Because diabetes mellitus (DM) poses harm to the reproductive systems of humans and mice on both the cellular and histological levels, we sought to understand the effects of streptozotocin-induced (STZ) diabetes mellitus on the spermatogenesis and Anti-Müllerian hormone (AMH) production of mice. The structural integrity of the testes is disrupted due to the high levels of oxidative stress caused by DM, which also hinders spermatogenesis. Advanced glycation end products (AGEs) have been found to induce autophagy in Sertoli cells, and the Sertoli cells of diabetic models have fewer tight junctions between them. Nicotine influences dopamine (DA) release in the nucleus accumbens, a brain region associated with reward processing and addiction. This study examined nicotine’s effects on DA signaling using fast-scan cyclic voltammetry (FSCV) in anesthetized rats. Electrodes were implanted to stimulate DA release in the medial forebrain bundle and record signals in the ventral striatum. After administering saline or nicotine, DA levels were monitored in real-time, and electrode placement was later verified through histological analysis. Saline-treated rats showed minor DA fluctuations, with a peak signal detected at 50 minutes, while technical difficulties prevented complete nicotine data collection. Background: The ventral tegmental area (VTA) contains dopamine-producing neurons projecting to various brain regions. VTA dysfunction is linked to addiction and depression. Delta opioid receptor upregulation relates to chronic pain, inflammation, cancer, and early adversity. Kappa receptors in the VTA influence stress, drug-seeking, mood, and reward. Notably, chronic substance abuse upregulates the κ-receptor/dynorphin system, and the opioid system regulates social interaction needs.

Hypothesis: Stress will upregulate opioid receptors.

Methods: We compared group-housed and individually-housed rats using stress tests, analyzing behavior and quantifying VTA opioid receptor expression.

Opioid use disorder occurs alongside impaired risk-related decision-making, but the underlying neural correlates are unclear. We developed an approach-avoidance conflict task using a modified conditioned place preference procedure to study neural signals of risky opioid seeking in the prefrontal cortex, a region implicated in executive decision-making. Following morphine conditioned place preference, rats underwent a conflict test in which fear-inducing cat odor was introduced in the previously drug-paired side of the apparatus. While the saline-exposed control group avoided cat odor, the morphine group included two subsets of rats that either maintained a preference for the paired side despite the presence of …

Perineuronal nets (PNNs) are specialized extracellular matrix structures that surround certain types of neurons in the central nervous system. Research from the past few decades has cemented PNNs as an integral player in the closure and onset of critical periods, memory modulation, and neural plasticity. These effects are thought to occur when PNNs form around a neuron and structurally stabilize its synaptic connections. PNNs have been directly linked to behavioral changes, making them of interest to psychologists as well as neuroscientists.
