Retiree/Part-Time Farmer Killed When Run Over By Bush Hog, Kentucky Injury Prevention And Research Center
Fatality Case Reports--Tractors/Logging
A 64-year-old male retiree and part-time farmer was killed when he fell from his tractor and was run over by the edge of the bush-hog attachment. The victim had been repairing a tractor tire earlier in the day. He was apparently trying to drive the tractor into a garage when he lost control and fell or was knocked from the tractor seat to the ground, falling between the tractor and the bush hog. The KY FACE investigator concluded that, in order to prevent similar occurrences, tractor operators should:
- Retrofit tractors with operator restraint systems (seatbelts) and Roll Over Protective Structures …

Transcriptional Activity Of Core Binding Factor-Alpha (Aml1) And Beta Subunits On Murine Leukemia Virus Enhancer Cores., Ari L. Zaiman, Amy F. Lewis, Barbara E. Crute, N. A. Speck, Jack Lenz
Dartmouth Scholarship
Core binding factor (CBF), also known as polyomavirus enhancer-binding protein 2 and SL3 enhancer factor 1, is a mammalian transcription factor that binds to an element termed the core within the enhancers of the murine leukemia virus family of retroviruses. The core elements of the SL3 virus are important genetic determinants of the ability of this virus to induce T-cell lymphomas and the transcriptional activity of the viral long terminal repeat in T lymphocytes. CBF consists of two subunits, a DNA binding subunit, CBF alpha, and a second subunit, CBF beta, that stimulates the DNA binding activity of CBF alpha. …

We identified a semidominant, chemically induced, mouse use mutation with a complex array of abnormal behaviors including bidirectional circling and hyperactivity, abnormal circadian rhythmicity and abnormal responses to light. In this report, we genetically and phenotypically characterized the circling/ waltzing component of the abnormal behavior. We mapped the locus controlling this trait by heterozygosity mapping of partially congenic lines carrying the mutagenized chromosome outcrossed to different inbred strains for three generations. Analysis of 68 PCR-based markers in 13 affected individuals indicated that the mutant locus, named Wheels (Whl), resides in the subcentromeric portion of mouse chromosome 4. The …

Learning Lessons From Sunk Costs, Brian H. Bornstein, Gretchin B. Chapman
Department of Psychology: Faculty Publications
Study participants rated the quality of several arguments for continuing an original plan in sunk cost situations in order to (a) avoid wasting resources, (b) learn to make better decisions, (c) punish poor decision making, and (d) appear consistent. The lesson-learning argument was perceived as most appropriate when adult teachers taught lessons to others, the original decision was carelessly made, or if it consumed comparatively more resources. Ratings of the lesson-learning argument were higher for teacher-learner than for adult-alone situations, regardless of whether the learner was a child or an adult. The implications for improving decision making and judging whether …

Altered Platelet Activating Factor Metabolism In Insulin Dependent Diabetes Mellitus, R A. Memon, S A. Saeed, A Jabbar, A Jafri, A H. Gilani, S Saleem, H Akbani
Department of Biological & Biomedical Sciences
Diabetes mellitus is associated with several abnormalities of platelet function. Recent studies have shown that the blood level of platelet activating factor (PAF), a potent inducer of platelet aggregation, is elevated in insulin dependent diabetes mellitus (IDDM) and remains unchanged in non-insulin dependent diabetes mellitus (NIDDM) patients. However, the mechanism of this increase in PAF levels has not been determined. In this study we have measured the activity of plasma PAF acetylhydrolase (an enzyme that regulates PAF levels) and lipoprotein levels in control subjects and diabetic patients. The data presented show that plasma PAF acetylhydrolase activity is significantly decreased in …

Increased Alpha 1(I) Procollagen Gene Expression In Tight Skin (Tsk) Mice Myocardial Fibroblasts Is Due To A Reduced Interaction Of A Negative Regulatory Sequence With Ap-1 Transcription Factor., Neena Philips, Reza I. Bashey, Sergio A. Jimenez
Department of Medicine Faculty Papers
The TSK mouse, a model of fibrosis, displays exaggerated connective tissue accumulation in skin and visceral organs including the heart. To study the mechanisms of myocardial fibrosis in TSK mice, we established several strains of TSK mice myocardial fibroblasts in culture and examined the regulation of collagen gene expression in these cells. These strains displayed increased collagen gene expression in comparison with myocardial fibroblasts established from normal mice. On an average, the TSK myocardial fibroblast cultures showed a 4-fold increase in collagen synthesis and 4.4- and 3.6-fold increases, respectively, in alpha 1(I) and alpha 1(III) collagen mRNA steady state levels. …

Protein-Peptide Interactions Analyzed With The Yeast Two-Hybrid System, Meijia Yang, Zining Wu, Stanley Fields
Protein-Peptide Interactions Analyzed With The Yeast Two-Hybrid System, Meijia Yang, Zining Wu, Stanley Fields
Dartmouth Scholarship
The yeast two-hybrid system was used to screen a library of random peptides fused to a transcriptional activation domain in order to identify peptides capable of binding to the retinoblastoma protein (Rb). Seven peptides were identified, allof which contain the Leu-X-Cys-X-Glu motif found in Rb-binding proteins, although their activity in the yeast assay variedover a 40-fold range. Mutagenesis of the DNA encoding two of these peptides followed by screening in the two-hybrid systemallowed the delineation of residues apart from the invariant Leu, Cys and Glu that affect binding to Rb. Binding affinities of a peptide and one of its variants …

Factors Which Predict The Initiation And Use Of Alcohol In Rural Adolescents, Charlotte Swint
Masters Theses & Specialist Projects
The purpose of this research was to determine whether a selected group of factors are significant when trying to predict the use of alcohol by adolescents who live in rural areas. This research is important because most of the studies concerning drug and alcohol use in adolescents have focused on adolescents who live in urban areas. The results of such studies may not be generalizable to rural populations. The factors investigated included the following: availability of alcohol, peer influence, parental influence, tobacco use, gender, and grade level. These factors were chosen based on their inclusion in four prominent theories regarding …

Partnerships Between Community Residents And Professionals: Issues Of Power And Social Class Across The Lifespan Of Neighbourhood Organizations, Barbara Derksen, Geoffrey Nelson
Psychology Faculty Publications
We described a qualitative sutdy of four neighbourhood centres jointly created by a sponsoring agency and community members to prevent problems in the lives of chidlren and adults in these neighbourhoods. Individual and focus group interviews with staff, community members, and other agency workers were used to develop an understanding of issues involved at various stages across the lifespan of these settings. Two tensions noted in this process were between community members and professionals from the sponsoring organization and between lower-class and middle-class community members. We discuss the implications of these findigns for practice in community development.

Athletic Identity And Sport Orientation Of Adolescent Swimmers With Disabilities, Jeffrey J. Martin, Carol Adams Mushett, Kari L. Smith
Kinesiology, Health and Sport Studies
Measures of athletic identity and sport orientation, developed from self-schema theory, social role theory, and achievement motivation theory, were used to examine international adolescent swimmers with disabilities. The multidimensional Athletic Identity Measurement Scale (Brewer, Van Raalte, & Linder, 1993) was used to assess self-identity, social identity, exclusivity, and negative affectivity. The Sport Orientation Questionnaire (Gill & Deeter, 1988) measured competitiveness, win orientation, and goal orientation. Swimmers reported (a) a strong self-identity, (b) a moderate to strong social identity, (c) negative affectivity with lower levels of exclusivity, (d) strong competitiveness and goal orientation, and (e) moderate win orientation. Self-identity was correlated …

A Change Of Il-2 And Il-4 Production In Patients With Helicobactor Pylori Infection, X. G. Fan, Javed Yakoob, X. J. Fan, P. W. N. Keeling
Section of Gastroenterology
Helicobactor pylori is the most common cause of gastroduodenal inflammation. However, the exact immune pathogenesis is not fully understood. To look for evidence of the immunological mechanism in H. pylori associated disease, we measured cytokine interleukin-2 (IL-2) and IL-4 levels produced by peripheral blood lymphocytes (PBL) and gastric biopsies in 20 subjects with or without H. pylori infection. H. pylori can stimulate IL-2 and IL-4 production from PBL in H. pylori negative as well as H. pylori positive individuals. The spontaneous IL-2 production by PBL and gastric biopsies was greater (p < 0.0025, <0.001)in H. pylori negative individuals than that in H. pylori infected patients. Increased IL-4 levels from PBL in H. pylori infected patients were found in the presence of H. pylori (p < 0.0025). An increased spontaneous production of IL-4 from gastric biopsies was also observed in H. pylori infected patients (p < 0.025). In conclusion, an enhanced type 2 cytokine production was observed in H. pylori infected patients, which may be responsible for H. pylori chronic infection.

Broth microdilution MICs were determined for pairs of strains isolated from five AIDS patients with polyclonal Mycobacterium avium infection. Four (80%) of the five patients were infected simultaneously with strains having different antimicrobial susceptibility patterns. These findings have implications for the interpretation of susceptibility data in M. avium prophylaxis and treatment trials.
