoding antigenomic minireplicons containing one or two transcription units. Venous thrombosis is a serious health problem affecting 1 in 1000 individuals annually. Until recently the pathogenic factors underlying thrombosis were associated with genetic defects in protein C, protein S and antithrombin III. However these were recognised in fewer than 5-10% of thrombotic patients. A breakthrough was made with the discovery of activated protein C resistance (APCr) which is associated in 90% of cases v/herein a G-->A transition in the factor V gene results in an abnormal molecule, Factor V Leiden. Four psychoactive drugs were studied using various in vivo nuclear magnetic resonance (NMR) techniques. Trifluoperazine (TFP) was given to several normal rats, whose brains were then studied using in vivo fluorine-19 NMR. Three peaks were detected in the in vivo fluorine spectra: one at -62 ppm, which was due to TFP, and two others at -55 and -75 ppm, which are due to unknown metabolites of TFP. Samples of skin, skull, muscle, and brain were obtained from these rats and extracted for in vitro analysis. The metabolites responsible for the in vivo peak at -75 ppm also appeared in the …

The aim of this study is to investigate the frequency of chromosomal abnormalities in men with severe andrological infertility, In 50 men with azoospermia and severe oligoasthenoteratozoospermia (OAT), who referred for our intracytoplasmic sperm injection program, a cytogenetic study has been prospectively carried out. Peripheral blood lymphocytes were cultured for chromosomal analysis and for each proband 30 metaphases were analyzed by GTG banding technique. Major chromosomal abnormalities were detected in 4 of cases (8%). There were 3 sex-chromosome abnormalities, 46XXY in all 3 cases, and one autosomal chromosomal abnormality 46 XY, t (1;7) (P32;Q32). Currently there are conflicting results regarding optic neuropathy in diabetes mellitus. Therefore, the aim of the present study was to evaluate optic neuropathy in diabetic patients. We studied visual evoked potentials (VEPs) in 20 diabetic patients (6 insulin dependent, 14 noninsulin dependent; 8 women, 12 men; mean age: 49.70± 18.22 years; mean duration of diabetes: 9.33 ± 5.75 years) and 20 age-, and sex-matched healthy subjects (7 women, 13 men; mean age: 52.40 ± 16.02 years) as a control group. In this study, the blood flow velocity waveforms of the leiomyoma, uterine artery and myometrium with color doppler ultrasound were analyzed in patients with postoperative diagnosis of benign uterine leiomyomas and 15 healthy volunteers as a control group were enrolled into the study. The lowest RI and PI values obtained from uterine leiomyomas were 0.39 and 0.93. Mean RI and PI values obtained from uterine leiomyomas were 0.39 and 0.93. The purpose of this study is to examine the morphological effect which appears in acini and ductus of palatine glands in patients wearing total prostheses. Acini had considerably wide lumens due to de-generation and desquamation in secretory epithelial cells in those who wear prostheses during varying periods. It was observed that, in regions adjacent to the apical membrane in most of the epithelial cells of acini and excretory canals, large vacuoles were formed, whereas in some others, cytoplasm was disarranged due to membrane destruction. The production and detection of a mannoprotein adhesin (MPA) of the hyphal-form cells of C. albicans on plastic petri dishes was investigated. Using Concanavalin A-coated latex microspheres, the MPA was detected on the plastic surface on which C. albicans produced germ tubes. The adhesin was extracted using dithiothreitol and iodoacetamide. It did not inhibit the adhesion of the yeast-form C. albicans to buccal epithelial cells (BEC). This suggested that the MPA of the hyphal-form cells on the plastic was different from that on the yeast cells responsible for attachment to the BECs.

We investigated limited joint mobility in 77 patients with type I diabetes mellitus. The patients were also assesed for neuropathy, retinopathy and pulmonary complications. The prevalence of limited joint mobility was 47%. Skin changes were present in 87% of patients with diabetes of more than 5 years' duration. No relationship was found between limited joint mobility and metabolic control, but there was a very strong correlation between limited joint mobility and other diabetic complications such as neuropathy, nephropathy and retinopathy. We did not find any differences in the pulmonary function tests of patients with and without limited joint mobility. We …

Hypervolemia is supposed to be the principal factor in hypertension in chronic hemodialysis adults. In this study, the role of intensive ultrafiltration (UF) treatment to reduce hypertension due to hypervolemia in hemodialysis children was investigated. The inferior vena cava (IVC) was also evaluated as an index of UF therapy. Twenty-three hemodialysis children with the mean age 10.6±3.1 years were classified due to their blood pressure status. Medullary thyroid carcinoma (MTC) may present in various clinical forms, either sporadic or a counterpart of hereditary endocrine disorders. The appropriate surgical procedure for MTC is total thyroidectomy combined with central neck clearance. The aim of this study was to document the initial clinical presentation, surgical treatment and clinical course of MTC in 27 patients. Thirteen (48.1%) cases were diagnosed as hereditary, either MEN 2a(7/27, 25.9%) or 2b syndrome (1/27, 3.7%) or familial non-MEN MTC (5/27, 18.5%), and fourteen cases were diagnosed as sporadic. In industry increased mechanisation results in increased noise levels. Operation of textile machines carries a high risk of hearing loss. In this study the evaluation of textile worker's noise induced hearing loss was reviewed cross sectionally. The hearing of 260 textile workers exposed to noise levels between 85-95 dB(A) in carpet and cotton textile factories was assessed by means of air and bone conductance audiograms obtained. The subjects were grouped into five hearing categories according to hearing thresholds at 125 to 8000 Hz with Klockhoffs classification. Endogenous leukotrienes (LT) levels in plasma and urine samples, and LT levels released from stimulated leukocytes in vitro were examined in children with steroid sensitive nephrotic syndrome (SSNS). Plasma LT concentrations were found to be higher in the SSNS group than in healthy subjects. Subjects with SSNS were also shown to have high levels of urinary cysteinyl-LTs and LTB 4 , while healthy children had only urinary LTE 4 levels. This project was initiated to determine if was possible to enrich either X- or Y- bearing sperm, and hence to preselect the sex of a child. Two of the possible reasons why couple might want to select the sex of a child are firstly because of a family history of an X-linked recessive genetic disorder, which usually only affect sons, and secondly families who have had several children of one sex. The aim of this research was to perform exclusion on a rare form of hereditary autosomal dominant polycystic kidney disease (ADPKD). To-date, two genes for ADPKD have been identified: PKDI which has been localized to the short arm of chromosome 16 and PKD2 which has been localized to the long arm of chromosome 4. However, a small number of families have been reported that have not shown linkage to either of these two loci, thus suggesting the existence of at least one additional locus (PKD3).
