Following injury, mesenchymal repair cells are activated to function as leader cells that modulate wound healing. These cells have the potential to differentiate to myofibroblasts, resulting in fibrosis and scarring. The signals underlying these differing pathways are complex and incompletely understood. The ex vivo mock cataract surgery cultures are an attractive model with which to address this question. With this model we study, concurrently, the mechanisms that control mesenchymal leader cell function in injury repair within their native microenvironment and the signals that induce this same cell population to acquire a myofibroblast phenotype when these cells encounter the environment of …

Inference And Analysis Of Multilayered Mirna-Mediated Networks In Cancer, Duc Do
Inference And Analysis Of Multilayered Mirna-Mediated Networks In Cancer, Duc Do
Dissertations (1934 -)
MicroRNAs (miRNAs) are small noncoding transcripts that can regulate gene expression, thereby controlling diverse biological processes. Aberrant disruptions of miRNA expression and their interactions with other biological agents (e.g., coding and noncoding transcripts) have been associated with several types of cancer. The goal of this dissertation is to use multidimensional genomic data to model two different gene regulation mechanisms by miRNAs in cancer. This dissertation results from two research projects. The first project investigates a miRNA-mediated gene regulation mechanism called competing endogenous RNA (ceRNA) interactions, which suggests that some transcripts can indirectly regulate one another's activity through their interactions with …
Understanding Relationships Between Early Life Toxic Stress, Childhood Socioeconomic Disadvantage, And Allostatic Load In Adolescence, Amanda King
Dissertations (1934 -)
Chronic disease prevalence among children and adolescents is rising, which is thought to result in part from elevations in allostatic load (AL). AL is the cumulative physiological dysregulation that results from exposure to biological, social and environmental stressors over time. Socioeconomic disparities in chronic disease and AL have been well-documented in adult populations, including links between childhood socioeconomic disadvantage (CSD) and AL, yet little is known as to whether CSD may begin to impact AL earlier in life. Differential exposure and vulnerability to stress among racial/ethnic minorities may increase risk for elevated AL among those experiencing CSD. Framed by the …

Pre-Death Grief, Resourcefulness And Perceived Stress Among Care Givers Of Partners With Young Onset Dementia, Karie Kobiske
Dissertations (1934 -)
Over 200,000 Americans are diagnosed with young onset dementia (YOD). YOD is the dementia diagnosed prior to the age of 65. Most persons of YOD are cared for by their partners. Caregiving for a partner diagnosed with YOD has unique challenges including multiple losses resulting from the functional, cognitive, and behavioral declines which can be demanding and stressful. These losses experienced by the caregiver of a partner diagnosed with YOD have been termed pre-death grief. Caregivers of partners with YOD often report high levels of burden and stress resulting in depression, anxiety, hopelessness, as well as increase morbidity and mortality. …
Change In Bone Mineral Density Among High Frequency Apheresis Blood Donors, Walter Bialkowski
Change In Bone Mineral Density Among High Frequency Apheresis Blood Donors, Walter Bialkowski
Dissertations (1934 -)
Exposure to citrate anticoagulant during apheresis blood donation induces significant decreases in serum ionized calcium with subsequent perturbations to parathyroid hormone, vitamin D, and markers of bone remodeling. Cross-sectional studies of bone mineral density (BMD) among apheresis donors exhibit conflicting results. Resolving the potential impact of the highest apheresis donation frequency represents a significant knowledge gap in ensuring adequate protections for volunteer apheresis blood donors. ALTRUYST (NCT02655055) was a randomized, longitudinal, controlled clinical trial designed to determine if repeated exposure to citrate through apheresis donation reduces BMD. Male donors, 18-65 years of age with no more than five previous apheresis …

Predictors And Outcomes Of Nurses’ Use Of Smartphones For Work Purposes, John Robert Bautista, Sonny Rosenthal, Trisha T.C. Lin, Yin Leng Theng
Research Collection College of Integrative Studies
Recent studies have indicated that nurses use their smartphones for work purposes to enhance productivity. However, few theory-driven quantitative studies have examined factors associated with such use. This study aims to address this research gap by developing and testing a model based on the theory of planned behavior, organizational support theory, and IT consumerization theory. Hypothesis testing used structural equation modeling of survey data from 517 staff nurses employed in 19 tertiary-level general hospitals in the Philippines. Results showed that injunctive norm, descriptive norm, and perceived behavioral control were positively associated with intention to use smartphones for work purposes. Moreover, …

Testing The Efficacy Of Transcranial Magnetic Stimulation (Tms) In Treating Depression In Patients With Cognitive Impairment, Daniel Robert Schaffer
Testing The Efficacy Of Transcranial Magnetic Stimulation (Tms) In Treating Depression In Patients With Cognitive Impairment, Daniel Robert Schaffer
Psychology Theses & Dissertations
The purpose of this study is to (1) examine the efficacy of Transcranial Magnetic Stimulation (TMS) in treating depression among individuals with cognitive impairment and (2) to examine if TMS is capable of facilitating cognitive improvements independent of mood improvements. Mild cognitive impairment (MCI) is often seen as a pre-clinical stage to dementia, and depressive disorders are highly prevalent among both MCI and dementia. There is a large body of research that has linked depressive disorders as a prodromal symptom of MCI and the later development of dementia. While some researchers debate whether or not this link between depression and …
Developmental Steps For A Functional Three-Dimensional Cell Culture System For The Study Of Asymmetrical Division Of Neural Stem Cells, Martina Zamponi
Developmental Steps For A Functional Three-Dimensional Cell Culture System For The Study Of Asymmetrical Division Of Neural Stem Cells, Martina Zamponi
Biomedical Engineering Theses & Dissertations
Stem cells are a cell type present during and following development, which possess self- renewal properties, as well as the ability to differentiate into specific cells. Asymmetrical division is the cellular process that allows stem cells to produce one differentiated and one un-differentiated daughter cell during the same mitotic event. Insights in the molecular mechanisms of such process are minimal, due to the absence of effective methods for its targeted study. Currently, traditional methods of investigation include monolayer cell culture and animal models. The first poses structural limitations to the accurate representation of human tissue and cell structures, while animal …

The DNA damage repair enzyme, O6-methylguanine DNA methyltransferase (MGMT) is overexpressed in breast cancer, correlating directly with estrogen receptor (ER) expression and function. In ER negative breast cancer the MGMT promoter is frequently methylated. In ER positive breast cancer MGMT is upregulated and modulates ER function. Here, we evaluate MGMT’s role in control of other clinically relevant targets involved in cell cycle regulation during breast cancer oncogenesis. We show that O6-benzylguanine (BG), an MGMT inhibitor decreases CDC2, CDC20, TOP2A, AURKB, KIF20A, cyclin B2, A2, D1, ERα and survivin and induces c-PARP and p21 and sensitizes ER …

Ferrochelatase (FECH) is an enzyme necessary for heme synthesis, which is essential for maintaining normal functions of endothelial nitric oxide synthase (eNOS) and soluble guanylyl cyclase (sGC). We tested the hypothesis that inhibition of vascular FECH to attenuate heme synthesis downregulates eNOS and sGC expression, resulting in impaired NO/cGMP-dependent relaxation. To this end, isolated bovine coronary arteries (BCAs) were in vitro incubated without (as controls) or with N-methyl protoporphyrin (NMPP; 10-5-10-7M; a selective FECH antagonist) for 24 and 72 hours respectively. Tissue FECH activity, heme, nitrite/NO and superoxide levels were sequentially measured. Protein expression of FECH, eNOS and sGC was …

Noninvasive Hemodynamic Monitoring In Emergency Trauma Patients, Ryan Boyd
Noninvasive Hemodynamic Monitoring In Emergency Trauma Patients, Ryan Boyd
Nurse Anesthesia MSNA Student Capstones
Noninvasive hemodynamic monitoring has evolved steadily over the past few decades in response to healthcare providers' preference for less invasive monitoring methods for their patients. Invasive monitoring has its place in the critically ill patient population as witnessed in intensive care units throughout the country. Even in this environment, providers are opting less for the pulmonary artery catheter which has been the gold standard for invasive cardiac output monitoring in the past. Providers are now utilizing less invasive monitoring techniques which offer fewer potential complications to the patient while providing rapid access to critically important hemodynamic data. Examination of different …
The Utilization Of Laryngeal Mask Airways Vs. Endotracheal Tubes In Pediatrics With Upper Respiratory Tract Infection In Reducing Airway Complications, Ruby Foster
Nurse Anesthesia MSNA Student Capstones
According to Tait & Malviya, (2005) children typically experience six to eight upper respiratory infections (URIs) per year with ninety-five percent being viral in etiology (Tait & Malvia, 2005). URIs commonly occur in the pediatric population (Bernando-Ocampo, 2012). An issue surrounding the debate on whether a child with upper respiratory infection (URI) should proceed with surgery or cancel surgery is a debate that has been ongoing for several years (Tait el al. 2005). This review examines whether children who have an URI scheduled for surgery are at an increased risk for adverse respiratory complications when the laryngeal mask airway (LMA) …

Investigating Sports Drinks. Are they worth the money?
The sports drinks industry is a huge global business which shows no signs of slowing down. The inspiration to conduct this investigation came from an observation that there is an increasing consumption of sports and energy drinks within our community, particularly amongst children. The message from the advertisers is clear- athletes should replace lost body fluid with drinks that contain electrolytes, such as sports drinks. However, there are conflicting suggestions from conducting literature reviews; that, not only are these drinks unhealthy in terms of the high sugar content, but instead water …

Classification Of Intensity-Modulated Proton Therapy Plans, Alice Liu
The International Student Science Fair 2018
Abstract
Title: Classification of IMPT Plans
Purpose:
Create a method to aid physicists and dosimetrists in the classification of Intensity Modulated Proton Therapy treatment plans.
Method:
A Python program was developed to read structure sets and dose grids from radiation treatment plans. The program outputs field-specific histograms of the number of voxels within the planning target volume that received dose and normalizes each histogram to the percent volume of the target that received each field’s contribution of the total dose. It then determines the number of Gaussian distributions in each histogram, which serves as an initial classification metric, and calculates …
The Effect Of Hyperthermia On Liver Cell Viability, Jada Grant
The Effect Of Hyperthermia On Liver Cell Viability, Jada Grant
The International Student Science Fair 2018
Hyperthermia is a treatment for liver cancer where the tumor is exposed to a relatively high heat temperature. This process is designed to make the tumor more susceptible to chemotherapy. My thought process was that if hyperthermia can kill cancerous cells, then there will be negative long term effects on the noncancerous cells surrounding the tumor. The project experimented on Kidney cells because the Kidney and Liver both go through the process of filtration. This means that the Kidney cells should give similar results that Liver cells would. Kidney cells were heated at 40 degrees Celsius for time amounts of …

The Effect Of The Oxytocin Hormone In Cocaine Addicted Rats Suffering From Abstinence Syndrome, Juan Padilla
The International Student Science Fair 2018
Rehabilitation from a history of drug abuse is a very difficult and life changing process. Abstinence syndrome forces the rehabilitating patient to go through anxiety like symptoms. The Oxytocin hormone has been believed to be a potential facilitator in the rehabilitation process. This hormone is very present during interactions of affection (hugs) and birth, it foments trust while it associates to a feeling of calmness. Oxytocin would theoretically be a calming agent to the anxiety like symptoms such as craving. The experiment aimed to test the calming effects of the hormone, consisted of a five-day systemic cocaine injections treatment schedule …

The Effect Of Kalanchoe Pinnata On The Cell Migration Of Astrocytomas, Jorge Rosa
The International Student Science Fair 2018
Astrocytomas, from cell line 1321N1,are cancerous astrocytes. These are a type of glial cell in the brain which is responsible for maintaining synaptic contact and the signaling capacities among neurons. They also represent a fundamental element in controlling blood flow and defining brain micro structure. Due to the fact that this variant is cancerous, a condition commonly described as an abnormal cell cycle and uncontrolled reproduction of cells, they have the ability to metastasize.
In the metastatic process of cancer cells the initial phase is know as tumor invasion. This stage consists mainly of the cells migrating away from the …

A Humanized Hypertrophic Cardiomyopathy Model To Elucidate Molecular Mechanism In Disease Pathology, Ragavi Vijayakumar, Maxine Hong
The International Student Science Fair 2018
Hypertrophic cardiomyopathy (HCM), that clinically manifests as an enlarged heart is a highly prevalent cardiac disorder with propensity towards arrhythmia-induced sudden cardiac death. The mechanism of HCM remains poorly defined, necessitating further understanding of the disease for improved therapeutic strategies. As it is challenging to obtain cardiac biopsies from human subjects, using induced pluripotent stem cells technology, we generated cardiomyocytes (CMs) in a dish from HCM patients. These HCM-CMs presented the clinical manifestation in that they were significantly larger in size in comparison to control (healthy)-CMs. Furthermore, gene expression profiling of cardiac ion channels revealed increased transcripts encoding for calcium …
