Bacteriophage Virus-Like Particles As Vaccine Platforms: From Heart Disease To Malaria, Erin Crossey
Bacteriophage Virus-Like Particles As Vaccine Platforms: From Heart Disease To Malaria, Erin Crossey
Biomedical Sciences ETDs
Virus-like particles (VLPs) make excellent vaccines. They are non-infectious, often easy to produce in bacterial expression systems, and highly immunogenic. The latter feature is granted by the regularity of the capsid structure, which presents viral (or other) epitopes as dense, highly repetitive arrays that strongly stimulate B cells. VLPs can be used as the basis for vaccines targeting the virus from which they were derived, or they can be used as platforms to display practically any short peptide epitope in a multivalent format. Foreign peptides displayed on VLPs exhibit the same high immunogenicity as unmodified VLPs. Even self-antigens, normally subject …
Stabilization Of Gp120 Outer Domain For Hiv-1-1 Vaccine Immunogen Design, Duoyi Hu
Stabilization Of Gp120 Outer Domain For Hiv-1-1 Vaccine Immunogen Design, Duoyi Hu
School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research
The sole envelope glycoprotein spike on the surface of Human Immunodeficiency Virus (HIV-1) is composed of two subunits: gp120 and gp41. The gp120 consists of three domains: the inner domain, outer domain and bridging sheet, to which the viral primary receptor CD4 binds and induces a conformational change in gp120 to expose the co-receptor binding site. Previous studies have found that the outer domain of gp120 is relatively more stable than the inner domain and bridging sheet. When superimposed, the co-crystal structures of CD4-gp120 complex and VRC01-gp120 complex revealed that the CD4-binding site (CD4-BS) antibody VRC01 actually mainly bound to …

Abstract Purpose: The purpose is to study whether pain and inflammation in knee joint osteoarthritis (OA) are associated with local synovial neuronal changes. Methods: Synovial biopsies were harvested from the medial and lateral knee compartments from OA patients undergoing total joint replacement surgery. All patients had predominant pain at the medial joint compartment. Pain and knee joint function were evaluated by knee society score (KSS). Synovial inflammation was analyzed by histopathological analysis and expression of growth associated protein-43 (GAP-43), sensory (SP, CGRP) and autonomic (NPY, VIP, TH) neuropeptides was studied by single and double immunohistochemistry techniques. Results: We observed reduced …

A Novel And Rapid Staphylococcus Aureus Bacterial Identification Method Utilizing Immunomagnetic Beads And Single Cell Laser-Light Scattering, Kaylagh Hollen
All NMU Master's Theses
Staphylococcus aureus is the most commonly isolated human associated bacterial pathogen. It plays an important role in skin and soft-tissue infections, pneumonia, endocarditis, osteomyelitis, foreign-body infections, and sepsis. S. aureus diagnosis and treatment requires a minimum of 24-48. With this in mind, previous studies suggest that faster pathogen identification has been linked to improved patient outcomes. Improved patient outcomes including a reduction in hospitalization time, decreased risk of nosocomial infections, and decreased in medical costs. The impact of faster identification on patient outcome has led us to develop an alternative method of S. aureus identification via ImmunoMagnetic Separation (IMS) and …

Examination Of Cd133 And Cd147 As Cancer Stem Cell Markers, Christopher Mcmahon
All NMU Master's Theses
To date, as many as 14,000 patients in the United States per year are diagnosed with glioblastoma, the most common and most malignant primary brain tumor. Glioblastomas are characterized by their ability to evade treatment on many fronts, thus a novel approach to curative therapies is imperative. A population of cells with stem cell-like properties are found within glioblastoma tumors and drive their initiation and progression. Identification of extracellular markers on these tumor stem cells is thus paramount. The cell surface glycoproteins CD133 and CD147 were examined as potential markers of cancer stem cells found in glioblastoma. Here we found …

Analysis Of Tumor Specific Protein Expression In Glioblastoma Multiforme (Gbms) Tumors Through Immunohistochemistry, Amanda M. Wigand
All NMU Master's Theses
GBM tumors are the most aggressive and, unfortunately, the most fatal form of brain cancer. GBM tumors with isocitrate dehydrogenase-1 (IDH1) mutation being expressed, lead to higher survival rates in patients that also have full resection of the tumor and chemotherapy. Without this mutation, it is thought that tumors have a higher expression of the protein Basigin and O6-methylguanine-DNA-methyltrasnferase (MGMT) present, causing it to be more aggressive and less responsive to standard care. The objective of this study was to understand the correlation between IDH1 mutation presence and the expression of Basigin and MGMT. The expression of these proteins was …

An Exploration Of The Effectiveness Of The Use Of Communication Apps Through Mobile Devices On Children With Autism Spectrum Disorders (Asd), Miriam O Sullivan
Theses
Autism Spectrum Disorder (ASD) is a complex neurological disorder which impacts on people in three primary areas: communication skills, social skills and behaviour skills. The diagnosis of ASD continues to rise with an estimated 1 in 100 receiving diagnoses of ASD in Ireland. There are many interventions promoted on a regular basis that claim positive effects on the basis of empirical research. However, the claims made by such studies are sometimes a little at odds with the level of methodological rigour and/or sample size employed. The research available is based on small scale international studies (America, Canada and Australia); however, …

UNLV Theses, Dissertations, Professional Papers, and Capstones
Background and purpose: Running is one of the most popular exercises but it is also an activity with a high incidence of injury. Running form modification involving a forward lean of the trunk and forefoot strike pattern has been shown to be effective in attenuating the impact forces of the lower extremity. However, it is currently unknown how these changes can be most effectively instructed and learned by the runners. Previous studies have shown that practicing a motor task with an external focus can facilitate learning and retention when compared to the more common internal focus instructions. The purpose of …

Methods: The creator of the app offered free use of their app to a physical therapy clinic. As the app is only compatible with Apple products, the clinic used the app with any patient that had an iPhone. Retrospective review was conducted to determine if differences in patient outcomes were observed. Patients who had access to an iPad or iPhone were considered part of the “app group” and used the mobile app to reference and report PT HEP compliance. Patients without access to an iPad or iPhone were considered part of the “non-app group” and received traditional PT HEP prescription …

Balance Confidence And Fear Of Falling Avoidance Behavior Are Most Predictive Of Falling In Older Adults: A Prospective Analysis, Sarrie Oscar, Jessica Sasaoka, Kyle Vaughn
UNLV Theses, Dissertations, Professional Papers, and Capstones
Background: Evidence suggests that there are several fall predictors in the elderly population, including previous fall history and balance impairment. To date, however, the role of psychological factors has not yet been thoroughly vetted in conjunction with physical factors as predictors of future falls. Objective: The purpose of this study was to determine which measures, physical and psychological, are most predictive of falling in older adults. Design: This was a prospective cohort study. Methods: Sixty-four participants (mean age=72.2 years, SD=7.2; 40 women, 24 men) with and without pathology (25 healthy, 17 with Parkinson disease, 11 with cerebrovascular accident, 6 with …

Heel-Raised Foot Posture And Weightlifting Shoes Do Not Affect Trunk And Lowerextremity Biomechanics During A Barbell Back Squat, Javier Ibarra, Derek Oldroyd, Ryan Zane
UNLV Theses, Dissertations, Professional Papers, and Capstones
Background and Purpose: It is claimed that wearing weightlifting shoes with a raised-heel can improve posture and leg muscle activation, and reduce the risk of back injuries during a barbell back squat. However, these proclaimed biomechanical effects have not been thoroughly investigated. The purpose of this study was to compare the thoracic, lumbar, and lower extremity biomechanics during barbell back squat in 3 foot posture conditions. Subjects: 14 healthy recreational weightlifters (7 male and 7 female) between the ages of 18-50 participated in the study. A minimum of 2 years weightlifting experience and regular training with the barbell back squat …

Assessing Clinical Significance Of The Immediate Post-Concussion Assessment And Cognitive Testing Battery When Comparing Adjusted And Unadjusted Rci Methods For Different Ranges Of Baseline Scores, Ashley N. Figaro
UNLV Theses, Dissertations, Professional Papers, and Capstones
The growing concern revolving around the dangers of sports-related concussions have led to the most recent implementation of neurocognitive (NC) test batteries as a means to objectively determine the presence of a cognitive defect. Whereas any other sports-related injury can be diagnosed with tools such as an x-ray or MRI, a concussion represents a metabolic disturbance that cannot be identified by these diagnostic tools. Many neurocognitive test batteries employ statistical techniques to derive cut off scores in order to represent significant or insignificant changes as compared to individual baseline scores, or pre-established normative values. If an individuals’ post-injury score exceeds …
Concurrent Mental Health And Sport Performance Enhancement In An Athlete Initiating Behavioral Intervention With No Assessed Pathology: A Case Examination Supporting Optimization, Yulia Gavrilova
UNLV Theses, Dissertations, Professional Papers, and Capstones
Collegiate athletes are often exposed to unique environmental stressors that may negatively impact the way they think, behave, and feel in athletic, academic, and social domains, potentially compromising their performance and mental health when cognitive and behavioral skills are relatively underdeveloped. Behavioral interventions have been shown to enhance cognitive and behavioral skills in persons with assessed mental health deficits in the general population. However, little is known about the relative effects of psychologically-based programs in persons who do not evidence pathology. Along these lines, The Optimum Performance Program in Sports (TOPPS), based on Family Behavior Therapy, was recently developed with …

Induced Stem Cells As A Novel Multiple Sclerosis Therapy., Chong Xie, Yan-Qun Liu, Yang-Tai Guan, Guang-Xian Zhang
Department of Neurology Faculty Papers
Stem cell replacement is providing hope for many degenerative diseases that lack effective therapeutic methods including multiple sclerosis (MS), an inflammatory demyelinating disease of the central nervous system. Transplantation of neural stem cells or mesenchymal stem cells is a potential therapy for MS thanks to their capacity for cell repopulation as well as for their immunomodulatory and neurotrophic properties. Induced pluripotent stem cell (iPSC), an emerging cell source in regenerative medicine, is also being tested for the treatment of MS. Remarkable improvement in mobility and robust remyelination have been observed after transplantation of iPSC-derived neural cells into demyelinated models. Direct …
Multiphoton Imaging Of Labeled Breast Cancer Cells To Quantify Intra And Extracellular Receptors, Sydney C. Wiggins
Multiphoton Imaging Of Labeled Breast Cancer Cells To Quantify Intra And Extracellular Receptors, Sydney C. Wiggins
Biomedical Engineering Undergraduate Honors Theses
Every year 200,000 women in the United States are diagnosed with breast cancer. Of the cases diagnosed, 10% -15% are classified as triple negative breast cancer (TNBC) due to the absence of estrogen, progesterone, and HER-2/Neu receptors. This breast cancer sub-type is markedly more aggressive and twice as likely to develop in premenopausal women. TNBC is resistant to endocrine therapies and current targeted agents, making clinical need for the development of validated therapeutics for TNBC a pressing matter. To initiate drug development, the internalization of directly immunolabeled epidermal growth factor receptors (EGFR) in SK-BR-3 human breast adenocarcinoma cells was quantitated …
