Protein Cofactors And Substrate Influence Mg2+-Dependent Structural Changes In The Catalytic Rna Of Archaeal Rnase P, Ila A Marathe, Stella M Lai, Walter J Zahurancik, Michael G Poirier, Vicki H Wysocki, Venkat Gopalan

Faculty, Staff and Student Publications

The ribonucleoprotein (RNP) form of archaeal RNase P comprises one catalytic RNA and five protein cofactors. To catalyze Mg2+-dependent cleavage of the 5' leader from pre-tRNAs, the catalytic (C) and specificity (S) domains of the RNase P RNA (RPR) cooperate to recognize different parts of the pre-tRNA. While ∼250-500 mM Mg2+ renders the archaeal RPR active without RNase P proteins (RPPs), addition of all RPPs lowers the Mg2+ requirement to ∼10-20 mM and improves the rate and fidelity of cleavage. To understand the Mg2+- and RPP-dependent structural changes that increase activity, we used pre-tRNA cleavage and ensemble FRET assays to …

Passing As White: The Experiences Of Bipoc Supervisees, Bridget Anton

Seton Hall University Dissertations and Theses (ETDs)

Cross-racial supervisory dyads are becoming more prevalent due to the increased diversity of graduate students in psychology. However, research has not adequately examined the narratives of BIPOC supervisees who pass as White. People who pass as White might not be seen as a person of color and their identities might be ignored because it is considered invisible. Due to the lack of research on BIPOC supervisees with a passing identity experience within the context of the supervisory dyad, this study explored the subjective experiences of BIPOC supervisees who pass as White in cross-racial supervisor dyads. Furthermore, an exploration into conversations …

Immune checkpoint blockade, an immunotherapy, has been applied in multiple systemic malignancies and has improved overall survival to a relatively great extent; whether it can be applied in breast cancer remains unknown. We endeavored to explore possible factors that may influence immunotherapy outcomes in breast cancer using several public databases. The possible treatment target TNF superfamily member 4 (TNFSF4) was selected from many candidates based on its abnormal expression profile, survival-associated status, and ability to predict immune system reactions. For the first time, we identified the oncogenic features of TNFSF4 in breast carcinoma. TNFSF4 was revealed to be closely related …

Effects Of Ecg Data Length On Heart Rate Variability Among Young Healthy Adults, En-Fan Chou, Michelle Khine, Thurmon Lockhart, Rahul Soangra

Physical Therapy Faculty Articles and Research

The relationship between the robustness of HRV derived by linear and nonlinear methods to the required minimum data lengths has yet to be well understood. The normal electrocardiography (ECG) data of 14 healthy volunteers were applied to 34 HRV measures using various data lengths, and compared with the most prolonged (2000 R peaks or 750 s) by using the Mann–Whitney U test, to determine the 0.05 level of significance. We found that SDNN, RMSSD, pNN50, normalized LF, the ratio of LF and HF, and SD1 of the Poincaré plot could be adequately computed by small data size (60–100 R peaks). …

Dark Side Of Cancer Therapy: Cancer Treatment-Induced Cardiopulmonary Inflammation, Fibrosis, And Immune Modulation, Boopathi Ettickan, Chellappagounder Thangavel

Department of Dermatology and Cutaneous Biology Faculty Papers

Advancements in cancer therapy increased the cancer free survival rates and reduced the malignant related deaths. Therapeutic options for patients with thoracic cancers include surgical intervention and the application of chemotherapy with ionizing radiation. Despite these advances, cancer therapy-related cardiopulmonary dysfunction (CTRCPD) is one of the most undesirable side effects of cancer therapy and leads to limitations to cancer treatment. Chemoradiation therapy or immunotherapy promote acute and chronic cardiopulmonary damage by inducing reactive oxygen species, DNA damage, inflammation, fibrosis, deregulation of cellular immunity, cardiopulmonary failure, and non-malignant related deaths among cancer-free patients who received cancer therapy. CTRCPD is a complex …

Department of Neurology Faculty Papers

We performed a genome-wide association study (GWAS) to identify genetic variation associated with common forms of idiopathic generalized epilepsy (GE) and focal epilepsy (FE). Using a cohort of 2220 patients and 14,448 controls, we searched for single nucleotide polymorphisms (SNPs) associated with GE, FE and both forms combined. We did not find any SNPs that reached genome-wide statistical significance (p ≤ 5 × 10−8) when comparing all cases to all controls, and few SNPs of interest comparing FE cases to controls. However, we document multiple linked SNPs in the PADI6-PADI4 genes that reach genome-wide significance and are associated …

Orthodontics and Endodontics Theses

Background and purpose: In recent years, questions regarding the genetic and environmental factors affecting variation in human craniofacial morphology have received increasing attention. Medical and clinical genetic research using family studies has proven foundational for our understanding of which genes affect craniofacial variation. On the other hand, cephalometricbased studies showed a relationship between the cranial base and midface characteristics among individuals with different skeletal pattern of malocclusion. The purpose of our pilot study was to analyze specific variants of facial genes that were suggested to be associated with cranial base width and depth in Class II and Class I malocclusions. …

Barriers To, And Facilitators Of, Patient Self-Management Of Chronic Conditions: An Integrative Literature Review, Shari Frueh

Nursing Masters Papers

The prevalence of adults with chronic medical conditions in the United States is growing. Adequate self-management of chronic conditions enables patients to manage unpleasant symptoms, improve quality of life, avoid life-threatening situations, and slow disease progression. Patients require support from healthcare providers to learn to optimally manage their chronic condition(s). The increasing volume of patients requiring assistance necessitates redesigned approaches to self-management support to meet the demand. The purpose of this inquiry is to gain a deeper understanding of the barriers to, and facilitators of, self-management in adults and to provide recommendations for nursing interventions and healthcare provider approaches that …

Among novel compounds under recent investigation as potential new antimalarial drugs are three independently developed inhibitors of the Plasmodium falciparum P-type ATPase (PfATP4): KAE609 (cipargamin), PA92, and SJ733. We assessed ex vivo susceptibilities to these compounds of 374 fresh P. falciparum isolates collected in Tororo and Busia districts, Uganda, from 2016 to 2019. Median IC50s were 65 nM for SJ733, 9.1 nM for PA92, and 0.5 nM for KAE609. Sequencing of pfatp4 for 218 of these isolates demonstrated many nonsynonymous single nucleotide polymorphisms; the most frequent mutations were G1128R (69% of isolates mixed or mutant), Q1081K/R (68%), G223S (25%), N1045K …

Where Epigenetics Meets Food Intake: Their Interaction In The Development/Severity Of Gout And Therapeutic Perspectives, Philippe T. Georgel, Philippe Georgel

Biological Sciences Faculty Research

Gout is the most frequent form of inflammatory arthritis in the world. Its prevalence is particularly elevated in specific geographical areas such as in the Oceania/Pacific region and is rising in the US, Europe, and Asia. Gout is a severe and painful disease, in which co-morbidities are responsible for a significant reduction in life expectancy. However, gout patients remain ostracized because the disease is still considered “self-inflicted”, as a result of unhealthy lifestyle and excessive food and alcohol intake. While the etiology of gout flares is clearly associated with the presence of monosodium urate (MSU) crystal deposits, several major questions …
