The remodeling of connexin43-containing gap junctions (GJs), as defined by changes in size, distribution, or function, is a prominent feature of both the developing and diseased myocardium. During postnatal development of the mammalian ventricle, there is a progressive remodeling of GJs into intercalated disks. This change in GJ quantity and distribution is correlated with alterations in the velocity and anisotropy of impulse spread within differentiating myocardium. Orderly distribution of GJs is perturbed in many cardiac diseases, and reemergence of immature patterns of myocyte coupling may be a factor in the arrhythmogenic potential attending such pathologies. The seven mammalian isotypes of beta tubulin are strikingly similar in amino acid sequence. The differences in isotypic sequence, although small, are nonetheless conserved in evolution, which suggests that they may confer distinct functional roles. If so, such roles should be reflected in the selective expression of isotypes by cell type, or even in the sorting of isotypes to within-cell pools. Hair cells of the vestibular sensory epithelia each possess a kinocilium, a microtubule-based organelle that could represent a distinct microtubule compartment, separate from the extensive microtubule network in the soma. There are seven isotypic forms of the microtubule protein beta tubulin in mammals, but not all isotypes are synthesized in every cell type. In the adult organ of Corti, each of the five major cell types synthesizes a different subset of isotypes. Inner hair cells synthesize only betaI and betaII tubulin, while outer hair cells make betaI and betaIV tubulin. Only betaII and betaIV tubulin are found in inner and outer pillar cells, while betaI, betaII, and betaIV tubulin are present in Deiters cells, and betaI, betaII and betaIII tubulin are found in organ of Corti dendrites. BACKGROUND: Platelets are thought to play a role in a variety of inflammatory conditions in the lung, some of which may lead to fibrosis. In the current study we tested the hypothesis that whole platelets and platelet lysate can mediate remodelling of extracellular matrix in vitro by affecting fibroblast-mediated contraction of a collagen gel. We also sought to determine to what extent platelet-derived growth factor (PDGF) and transforming growth factor-beta (TGF-beta) contribute to this effect.

ABSTRACT

Peptide toxins with disulfide-stabilized structures have been used as molecular calipers to probe the outer vestibule structure of K channels. We want to apply this approach to the human ether-a-go-go-related gene (HERG) channel, whose outer vestibule is unique in structure and function among voltage-gated K channels. Our focus here is BeKm-1, a HERG-specific peptide toxin that can suppress HERG in the low nM concentration range. Although BeKm-1 shares the three-dimensional scaffold with the well-studied charybdotoxin, the two use different mechanisms in suppressing currents through their target K channels. This study was conducted at the Aga Khan University Hospital, Karachi, Pakistan to evaluate factors that affect outcome following revascularisation in patients with renovascular hypertension. We included all the patients diagnosed to have renovascular hypertension, confirmed by renal angiography, between July 1997 and September 2000. Of the total 15 patients, nine were males and six were females. Eleven patients received venous grafts, three received polytetraflouroethyline (PTFE) grafts while one patient underwent angioplasty and stenting. All were followed-up for a period of nine months (median) with the range from 2 to 84 months. A total of 33.3% of the study patients …

Wegener's Granulomatosis (WG) is a disorder characterized by necrotizing granulomatous vasculitis that primarily affects the upper and lower respiratory tracts and the kidneys. Although the cause is unknown, there is strong evidence that this is an autoimmune disease and immunosuppressive therapy with cyclophosphamide and corticosteroids efficiently relieves the symptoms and, prevents relapses. We report on four cases of WG that presented with unusual features. The first patient presented with vague joint pain, blackish discoloration of the skin and leg ulcers. In this paper, I review the impact of HIV/AIDS on families and development in sub-Saharan Africa. I begin by showing the magnitude of the problem and the factors that have led to rapid spread of HIV/AIDS in this sub-continent. I discuss gender inequality, poverty, social inequality, and globalization, and show how these facilitate the rapid spread of the epidemic. HIPAA - the Health Insurance Portability and Accountability act of 1996 is the largest government action in healthcare since Medicare. Since the passage of the HIPAA legislation, estimated cost of implementation has ranged from $3.8 billion to $45 billion for the implementation of the privacy rules. This study examines the cost on implementing HIPAA in private physicians' offices in South Carolina. The study selected the medical specialties of family practice, general practice, obstetrics & gynecology, and pediatrics. The study compares and analyzes cost of rural vs. urban physician practices, by specialty, and by type of entity when implementing privacy rules.

In the 2000 case Pegram v. Herdrich, the U.S. Supreme Court decided that administrators of health benefits plans covered by the Employee Retirement Income Security Act (ERISA) of 1974 do not have a fiduciary duty towards beneficiaries when making "mixed decisions" involving treatment and eligibility components. This thesis, Rethinking Pegram: Why ERISA Health Benefits Administrators Should Have a Comprehensive Legal Requirement of Fiduciary Duty, challenges the Court's opinion on grounds of legal precedent and public health ethics. Background: The purpose of this study is to review cases of renal cortical necrosis (RCN) which we came across during a period of five years, confirmed on needle biopsy specimens, with literature available so far.
Methods: All renal biopsy records between January 1996 to December 2001 at the Aga Khan University Hospital, Karachi, Pakistan were reviewed to identify patients with histologically proven renal cortical necrosis. Seven patients were also referred by the department of Nephrology, Lady Reading Hospital, Peshawar, Pakistan. Patients' medical records were reviewed for demographic, clinical, and follow up data.

Introduction: Vision loss resulting from retinal ischemia is associated with several systemic and ocular diseases. In the retina, ischemia is thought to lead to neuronal cell death by initiating a series of events that include oxidative stress, glutamate excitotoxicity, and intracellular Ca2+ overload. Attempts to prevent ischemic retinal degeneration have included the use of NMDA receptor antagonists, Ca2+ channel blockers, and antioxidants. These treatments have had limited success due to incomplete retinal protection and development of adverse side-effects. Knee injuries are among the most common injuries seen in a physical therapy clinic. Nearly all rehabilitation programs following a knee injury involve quadriceps muscle strengthening. One popular and safe method of quadriceps strengthening often used is the wall slide. The human cytomegalovirus-encoded chemokine receptor US28 induces arterial smooth muscle cell (SMC) migration; however, the underlying mechanisms involved in this process are unclear. We have previously shown that US28-mediated SMC migration occurs by a ligand-dependent process that is sensitive to proteintyrosine kinase inhibitors. We demonstrate here that US28 signals through the non-receptor protein-tyrosine kinases Src and focal adhesion kinase (FAK) and that this activity is necessary for US28-mediated SMC migration. In the presence of RANTES (regulated on activation normal T cell expressed and secreted), US28 stimulates the production of a FAK Src kinase complex. Protein tyrosine kinases (PTK) are key enzymes of mammalian signal transduction. For the fidelity of signal transduction, each PTK phosphorylates only one or a few proteins on specific Tyr residues. Substrate specificity is thought to be mediated by PTK–substrate docking interactions and recognition of the phosphorylation site sequence by the kinase active site. However, a substrate-docking site has not been determined on any PTK. C-terminal Src kinase (Csk) is a PTK that specifically phosphorylates Src family kinases on a C-terminal Tyr. NAD⁺-dependent DNA ligases are essentially restricted to eubacteria, thus constituting an attractive target in the development of novel antibiotics. Formin-homology-domain-containing proteins interact with Rho-family GTPases and regulate actin cytoskeleton organization and gene transcription. FHOD1 is a member of this family, interacts with Rac1 and induces transcription from the serum response element. In this study, we examined the effects of FHOD1 expression on cytoskeletal organization and function in mammalian cells. FHOD1 proteins were stably expressed in WM35 melanoma cells and NIH-3T3 fibroblasts. Cells expressing full-length FHOD1 demonstrated an elongated phenotype compared with vector-transfected cells and cells expressing a truncated FHOD1 (1-421) that lacks the conserved FH1 and FH2 domains. Usually, people's emotions arise from their perceptions of their circumstances-immediate, imagined, or remembered. This idea has been implicit in many philosophical treatments of emotions (e.g., in Aristotle, Spinoza, and even Descartes and James; see Ellsworth 1994a; Gardiner, Clark-Metcalf, & Beebe-Centa, 1980; Scherer, 2000) and explicit in some (e.g., Hume and Hobbes), and it is the central emphasis of current appraisal theories of emotion.
