Drug targeting is an active area of research and nano-scaled drug delivery systems hold tremendous potential for the treatment of neoplasms. In this study, a novel cyclodextrin (CD)-based nanoparticle drug delivery system has been assembled and characterized for the therapy of folate receptor-positive [FR(+)] cancer. Water-soluble folic acid (FA)-conjugated CD carriers (FACDs) were successfully synthesized and their structures were confirmed by 1D/2D nuclear magnetic resonance (NMR), matrix-assisted laser desorption ionization time-of-flight mass spectrometer (MALDI-TOF-MS), high performance liquid chromatography (HPLC), Fourier transform infrared spectroscopy (FTIR), and circular dichroism. Drug complexes of adamatane (Ada) and cytotoxic doxorubicin (Dox) with FACD were readily …

Continuous Correction Of Differential Path Length Factor In Near-Infrared Spectroscopy, Tanveer Talukdar, Jason H. Moore, Solomon G. Diamond May 2013

Continuous Correction Of Differential Path Length Factor In Near-Infrared Spectroscopy, Tanveer Talukdar, Jason H. Moore, Solomon G. Diamond

Dartmouth Scholarship

In continuous-wave near-infrared spectroscopy (CW-NIRS), changes in the concentration of oxyhemoglobin and deoxyhemoglobin can be calculated by solving a set of linear equations from the modified Beer-Lambert Law. Cross-talk error in the calculated hemodynamics can arise from inaccurate knowledge of the wavelength-dependent differential path length factor (DPF). We apply the extended Kalman filter (EKF) with a dynamical systems model to calculate relative concentration changes in oxy- and deoxyhemoglobin while simultaneously estimating relative changes in DPF. Results from simulated and experimental CW-NIRS data are compared with results from a weighted least squares (WLSQ) method. The EKF method was found to effectively …

The Development And Applications Of The Hint Scoring Function: Exploring Colchicine-Site Anticancer Agents And Tautomerism, Chenxiao Da May 2013

The Development And Applications Of The Hint Scoring Function: Exploring Colchicine-Site Anticancer Agents And Tautomerism, Chenxiao Da

Theses and Dissertations

The overall aim of this work was to apply HINT, an empirical scoring function based on the understanding of hydrophobicity, to analyze and predict the binding affinities and biological activities of colchicine-site anticancer agents. The second, concurrent aim was to improve the scoring function by incorporating tautomerism within the modeling process. Our belief is that proper evaluation of tautomeric forms for small molecules will improve performance of virtual screening. The novel pyrrole-based compounds targeting the colchicine site were docked into the receptor using HINT as a rescoring function. Two distinct binding modes dictated by the size and shape of a …

Estimating The Impact Of Lost To Follow-Up On Breast Cancer Patients' Disease-Free Survival, Debbie Yan Qun Huang

Statistics

Background The 5-year survival rate for patients with breast cancer is much higher than patients with other types of cancer. Due to this longer survival period, breast cancer patients also tend to have increased rates of lost to follow-up, when compared to other cancers. When a patient becomes lost, the occurrence of distant metastasis cannot be reliably ascertained, unless the patient had a breast cancer-specific (BC) death. The impact of lost patients on recurrence rates and disease-free survival (DFS) was explored in breast cancer patients seen at the City of Hope from 1997 to 2012.

Methods Female breast cancer patients …

Mmp-3 Mediates Psychosine-Induced Globoid Cell Formation: Implications For Leukodystrophy Pathology, Kumiko Ijichi, Graham D. Brown, Craig S. Moore, Paige N. Winokur, Roberto Pagarigan, Stephen J. Crocker

UCHC Articles - Research

Globoid cell leukodystrophy (GLD) or Krabbe disease, is a fatal demyelinating disease attributed to mutations in the galactocerebrosidase (GALC) gene. Loss of function mutations in GALC result in accumulation of the glycolipid intermediate, galactosylsphingosine (psychosine). Due to the cytotoxicity of psychosine, it has been hypothesized that accumulated psychosine underlie the pathophysiology of GLD. However, the cellular mechanisms of GLD pathophysiology remain unclear. Globoid cells, multinucleated microglia/macrophages in the central nervous system (CNS), are a defining characteristic of GLD. Here we report that exposure of primary glial cultures to psychosine induces the expression and the production of matrix metalloproteinase …

Phenethylisothiocyanate Alters Site- And Promoter-Specific Histone Tail Modifications In Cancer Cells, Yi Liu, Suvobrata Chakravarty, Moul Dey

Chemistry and Biochemistry Faculty Publications

Site-specific histone modifications are important epigenetic regulators of gene expression. As deregulation of genes often results in complex disorders, corrective modulation of site-specific histone marks could be a powerful therapeutic or disease-preventive strategy. However, such modulation by dietary compounds and the resulting impact on disease risk remain relatively unexplored. Here we examined phenethylisothiocyanate (PEITC), a common dietary compound derived from cruciferous vegetables with known chemopreventive properties under experimental conditions, as a possible modulator of histone modifications in human colon cancer cells. The present study reports novel, dynamic, site-specific chemical changes to histone H3 in a gene-promoter-specific manner, associated with PEITC …

The Effect Of Skeletal Muscle Mass On Basal Metabolic Rate In College-Age Males 2013, Chase Vaughn

Master's Theses

There are many misconceptions within the fitness community. One such misconception is that an increase in muscle mass will have a large increase in basal metabolic rate, yet this may not be the best way for successful weight loss. The purpose of this study was to determine the strength of the correlation between skeletal muscle mass and basal metabolic rate. The hypothesis tested was that basal metabolic rate is strongly correlated with skeletal muscle mass. The participants were college-aged males (n =17). Skeletal muscle mass (SMM) was estimated using the skin-fold circumference model (SFCM): SMM (kg) = Ht (0.00744 CAG2 …

Abstract

Community building is an important concept for all healthcare providers to understand in order to implement effective health promotion and empower members of a community. The World Health Organization (WHO) states that community building involves action at three levels to improve health. These actions include the advancement of skills among practitioners, expansion of support and infrastructure for health promotion, and the development of cohesiveness and partnerships within communities. If implemented effectively, the benefits of community building include enhancing quality of life, promoting health and well being, empowering individuals, and creating sustainable health care organizations that conduct and maintain successful …
