Publications and Research
Inhibitory components in myelin are largely responsible for the lack of regeneration in the mammalian CNS. Myelin-associated glycoprotein (MAG), a sialic acid binding protein and a component of myelin, is a potent inhibitor of neurite outgrowth from a variety of neurons both in vitro and in vivo. Here, we show that MAG’s sialic acid binding site is distinct from its neurite inhibitory activity. Alone, sialic acid–dependent binding of MAG to neurons is insufficient to effect inhibition of axonal growth. Thus, while soluble MAG-Fc (MAG extracellular domain fused to Fc), a truncated form of MAG-Fc missing Ig-domains 4 and 5, MAG(d1-3)-Fc, …

All 50 states have laws requiring mental health and other professionals to report suspected maltreatment. Unfortunately, many professionals who are mandated to report suspicions of child maltreatment often fail to recognize potential maltreatment or fail to report their suspicions. The present study examines several factors that may influence identification and reporting of child maltreatment. Subjects were licensed psychologists in the Midwest and certified Masters social workers in Nebraska. Child maltreatment included neglect, physical abuse, psychological maltreatment, and sexual abuse. Characteristics associated with the family or “case” (race, socioeconomic status of family, age of victim, type of maltreatment) were manipulated and …

The purpose of this study was to determine the relative importance of the metabolic effects of insulin for diabetes prevention by administering insulin or an inactive insulin analog by daily subcutaneous injections to prediabetic mice. A recombinant monomeric human insulin analog, which does not bind to the insulin receptor as a consequence of an alteration of a single amino acid at position 25 of the B chain, was shown to be equally effective at diabetes prevention as was intact insulin. In contrast to native insulin, the insulin analog did not cause hypoglycemia after subcutaneous injection. The insulin analog, however, protected …

Agriculture has one of the highest occupational fatality rates of all industries in the United States (1). Tractors and other types of agricultural equipment account for a large proportion of these fatalities, and farm-tractor rollovers account for approximately 130 work-related deaths each year in the United States (2). Although rollover protective structures (ROPS) are effective in protecting tractor operators from fatal injuries during rollovers (3-5), most tractors in the United States are not equipped with ROPS (4-7). Beginning in 1985, tractor manufacturers in the United Sates agreed to sell only tractors with ROPS; however, many older tractors without ROPS remain …

Mitotic Spindle Poles Are Organized By Structural And Motor Proteins In Addition To Centrosomes, Tirso Gaglio, Mary A. Dionne, Duane A. Duane A. Compton

The focusing of microtubules into mitotic spindle poles in vertebrate somatic cells has been assumed to be the consequence of their nucleation from centrosomes. Contrary to this simple view, in this article we show that an antibody recognizing the light intermediate chain of cytoplasmic dynein (70.1) disrupts both the focused organization of microtubule minus ends and the localization of the nuclear mitotic apparatus protein at spindle poles when injected into cultured cells during metaphase, despite the presence of centrosomes. Examination of the effects of this dynein-specific antibody both in vitro using a cell-free system for mitotic aster assembly and in …

Significant numbers of myocytes die by apoptosis during myocardial infarction. The molecular mechanism of this process, however, remains largely unexplored. To facilitate a molecular genetic analysis, we have developed a model of ischemia-induced cardiac myocyte apoptosis in the mouse. Surgical occlusion of the left coronary artery results in apoptosis, as indicated by the presence of nucleosome ladders and in situ DNA strand breaks. Apoptosis occurs mainly in cardiac myocytes, and is shown for the first time to be limited to hypoxic regions during acute infarction. Since hypoxia- induced apoptosis in other cell types is dependent on p53, and p53 is …

Minimizing Infant Exposure To And Risks From Medications While Breastfeeding, Sandra Banta-Wright
Faculty Publications - College of Nursing
The advantages of breastfeeding to the mother and newborn are many. Lactating mothers frequently ask about the safety of taking medications and the risk to their newborn. It is well established that all drugs are excreted into breast milk. However, most medications appear in only small amounts within the breast milk. With the availability of numerous resources on drug use while breastfeeding, a medication can be identified as contraindicated or compatible with breastfeeding. By understanding the anatomy of the breast, principles of lactation, and drug passage into breast milk, an approach to minimize the transfer of the medications in the …

Attitudes Toward Sexuality And Sexual Behaviors Of Asian-American Adolescents: Implications For Risk Of Hiv Infection, Connie S. Chan
Attitudes Toward Sexuality And Sexual Behaviors Of Asian-American Adolescents: Implications For Risk Of Hiv Infection, Connie S. Chan
Institute for Asian American Studies Publications
Until 1990, Asian Americans represented an ethnic minority group that was perceived to be at lower risk than African Americans or Hispanics/Latinos for HIV infection, the presumed causal agent for AIDS. Reasons cited for this perception include behavioral differences in intravenous drug use, sexual behavioral habits, and underidentification of AIDS cases. However, in urban areas such as San Francisco, Toronto, New York, Boston, Los Angeles, and Seattle, where Asians have immigrated and settled in large numbers, cases of HIV infection and AIDS have begun to increase dramatically, perhaps reflecting the rise in the number of AIDS cases in Asia. In …

Potential Role Of Tnfα And Lipoprotein Lipase As Candidate Genes For Obesity, Philip A. Kern
Potential Role Of Tnfα And Lipoprotein Lipase As Candidate Genes For Obesity, Philip A. Kern
Clinical and Translational Science Faculty Publications
To maintain body weight, metabolic efficiency was promoted during evolution; two candidate genes for body weight regulation are lipoprotein lipase (LPL) and tumor necrosis factor-α [TNFα). Human fat cells do not synthesize lipid, but rely on LPL-mediated plasma triglyceride hydrolysis. Adipose LPL is elevated in obesity. Following weight loss, LPL is elevated further, suggesting attempts to maintain lipid stores during fasting and to replenish lipid stores during refeeding. Muscle LPL is regulated inversely to adipose LPL. Thus, an increased adipose/muscle LPL ratio would partition dietary lipid into adipose tissue and would explain some of the variability in weight gain when …

Thyroid hormone plays an essential role in mammalian brain maturation and function, in large part by regulating the expression of specific neuronal genes. In this tissue, the type 2 deiodinase (D2) appears to be essential for providing adequate levels of the active thyroid hormone 3,5,3′-triiodothyronine (T3) during the developmental period. We have studied the regional and cellular localization of D2 mRNA in the brain of 15-day-old neonatal rats. D2 is expressed in the cerebral cortex, olfactory bulb, hippocampus, caudate, thalamus, hypothalamus, and cerebellum and was absent from the white matter. At the cellular level, D2 is expressed predominantly, if not …

A novel polymorphism in the extracellular domain 2 (EC2) of FcgammaRIIIA affects ligand binding by natural killer (NK) cells and monocytes from genotyped homozygous normal donors independently of receptor expression. The nonconservative T to G substitution at nucleotide 559 predicts a change of phenylalanine (F) to valine (V) at amino acid position 176. Compared with F/F homozygotes, FcgammaRIIIa expressed on NK cells and monocytes in V/V homozygotes bound more IgG1 and IgG3 despite identical levels of receptor expression. In response to a standard aggregated human IgG stimulus, FcgammaRIIIa engagement on NK cells from V/V (high-binding) homozygotes led to a larger …

Recommendations For Elimination Of Bovine Tuberculosis In Free-Ranging White-Tailed Deer In Michigan, K. L. Cool, James Haveman, Dan Wyant
Michigan Bovine Tuberculosis Bibliography and Database
A significant infection rate of bovine TB in the deer population of the northeastern lower peninsula poses a potential risk to several important values including public health, United States Department of Agriculture (U.S.D.A.) TB-free accreditation for Michigan cattle, wildlife health, wildlife-related recreation and tourism and economic stability in several sectors. A risk assessment study by the U.S. D.A. Centers for Epidemiology and Animal Health (Fort Collins, CO) predicted that if no changes were made in the management of the affected free-ranging deer population, the TB prevalence (compared to the current prevalence of 2.3%). Although the current annual risk of TB …

Analysis Of Bovine Herpesvirus 1 Transcripts During A Primary Infection Of Trigeminal Ganglia Of Cattle, Luis M. Schang, Clinton J. Jones
School of Veterinary and Biomedical Sciences: Faculty Publications
During an infection of nonneuronal cells, bovine herpesvirus 1 (BHV-1) gene expression proceeds in a well-defined cascade. Products of immediate-early (IE) genes are expressed first, and they activate expression of early (E) and late (L) genes. Although the same cascade is assumed to occur during an infection of neurons in trigeminal ganglia (TG) of cattle, no experimental data is available to support this hypothesis. Consequently, we analyzed BHV-1 gene expression in bovine TG at 1, 2, 4, 7, and 15 days postinfection (dpi). Infectious virus was detected in ocular swabs from 1 to 7 dpi but not 15 dpi. By …

Intimacy Deficits, Fear Of Intimacy, And Loneliness Among Sexual Offenders, Kurt M. Bumby, David J. Hansen
Department of Psychology: Faculty Publications
Intimacy deficits and loneliness recently have been theorized as influential factors in the etiology and maintenance of sexually offending behaviors, although to date there has been a lack of empirical research conducted to address this proposition. The present study examined intimacy defi cits, fear of intimacy, and loneliness among intrafamilial child molesters, rapists, nonsexually offending inmates, and a community sample of adult males. The child molesters and rapists reported greater overall intimacy deficits than did the nonsexually offending inmates and community controls, with rapists reporting the greatest intimacy deficits. Fear of intimacy was found to be a particularly salient characteristic …

Sqt1, Which Encodes An Essential Wd Domain Protein Of Saccharomyces Cerevisiae, Suppresses Dominant-Negative Mutations Of The Ribosomal Protein Gene Qsr1., Dominic P. Eisinger, Frederick A. Dick, Elke Denke, Bernard L. Trumpower
Dartmouth Scholarship
QSR1 is an essential Saccharomyces cerevisiae gene, which encodes a 60S ribosomal subunit protein required for joining of 40S and 60S subunits. Truncations of QSR1 predicted to encode C-terminally truncated forms of Qsr1p do not substitute for QSR1 but do act as dominant negative mutations, inhibiting the growth of yeast when expressed from an inducible promoter. The dominant negative mutants exhibit a polysome profile characterized by 'half-mer' polysomes, indicative of a subunit joining defect like that seen in other qsr1 mutants (D. P. Eisinger, F. A. Dick, and B. L. Trumpower, Mol. Cell. Biol. 17:5136-5145, 1997.) By screening a high-copy …

Increased Focal Adhesion Kinase- And Urokinase-Type Plasminogen Activator Receptor-Associated Cell Signaling In Endothelial Cells Exposed To Asbestos, A Barchowsky, B M. Lannon, L C. Elmore, M D. Treadwell
Dartmouth Scholarship
Exposure of low-passage endothelial cells in culture to nonlethal amounts of asbestos, but not refractory ceramic fiber-1, increases cell motility and gene expression. These changes may be initiated by the fibers mimicking matrix proteins as ligands for receptors on the cell surface. In the present study, 1- to 3-hr exposures of endothelial cells to 5 mg/cm2 of chrysotile asbestos caused marked cell elongation and motility. However, little morphological change was seen when chrysotile was added to cells pretreated with either mannosamine to prevent assembly of glycophosphatidylinositol (GPI)-anchored receptors or with herbimycin A to inhibit tyrosine kinase activity. Affinity purification of …

Situation awareness (SA) is most commonly defined as "the perception of the elements in the environment within a volume of time and space, the comprehension of their meaning, and the projection of their status in the near future" (Endsley, 1995, p. 36). While researching this construct, we were continuously faced with several terms that, like SA, have themselves never been uniformly defined. For example, the following terms have, in some way, been associated with the construct known as SA: device model, domain model, mental model, script, schema, and "the picture" among others (c.f., Mogford, 1994; Endsley, 1995; Garland & Hopkin, …
