Integronlike Structures In Campylobacter Spp. Of Human And Animal Origin, Brigid Lucey, D. Crowley, P. Moloney, B. Cryan, M. Daly, F. O'Halloran, E. J. Threlfall, S. Fanning

Department of Biological Sciences Publications

Resistance to antimicrobial agents used to treat severe Campylobacter spp. gastroenteritis is increasing worldwide. We assessed the antimicrobial resistance patterns of Campylobacter spp. isolates of human and animal origin. More than half (n = 32) were resistant to sulphonamide, a feature known to be associated with the presence of integrons. Analysis of these integrons will further our understanding of Campylobacter spp. epidemiology.

Naturally Occurring Tap-Dependent Specific T-Cell Tolerance For A Variant Of An Immunodominant Retroviral Cytotoxic T-Lymphocyte Epitope, Victor Kim, Jonathan W. Yewdell, William R. Green Jan 2000

Naturally Occurring Tap-Dependent Specific T-Cell Tolerance For A Variant Of An Immunodominant Retroviral Cytotoxic T-Lymphocyte Epitope, Victor Kim, Jonathan W. Yewdell, William R. Green

Dartmouth Scholarship

Upon immunization and restimulation with tumors induced by the endogenous AKR/Gross murine leukemia virus (MuLV), C57BL/6 mice generate vigorous H-2K(b)-restricted cytotoxic T-lymphocyte (CTL) responses to a determinant (KSPWFTTL) derived from the p15E transmembrane portion of the viral envelope glycoprotein. By contrast, the highly homologous determinant RSPWFTTL, expressed by tumor cells induced by Friend/Moloney/Rauscher (FMR) MuLV, is not immunogenic, even when presented to the immune system as vaccinia virus-encoded cytosolic or endoplasmic reticulum (ER)-targeted minigene products. Such minigene products are usually highly immunogenic since they bypass the need for cells to liberate the peptide or transport the peptide into the ER …

Conditioned Place Preference: What Does It Add To Our Preclinical Understanding Of Drug Reward?, M. T. Bardo, Rick A. Bevins

Department of Psychology: Faculty Publications

Rationale: Among the various experimental protocols that have been used to measure drug reward in laboratory animals, conditioned place preference (CPP) has been one of the most popular. However, a number of controversial issues have surrounded the use of this experimental protocol.
Objective: The present review provides a theoretical overview of some critical issues relevant to CPP. The advantages and limitations of CPP are also covered.
Results: Based on modern and traditional theoretical formulations of Pavlovian conditioning, CPP appears to reflect a preference for a context due to the contiguous association between the context and a drug stimulus. Within this …

Reshaping Ahcs' Role In Biomedical Research, Karen Matherlee Jan 2000

Reshaping Ahcs' Role In Biomedical Research, Karen Matherlee

National Health Policy Forum

This issue brief explores the reconfiguration of academic health centers (AHCs) in response to health marketplace and other pressures. It reviews four roles AHCs play in medical innovation: (a) development of new drugs, devices, diagnostic techniques, and therapeutic procedures; (b) adoption of new technologies, instruments, and drugs; (c) evaluation of new technologies; and (d) assessment of the need for new modalities and monitoring of their initial uses. The paper examines ways in which these roles are enhanced or threatened by evolving economic forces in the public and private sectors.

Phosphorylation Of The Neurospora Clock Protein Frequency Determines Its Degradation Rate And Strongly Influences The Period Length Of The Circadian Clock, Yi Liu, Jennifer Loros, Jay C. Dunlap Jan 2000

Phosphorylation Of The Neurospora Clock Protein Frequency Determines Its Degradation Rate And Strongly Influences The Period Length Of The Circadian Clock, Yi Liu, Jennifer Loros, Jay C. Dunlap

Dartmouth Scholarship

Under free running conditions, FREQUENCY (FRQ) protein, a central component of the Neurospora circadian clock, is progressively phosphorylated, becoming highly phosphorylated before its degradation late in the circadian day. To understand the biological function of FRQ phosphorylation, kinase inhibitors were used to block FRQ phosphorylation in vivo and the effects on FRQ and the clock observed. 6-dimethylaminopurine (a general kinase inhibitor) is able to block FRQ phosphorylation in vivo, reducing the rate of phosphorylation and the degradation of FRQ and lengthening the period of the clock in a dose-dependent manner. To confirm the role of FRQ phosphorylation in this clock …

Effect Of Ceftazidime On Systemic Cytokine Concentrations In Rats, Khalid M. Alkharfy, John A. Kellum, Reginald F. Frye, Gary R. Matzke Jan 2000

Effect Of Ceftazidime On Systemic Cytokine Concentrations In Rats, Khalid M. Alkharfy, John A. Kellum, Reginald F. Frye, Gary R. Matzke

Pharmacotherapy and Outcomes Science Publications

The effect of a single dose of ceftazidime on circulating concentrations of interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) in a rat model of sepsis was studied. IL-6 concentrations were significantly elevated (100 to 200 times the baseline) 6 h after ceftazidime administration in both septic and nonseptic (control) rats. TNF-α concentrations increased significantly in nonseptic (∼40 times the baseline) rats but not septic (∼2 to 3 times the baseline) rats. Ceftazidime administration was not associated with an increase in endotoxin concentrations. These findings suggest that ceftazidime modulation of proinflammatory cytokine concentrations may be independent of its antimicrobial properties.

The Phantom Burster Model For Pancreatic Β-Cells, Richard Bertram, Joseph Previte, Arthur Sherman, Tracie A. Kinard, Leslie S. Satin Jan 2000

The Phantom Burster Model For Pancreatic Β-Cells, Richard Bertram, Joseph Previte, Arthur Sherman, Tracie A. Kinard, Leslie S. Satin

Pharmacology and Toxicology Publications

Abstract

Pancreatic β-cells exhibit bursting oscillations with a wide range of periods. Whereas periods in isolated cells are generally either a few seconds or a few minutes, in intact islets of Langerhans they are intermediate (10–60 s). We develop a mathematical model for β-cell electrical activity capable of generating this wide range of bursting oscillations. Unlike previous models, bursting is driven by the interaction of two slow processes, one with a relatively small time constant (1–5 s) and the other with a much larger time constant (1–2 min). Bursting on the intermediate time scale is generated without need …

Background: Timely recognition and prevention of health problems among elderly people have been shown to improve their health. In this randomized controlled trial the authors examined the impact of preventive home visits by a nurse compared with usual care on the outcomes of frail elderly people living in the community. Methods: A screening questionnaire identified eligible participants (those aged 70 years or more at risk of sudden deterioration in health). Those randomly assigned to the visiting nurse group were assessed and followed up in their homes for 14 months. The primary outcome measure was the combined rate of deaths and …

Elements Of Success In Telemedicine Programs: Advice From Telemedicine Leaders, Mary Moore Phd Jan 2000

Elements Of Success In Telemedicine Programs: Advice From Telemedicine Leaders, Mary Moore Phd

Faculty Research, Publications, and Presentations

Telemedicine projects in the past have demonstrated the potential of telecommunication services to improve healthcare and reduce costs by providing patient consultations and continuing education to remote healthcare practitioners. With the lowered costs of related technologies and increased network capabilities it is likely that telemedicine could transform health care, even for non-isolate, general populations.

It is difficult, however, to identify information on elements affecting the success of past telemedicine projects. This may be because information on these projects is either unpublished, or is hidden in obscure government reports (Brown, 1982). If telemedicine services are to be routinely provided, it is …

Remote Episodic Memory Deficits In Patients With Unilateral Temporal Lobe Epilepsy And Excisions, Indre Viskontas, M. P. Mcandrews, M. Moscovitch Jan 2000

Remote Episodic Memory Deficits In Patients With Unilateral Temporal Lobe Epilepsy And Excisions, Indre Viskontas, M. P. Mcandrews, M. Moscovitch

Psychology

The nature of remote memory impairment in patients with medial temporal lobe damage is the subject of some debate. While some investigators have found that retrograde amnesia in such patients is temporally graded, with relative sparing of remote memories (Squire and Alvarez, 1995), others contend that impairment is of very long duration and that remote memories are not necessarily spared (Sanders and Warrington, 1971; Nadel and Moscovitch, 1997). In this study, remote memory was assessed in 25 patients with unilateral temporal lobe epilepsy and 22 non-neurologically impaired controls using the Autobiographical Memory Interview (Kopelman et al., 1989). Results indicate that …

VCU Health Publications

Background

Chronic hepatitis C virus (HCV) infection in patients with cirrhosis is difficult to treat. In patients with chronic hepatitis C but without cirrhosis, once-weekly administration of interferon modified by the attachment of a 40-kd branched-chain polyethylene glycol moiety (peginterferon alfa-2a) is more efficacious than a regimen of unmodified interferon. We examined the efficacy and safety of peginterferon alfa-2a in patients with HCV-related cirrhosis or bridging fibrosis.

Methods

We randomly assigned 271 patients with cirrhosis or bridging fibrosis to receive subcutaneous treatment with 3 million units of interferon alfa-2a three times weekly (88 patients), 90 µg of peginterferon alfa-2a once …

Attributional Style And Disability Outcome, Craig Ravesloot Ph.D., Quincy-Robyn Young, Tom Seekins Ph.D., Ann Szalda-Petree Ph.D., University Of Montana Rural Institute

Health and Wellness

Disability is both a very personal and social issue. In 1999, The World Health Organization (WHO) published their latest revisions to the ICIDH-2. This document includes the latest conceptual developments in understanding the structure and process of disabling conditions. The document highlights that disability outcome occurs within a dynamic system characterized by the impairment itself, environmental and personal factors (World Health Organization, 1999).

Isolation Of Current Components And Partial Reaction Cycles In The Glial Glutamate Transporter Eaat2, Thomas S. Otis, Michael Kavanaugh Jan 2000

Isolation Of Current Components And Partial Reaction Cycles In The Glial Glutamate Transporter Eaat2, Thomas S. Otis, Michael Kavanaugh

Biomedical and Pharmaceutical Sciences Faculty Publications

The kinetic properties of the excitatory amino acid transporter EAAT2 were studied using rapid applications of l-glutamate to outside-out patches excised from transfected human embryonic kidney 293 cells. In the presence of the highly permeant anion SCN−, pulses of glutamate rapidly activated transient anion channel currents mediated by the transporter. In the presence of the impermeant anion gluconate, glutamate pulses activated smaller currents predicted to result from stoichiometric flux of cotransported ions. Both anion and stoichiometric currents displayed similar kinetics, suggesting that anion channel gating and stoichiometric charge movements are linked to early transitions in the transport cycle. …

Pentameric Assembly Of A Neuronal Glutamate Transporter, Sepehr Eskandari, Michael Kreman, Michael Kavanaugh, Ernest M. Wright, Guido A. Zampighi

Biomedical and Pharmaceutical Sciences Faculty Publications

Freeze-fracture electron microscopy was used to study the structure of a human neuronal glutamate transporter (EAAT3). EAAT3 was expressed in Xenopus laevis oocytes, and its function was correlated with the total number of transporters in the plasma membrane of the same cells. Function was assayed as the maximum charge moved in response to a series of transmembrane voltage pulses. The number of transporters in the plasma membrane was determined from the density of a distinct 10-nm freeze-fracture particle, which appeared in the protoplasmic face only after EAAT3 expression. The linear correlation between EAAT3 maximum carrier-mediated charge and the total number …
