Dartmouth Scholarship
During external beam radiotherapy (EBRT), in vivo Cherenkov optical emissions can be used as a dosimetry tool or to excite luminescence, termed Cherenkov-excited luminescence (CEL) with microsecond-level time-gated cameras. The goal of this work was to develop a complete theoretical foundation for the detectable signal strength, in order to provide guidance on optimization of the limits of detection and how to optimize near real time imaging. The key parameters affecting photon production, propagation and detection were considered and experimental validation with both tissue phantoms and a murine model are shown. Both the theoretical analysis and experimental data indicate that the …

Engineering Faculty Articles and Research
We describe a virtual reality environment, Bob’s Fish Shop, which provides a system where users diagnosed with Autism Spectrum Disorder (ASD) can practice social interactions in a safe and controlled environment. A case study is presented which suggests such an environment can provide the opportunity for users to build the skills necessary to carry out a conversation without the fear of negative social consequences present in the physical world. Through the repetition and analysis of these virtual interactions, users can improve social and conversational understanding.

Targeting Primary Cilia Immune Receptor Proteins For The Treatment Of Polycystic Kidney Disease Mechanisms, Nedaa Alomari
Targeting Primary Cilia Immune Receptor Proteins For The Treatment Of Polycystic Kidney Disease Mechanisms, Nedaa Alomari
Pharmaceutical Sciences (MS) Theses
Background: Primary cilia are cellular organelles project from the cell surface of mammalian cell and play important roles in vertebrate development, organogenesis, health, and others genetic diseases. Primary cilium functions as a mechano-sensor and chemo-sensor. Defect in primary cilia causes the progression of polycystic kidney disease (PKD) which further leads to the inflammatory responses. We, therefore, investigated the role of Toll-like receptors 4 and 9 (TLR) in primary cilia towards PKD.
Purpose: The main purpose of the proposed study is to identify and target the immune reactive proteins i.e. TLRs in the primary cilia. By targeting those primary cilia immune …

Post menopause is considered a critical period for bone-mass loss. Impaired bone metabolism during this phase can increase the risk of fractures in old age. Inflammation is a risk factor for bone health, and diet is a potential source of inflammation. However, few studies have examined the association between the dietary inflammatory index (DII®) and bone-mineral density (BMD) in postmenopausal women in Korea. The objective of this study was to determine, by means of a Korean cross-sectional investigation, whether higher DII scores are associated with decreased BMD in postmenopausal women. To that end, the raw data from the …

The Correlation Between The Microbiome And Neurodegenerative Disorders Such As Alzheimer Disease, Aurora Mokris
Honors Theses
Recent studies suggest that there are 1.3 times as many microbial cells as human cells in the body (Abbot, 2016). Another study claims that the combined genomes of this microbiota is 150 times that of the human genome (Zhuang & Shen, 2018). If you have ever had the feeling that you are not alone in an empty room you are most certainly correct. Your body is teeming with millions of organisms living in mutualistic symbiosis with you and surrounding microbes, but at times, this seemingly harmonious relationship can be disturbed, resulting in serious physical and psychological changes to the human …

Heat shock protein 90 (Hsp90) is a eukaryotic chaperone responsible for the folding and functional activation of numerous client proteins, many of which are oncoproteins. Thus, Hsp90 inhibition has been intensely pursued, resulting in the development of many potential Hsp90 inhibitors, not all of which are well-characterized. Hsp90 inhibitors not only abrogate its chaperone functions, but also could help us gain insight into the structure-function relationship of this chaperone. Here, using biochemical and cell-based assays along with isothermal titration calorimetry, we investigate KU-32, a derivative of the Hsp90 inhibitor novobiocin (NB), for its ability to modulate Hsp90 chaperone function. Although …

Research outputs 2014 to 2021
The use of antenatal colostrum expression in the weeks prior to birth may help improve long-term breastfeeding, but few large-scale studies exist. Typically, antenatal colostrum expression instruction relies on face-to-face education, making large interventions costly. We aimed to determine whether an expert online instructional video can improve knowledge and confidence around antenatal colostrum expressing. Pregnant women were asked to complete a questionnaire pre- and post-watching the instructional video online. Ninety five pregnant women completed both pre- and post-questionnaires. Total antenatal colostrum expression knowledge scores improved after watching the video, from a mean of 3.05 ± 1.70 correct out of a …

Department of Biochemistry and Molecular Biology Posters
Many people pursue direct-to-consumer (DTC) DNA testing to learn about health risks. While DTC testing for breast cancer genes BRCA1 and BRCA2 may provide vital information to those testing positive, it is unclear whether individuals testing negative understand the implications of these results. Furthermore, it is unknown how such findings may influence future cancer risk management. Ninety-six individuals who underwent BRCA testing through the DTC service, 23andMe, completed a survey designed to assess their understanding of a negative result and how this result may impact cancer risk management. Respondents were recruited via social media sources Reddit and Facebook. All participants …

Manuscripts, Articles, Book Chapters and Other Papers
Multiple reports of uncoupling protein 1 (UCP1) expression have established its presence in human epicardial adipose tissue (eAT). Its functional relevance to eAT, however, remains largely unknown. In a recent study, we reported that adrenergic stimulation of eAT was associated with downregulation of secreted proteins involved in oxidative stress-related and immune-related pathways. Here, we explored the UCP1-associated features of human eAT using next-generation deep sequencing. Paired biopsies of eAT, mediastinal adipose tissue (mAT), and subcutaneous adipose tissue (sAT) obtained from cardiac surgery patients, with specific criteria of high and low expression of UCP1 in eAT, were subjected to RNA sequencing. …

The analysis of molecular states of individual cells, as defined by their mRNA expression profiles and protein composition, has gained widespread interest in studying biological phenomena ranging from embryonic development to homeostatic tissue function and genesis and evolution of cancers. Although the molecular content of individual cells in a tissue can vary widely, their molecular states tend to be constrained within a transcriptional landscape partly described by the canonical archetypes of a population of cells. In this study, we sought to characterize the effects of an acute (partial hepatectomy) and chronic (alcohol consumption) perturbation on the molecular states of individual …

Speech-Language Pathologist Preferences For Treatment Types For Childhood Apraxia Of Speech, Meeghan Hoose
Honors Theses
Childhood Apraxia of Speech (CAS), also known as developmental apraxia is, “a motor speech disorder that makes it hard for children to speak” (American Speech-Language-Hearing Association). While the child knows what he/she wants to say, the signals going from the brain to the mouth are not delivered correctly, resulting in the mouth not moving appropriately to produce the desired sounds. There are several methods used by speech-language pathologists to treat CAS including moto-programming, a linguistic approach, a combination of motor-programming and linguistic methods, sensory cuing and rhythmic approaches. Through surveying local practicing school speech-language pathologists, I will learn their preferred …
Exploring A Child’S History For Optimal Treatment Of Food Selectivity., Briana Lear
Exploring A Child’S History For Optimal Treatment Of Food Selectivity., Briana Lear
Honors Theses
Food selectivity is defined as “food refusal based on type, texture, temperature, or appearance of food, leading to a self-restricted diet” (Suarez, 2015). Food selectivity affects children of all ages. Food selectivity refers to when a child is eating as few as 5-10 different foods and demonstrates food refusal behaviors, which can make mealtime a challenge for children and their families (Suarez, 2015). Occupational therapy addresses activities of daily living (ADLS), therefore feeding, including food selectivity is included within the OT scope of practice. There is a need to create a comprehensive and thorough document to collect a client’s history …

Circular RNAs (circRNAs) are non-coding RNAs that form covalently closed continuous loops, and act as gene regulators in physiological and disease conditions. To test our hypothesis that proatherogenic lipid lysophosphatidylcholine (LPC) induce a set of circRNAs in human aortic endothelial cell (HAEC) activation, we performed circRNA analysis by searching our RNA-Seq data from LPC-activated HAECs, and found: (1) LPC induces significant modulation of 77 newly characterized cirRNAs, among which 47 circRNAs (61%) are upregulated; (2) 34 (72%) out of 47 upregulated circRNAs are upregulated when the corresponding mRNAs are downregulated, suggesting that the majority of circRNAs are upregulated presumably via …

Different Effects Of Static And Vibrating Foam Rollers On Ankle Plantar Flexion Flexibility And Neuromuscular Activation, Brianna G. Mazzei
Honors College Theses
The relatively new implementation of vibration into foam rollers was developed in response to the positive feedback of whole-body vibration treatment and foam rolling therapy. The purpose of the study is to research the changes in range of motion and myoelectric activity of the ankle dorsiflexors in female NCAA Division I swimmers when using a vibrating foam roller in comparison to a static foam roller. Combining the self-myofascial release with the increased blood flow and muscle temperature exerted from vibration could possibly enhance the effects of foam-rolling treatment. The different effects of ankle flexibility and motor unit activation after static …

Sustainability In Quality Improvement And Patient Safety Initiatives, Angelo P. Giardino, Eileen R. Giardino
Journal of Nursing & Interprofessional Leadership in Quality & Safety
The question often asked of a quality improvement initiative is whether the improvement process has been sustained within the organization. Rarely is the question answered satisfactorily. The sustainability of an improvement process is important as it justifies the investment of human and financial capital. The term 'evaporation of improvements' addresses the dilemma that between 33% to 70% of all innovations are reportedly not sustained (Fleiszer, Semenic, Ritchie, Richer, & Denis, 2015) . This evaporation of improvement captures the frustrating inability of many institutions to maintain the achieved improvement after the newness of the initial effort wears off (Buchanan, Fitzgerald, & …
