Temporal, Perspectives, Dispositional Styles, And Subjective Well-Being, Mary Naeger
Masters Theses & Specialist Projects
This study investigated the relationship between time perspective (TP) and the personality dispositions of optimism, pessimism, and realism with regard to their proposed influence on three measures that collectively assessed subjective well-being (SWB). The Depression-Happiness Scale (McGreal & Joseph, 1993) assessed happiness or the presence of positive affect and the absence of negative affect, the cognitiveaffective components of SWB. The Satisfaction with Life Scale (SWLS) (Diener, Emmons, Larsen, & Griffin, 1985) provided the measurement of subjective life satisfaction, the essential final cognitive-judgmental component of SWB. The Life Orientation Scale-Revised (LOT-R) (Scheier, Carver, and Bridges, 1994) and a Reality Scale, composed …

Inheritance Of Dorsal Fin Coloration In The Metriaclima Species Complex (Teleostei: Cichlidae) Of Lake Malawi, Paulette Reneau
Masters Theses & Specialist Projects
The rock-dwelling cichlids (mbuna) of Lake Malawi have undergone an explosive evolution, giving rise to an assemblage of 300-500 species within the last one million years. Most widespread mbuna 'species' are characterized by the presence of local endemic populations, differing primarily in coloration and often of uncertain taxonomic rank. The recency and rapidity of speciation within the mbuna have led to difficulties in reconstructing an accurate species-level phylogeny, in turn limiting our ability to elucidate the evolutionary dynamics associated with divergence of coloration and other characters. Based on morphometric and meristic characters, Stauffer et al. (1997) erected a new genus …

CGU Faculty Publications and Research
This paper describes the development and testing of the Medical Concept Mapper, a tool designed to facilitate access to online medical information sources by providing users with appropriate medical search terms for their personal queries. Our system is valuable for patients whose knowledge of medical vocabularies is inadequate to find the desired information, and for medical experts who search for information outside their field of expertise. The Medical Concept Mapper maps synonyms and semantically related concepts to a user's query. The system is unique because it integrates our natural language processing tool, i.e., the Arizona (AZ) Noun Phraser, with human-created …

Pretreatment Characteristics Of Legally Coerced Drug Treatment Seekers, Bridgett Augustino
Masters Theses & Specialist Projects
This study examines the sociodemographics, drug use, criminal, and treatment histories of 598 residential and outpatient legally coerced drug treatment seekers in the Kentucky Treatment Outcome and Performance Pilot Studies Enhancement Project. Analyses examined whether users/addicts entering chemical dependency treatment under legal coercion differed from nonlegally coerced treatment seekers. Stanley Cohen's theoretical model of social control provided the theoretical framework for the study. Results showed demographic and behavioral differences were noted between respondents under no coercion and those under legal coercion on gender, age, educational status, pretreatment criminality and current treatment modality. In addition, differences between the legally coerced and …
Effects Of Antioxidant Supplementation With Vitamin E And Selenium On Cytokine Production In Mice Infected With Trypanosoma Cruzi, Dong Chen
Masters Theses & Specialist Projects
Dietary supplementation with the antioxidants vitamin E and selenium (Se) has been shown to be beneficial against the intracellular parasite Trypanosoma cruzi during experimental Chagas' disease. Supplementation of mice with vitamin E and selenium at levels well above the recommended daily allowance for these two substances results in significantly decreased parasitemia levels and increased longevity. However, the mechanistic role of the antioxidants in this process is not well understood. It is known that vitamin E and selenium can reduce oxidative stress and improve host immune responses. In addition, the synthesis of many regulatory cytokines is known to be influenced by …

Purpose: A specific TLS-CHOP fusion gene resulting from the t(12;16) is present in at least 95% of myxoid liposarcomas (MLS). Three common forms of the TLS-CHOP fusion have been described, differing by the presence or absence of TLS exons 6-8 in the fusion product. Type 5-2 (also known as type II) consists of TLS exons 1-5 fused to CHOP exon 2; type 7-2 (also known as type I) also includes TLS exons 6 and 7 in the fusion, whereas type 8-2 (also known as type III) fuses TLS exons 1-8 to CHOP exon 2. We sought to determine the impact …

Dextran-Methylprednisolone Succinate As A Prodrug Of Methylprednisolone: Plasma And Tissue Disposition, Xiaoping Zhang, Reza Mehvar
Pharmacy Faculty Articles and Research
Plasma and tissue disposition of a macromolecular prodrug of methylprednisolone (MP), dextran (70 kDa)–methylprednisolone succinate (DMP), was studied in rats. Single 5‐mg/kg doses of DMP or unconjugated MP were administered into the tail veins of different groups of rats (n = 4/group/time point). Blood (cardiac puncture) and tissues (liver, spleen, kidney, heart, lung, thymus, and brain) were collected at various times after DMP (0–96 h) or MP (0–2 h) injections. Concentrations of DMP and MP in samples were analyzed by size‐exclusion chromatography (SEC) and reversed‐phase high‐performance liquid chromatography (HPLC), respectively. Conjugation of MP with 70‐kDa dextran resulted in 22‐, …

A Decentralized, Patient-Centered Approach To Diabetes Disease Management In The Primary Care Setting, Michael Toscani, John P. O'Connor, David B. Nash
College of Population Health Faculty Papers
Although many disease management programs have been developed for diabetes, no single design has proved best for all providers and patient populations. Cost effectiveness is especially relevant to diabetes programs because significant costs of the disease may come from complications that occur later in life, while the costs of the program are incurred immediately. For this reason, diabetes disease management programs with positive outcomes and low implementation costs are of particular importance. We report here on the outcomes of a pilot test of the Steps to Health program developed by Abbott Laboratories. The Steps to Health program was designed to …

During the past decade, the profession of pharmacy has changed dramatically. The Doctor of Pharmacy degree has replaced the Bachelor of Science degree as the first professional degree offered at most accredited U.S. pharmacy schools. Advanced clinical training is now a mainstay of pharmacy training, and this has enabled pharmacists to contribute to disease management efforts. In addition, technological improvements in prescription processing have afforded pharmacists more time to participate in disease management activities. This paper describes how the role of the pharmacist has changed and reviews the results of programs involving pharmacists as disease management providers in the areas …

Construction And Screening Of A Cdna Library For The C3 Gene(S) Of The Nurse Shark (Ginglymostoma Cirratum), Janette Cristina Builes
FIU Electronic Theses and Dissertations
Mammalian C3 is a complement protein which consists of an α chain (125kDa) and β chain (75kDa) held together by a disulfide bond. The a chain contains a conserved thiolester site which provides the molecule with opsonic properties. The protein is synthesized as a single pro-C3 molecule which is post-translationally modified. C3 genes have been identified in organisms from different phyla, however, the shark C3 gene remains to be cloned. Sequence data from the shark will contribute to understanding further the evolution of this key protein. To obtain additional sequence data for shark C3 genes a cDNA library was constructed …

Department of Microbiology and Immunology Faculty Papers
CD44 is a widely distributed cell surface adhesion molecule and is implicated in diverse biological processes. However, the nature of intracellular signaling triggered by CD44 remains to be elucidated. Here, we show that CD44 undergoes sequential proteolytic cleavage in the ectodomain and intracellular domain, resulting in the release of a CD44 intracellular domain (ICD) fragment. Consequently, CD44ICD acts as a signal transduction molecule, where it translocates to the nucleus and activates transcription mediated through the 12-O-tetradecanoylphorbol 13-acetate-responsive element, which is found in numerous genes involved in diverse cellular processes. Expression of an uncleavable CD44 mutant as well as metalloprotease inhibitor …

New Procedures To Assess Executive Functions In Preschool Children, K. A. Espy, P. M. Kaufmann, M. L. Glisky, M. D. Mcdiarmid
Developmental Cognitive Neuroscience Laboratory: Faculty and Staff Publications
Executive functions are difficult to assess in preschool children, yet the preschool period is particularly important, both in the development of behavioral control and of the brain, particularly the prefrontal cortex. Several tasks were adapted from developmental and neuroscience literature and then administered to 98 preschool children (30-, 36-, 42-, 48- and 60-month age groups). Executive function task performance was related largely to age group, but not to sex or intelligence. These tasks, then, were sensitive in this age range and may be useful to delineate distinct cognitive profiles among preschool children with various neurological and developmental disorders.

The use of culture media to support the development of preimplantation embryos to the blastocyst stage is often associated with detrimental effects on normal development. These effects have been uncovered largely by investigating the phenotypic abnormalities displayed by fetuses and newborns derived from cultured preimplantation embryos. Research to understand the impact of culture on the embryonic developmental programme has focused on embryo metabolism, gene expression and genomic imprinting. We have used differential display RT-PCR to examine culture influences on global transcript pools in bovine embryos. Others have examined culture influences on candidate "marker genes" in cultured murine, ovine and bovine …

Meeting Employees' Information Needs In An Evolving Health Care Marketplace, Lise Rybowski
National Health Policy Forum
This background paper explores the role of information in an evolving health care marketplace. It notes that, in a softening economy, many employers seeking relief from escalating health care expenses shift costs onto employees. Some try to make their retreat more palatable by offering employees both more options for how and where they receive health care and more control over how and where the money is spent. This paper discusses the changes that are likely to occur over the next decade in the ways that people make decisions about health plans and providers, the implications for consumers' information needs, and …

Over the past few years, some disadvantages to alternative spikes have surfaced. The amount of damage to putting surfaces is in question, but the damage may not be noticed immediately, as it is with the conventional 8-mm length metal spike. The present study questions the effect of alternative spikes on putting green quality and ball roll.

Dartmouth Scholarship
Osteoarthritic chondrocytes secrete matrix metalloproteinase-13 (MMP-13) in response to interleukin-1 (IL-1), causing digestion of type II collagen in cartilage. Using chondrocytic cells, we previously determined that IL-1 induced a strong MMP-13 transcriptional response that requires p38 MAPK, JNK and the transcription factor NF-κB. Now, we have studied the tissue-specific transcriptional regulation of MMP-13. Constitutive expression of the transcription factor Runx-2 correlated with the ability of a cell type to express MMP-13 and was required for IL-1 induction; moreover, Runx-2 enhanced IL-1 induction of MMP-13 transcription by synergizing with the p38 MAPK signaling pathway. Transiently transfected MMP-13 promoters were not IL-1 …

Type 2 iodothyronine deiodinase (D2) is a selenoenzyme, the product of the recently cloned cAMP-dependent Dio2 gene, which increases 10- to 50-fold during cold stress only in brown adipose tissue (BAT). Here we report that despite a normal plasma 3,5,3′-triiodothyronine (T3) concentration, cold-exposed mice with targeted disruption of the Dio2 gene (Dio2–/–) become hypothermic due to impaired BAT thermogenesis and survive by compensatory shivering with consequent acute weight loss. This occurs despite normal basal mitochondrial uncoupling protein 1 (UCP1) concentration. In Dio2–/– brown adipocytes, the acute norepinephrine-, CL316,243-, or forskolin-induced increases in lipolysis, UCP1 mRNA, and …

An interview questionnaire was presented to a multiethnic sample of 511 women, age 18–64 years, at public and private specialty clinics to determine the frequency, type, and perpetrator of abuse toward women with physical disabilities. The four-question Abuse Assessment Screen–Disability (AAS-D) instrument detected a 9.8% prevalence (50 of 511) of abuse during the previous 12 months. Using two standard physical and sexual assault questions, 7.8% of the women (40 of 511) reported abuse. The two disability-related questions detected an additional 2.0% of the women (10 of 511) as abused. Women defining themselves as other than black, white, or Hispanic (i.e., …

We investigated the molecular mechanisms by which pH-sensitive liposomes surpass the cytoplasmic and endosomal membranes to deliver their aqueous contents into the cytoplasm. Various liposome formulations were evaluated for their efficacy to mediate intracellular delivery of encapsulated material, including a novel sterically stabilized pH-sensitive formulation ((DOPE:CHEMS:DSPE-PEG2000 (6:4:0.3)) that was previously developed in our laboratories. In an attempt to fully characterize the nature of liposome-cell interactions different approaches based on a dual-labeling fluorescence assay were used. Our results indicate that the efficacy of interaction of pH-sensitive liposomes, both plain and sterically stabilized, with cells is strongly determined by the inclusion of …
