Chlamydia Trachomatis Infection In The Female Reproductive Tract Of The Rat: Influence Of Progesterone On Infectivity And Immune Response, Charu Kaushic, Andrew D. Murdin, Brian J. Underdown, Charles R. Wira
Dartmouth Scholarship
As the most common cause of sexually transmitted disease in women, chlamydial infections can lead to pelvic inflammatory disease, infertility, and ectopic pregnancy. To better understand the role played by sex hormones in modulating the immune response of the genital tract to microbial infections, we have developed a rat model to study Chlamydia trachomatis infection. Inbred female Lewis rats were primed with progesterone and inoculated by intrauterine instillation of C. trachomatis (mouse pneumonitis strain MoPn) into each uterine horn. When infected animals were examined for the presence of chlamydial antigens 14 days postinfection, both the uterus and vagina were found …

Development Of A Blocking Enzyme-Linked Immunosorbent Assay For Detection Of Serum Antibodies To O157 Antigen Of Escherichia Coli., W Laegreid, M Hoffman, J Keen, R Elder, J Kwang
Manuscripts, Articles, Book Chapters and Other Papers
The O157 antigen of Escherichia coli shares structural elements with lipopolysaccharide (LPS) antigens of other bacterial species, notably Brucella abortus and Yersinia enterocolitica 09, a fact that confounds the interpretation of assays for anti-O157 antibodies. To address this problem, a blocking enzyme-linked immunosorbent assay (bELISA) was designed with E. coli O157:H7 LPS as the antigen and a monoclonal antibody specific for E. coli O157, designated 13B3, as the competing antibody. The bELISA had equivalent sensitivity to, and significantly higher specificity than, the indirect ELISA (iELISA), detecting anti-O157 antibodies in sera from cattle experimentally inoculated with O157:H7. Only 13% of sera …

Subcellular Localization Of Mineralocorticoid Receptors In Living Cells: Effects Of Receptor Agonists And Antagonists, Géza Fejes-Tóth, David Pearce, Anikó Náray-Fejes-Tóth
Dartmouth Scholarship
Results on the subcellular localization of the mineralocorticoid receptor (MR) have been controversial. To determine the subcellular distribution and trafficking of the MR in living cells after binding of agonists and antagonists, we expressed a MR-green fluorescent protein (GFP) chimera in mammalian cells lacking endogenous MR. The GFP-tagged MR (GFP-MR) remained transcriptionally active, as determined in cotransfection experiments with the MR-responsive reporter, TAT3-LUC. The subcellular localization of GFP-MR was monitored by fluorescence time-lapse microscopy. In the absence of hormone, MR was present both in the cytoplasm and nucleus. Aldosterone induced a rapid nuclear accumulation of the MR. Aldosterone-bound GFP-MR was …

Predicting Prognosis In Lung Cancer: Use Of Proliferation Marker, Ki67 Monoclonal Antibody, I N. Soomro, J Holmes, W F. Whimster
Department of Pathology and Laboratory Medicine
An investigation was carried out to assess the prognostic significance of proliferation marker Ki67 in a group of lung cancer patients treated by surgery (limited disease). Tissue was not available for Ki67 immunostaining in inoperable group. The diagnosis is established by bronchial biopsy which does not carry enough tissue for frozen section and counting. This study is supplemented by estimating the prognostic significance of histological sub-types in the operable group and in a group of inoperable patients with extensive disease. These are usually treated by radiotherapy and/or chemotherapy. In all, 267 patients were studied including 105 treated by surgery. These …

Accuracy Of The Ovulon Fertility Monitor To Predict And Detect Ovulation, Richard J. Fehring, William D. Schlaff
College of Nursing Faculty Research and Publications
The purpose of this pilot study was to correlate the three biologic markers of the Ovulon fertility monitor (a long-term predictive peak about 6 days before ovulation, a short-term predictive peak about 1 day before ovulation, and a nadir at the time of ovulation) with the peak in cervical mucus and the luteinizing hormone (LH) surge in the urine. Ten volunteer subjects (mean age 30.2 years) monitored their cervical-vaginal mucus, the surge of LH in the urine with a home assay test, and their vaginal electrical readings (with Ovulon monitors) on a daily basis for one to four menstrual cycles. …

To determine the developmental periods, fecundity and survival of Amblyomma variegatum and Boophilus decoloratus and the effect of host resistance, a study was carried out in the field at Abemossa ranch in Ethiopia using three breeds of cattle: Arssi, Baran and Baran X Friesian crossbreed. The investigation took place between October 1991 and September 1992. The developmental periods of both tick species were recorded. The hatching periods of larvae, engorgement, larvo-nymphal and nympho-adult moulting periods during various climatic seasons in shaded and dry areas were also recorded. While the engorgement periods recorded on the animal were relatively constant for each …

Repeated Testing In Eyewitness Memory: A Means To Improve Recall Of A Negative Emotional Event, Brian H. Bornstein, Lesley M. Liebel, Nikki C. Scarberry
Department of Psychology: Faculty Publications
Participants viewed either a violent, arousing film or a non-violent, control version of the same film. After viewing the film, they made three successive attempts to recall details of the event. Participants who were exposed to the negative emotional event were better than control participants at recalling details of the event itself, but they were worse at recalling details that preceded or followed the violence. Both groups of participants recalled significantly more information over successive recall attempts, suggesting that memory impairment due to arousal can be alleviated by repeated testing. Repeated testing was also associated with a small but reliable …

Fornix Lesions Can Facilitate Acquisition Of The Transverse Patterning Task: A Challenge For "Configural" Theories Of Hippocampal Function., T J Bussey, E Clea Warburton, J P Aggleton, J L Muir
Brain and Mind Institute Researchers' Publications
Configural theories of hippocampal function predict that hippocampal dysfunction should impair acquisition of the transverse patterning task, which involves the concurrent solution of three discrimination problems: A+ versus B-; B+ versus C-; and C+ versus A-. The present study tested this prediction in rats using computer-graphic stimuli presented on a touchscreen. Experiment 1 assessed the effects of fornix lesions when the three problems were introduced sequentially (phase 1: A+ vs B-; phase 2: A+ vs B-, B+ vs C-; phase 3: A+ vs B-, B+ vs C-, C+ vs A-). Fornix lesions significantly facilitated acquisition of the complete transverse patterning …

The Greatest Memory Teacher Ever: Review Of Memory’S Ghost: The Nature Of Memory And The Strange Tale Of Mr. M., By Philip J. Hilts, Brian H. Bornstein
Department of Psychology: Faculty Publications
Although psychologists and neurologists have been familiar with the case of H.M. for years, this small volume makes his story—and what it can teach us about the functioning of memory—accessible to a much wider audience. Hilts has done a good job of piecing together H.M.’s early life, in the absence of any living relatives and all but a few written records. He recreates the life of a very normal youth in the 1930s and 1940s; normal, that is, until H.M.’s first epileptic seizure. In 1954, after several other treatments for the epilepsy had failed, H.M. (at age 27) had a …

The repellency of the essential oil of the previously reported anti- tick pasture shrub Gynandropsis gynandra and identified constituents of the oil were evaluated against the livestock tick, Rhipicephalus appendiculatus. In a tick-climbing repellency bioassay, the oil of G. gynandra exhibited a repellency effect which at the highest treatment levels was higher than that of the commercial arthropod repellent N,N-diethyltoluamide. Twenty eight compounds were identified in the G. gynandra oil by GC, GC-MS and coinjection with authentic samples. Carvacrol was found to occur in largest quantity (29.2%), followed by trans-phytol (24.0%), linalool (13.3%), trans-2- methylcyclopentanol (7.2%) and β-caryophyllene (4.4%). m-Cymene, …

Bordetella Holmesii-Like Organisms Associated With Septicemia, Endocarditis, And Respiratory Failure, Perry J. Severance, Yi-Wei Tang, Marlene K. Hopkins, Christopher P. Kolbert, Paul A. Hartley, David H. Persing
Articles
We recovered an unusual bacterial strain from blood or sputum of three patients with septicemia, endocarditis, and/or respiratory failure. The three isolates were thin, curved, gram-negative, light brown, pigment-producing bacilli with variable catalase activity. They were asaccharolytic, oxidasenegative, nonmotile, and fastidious. Identification was not possible on the basis of these characteristics alone or in combination with cellular fatty acid profiles. Nucleic acid amplification and sequence analysis of the 16S rRNA gene revealed that all three isolates were identical and most closely related to the emerging pathogen Bordetella holmesii, diverging from the published sequence at three nucleotide positions (99.8% similarity). …

Site Visit To Northern California — Risk, Accountability, And Staying Power In Next-Generation Managed Care, Lisa Sprague, Sandra Foote
National Health Policy Forum
This three-day site visit looked at private health insurance and delivery of care in a market notable for high managed care penetration, relatively low premium costs, and sophisticated provider groups. Federal congressional and regulatory staff heard from health insurance purchasers (both public and private), health plans, hospital systems, and physician groups about the changing relationships and accountabilities in this market and the ways various organizations were positioning themselves to survive the turmoil and emerge the stronger for it. Participants also heard from two county health programs about how competition and realignment in the private market were impacting safety-net services for …

Health Care Quality: From Data To Accountability, Mary Darby
National Health Policy Forum
This paper discussed the evolution of quality measurement and reporting activities as well as efforts to develop accountability for delivering high-quality care. It examined (a) initiatives in the professional arena, such as accreditation and performance measurement by independent accrediting bodies; (b) activities of market proponents, who have preferred to concentrate on making data available to purchasers and consumers that would enable them to choose high- quality plans and providers; and (c) attempts by the public sector to secure quality through regulation and oversight. An exploration of the successes, limitations, overlaps, and gaps in quality assurance initiatives, the paper was published …

In this study, adult volunteers were fed tcpA and mshA deletion mutants of V. cholerae O139 strain CVD 112 to determine the role of toxin-coregulated pili (TCP) and mannose-sensitive hemagglutinin (MSHA) in intestinal colonization. Eight of 10 volunteers who received CVD 112 or CVD 112 ΔmshA shed the vaccine strains in their stools; the geometric mean peak excretion for both groups was 1.4 × 105 CFU/g of stool. In contrast, only one of nine recipients of CVD 112 ΔtcpA shed vibrios in his stool (P < 0.01); during the first 24 h after inoculation, 3 × 102 CFU/g was recovered from this volunteer. All recipients of CVD …

Effects Of First And Second Trimester Vaginal Bleeding On Pregnancy Outcome, S A. Karim, I Bakhtawar, A T. Butta, M Jalil
Department of Obstetrics & Gynaecology
The effects of vaginal bleeding during first and second trimester on pregnancy outcomes was assessed in a hospital-based population of 268 non-diabetic women. The group of non-bleeders comprised 173 females whereas, there were 71 females with first and 24 with second trimester bleeding. Fetal loss (abortion) occurred in 34% of first trimester and 25% of second trimester bleeders. Low birth weight and preterm delivery were significantly associated with second trimester haemorrhage. The results suggest that first and second trimester vaginal bleeding correlates with adverse infant outcomes.

Homotypic vacuole fusion in yeast requires Sec18p (N-ethylmaleimide-sensitive fusion protein [NSF]), Sec17p (soluble NSF attachment protein [alpha-SNAP]), and typical vesicle (v) and target membrane (t) SNAP receptors (SNAREs). We now report that vacuolar v- and t-SNAREs are mainly found with Sec17p as v-t-SNARE complexes in vivo and on purified vacuoles rather than only transiently forming such complexes during docking, and disrupting them upon fusion. In the priming reaction, Sec18p and ATP dissociate this v-t-SNARE complex, accompanied by the release of Sec17p. SNARE complex structure governs each functional aspect of priming, as the v-SNARE regulates the rate of Sec17p release and, …
