Macrophages are susceptible to human immunodeficiency virus type 1 (HIV-1) infection despite abundant expression of antiviral proteins. Perhaps the most important antiviral protein is the restriction factor sterile alpha motif domain and histidine/aspartic acid domain-containing protein 1 (SAMHD1). We investigated the role of SAMHD1 and its phospho-dependent regulation in the context of HIV-1 infection in primary human monocyte-derived macrophages and the ability of various interferons (IFNs) and pharmacologic agents to modulate SAMHD1. Here we show that stimulation by type I, type II, and to a lesser degree, type III interferons share activation of SAMHD1 via dephosphorylation at threonine-592 as a …

K2 And K2*: Efficient Alignment-Free Sequence Similarity Measurement Based On Kendall Statistics, Jie Lin, Donald A. Adjeroh, Bing-Hua Jiang, Yue Jiang

Clinical and Translational Science Institute

Motivation: Alignment-free sequence comparison methods can compute the pairwise similarity between a huge number of sequences much faster than sequence-alignment based methods. Results: We propose a new non-parametric alignment-free sequence comparison method, called K2, based on the Kendall statistics. Comparing to the other state-of-the-art alignment-free comparison methods, K2 demonstrates competitive performance in generating the phylogenetic tree, in evaluating functionally related regulatory sequences, and in computing the edit distance (similarity/dissimilarity) between sequences. Furthermore, the K2 approach is much faster than the other methods. An improved method, K 2 , is also proposed, which is able to determine the appropriate algorithmic parameter …

As the senior population in the United States increases, the aging LGBT (lesbian, gay, bisexual, transgender) population should also see comparable growth rates. Health care providers and social service organizations will care for more LGBT seniors with special needs beyond the general population of older adults as they are more at risk for certain conditions. This article identifies some specific health disparities and examines several organizations that work to improve LGBT senior health by providing critical health information to LGBT older adults, caregivers, and health care professionals.

Faculty Research 2018

Congenital diaphragmatic hernia (CDH), characterized by malformation of the diaphragm and hypoplasia of the lungs, is one of the most common and severe birth defects, and is associated with high morbidity and mortality rates. There is growing evidence demonstrating that genetic factors contribute to CDH, although the pathogenesis remains largely elusive. Single-nucleotide polymorphisms have been studied in recent whole-exome sequencing efforts, but larger copy number variants (CNVs) have not yet been studied on a large scale in a case control study. To capture CNVs within CDH candidate regions, we developed and tested a targeted array comparative genomic hybridization platform to …

Neural Responses To Naturalistic Clips Of Behaving Animals Under Two Different Task Contexts, Samuel A. Nastase, Yaroslav O. Halchenko, Andrew C. Connolly, M. Ida Gobbini, James V. Haxby

Dartmouth Scholarship

The human brain rapidly deploys semantic information during perception to facilitate our interaction with the world. These semantic representations are encoded in the activity of distributed populations of neurons (Haxby et al., 2001; McClelland and Rogers, 2003; Kriegeskorte et al., 2008b) and command widespread cortical real estate (Binder et al., 2009; Huth et al., 2012). The neural representation of a stimulus can be described as a location (i.e., response vector) in a high-dimensional neural representational space (Kriegeskorte and Kievit, 2013; Haxby et al., 2014). This resonates with behavioral and theoretical work describing mental representations of objects and actions as being …

Fatty Acid Synthase (FASN), a key enzyme of de novo lipogenesis, is upregulated in many cancers including colorectal cancer (CRC); increased FASN expression is associated with poor prognosis. Potent FASN inhibitors (TVBs) developed by 3-V Biosciences demonstrate anti-tumor activity in vitro and in vivo and a favorable tolerability profile in a Phase I clinical trial.

However, CRC characteristics associated with responsiveness to FASN inhibition are not fully understood. We evaluated the effect of TVB-3664 on tumor growth in nine CRC patient-derived xenografts (PDXs) and investigated molecular and metabolic changes associated with CRC responsiveness to FASN inhibition.

CRC cells and PDXs …

Physical Therapy Faculty Articles and Research

Background

Low back pain (LBP) is commonly associated with paraspinal muscle dysfunctions. A method to study deep lumbar paraspinal (i.e. multifidus) muscle function and neuromuscular activation pattern is intramuscular electromyography (EMG). Previous studies have shown that the procedure does not significantly impact muscle function during activities involving low-level muscle contractions. However, it is currently unknown how muscular function and activation are affected during high-exertion contractions.

Objective

To examine the effects of insertion and presence of fine-wire EMG electrodes in the lumbar multifidus on muscle strength, endurance, and activation profiles during high-exertion spinal extension muscle contractions.

Design

Single-blinded, repeated measures intervention …

Implementing Dietary And Physical Activity Guidelines In A College Health Center, Terry Lynn Bartmus May 2018

Implementing Dietary And Physical Activity Guidelines In A College Health Center, Terry Lynn Bartmus

UNLV Theses, Dissertations, Professional Papers, and Capstones

Obesity has been classified as a major national health care problem that results in a significant increase in morbidity, mortality, and health care related costs. Seventy percent of all adults age 18 and older have a body mass index (BMI) of 25 or greater, indicating they are overweight or obese. Thirty-six percent of college students are overweight or obese. Research has shown that lack of time has further decreased weight-related counseling by primary-care providers. Printed materials have been shown to be effective in cueing individuals to engage in healthy behaviors such as following physical activity and recommended nutritional guidelines. Currently, …

Using Pictorial Action Instructions To Train Low-Literacy Adults To Construct A Basic Humanitarian Engineering Project, Kathleen Paco Cadman May 2018

Using Pictorial Action Instructions To Train Low-Literacy Adults To Construct A Basic Humanitarian Engineering Project, Kathleen Paco Cadman

UNLV Theses, Dissertations, Professional Papers, and Capstones

Low- and middle-income countries (LMICs) are often disproportionately faced with environmental hazards, which can cause adverse health outcomes. Although humanitarian engineering projects have been created to mitigate these hazards, LMICs are also frequently impacted by literacy deficits, creating a barrier in training low-literacy individuals to construct these projects. The purpose of this dissertation was to develop, refine, and test the usability of pictorial action instructions (PAI) in training low-literacy individuals to construct a humanitarian engineering project. A concept analysis was completed, including the creation of an operational definition, for “lay worker health literacy.” This analysis and definition set a foundation …

Mate Preferences Among Single Mothers, Carol Yvette Franco May 2018

Mate Preferences Among Single Mothers, Carol Yvette Franco

UNLV Theses, Dissertations, Professional Papers, and Capstones

As individuals become parents, they shift time and energy towards parenting to ensure the survival and wellbeing of their offspring. However, it is not uncommon for a romantic partnership between parents to dissolve, which may lead a single mother to pursue a new partner in the future. But how does having a dependent child influence whom she will choose for her new partner? And how will resource availability and assistance from family influence the decisions she makes? A significant amount of research has been conducted on female mate preferences in the United States and elsewhere, yet little is specifically focused …

Clinical Instructors' Cultivation Of Pre-Licensure Nursing Students' Pharmacology Ksas, Rieneke Holman May 2018

Clinical Instructors' Cultivation Of Pre-Licensure Nursing Students' Pharmacology Ksas, Rieneke Holman

UNLV Theses, Dissertations, Professional Papers, and Capstones

Adverse drug events affect up to 5% of inpatients; half of these events are caused by preventable medication errors (Agency for Healthcare Research and Quality, 2015). Nurses are the final checkpoint between a medication being ordered and the patient taking it. This profound responsibility requires nurses to possess well-developed pharmacology knowledge, skills, and attitudes (KSAs). To acquire these abilities, they must not only receive pharmacology content as part of their educational preparation, but also translate what they have learned during student clinical experiences. This is important in bridging the gap between what is learned in the

classroom and clinical practice. …

Effect Of Bmp Treatment On Periostin Gene Expression In Pre-Osteoblastic Mc3t3-E1 Mouse Cells, Vincent Lee Khang May 2018

Effect Of Bmp Treatment On Periostin Gene Expression In Pre-Osteoblastic Mc3t3-E1 Mouse Cells, Vincent Lee Khang

UNLV Theses, Dissertations, Professional Papers, and Capstones

Periostin is a secreted, extracellular matrix (ECM) protein widely expressed within collagen-rich fibrous connective tissues of the body including the periodontal ligament (PDL), bone, skin, heart, and cornea. Periostin has been shown to serve many important regulatory functions including cell adhesion, cell motility, wound healing and of particular importance to the dental field, differentiation of osteoblasts. The deletion of periostin compromises osteoblast attachment to bone matrix and induces a reduction in mineralization and expression of bone markers, including type I collagen, osteocalcin, osteopontin and alkaline phosphatase. Periostin has also been shown to play a significant role in collagen fibrillogenesis by …
