Permutation Inference For The General Linear Model, Anderson M. Winkler, Gerard R. Ridgway, Matthew A. Webster, Stephen M. Smith, Thomas E. Nichols
School of Medicine Publications
Permutation methods can provide exact control of false positives and allow the use of non-standard statistics, making only weak assumptions about the data. With the availability of fast and inexpensive computing, their main limitation would be some lack of flexibility to work with arbitrary experimental designs. In this paper we report on results on approximate permutation methods that are more flexible with respect to the experimental design and nuisance variables, and conduct detailed simulations to identify the best method for settings that are typical for imaging research scenarios. We present a generic framework for permutation inference for complex general linear …

Arts & Sciences Graduate Student Theses and Dissertations
A primary goal of deaf education teacher preparation programs is to help students acquire a sufficient body of factual and pedagogical knowledge and retain it for future application while serving children with hearing loss and their families. One potential way to improve teacher preparation is through the implementation of retrieval practice, a strategy to promote learning and retention of material over time. This study examined whether retrieval practice could be used to improve learning in an authentic educational environment, using real content, and real materials. While this study provided important information regarding the implementation of retrieval practice authentic classrooms, it …

Discovering Behavioral Intervention: A Parent’S Interactive Guide To Aba, Richard Fleming, Carol Curtin, Cheryl A. Gray, Charles D. Hamad
Current Issues in Emerging eLearning
Autism spectrum disorders (ASD) affect one in 110 children in the U.S. Parents of children with ASD need clear and accurate information to communicate with professionals as they seek appropriate services, including applied behavior analysis (ABA) based intervention. Behavioral professionals can assist parents in this endeavor by recommending resources, including online courses. This paper describes the development and evaluation of an online course on ABA for parents of children with ASD. Parents completing a summative field test (N=21) made significant gains in knowledge and reported high levels of satisfaction. Implications include the potential for enhanced parent-professional collaboration in treatment decision-making.

Postoperative Non-Steroidal Anti-Inflammatory Drugs And Risk Of Bleeding In Pediatric Intracapsular Tonsillectomy, Jill N. D'Souza, Md, Julie P. Adelman, Bs, Richard J. Schmidt, Md, Heather C. Nardone, Md
Department of Otolaryngology - Head and Neck Surgery Presentations and Grand Rounds
Tonsillectomy with or without adenoidectomy is one of the most frequently performed surgeries in the United States, with over 500,000 performed annually. Post-tonsillectomy hemorrhage is one of the most feared complications; thus, medications that could increase the risk of postoperative bleeding traditionally have been avoided. With recent FDA guidelines encouraging a departure from codeine-based medications in pediatric patients undergoing tonsillectomy, we examined the use of ibuprofen for post-tonsillectomy pain control. The records of 449 children who underwent tonsillectomy and received ibuprofen for postoperative pain control were reviewed and compared to a cohort of 1731 children who received codeine for pain …
Injection Site Massage To Improve The Pharmacokinetics Of Aspart In Obese Adolescents With Diabetes, Hiwot Ketema Girma
Injection Site Massage To Improve The Pharmacokinetics Of Aspart In Obese Adolescents With Diabetes, Hiwot Ketema Girma
Yale School of Medicine Physician Associate Program Theses
The invention of rapid-acting insulin analogs, such as aspart, was a great step forward in achieving optimal control of blood glucose in patients with type 1 diabetes. Aspart’s action resembles the physiologic endogenous post-meal insulin action; however, the slow rate of absorption through subcutaneous tissue leads to a delay in the time to peak levels and action of pre-meal insulin injection and suboptimal control of postprandial blood glucose excursions. We propose that massaging the site of aspart injection will significantly accelerate insulin action and mitigate postprandial blood glycemic excursions. The study will investigate the effect of injection site massage on …

Quality control operates at different steps in translation to limit errors to approximately one mistranslated codon per 10,000 codons during mRNA-directed protein synthesis. Recent studies have suggested that error rates may actually vary considerably during translation under different growth conditions. Here we examined the misincorporation of Phe at Tyr codons during synthesis of a recombinant antibody produced in tyrosine-limited Chinese hamster ovary (CHO) cells. Tyr to Phe replacements were previously found to occur throughout the antibody at a rate of up to 0.7% irrespective of the identity or context of the Tyr codon translated. Despite this comparatively high mistranslation rate, …

Epichromatin, the surface of chromatin facing the nuclear envelope in an interphase nucleus, reveals a “rim” staining pattern with specific mouse monoclonal antibodies against histone H2A/H2B/DNA and phosphatidylserine epitopes. Employing a modified ChIP-Seq procedure on undifferentiated and differentiated human leukemic (HL-60/S4) cells, >95% of assembled epichromatin regions overlapped with Alu retrotransposons. They also exhibited enrichment of the AluS subfamily and of Alu oligomers. Furthermore, mapping epichromatin regions to the human chromosomes revealed highly similar localization patterns in the various cell states and with the different antibodies. Comparisons with available epigenetic databases suggested that epichromatin is neither “classical” heterochromatin nor highly …

Human Antimicrobial Peptides And Proteins, Guangshun Wang
Journal Articles: Pathology and Microbiology
As the key components of innate immunity, human host defense antimicrobial peptides and proteins (AMPs) play a critical role in warding off invading microbial pathogens. In addition, AMPs can possess other biological functions such as apoptosis, wound healing, and immune modulation. This article provides an overview on the identification, activity, 3D structure, and mechanism of action of human AMPs selected from the antimicrobial peptide database. Over 100 such peptides have been identified from a variety of tissues and epithelial surfaces, including skin, eyes, ears, mouths, gut, immune, nervous and urinary systems. These peptides vary from 10 to 150 amino acids …

The hypoxia inducible factors (Hifs) are evolutionarily conserved transcriptional factors that control homeostatic responses to low oxygen. In developing bone, Hif-1 generated signals induce angiogenesis necessary for osteoblast specification, but in mature bone, loss of Hif-1 in osteoblasts resulted in a more rapid accumulation of bone. These findings suggested that Hif-1 exerts distinct developmental functions and acts as a negative regulator of bone formation. To investigate the function of Hif-1α in osteoanabolic signaling, we assessed the effect of Hif-1α loss-of-function on bone formation in response to intermittent parathyroid hormone (PTH). Mice lacking Hif-1α in osteoblasts and osteocytes form more bone …

It is estimated that the divorce rate for marriages in which at least one party is a physician is 10-20% higher than the general population. Predicting the variables that contribute to marital satisfaction, identifying the skills associated with those behaviors and developing a residency curriculum to teach the skills to resident physicians is the focus of this study. Intimacy, conflict resolution and emotional expressiveness have all been identified as important aspects of marital satisfaction. We will determine how each of these variables contributes to marital satisfaction using multilevel logistic regression. The results will be used to develop a skills based …

Distress In Women With Ovarian Cancer, Judith Dellaripa
Theses and Dissertations
Clinicians and researchers know that women experience distress related to the diagnosis of and treatment for ovarian cancer. A review of the literature revealed that while there is interest in the topic, distress is inconsistently defined and measured. Women have been reported to have a variety of distress experiences including the challenges of late diagnosis and the treatment regimen, communication difficulties with healthcare providers, and concern about the effect of their diagnosis on their loved ones. Without information directly from women, assumptions predominate about what the experience is like and what they would find helpful from support persons. Women’s perceptions …

Avian influenza (AI), commonly known as bird flu, typically causes little or no harm to its wild waterfowl hosts. In 2003-2004, however, nine Asian countries reported unprecedented outbreaks of high-mortality AI in domestic poultry. This viral lineage, subtype H5N1, has spread to over 60 countries, and is considered endemic in at least four.
In India, H5N1 outbreaks were first recorded in 2006, and have continued each year since, reaching a total of 86 by 2012. Millions of India’s chickens and ducks have been culled in efforts to contain and eliminate the virus. India has declared itself free of AI a …

HIV-1 infection results in a chronic but incurable illness since long-term HAART can keep the virus to an undetectable level. However, discontinuation of therapy rapidly increases viral burden. Moreover, patients under HAART frequently develop various metabolic disorders and HIV-associated neuronal disease. Today, the main challenge of HIV-1 research is the elimination of the residual virus in infected individuals. The current HIV-1 diagnostics are largely comprised of serological and nucleic acid based technologies. Our goal is to integrate the nanotrap technology into a standard research tool that will allow sensitive detection of HIV-1 infection. This study demonstrates that majority of HIV-1 …

The etoposide-induced protein Ei24 was initially identified as a p53-responsive, proapoptotic factor, but no clear function has been described. Here, we use a nonbiased proteomics approach to identify members of the importin (IMP) family of nuclear transporters as interactors of Ei24 and characterize an IMPβ-binding-like (IBBL) domain within Ei24. We show that Ei24 can bind specifically to IMPβ1 and IMPα2, but not other IMPs, and use a mutated IMPβ1 derivative to show that Ei24 binds to the same site on IMPβ1 as the IMPα IBB. Ectopic expression of Ei24 reduced the extent of IMPβ1- or IMPα/β1-dependent nuclear protein import specifically, …

Effects Of An Apn-Led Amiodarone Clinic On Adherence To Recommended Monitoring Guidelines, Melissa Bartoszewicz
Evidence-Based Practice Project Reports
Evidence shows that patients receiving amiodarone therapy have not adhered to monitoring guidelines set forth by the Heart Rhythm Society. Uncertainty with responsibility for monitoring has led to the development of pharmacist-managed or multidisciplinary-managed outpatient amiodarone clinics. Some limitations have been identified in the pharmacist-managed outpatient clinics that may be overcome by advanced practice nurse (APN)-managed clinics. The purpose of this EBP project was to determine what effects an APN-led amiodarone clinic would have on adherence to amiodarone monitoring guidelines. Using the PICOT format, the clinical question was developed: Does enrollment in an amiodarone clinic compared with “usual care” change …

Recent immune correlates analysis from the RV144 vaccine trial has renewed interest in the role of non-neutralizing antibodies in mediating protection from infection. While neutralizing antibodies have proven difficult to induce through vaccination, extra-neutralizing antibodies, such as those that mediate antibody-dependent cellular cytotoxicity (ADCC), are associated with long-term control of infection. However, while several non-neutralizing monoclonal antibodies have been tested for their protective efficacy in vivo, no studies to date have tested the protective activity of naturally produced polyclonal antibodies from individuals harboring potent ADCC activity. Because ADCC-inducing antibodies are highly enriched in elite controllers (EC), we passively transferred …
