Skeletal muscle adapts to external stimuli such as increased work. Muscle progenitors (MPs) control muscle repair due to severe damage, but the role of MP fusion and associated myonuclear accretion during exercise are unclear. While we previously demonstrated that MP fusion is required for growth using a supra-physiological model (Goh and Millay, 2017), questions remained about the need for myonuclear accrual during muscle adaptation in a physiological setting. Here, we developed an 8 week high-intensity interval training (HIIT) protocol and assessed the importance of MP fusion. Production of 3-dimensional neural progenitor cultures from human pluripotent stem cells offers the potential to generate large numbers of cells. We utilised our nanobridge system to generate 3D hPSC aggregates for differentiation towards the neural lineage, and investigate the ability to passage aggregates while maintaining cells at a stem/progenitor stage. Over 38 days, aggregate cultures exhibited upregulation and maintenance of neural-associated markers and demonstrated up to 10 fold increase in cell number. Aggregates undergoing neural induction in the presence or absence of nanobridges demonstrated no differences in marker expression, proliferation or viability. To become activated and perform effector functions, naive T cells move within lymph nodes to scan dendritic cells presenting antigen. Previous work has shown naive T cell activation is promoted by interleukin 7 in vivo, and it does so by regulating the T cell:dendritic cell interaction. Our work identifies a novel role for IL-7 in mediating naive T cell motility in the lymph node and promoting T cell association with DCs. Additionally, we have developed a method for analyzing microscopy images, regionalized normalized mutual information, to assess intercellular associations between T cells and other cells within the lymph node. The National Library of Medicine (NLM) established the Disaster Information Management Research Center (DIMRC) in 2008 based on its history of providing health information for disaster preparedness, response, and recovery. The NLM long range plan (Charting a Course for the 21st Century: NLM's Long Range Plan 2006-2016) called for creation of a center to show "a strong commitment to disaster remediation and to provide a platform for demonstrating how libraries and librarians can be part of the solution to this national problem". Persistent quadriceps weakness in individuals with anterior cruciate ligament reconstruction (ACLR) has been attributed to arthrogenic muscle inhibition (AMI), which is partially due to the reciprocal inhibition of the quadriceps with the over-excitation of the hamstrings. The objective of this study was to investigate the effectiveness of the hamstrings fatigue immediately prior to quadriceps strength training on reducing AMI. Eight subjects with ACLR (time since injury: 46.5±23.6 months, age: 21.4±1.4 years) and eight healthy subjects (age: 22.5±2.7 years) with no previous history of knee injury participated. Culture and sequencing have produced divergent hypotheses about cystic fibrosis (CF) lung infections. Culturing suggests that CF lungs are uninfected before colonization by a limited group of CF pathogens. Sequencing suggests diverse communities of mostly oral bacteria inhabit lungs early on and diversity decreases as disease progresses. We studied the lung microbiota of CF children using bronchoscopy and sequencing, with measures to reduce contamination. We found no evidence for oral bacterial communities in lung lavages that lacked CF pathogens. Lavage microbial diversity varied widely, but decreases in diversity appeared to be driven by increased CF pathogen abundance, which reduced the …

A key distinction of Doctors of Osteopathic Medicine (D.O.s) is their recognition of each patient as a whole person rather than just addressing his ailment. This focus previously highlighted physical manipulation over medications; however, the osteopathic profession has evolved significantly over the years. As this field is no longer identified by its original rejection of pharmaceuticals, other original principles of osteopathic medicine have impacted the growth of the field. Background: The colorectal cancer antigen GUCY2C exhibits unique split tolerance, evoking antigen-specific CD8+, but not CD4+, T-cell responses that deliver anti-tumor immunity without autoimmunity in mice. Here, the cancer vaccine Ad5-GUCY2C-PADRE was evaluated in a first-in-man phase I clinical study of patients with early-stage colorectal cancer to assess its safety and immunological efficacy.
Methods: Ten patients with surgically-resected stage I or stage II (pN0) colon cancer received a single intramuscular injection of 1011 viral particles (vp) of Ad5-GUCY2C-PADRE. Women's health care professionals, such as general physicians, obstetricians and gynecologists, midwives, nurses, and doulas, in the US need to be aware of cultural issues and disparities. Minorities and migrant women experience cultural challenges and disparities when receiving health care in the US. Without cultural sensitivity, patient care is compromised. Pregnancy and childbirth practices vary widely by culture, and potential differences in perspectives, beliefs, and treatment of these are critical issues for women’s health care professionals to study. Female genital cutting (FGC), obstetric fistulas (OF), and female cancer are also discussed in this paper.

Objective. To develop software to assess the potential aggressiveness of an incidentally detected renal mass using images. Methods. Thirty randomly selected patients who underwent nephrectomy for renal cell carcinoma (RCC) had their images independently reviewed by engineers. Tumor “Roughness” was based on image algorithm of tumor topographic features visualized on computed tomography (CT) scans. Univariant and multivariant statistical analyses are utilized for analysis. Results. We investigated 30 subjects that underwent partial or radical nephrectomy. Background
Children who undergo surgery experience significant pain in the post anesthesia care unit. Nurse and parent behaviors in the post anesthesia care unit directly impact child postoperative pain. Therefore, we have developed and evaluated (Phase 1) and then tested (Phase 2) the feasibility of a new intervention (Nurse and Parent Training in Postoperative Stress) to alter parent and nurse behaviors in a way consistent with reducing child postoperative pain.

Magnetotactic bacteria biomineralize intracellular magnetic nanocrystals surrounded by a lipid bilayer called magnetosomes. Due to their unique characteristics, magnetite magnetosomes are promising tools in Biomedicine. However, the uptake, persistence, and accumulation of magnetosomes within mammalian cells have not been well studied. Here, the endocytic pathway of magnetite magnetosomes and their effects on human cervix epithelial (HeLa) cells were studied by electron microscopy and high spatial resolution nano-analysis techniques. Transmission electron microscopy of HeLa cells after incubation with purified magnetosomes showed the presence of magnetic nanoparticles inside or outside endosomes within the cell, which suggests different modes of internalization, and that …

Because power training has been known to augment stability, the purpose of this study was to assess whether the removal of visual input affects lower limb muscle power production in young women who are resistance trained to the same degree it affects the untrained. This provided insight as far as the need for resistance training protocols in a largely untrained visually impaired population. To study this, fourteen college-aged female participants (18-23 years) performed a seated double-leg press on a leg sled machine, isolating power production of the lower limbs. Medial Tibial Stress Syndrome (MTSS) is a chronic lower-limb injury that effects a large population of athletes and exercisers. There is no definitive knowledge on what causes this injury, making prevention and treatment difficult. The purpose of this thesis is to identify the prevention and treatment methods for MTSS that seem the most promising. The risk factors of MTSS and possible preventative methods are first presented. Following this is an overview of both traditional and cutting-edge treatment options. PURPOSE: To update the American Society of Clinical Oncology (ASCO)/American Society of Hematology (ASH) recommendations for use of erythropoiesis-stimulating agents (ESAs) in patients with cancer.
METHODS: PubMed and the Cochrane Library were searched for randomized controlled trials (RCTs) and meta-analyses of RCTs in patients with cancer published from January 31, 2010, through May 14, 2018. For biosimilar ESAs, the literature search was expanded to include meta-analyses and RCTs in patients with cancer or chronic kidney disease and cohort studies in patients with cancer due to limited RCT evidence in the cancer setting. Proactive policing, the strategic targeting of people or places to prevent crimes,is a well-studied tactic that is ubiquitous in modern law enforcement. A 2017 National Academies of Sciences report reviewed existing literature, entrenched in deterrence theory, and found evidence that proactive policing strategies can reduce crime. The existing literature, however, does not explore what the short and long-term effects of police contact are for young people who are subjected to high rates of contact with law enforcement as a result of proactive policing. A 31-year-old male presented with a complaint of chronic pruritus with diffuse urticarial wheals for the past seven months. He underwent extensive workup and was found to have positive Helicobacter pylori (H. pylori) stool antigen test. He was treated with bismuth subsalicylate, metronidazole, tetracycline, and omeprazole for two weeks. Pruritus and urticarial wheals disappeared in four weeks after therapy was started.

Significant increases in age-related cognitive impairment (ARCI) associated with aging have been observed among older adults, negatively affecting measures of Quality of Life (QOL). Purpose: To examine the effects of a 12-week aerobic and cognitive training intervention on cognitive function in apparently healthy older adults. Methods: Six participants (40-75 years) were randomly placed into one of three training groups: 1) aerobic, cognitive, and flexibility (AER/COG); 2) aerobic and flexibility (AER); or 3) cognitive and flexibility (COG). The purpose of this paper is to measure the effectiveness of existing employment accommodations required by the Americans with Disabilities Act for employees who are Deaf or hard of hearing. Participants completed an online survey in which they identified with one of four levels of hearing loss and selected from descriptions of workplace accommodations. Each selection was ranked according to perceived importance and satisfaction. Accommodations that showed any significance of importance were endorsed by 18% or less of the respondents. The most important accommodations were computer assisted note-taking (18%) and flashing alarms (11%). Participants reported high satisfaction with most of …

This project examined the relationships between pediatric cochlear implantation and Deaf Culture. More specifically, this research investigated how/if parents are educated about or exposed to Deaf Culture during the cochlear implant candidacy evaluation process, and what type of interactions (if any) the child or caregiver had with members of the Deaf community after the child received a cochlear implant. A short survey was distributed to caregivers of pediatric cochlear implantees. This research study was designed to explore dog owners’ views on how often they expose their dogs to loud noises and whether or not the owners believe noise exposure affects their dogs’ hearing. Many humans are not aware that dogs, too, can suffer from hearing loss. To determine dog owners’ perceptions of the amount of noise exposure their dogs face and if their hearing is affected as a result, I created a survey for dog owners in Colorado. Despite enhanced attention to and advances in patient experience, many of the published initiatives focus on in-hospital care and opportunities have been identified to continue to strategically address patient experience in the ambulatory setting. Family and friend caregivers (i.e., unpaid carers) play a critical role in meeting the needs of people across various ages and illness circumstances. Caregiver experiences and expertise, which are currently overlooked, should be considered in practice (such as designing and evaluating services) and when designing and conducting research.
