The Thai Journal of Veterinary Medicine

The clinical benefits of Coenzyme Q10 (CoQ10) have been well documented in heart failure patients. In veterinary medicine, however, the clinical benefits of CoQ10 have not been well established in dogs with heart diseases. This study aimed to determine the effects of CoQ10 supplementation on 1) preventing myocardial injury 2) cardiac function 3) autonomic balance in dogs with myxomatous degenerative mitral valve disease (MMVD). Thirteen dogs with MMVD at ACVIM stage C were recruited in this study. Dogs were further divided into group I (Body weight [BW] < 6 kg.; n = 7) and group II (BW ≥ 6 kg.; n = 6). Cardiac troponin I (cTnI), echocardiographic examination, and 2 hours of Holter recording were performed on Day 0 and after receiving CoQ10 at 100 mg/dog twice daily for 28 days. CoQ10 caused a reduction of cTnI level in 71% of the dogs. Median cTnI tended to cause tended to decline but not significantly. Systolic function [i.e. fractional shortening (FS) and ejection fraction (EF)] increased significantly by 8 % and 9% (P < 0.05), respectively in group I after CoQ10 supplementation. Other echocardiographic parameters were not altered in either group. HRV analysis revealed no change in autonomic function and balance. cTnI negatively correlated with FS (P < 0.01) and positively correlated with LF/HF ratio (P < 0.05). In conclusion, this study demonstrated the clinical benefits of CoQ10 supplementation in dogs with MMVD. CoQ10 improves cardiac function in small dogs with MMVD. Finally, supplementation of CoQ10 in dogs should be based on body weight.

All Works

The purpose of this study is to provide recommendations to bridging the "theoretical" with the "practical" in developing community-based participatory research (CBPR) health communication projects. As illustrated through a review of several case studies from health campaigns using CBPR, often times the theoretical orientations of CBPR become secondary to its praxis, with unspoken motives and agendas become motivating factors in guiding the initiatives. These motives may come in the form of funding organization priorities, funded grant proposal constraints, and the desire to continue relationships that are fostered in the development of CBPR projects. In response, this essay reintroduces the culture-centered …

A Disturbance In The Force: Cellular Stress Sensing By The Mitochondrial Network, Robert Gilkerson Sep 2018

A Disturbance In The Force: Cellular Stress Sensing By The Mitochondrial Network, Robert Gilkerson

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

As a highly dynamic organellar network, mitochondria are maintained as an organellar network by delicately balancing fission and fusion pathways. This homeostatic balance of organellar dynamics is increasingly revealed to play an integral role in sensing cellular stress stimuli. Mitochondrial fission/fusion balance is highly sensitive to perturbations such as loss of bioenergetic function, oxidative stress, and other stimuli, with mechanistic contribution to subsequent cell-wide cascades including inflammation, autophagy, and apoptosis. The overlapping activity with m-AAA protease 1 (OMA1) metallopeptidase, a stress-sensitive modulator of mitochondrial fusion, and dynamin-related protein 1 (DRP1), a regulator of mitochondrial fission, are key factors that shape …

Early Investigations And Recent Advances In Intraperitoneal Immunotherapy For Peritoneal Metastasis., Anusha Thadi, Marian Khalili, William F. Morano, Scott D. Richard, Steven C. Katz, Wilbur B. Bowne

Department of Obstetrics and Gynecology Faculty Papers

Peritoneal metastasis (PM) is an advanced stage malignancy largely refractory to modern therapy. Intraperitoneal (IP) immunotherapy offers a novel approach for the control of regional disease of the peritoneal cavity by breaking immune tolerance. These strategies include heightening T-cell response and vaccine induction of anti-cancer memory against tumor-associated antigens. Early investigations with chimeric antigen receptor T cells (CAR-T cells), vaccine-based therapies, dendritic cells (DCs) in combination with pro-inflammatory cytokines and natural killer cells (NKs), adoptive cell transfer, and immune checkpoint inhibitors represent significant advances in the treatment of PM. IP delivery of CAR-T cells has shown demonstrable suppression of tumors …

OBJECTIVE: Unreliable antibodies often hinder the accurate detection of an endogenous protein, and this is particularly true for the cardiac and smooth muscle cofactor, MYOCD (myocardin). Accordingly, the mouse Myocd locus was targeted with 2 independent epitope tags for the unambiguous expression, localization, and activity of MYOCD protein. APPROACH AND RESULTS: 3cCRISPR (3-component clustered regularly interspaced short palindromic repeat) was used to engineer a carboxyl-terminal 3xFLAG or 3xHA epitope tag in mouse embryos. Western blotting with antibodies to each tag revealed a protein product of approximately 150 kDa, a size considerably larger than that reported in virtually all publications. MYOCD …

Pathogenicity And Protection Mediated By A Single Tcrβ In Experimental Autoimmune Encephalomyelitis, Tianhua Wu Sep 2018

Pathogenicity And Protection Mediated By A Single Tcrβ In Experimental Autoimmune Encephalomyelitis, Tianhua Wu

Theses and Dissertations (ETD)

How the TCR repertoire, together with risk-associated major histocompatibility complex (MHC), imposes susceptibility for autoimmune disease is not fully understood. A small fraction of TCR α or β chains are “public”, and are shared by most individuals.High-throughput sequencing of the mouse TCRβ repertoire during myelin oligodendrocyte glycoprotein (MOG)-induced experimental autoimmuneencephalomyelitis (EAE) identified a public TCRβ chain, TCRβ1, which was highly shared among individual mice and preferentially deployed during EAE. Retrogenic expression of TCRβ1 resulted in spontaneous early-onset EAE in mice with high penetrance and severity, despite being paired with a diverse endogenous TCRα repertoire. To further study autoimmunity conferred by …

Aging Suppresses Skin-Derived Circulating Sdf1 To Promote Full-Thickness Tissue Regeneration, Mailyn A. Nishiguchi, Casey A. Spencer, Denis H. Y. Leung, Thomas H. Leung

Research Collection School Of Economics

Physicians have observed that surgical wounds in the elderly heal with thinner scars than wounds in young patients. Understanding this phenomenon may reveal strategies for promoting scarless wound repair. We show that full-thickness skin wounds in aged but not young mice fully regenerate. Exposure of aged animals to blood from young mice by parabiosis counteracts this regenerative capacity. The secreted factor, stromal-derived factor 1 (SDF1), is expressed at higher levels in wounded skin of young mice. Genetic deletion of SDF1 in young skin enhanced tissue regeneration. In aged mice, enhancer of zeste homolog 2 (EZH2) and histone H3 lysine 27 …

Propofol Inhibits Prokaryotic Voltage-Gated Na+ Channels By Promoting Activation-Coupled Inactivation, Elaine Yang, Daniele Granata, Roderic G. Eckenhoff, Vincenzo Carnevale, Manuel Covarrubias

Department of Neuroscience Faculty Papers

Propofol is widely used in the clinic for the induction and maintenance of general anesthesia. As with most general anesthetics, however, our understanding of its mechanism of action remains incomplete. Local and general anesthetics largely inhibit voltage-gated Na+ channels (Navs) by inducing an apparent stabilization of the inactivated state, associated in some instances with pore block. To determine the biophysical and molecular basis of propofol action in Navs, we investigated NaChBac and NavMs, two prokaryotic Navs with distinct voltage dependencies and gating kinetics, by whole-cell patch clamp electrophysiology in the absence and presence of propofol at clinically relevant concentrations …
