Walden Dissertations and Doctoral Studies

Lesbian, gay, bisexual and transgender (LGBT) members live in the community, yet they are prone to various types of oppression, mistreatments, and denial of services from most institutions including health care settings. Concerns, such as being denied access to caregivers or being given a lower quality of care continue despite the community’s fighting for their voices to be heard across the globe and various movements that have been established, especially the United States and the United Kingdom, and laws that call for equitable healthcare. The purpose of this project was to explore whether a change in knowledge and attitudes would …

Physiological Measurements For Real-Time Fatigue Monitoring In Train Drivers: Review Of The State Of The Art And Reframing The Problem, Bojana Bjegojevic, Maria Chiara Leva, Nora Balfe, Sam D. Cromie, Luca Longo Jan 2021

Physiological Measurements For Real-Time Fatigue Monitoring In Train Drivers: Review Of The State Of The Art And Reframing The Problem, Bojana Bjegojevic, Maria Chiara Leva, Nora Balfe, Sam D. Cromie, Luca Longo

Conference papers

The impact of fatigue on train drivers is one of the most important safety-critical issues in rail. It affects drivers’ performance, significantly contributing to railway incidents and accidents. To address the issue of real-time fatigue detection in drivers, most reliable and applicable psychophysiological indicators of fatigue need to be identified. Hence, this paper aims to examine and present the current state of the art in physiological measures for real-time fatigue monitoring that could be applied in the train driving context. Three groups of such measures are identified: EEG, eye-tracking and heart-rate measures. This is the first paper to provide the …

Endolysins of a number of bacteriophages, including coliphages T5, RB43, and RB49, target the peptidoglycans of the bacterial cell wall. The backbone of these bacterial peptidoglycans consist of alternating N-acetylglucosamine and N-acetylmuramic acid residues that is further “reinforced” by the peptide subunits. Because of the mesh-like structure and insolubility of peptidoglycans, the processes of the peptidoglycan binding and hydrolysis by enzymes cannot be studied by spectral methods. To overcome these issues we synthesized and analyzed here one of the simplest water soluble peptidoglycan mimetics.

A compound has been synthesized that mimics the peptidoglycan fragment of the bacterial cell wall, N-acetylglucosaminyl-β(1-4)-N-acetylmuramoyl-l-alanyl-γ-d-glutamyl-l-alanyl-d-alanine. …

In recent years, there has been tremendous enthusiasm with respect to detailing the genetic basis of many neoplasms, including conjunctival melanoma (CM). We aim to analyze five proteins associated with CM, namely BRAF, NRAS, c-KIT, NF1, and PTEN. We evaluated each protein for its intrinsically disordered protein regions (IDPRs) and its protein-protein interactions (PPI) with the Predictor of Natural Disordered Protein Regions (PONDR®) and the Search Tool for the Retrieval of Interacting Genes (STRING®). Our PONDR® analysis found high levels of IDPRs in all five proteins with mutations linked to CM. The highest levels of IDPRs were in BRAF (45.95%), …

The coil to globule transition of the polypeptide chain is the physical phenomenon behind the folding of globular proteins. Globular proteins with a single domain usually consist of about 30 to 100 amino acid residues, and this finite size extends the transition interval of the coil-globule phase transition. Based on the pedantic derivation of the two-state model, we introduce the number of amino acid residues of a polypeptide chain as a parameter in the expressions for two cooperativity measures and reveal their physical significance. We conclude that the 𝑘2 measure, defined as the ratio of van ’t Hoff and calorimetric …

For proteins, the sequence → structure → function paradigm applies primarily to enzymes, transmembrane proteins, and signaling domains. This paradigm is not universal, but rather, in addition to structured proteins, intrinsically disordered proteins and regions (IDPs and IDRs) also carry out crucial biological functions. For these proteins, the sequence → IDP/IDR ensemble → function paradigm applies primarily to signaling and regulatory proteins and regions. Often, in order to carry out function, IDPs or IDRs cooperatively interact, either intra- or inter-molecularly, with structured proteins or other IDPs or intermolecularly with nucleic acids. In this IDP/IDR thematic collection published in Cell Communication …

Nanoscale miRNAs regulate the synthesis of most human proteins involved in differentiation, proliferation, cell cycle, apoptosis, and other processes associated with the growth and the development of an organism. miRNAs also play a number of important roles in the development of gastric cancer. In this work, we studied the quantitative characteristics of miRNA interactions with 69 candidate gastric cancer genes using bioinformatics approaches. To this end, the MirTarget program was used, which determines the characteristics of miRNA binding to mRNA in the 5′UTR, CDS, and 3′UTR. Associations of miRNAs with alternative target genes and associations of genes with alternative miRNAs …

Many viruses that cause serious diseases in humans and animals, including the betacoronaviruses (beta-CoVs), such as SARS-CoV, MERS-CoV, and the recently identified SARS-CoV-2, have natural reservoirs in bats. Because these viruses rely entirely on the host cellular machinery for survival, their evolution is likely to be guided by the link between the codon usage of the virus and that of its host. As a result, specific cellular microenvironments of the diverse hosts and/or host tissues imprint peculiar molecular signatures in virus genomes. Our study is aimed at deciphering some of these signatures. Using a variety of genetic methods we demonstrated …

Chandipura virus (CHPV, a member of the Rhabdoviridae family) is an emerging pathogen that causes rapidly progressing influenza-like illness and acute encephalitis often leading to coma and death of the human host. Given several CHPV outbreaks in Indian sub-continent, recurring sporadic cases, neurological manifestation, and high mortality rate of this infection, CHPV is gaining global attention. The ‘dark proteome’ includes the whole proteome with special emphasis on intrinsically disordered proteins (IDP) and IDP regions (IDPR), which are proteins or protein regions that lack unique (or ordered) three-dimensional structures within the cellular milieu. These proteins/regions, however, play a number of vital …

Two adenovirus-based vaccines, ChAdOx1 nCoV-19 and Ad26.COV2.S, and two mRNA-based vaccines, BNT162b2 and mRNA.1273, have been approved by the European Medicines Agency (EMA), and are invaluable in preventing and reducing the incidence of coronavirus disease-2019 (COVID-19). Recent reports have pointed to thrombosis with associated thrombocytopenia as an adverse effect occurring at a low frequency in some individuals after vaccination. The causes of such events may be related to SARS-CoV-2 spike protein interactions with different C-type lectin receptors, heparan sulfate proteoglycans (HSPGs) and the CD147 receptor, or to different soluble splice variants of the spike protein, adenovirus vector interactions with the …

Signal sequences are short peptides at the N terminus of proteins destined for the secretion pathway. Typically, after cleavage by peptidases, signal peptides are degraded by intra-membrane proteases. In some cases, however, signal peptides can be processed further and released into the endoplasmic reticulum, secretion pathways, or cytoplasm. The consequences of these processes remain unclear, in particular considering that dysregulated signal peptides could potentially aggregate and induce cytotoxicity. To investigate this problem, we study the signal peptide of the amyloid precursor protein (APP), which originates the Alzheimer’s β-amyloid (Aβ) peptide. Our results show that this signal peptide (residues 1–17 of …

Discovery Of Genetic Pleiotropy For Obesity And Inflammation And Mediation In Identified Variants Through Obesity-Related Pathways, Yiqian Wenren Jan 2021

Discovery Of Genetic Pleiotropy For Obesity And Inflammation And Mediation In Identified Variants Through Obesity-Related Pathways, Yiqian Wenren

Public Health Theses

Genetic data suggest that obesity and inflammation shared a portion of their genetic architectures. Meanwhile, epidemiologic data suggest a causal link between obesity and inflammation. Therefore, we hypothesized that there are variants in the human genome that display pleiotropy for obesity and inflammation and that the effects of at least some of those variants could be mediated by obesity-related pathways. To test these hypotheses, we conducted univariate genome-wide association analyses of obesity-related traits (BMI and WHRadjBMI) and inflammation (CRP) in the UK Biobank (n= 291,396 Caucasian subjects), searched for overlapping signals, fine-mapped the regions to identify putative causal SNPs, and …

Evidence indicates family caregivers of individuals living with dementia (ILwD) are at risk for diminished physical and mental health; which may decrease their quality of life and directly impact their ability to provide care. An interdisciplinary approach to self-care and skill-building for caregivers is provided in a virtual support group offered by Council on Aging in Sonoma County, CA. As part of the nonprofit’s Adult Day Program, the group is offered to client caregivers and has two goals: First, creating a communitybased, long-term support system for ILwD who are agingin-place; second, fostering a safe and supportive community for family caregivers, …

Nonprofits And Government Agencies Addressing The Needs Of Mental Health Community, Wyatt Ulrich Jan 2021

Nonprofits And Government Agencies Addressing The Needs Of Mental Health Community, Wyatt Ulrich

School of Professional and Continuing Studies Nonprofit Studies Capstone Projects

Nonprofit organizations have been utilizing cross-sector collaboration to address problems that are not able to be solved on their own. A partner to make said collaboration can be with any organization, business, or volunteer who has the same goals as the nonprofit organization. Mental health nonprofit organizations specifically need more partners due to how little the world knows about mental health and mental illnesses. The government is one partner many mental health organizations aim to collaborate with. Government contracts have been known to help with the many challenges nonprofit organizations face like funding and services. Previous research has suggested this …

A Study Of Factors That Influence Symbol Selection On Augmentative And Alternative Communication Devices For Individuals With Autism Spectrum Disorder, William Todd Dauterman

CCAC Theses and Dissertations

According to the American Academy of Pediatrics (AAP), 1 in 59 children are diagnosed with Autism Spectrum Disorder (ASD) each year. Given the complexity of ASD and how it is manifested in individuals, the execution of proper interventions is difficult. One major area of concern is how individuals with ASD who have limited communication skills are taught to communicate using Augmentative and Alternative Communication devices (AAC). AACs are portable electronic devices that facilitate communication by using audibles, signs, gestures, and picture symbols. Traditionally, Speech-Language Pathologists (SLPs) are the primary facilitators of AAC devices and help establish the language individuals with …

Neuroimaging On Brain Aging And Neurodegeneration, Katelynn Nguyen

Scripps Senior Theses

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder that usually affects but not limited to the elder population. We see that when an individual develops AD, the brain cells are degenerating and dying at rates that are uncontrollable. Worldwide AD has affected at least 50 million people and we will continue to see this number increase. Although the research done on AD has made great strides, much is still unknown and being studied. Previous studies have allowed us to understand that many of the impacts of AD are correlated to various regions of the brain experiencing atrophy. This causes an …

Death, Discipline, And The Dead: Biopolitical Rhetoric In Early Modern English Texts, Leslie Raybuck Malland Jan 2021

Death, Discipline, And The Dead: Biopolitical Rhetoric In Early Modern English Texts, Leslie Raybuck Malland

Theses and Dissertations--English

Death, Discipline, and the Dead: Biopolitical Rhetoric in Early Modern English Texts locates allusions to the biopolitical culture of Early Modern England within popular English texts. Through my examination of the period’s fascination with death—public executions, newly-authorized anatomies—and the ways in which death, as well as the treatment of the dead, was authorized by and supported the ideological aims of the state, my research identifies how those themes carry over into the most popular works of the day, reviewing instances of both verbal and nonverbal rhetoric across genres to find allusions to biopower — or, state control of the biological. …

The Role Of Community Belongingness In The Mental Health And Well-Being Of Black Lgbtq Adults, Keith Justin Watts

Theses and Dissertations

The impact of racial and sexual minority stigma, prejudice, and discrimination on the mental health and well-being of Black and LGBTQ individuals, respectively, has been well documented in the literature. Research on these relationships for Black LGBTQ individuals who are multiply marginalized due to their position at the social intersections of gender identity, sexual orientation, and race/ethnicity is less common. Belongingness to identity-based communities can protect against the negative impact of these minority stressors for Black and LGBTQ individuals and aid coping processes. However, Black LGBTQ individuals often experience stigma and discrimination in their racial, sexual, and gender minority communities …
