Blood-mimicking fluids (BMFs) are often used to investigate blood flow using physical replicas of vessels with cardiovascular disease. Particle image velocimetry (PIV) is used with silicone poly-dimethyl siloxane (PDMS) vascular models to visualize this flow. The challenge is creating a blood-mimicking fluid that matches the viscosity profile, viscoelasticity, and density of whole blood while also matching the refractive index (RI) of PDMS. Water-glycerol solutions are commonly used with sodium iodide (NaI) added to increase the RI without changing viscosity. RATIONALE: Pulmonary hypertension is characterized by increased pressure in the pulmonary artery, and is a key contributor to worsening symptoms in individuals with chronic obstructive pulmonary disease (COPD). The pulmonary artery to aorta diameter ratio (PA:Ao), measured using computed tomography (CT), is a biomarker of pulmonary hypertension; however, longitudinal changes in this measurement and its relationship to pulmonary vascular and airway structural changes is not well understood. Our objective was to investigate longitudinal changes in PA:Ao and its relationship with CT pulmonary vascular and airway abnormalities, airflow limitation and exercise-capacity.

Tendons join muscle to bone and are essential for posture and movement. Tenocytes are the resident tendon cells. Isolating RNA from small animal models such as mice can be difficult, especially from tendon, as much of the time, they are extremely small samples. Downstream analysis such as qPCR and especially RNA sequencing require greater quantity and quality of RNA than is often achievable. RNA degradation is extremely rapid following tendon isolation, with RNA quality decreasing as time increases between the collection of tendons and RNA isolation. Microvascular mechanisms that regulate the microcirculation can be observed within blood flow as coupled oscillations operating over a wide range of different frequencies and time scales. Abnormally discordant interactions between regulation mechanisms can cause derangement of microvascular flow patterns, which has been suggested to be indicative of increased cardiovascular disease risk. Based on the premise that interactions between microvascular mechanisms are key to understanding flow regulation and disease progression, wavelet-based phase coherence was used to characterize the relationship between cardiac, respiratory, myogenic, neurogenic, and metabolic regulation across different microvascular beds and physiological conditions.

Background: The rapid reduction in arterial pressure (AP) during VVS is associated with a decline in diastolic cerebral blood velocity (CBV) and maintained systolic CBV. We tested the hypothesis that this response may represent increased cerebrovascular compliance (Ci) in the presence of rapid hypotension.

Methods: AP and CBV were recorded from 14 otherwise healthy patients (34 ± 11 years) at rest and during head-up tilt (80°, 5-28 min). Baseline, mid-tilt, and VVS were analyzed. Individual AP and CBV waveforms were input into a modified Windkessel model to calculate Ci.

Faecal microbiota transplantation (FMT) involves the administration of donor faecal matter to a diseased recipient with the goal of remodeling the host microbiome to provide health benefits. In recent years, FMT has emerged as a potential therapy for a variety of microbiome-associated diseases such as atherosclerosis. Trimethylamine (TMA) is an atherosclerosis-linked metabolite generated by the gut microbiota from dietary precursors which is then oxidized to trimethylamine N-oxide (TMAO) by the liver, contributing to increased gut permeability. Intratumoral modulation therapy (IMT) is a novel electrotherapy used to treat brain cancer tumours using electric fields applied directly to the tumours through implanted electrodes. Previous research has validated IMT's effectiveness and provided computer-simulated optimizations for IMT electric fields. This work validates these computer optimizations in-vitro, using a PCB construct to deliver electric fields, and bioluminescence imaging to assess cell viability.

We found electric field strength to correlate with cell viability, and found that rotating (phase-shifted) electric fields did not produce significant improvements in IMT efficacy. Future work will investigate different IMT frequencies and other parameters, while providing biological replicates …

The placenta is critical for nurturing fetal growth and development, with dysregulated placentation associated with adverse pregnancy outcomes. The main fetal and maternal placental components consist of trophoblasts and modified endometrial stromal cells known as decidual cells, respectively. Since investigating in vivo placentas in humans through non-invasive methods is challenging, comprehensive in vitro placental models are needed for in-depth studies. However, emphysema progression is able to cause increasing unventilated lung areas which likely excludes the estimates of the largest apparent diffusion coefficient (ADC). As such, longitudinal observation of the emphysema progression using hyperpolarized gas MRI-based ADC can be problematic, masking the severity of emphysema. Translation, or the production of protein from an mRNA blueprint, is among the most fundamental processes to life as we know it. tRNAs are essential to accurate translation, as they decode the codons of mRNA and recruit corresponding amino acids. Variant tRNAs with anticodon mutations can decrease translational fidelity by recruiting the incorrect amino acid, an aberrant process known as mistranslation. Cisplatin is a chemotherapeutic agent complicated by its dose-limiting nephrotoxicity. This commonly presents as acute kidney injury (AKI), which is characterized by a reversible decline in kidney function. AKI is currently diagnosed by increased serum creatinine (SCr) levels, but this rise in SCr is delayed limiting diagnosis to when significant renal damage has already occurred. Currently, there are no known biomarkers capable of predicting or diagnosing early-stage cisplatin-induced AKI. When someone learns a new motor skill (i.e. throwing a football to a wide receiver) the brain must make and map an internal model of the forces required for that movement. Sensory filtering is the process of separating and distilling relevant sensory information from irrelevant, which in turn greatly reduces the quantity of sensory information that is fully processed and leads to significant increases in efficiency. Atypical sensory filtering can result in sensory hypo- or hypersensitivity — atypical sensory filtering and hypo/hypersensitivity have been observed in people with autism. Atypical sensory filtering contributes to canonical symptoms in Autism.

Type 1 Diabetes Mellitus (T1DM) is an autoimmune disorder that results in insufficient endogenous insulin production. Regular exercise has numerous health benefits for individuals with T1DM, however, most insulin-dependent diabetics avoid physical activity due to the fear of exercise-induced hypoglycemia (low blood glucose/BG).

The risk of hypoglycemia in this population may be partly due to lower liver glycogen stores which is a major source of blood glucose during exercise. However, the mechanism that leads to lower glycogen stores in T1DM is unknown.

However, a barrier to regular exercise for this population is the fear of low blood glucose (BG) levels, also known as hypoglycemia. MRI can visualize and quantify the movement of water in real time. In this study, MRI was used to observe the water penetration through clay. When bound to an agonist, such as neurotransmitters or hormones, the receptor adopts an active state to allow these biochemical pathways to occur. Adult-onset leukoencephalopathy with axonal spheroids (ALAS) is a group of hereditary, progressive, neurodegenerative disorders involving primarily the central nervous system white matter (WM). ALAS is characterized by patchy, asymmetrical myelin loss and axonal destruction in the WM, predominantly involving the frontoparietal regions. However, the asymmetrical and heterogenous involvement of different brain regions remains poorly characterized.

In this study, digital spatial profiling was performed to investigate the region-specific expressions of 60 proteins. Sepsis is characterized by the widespread inflammation of the body. Systemic inflammation activates and recruits inflammatory cells (e.g., leukocytes) and platelets to the affected organs.

During these inflammatory conditions, human brain microvascular endothelial cells (hBMEC) and platelets both upregulate adhesive molecules rendering platelets to adhere to hBMEC.

Although carbon monoxide is thought of as a toxic molecule to many, previous work shows its anti-inflammatory properties. Acquired or innate resistance to chemotherapy poses a significant challenge in the treatment of head and neck squamous cell carcinomas (HNSCC). HNSCCs are often treated with a combination of surgery, radiation and chemotherapy, with cisplatin being a standard-of-care chemotherapy drug. Human Papillomavirus (HPV) associated HNSCCs have better treatment outcomes and prognosis when compared to HPV- HNSCCs.
