Concurrent chemoradiotherapy (cCRT) is the mainstay of treatment for patients diagnosed with locally advanced non-small cell lung cancer (NSCLC). One significant challenge in the effectiveness of this therapy is the potential development of resistance mechanisms, where autophagy up-regulation has been proposed as a key contributing factor. However, there is a lack of reliable biomarkers to predict outcomes on these patients. Interestingly, for addressing this gap, extracellular vesicles (EVs) and circulating tumor cells (CTCs) have emerged as potential sources of such biomarkers. In this study, we investigated EV-associated miRNAs and presence of autophagic CTCs in prospectively collected serial samples from 38 …

Alzheimer's disease (AD) is associated with changes in large-scale functional brain network organization. Individuals with AD exhibit less segregated resting-state brain networks compared with individuals without dementia. However, declines in brain network segregation are also evident as adult individuals grow older. Determining whether these observations reflect unique or overlapping alterations on the functional connectome of the brain is essential for understanding the impact of AD on network organization and incorporating measures of functional brain network organization toward AD characterization. Relationships between AD dementia severity and participant's age on resting-state brain system segregation were examined in 326 cognitively healthy and 275 …

SARS-CoV-2 mRNA vaccines are administered as effective prophylactic measures for reducing virus transmission rates and disease severity. To enhance the durability of post-vaccination immunity and combat SARS-CoV-2 variants, boosters have been administered to two-dose vaccinees. However, long-term humoral responses following booster vaccination are not well characterized. A 16-member cohort of healthy SARS-CoV-2 naïve participants were enrolled in this study during a three-dose BNT162b2 vaccine series. Serum samples were collected from vaccinees over 420 days and screened for antigen (Ag)-specific antibody titers, IgG subclass distribution, and neutralizing antibody (nAb) responses. Vaccine boosting restored peak Ag-specific titers with sustained α-RBD IgG and …

BACKGROUND: There are few assessments evaluating associations between birth defects with neural crest cell developmental origins (BDNCOs) and embryonal tumors, which are characterized by undifferentiated cells having a molecular profile similar to neural crest cells. The effect of BDNCOs on embryonal tumors was estimated to explore potential shared etiologic pathways and genetic origins.
METHODS: With the use of a multistate, registry-linkage cohort study, BDNCO-embryonal tumor associations were evaluated by generating hazard ratios (HRs) and 95% confidence intervals (CIs) with Cox regression models. BDNCOs consisted of ear, face, and neck defects, Hirschsprung disease, and a selection of congenital heart defects. Embryonal …

Congenital defects in the pancreas can cause severe health issues such as pancreatic cancer and diabetes which require lifelong treatment. Regenerating healthy pancreatic cells to replace malfunctioning cells has been considered a promising cure for pancreatic diseases including birth defects. However, such therapies are currently unavailable in the clinic. The developmental gene regulatory network underlying pancreatic development must be reactivated for in vivo regeneration and recapitulated in vitro for cell replacement therapy. Thus, understanding the mechanisms driving pancreatic development will pave the way for regenerative therapies. Pancreatic progenitor cells are the precursors of all pancreatic cells which use epigenetic changes …

A Paracrine Circuit Of Il-1Β/Il-1r1 Between Myeloid And Tumor Cells Drives Genotype-Dependent Glioblastoma Progression, Zhihong Chen, David H. Gutmann, Et Al.
2020-Current year OA Pubs
Monocytes and monocyte-derived macrophages (MDMs) from blood circulation infiltrate glioblastoma (GBM) and promote growth. Here, we show that PDGFB-driven GBM cells induce the expression of the potent proinflammatory cytokine IL-1β in MDM, which engages IL-1R1 in tumor cells, activates the NF-κB pathway, and subsequently leads to induction of monocyte chemoattractant proteins (MCPs). Thus, a feedforward paracrine circuit of IL-1β/IL-1R1 between tumors and MDM creates an interdependence driving PDGFB-driven GBM progression. Genetic loss or locally antagonizing IL-1β/IL-1R1 leads to reduced MDM infiltration, diminished tumor growth, and reduced exhausted CD8+ T cells and thereby extends the survival of tumor-bearing mice. In contrast …

UNLABELLED: The malignant Hodgkin and Reed Sternberg (HRS) cells of classical Hodgkin lymphoma (cHL) are scarce in affected lymph nodes, creating a challenge to detect driver somatic mutations. As an alternative to cell purification techniques, we hypothesized that ultra-deep exome sequencing would allow genomic study of HRS cells, thereby streamlining analysis and avoiding technical pitfalls. To test this, 31 cHL tumor/normal pairs were exome sequenced to approximately 1,000× median depth of coverage. An orthogonal error-corrected sequencing approach verified >95% of the discovered mutations. We identified mutations in genes novel to cHL including: CDH5 and PCDH7, novel stop gain mutations in …

Differential Contribution Of Cardiac And Peripheral Blood Flow To Metaboreceptor-Mediated Pressor Responses In Autism Spectrum Disorder, Matthew Pizano Gomez, Gabriel B. Carlota, Stephanie S. Wang, Areum K. Jensen
International Journal of Exercise Science: Conference Proceedings
Young adults with Autism Spectrum Disorder (ASD) experience a higher prevalence of hypertension than the general population. Abnormal blood pressure (BP) response to acute exercise is seen in hypertension. BP response evoked during exercise is the outcome of a central interaction between neural inputs such as the exercise pressor reflex. However, it is unclear if this neurocardiovascular interaction is altered differently to regulate BP during exercise in ASD. PURPOSE: To determine whether contributions of cardiac and peripheral blood flow to metaboreceptor-mediated pressor responses are different during exercise in ASD. METHODS: 21 young adults with and without ASD completed the study. …

Pickleball is an emerging sport that is continuing to gain popularity among people of all ages. Little research has been completed on the performance aspects of the game. PURPOSE: Lower body force production is an important aspect of any sport. A countermovement jump is a useful way to test that characteristic. Acute decreases in force production are an indicator of fatigue. No previous research of force production and fatigue has been completed on pickleball. Therefore the purpose of this study was to test countermovement jump performance before and after a match of doubles pickleball to determine if neuromuscular fatigue …

Walking With Blood Flow Restriction Elicits Postexercise Hypotension In Prehypertensive Individuals, Dae Sik Song, Geoffrey Dorsett, Justine Wills, Iridian Valdivia, Isaac Sumaran, Jong-Kyung Kim
International Journal of Exercise Science: Conference Proceedings
It is well documented that prehypertension is associated with increased risk of developing into the future hypertension and cardiovascular events. Exercise is well known as one of the best nonpharmacological treatments for prehypertension to prevent future hypertension development. Blood flow restriction (BFR) training have been known to improve health-related components of physical fitness such as body composition, muscular strength and endurance, and aerobic capacity. However, there is still paucity of research that BFR exercise induces postexercise hypotension. PURPOSE: accordingly, the goal of this study was to assess that BFR exercise is more effective in reducing blood pressure (BP) compared to …

A Novel Waveform To Extract Exercise Gas Exchange Response Dynamics: The Chirp Waveform, Michele Girardi, Michael A. Roman, Janos Porszasz, William W. Stringer, Carrie Ferguson, Harry B. Rossiter, Richard Casaburi
International Journal of Exercise Science: Conference Proceedings
Characterizing exercise gas exchange response dynamics reveals important information about physiological control processes and cardiopulmonary dysfunction. However, current methods for extracting exercise response dynamics typically use multiple step-wise transitions, limiting applicability of this technique. PURPOSE: We designed a new protocol (chirp waveform) to extract exercise gas exchange response dynamics in a single visit. We tested the hypothesis that gas exchange response dynamics extracted from chirp forcing would be similar to those extracted from step-wise transitions. METHODS: Thirty-one participants (14 young healthy, 7 older healthy, and 10 patients with chronic obstructive pulmonary disease) visited the laboratory on three occasions. …
Low Back Demand Of Equipment Carriage Tasks In Golf, Guanrong Cai, Jacob Cho, Yunsheng Zou, Jordan Cannon, George Salem
Low Back Demand Of Equipment Carriage Tasks In Golf, Guanrong Cai, Jacob Cho, Yunsheng Zou, Jordan Cannon, George Salem
International Journal of Exercise Science: Conference Proceedings
Low back injury is a common concern in golfers and caddies. The literature often points to the biomechanics of the golf swing as the major contributing factor; however, golfers and caddies can regularly walk upward of 10,000 steps on uneven terrain while carrying equipment, which can contribute to the cumulative load on the low back. PURPOSE: To examine the low back biomechanical demands during golf equipment carriage and pick-up tasks. METHODS: Sixteen golfers (11M/5F, 27.4±3.5yrs) participated in 3D motion capture of a golf bag pick-up task and 3 walking tasks (1. unloaded walking [UW], 2. unilateral carrying on …

The Effect Of Aicar-Induced Ampk Activation On Gene Expression In Sarcopenic Muscle, Daniel M. Rasch, Steven D. Dray, Jouber Dos Santos, Shalene E. Wilcox, David M. Thomson
International Journal of Exercise Science: Conference Proceedings
The loss of muscle mass and function (sarcopenia) afflicts 11-74% of all aging adults, with prevalence increasing with age. Exercise training is clearly effective in preventing or attenuating sarcopenia. The cellular mechanisms of exercise’s protective effects are not entirely clear, but AMP-activated protein kinase (AMPK) is thought to play an important role, in part by regulating gene expression. PURPOSE: To determine the effect of chronic pharmacological AMPK activation on skeletal muscle gene expression in sarcopenic muscle. METHODS: 24-month-old C57Bl/6J mice received either one acute injection or chronic daily saline injections of the AMPK-activating drug AICAR for 31 days. …

International Journal of Exercise Science: Conference Proceedings
Evidence suggested heart rate recovery (HRR) is a useful maker to evaluate cardiac sympathetic function. Prehypertension is characterized by overactive sympathetic output and dysfunction of cardiac parasympathetic activity and associated with delayed HRR. Even though dietary supplementation with grape seed extract (GSE) elicits the production of nitric oxide, limited evidence has reported regarding the potential beneficial effects of this extract on HRR. Purpose: The aim of this study was to compare effects of 7 days of GSE supplementation and placebo (PL) on HRR for 5 min after graded exercise. Methods: Subjects were randomly assigned via a double-blind, cross-over design to …

International Journal of Exercise Science: Conference Proceedings
Battle rope exercises are commonly used among cross-fitters to build overall muscular strength and cardiorespiratory endurance. Although this training method is physically demanding, no studies have examined the frontal plane movements in the lower extremities when performing battle ropes exercises in their different variations. PURPOSE: The objective of this research was to compare the frontal plane range of motion (ROM) along with the peak joint angles of the ankle, knee, and hip when performing bilateral and unilateral battle ropes exercises. METHODS: Participants with at least six months of resistance training experience performed four sets of bilateral and unilateral battle ropes …

Low energy availability (LEA) is the underlying factor for Relative Energy Deficiency in Sport which includes a facet of bone health. Stress fractures (SF) are common injuries among runners. The prevalence of coexisting LEA and multiple SF in recreational runners is unknown. PURPOSE: The purpose is 1) to assess the prevalence of multiple SF and risk of LEA among recreational female runners 2) to compare current eating disorders, body weight dissatisfaction, and restrictive eating behaviors between females with a history of SF and those with ≤ 1 SF. METHODS: Female noncompetitive, recreational runners (running > 10 miles/week, 18-25 years …

International Journal of Exercise Science: Conference Proceedings
The battle rope exercise recently has increased its popularity due to cross fit gyms implementing it into the workout programs. Individuals use this workout to strengthen their upper body, however it is unknown how different modalities of using the battle ropes affects the lower extremity joint force required to stabilize the body. PURPOSE: To examine the effects of unilateral and bilateral battle ropes exercises on lower extremity joint force. METHODS: Subjects performed four bilateral battle rope trials and four unilateral battle rope trials. The trials consisted of twenty seconds of continuous activity with three minutes of rest in between …
The Effects Of Fatigue On Center Of Pressure Measures During Front Lunges, Logan C. Randall, Michele Leblanc
The Effects Of Fatigue On Center Of Pressure Measures During Front Lunges, Logan C. Randall, Michele Leblanc
International Journal of Exercise Science: Conference Proceedings
Front lunges are a common exercise in strength and conditioning programs. Extensive research on front lunge biomechanical movement exists previously, however, the existing research lacks information regarding fatigue or center of pressure (COP). PURPOSE: The aim of this study was to determine the effect of fatigue on COP behavior in front lunges. METHODS: Participants were ages 18 to 30 and recreationally active. Participants were guided through sufficient warm up and practiced the lunge movement, barefoot, at the cadence of 15 repetitions/minute. Step length was assigned as the length from their greater trochanter to lateral malleolus. Participants completed front lunges …

International Journal of Exercise Science: Conference Proceedings
bioDensity training is a novel type of resistance exercise, which includes four basic isometric exercises in a subsequent fashion: chest press, leg press, core pull, and vertical lift. Although it’s primarily targeted on bone strength, a previous study showed that it improved HbA1C, fasting glucose, HDL, LDL and total cholesterol in elderly patients with Type 2 diabetes (Zheng et al. 2019). To date, the research on bioDensity training is limited, and its effects on a healthy population are unknown. PURPOSE: We are looking to examine the effects of 12-week bioDensity training on lipid profiles in young women. METHODS: …
