Objectives: Vasculopathy emerges early in systemic sclerosis (SSc) and links to endothelial cell (EC) injury and angiogenesis. Understanding EC transcriptomes and epigenomes is crucial for unravelling the mechanisms involved.

Methods: Transcriptomes and chromatin accessibility were assessed by single-cell RNA sequencing and single-nucleus transposase-accessible chromatin sequencing. Immunofluorescent staining of skin and proteomics assay were employed to confirm the altered SSc EC phenotypes. Gain-of-function assay was used to evaluate the effects of ETS transcription factors on human dermal ECs (hDECs).

Results: Both control and SSc ECs shared transcriptomic signatures of vascular linages (arterial, capillary and venous ECs) and lymphatic ECs. Arterial ECs …

Oral potentially malignant disorders (OPMDs) with genomic alterations have a heightened risk of evolving into oral squamous cell carcinoma (OSCC). Currently, genomic data are typically obtained through invasive tissue biopsy. However, brush biopsy is a non-invasive method that has been utilized for identifying dysplastic cells in OPMD but its effectiveness in reflecting the genomic landscape of OPMDs remains uncertain. The non-compounded prescription drug service via telemedicine became an alternative solution to continue providing pharmaceutical services while simultaneously controlling physical contact in hospitals during the COVID-19 pandemic. The high demand for ambulance services at the 119 Public Safety Center of the DKI Jakarta Health Office, including in the Administrative District of the Kepulauan Seribu, highlights the importance of providing water ambulances. This study aims to analyze the implementation of the Water Ambulance Service by the DKI Jakarta Health Office using the Context, Input, Process, Product (CIPP) program evaluation approach. Using a qualitative approach, primary and secondary data were collected through observations, in-depth interviews, and document review. BACKGROUND: SARS CoV-2 virus (COVID-19) impacted the practice of healthcare in the United States, with technology being used to facilitate access to care and reduce iatrogenic spread. Since then, patient message volume to primary care providers has increased. However, the volume and trend of electronic communications after lockdown remain poorly described in the literature.

BACKGROUND: This pilot study evaluates the effectiveness of mobile talk-time incentives in maintaining participation in a longitudinal mobile health (mHealth) data collection program among people living with HIV in Lusaka, Zambia. While mHealth tools, such as mobile phone surveys, provide vital health feedback, optimal incentive strategies to ensure long-term engagement remain limited. This study explores how different incentive levels affect response rates in multiple survey rounds, providing insights into effective methods for encouraging ongoing participation, especially in the context of Zambia's prepaid mobile system and multi-SIM usage, a common practice in sub-Saharan Africa.

Helicobacter pylori (H. pylori) is a bacterium known to be associated with a significant risk of gastric cancer in addition to chronic gastritis, peptic ulcer, and MALT lymphoma. Although only a small percentage of patients infected with H. pylori develop gastric cancer, Gastric cancer causes more than 750,000 deaths worldwide, with 90% of cases being caused by H. pylori. The eradication of this bacterium rests on multiple drug regimens as guided by various consensus. However, the efficacy of empirical therapy is decreasing due to antimicrobial resistance. BACKGROUND: Both clomiphene citrate and its isomer, enclomiphene, have become widespread within urologic practice; thus, understanding these medications' comparative benefits and risks is crucial for optimizing treatment and providing improved therapeutic options. We sought to investigate the longitudinal benefits and risks associated with enclomiphene, compared to clomiphene, and to provide valuable insights for clinicians when making treatment decisions in the management of hypogonadism.

BACKGROUND: Fibroids are non-cancerous uterine growths that can cause symptoms impacting quality of life. The breadth of treatment options allows for patient-centered preference. While conversation aids are known to facilitate shared decision making, the implementation of these aids for uterine fibroids treatments is limited. We aimed to develop two end-user-acceptable uterine fibroids conversation aids for an implementation project. Our second aim was to outline the adaptations that were made to the conversation aids as implementation occurred.

African American Women: Barriers Encountered When Accessing Substance Use Disorder Treatment, Alexis Jones, Lindsay Gietzen, Madhu Sundarrajan

This study identifies barriers that African American women encounter while accessing substance use disorder treatment in San Joaquin County and Sacramento County. There is a gap in research identifying barriers that African American women encounter while accessing substance use disorder treatment. The researcher conducted semi-structured interviews with 5 women. Three themes emerged; perceived racial bias, lack of family support, and believing they did not need treatment. It is recommended that healthcare providers provide more treatment options for African American women and cultural competency training for substance use disorder providers.

The need for behavioral health integration (BHI) in primary care continues to grow to address the behavioral health needs of patients and families. Developing a competent interprofessional workforce to operate in primary care behavioral health settings necessitates BHI-literate clinical learning environments, as well as tools to evaluate and assist in the integration of services appropriate for BHI training and supervision. Health inequities refer to avoidable, unjust differences in health outcomes caused by systemic disadvantages rooted in social, economic, and environmental contexts (Braveman, 2014). Health disparities refer to differences in health outcomes between groups, often measured by prevalence, morbidity, or mortality (Centers for Disease Control and Prevention [CDC], 2020). Health inequalities are measurable differences in health status that may arise from biological, social, or environmental factors (World Health Organization [WHO], 2019). Health inequities, however, are specifically preventable and unjust differences.

Menopause is characterized by the cessation of menstruation for 12 consecutive months. In this narrative review, we describe the transitional stages of menopause, clinical presentation, pharmacological management of symptoms, and effects on fluctuating hormone levels. To standardize the stages of menopause, the Stages of Reproductive Aging Workshop + 10 (STRAW+10) system was designed with five distinct categories corresponding to symptom presentation and numerical years. Common clinical presentations consist of vasomotor hot flashes, mood changes, decreased libido, osteoporosis, and genitourinary changes, all of which are associated with changes in hormone levels. Community science has increased in popularity in communities where residents hope to investigate the relationship between environmental issues and personal health. This study partnered with neighborhoods in the most polluted residential zip code in the US to conduct community science air quality monitoring. We conducted 60 semi-structured interviews after two monitoring deployments to understand participants’ subjective experiences of pollution exposure, their engagement with low-cost air quality monitors, and their data interpretation. BACKGROUND: Fragile X syndrome, with an approximate incidence rate of 1 in 4000 males to 1 in 8000 females, is the most prevalent genetic cause of heritable intellectual disability and the most common monogenic cause of autism spectrum disorder. The full mutation of the Fragile X Messenger Ribonucleoprotein-1 gene, characterized by an expansion of CGG trinucleotide repeats (>200 CGG repeats), leads to fragile X syndrome. Suicide is a prevalent issue that widely affects all demographics in South Korea. Among adolescents, it is the leading cause of mortality with an average rate of 7 deaths per 100,000 people. Some of the most significant factors that influence the risk of suicidal ideation and attempts are cultural stigma and academic stress. Other factors, such as bullying and family structure, also increase the likelihood of suicide, while specific characteristics including internet addiction, socioeconomic status, and substance abuse have been found to be predictors of suicide. It is uncertain whether the prognostic power of white matter hyperintensity (WMH) on post-stroke outcomes is modulated as a function of initial neurological severity, a critical determinant of outcome after stroke. This multi-center MRI study tested if higher WMH quintiles were associated with 3-month poor functional outcome (modified Rankin Scale ≥ 3) for mild versus moderate-to-severe ischemic stroke. Mild and moderate-to-severe stroke were defined as admission National Institute of Health Stroke Scale scores of 1-4 and ≥ 5, respectively. Background: The diverse clinical characteristics of erosive esophagitis (EE) and symptom perception in patients with gastroesophageal reflux disease (GERD) remain a major challenge in understanding their underlying pathogenesis. This study aimed to investigate the association between the levels of IL-8, TNF-α, and IL-17 in serum and the presence of erosive esophagitis and symptoms related to GERD. Chemical modifications to mRNA respond dynamically to environmental cues and are important modulators of gene expression. Nanopore direct RNA sequencing has been applied for assessing the presence of pseudouridine (ψ) modifications through basecalling errors and signal analysis. These approaches strongly depend on the sequence context around the modification, and the occupancies derived from these measurements are not quantitative. In this work, we combine direct RNA sequencing of synthetic RNAs bearing site-specific modifications and supervised machine learning models (ModQuant) to achieve near-analytical, site-specific ψ quantification. Background

Antimicrobial resistance (AMR) poses an important global health challenge in the 21st century. A previous study has quantified the global and regional burden of AMR for 2019, followed with additional publications that provided more detailed estimates for several WHO regions by country. To date, there have been no studies that produce comprehensive estimates of AMR burden across locations that encompass historical trends and future forecasts.
