The Effect Of Different Rations On Some Rumen Fluid And Blood Parameters In Sheep, Taylan Aksu, Suphi̇ Deni̇z

The objective of this study was to evaluate the effects of different combinations of energy (barley and dried molassed sugar beet pulp (DMSBP)) and protein sources (soybean meal (SBM), cotton-seed meal (CSM), SBM-treated with formaldehyde (FTSBM), CSM-treated with formaldehyde (FTCSM), and urea) on some rumen fluid and blood parameters. In this study, four rumen fistulated Morkaraman rams were used. Rumen pH values were around the optimal pH values suggested for rumen fluid (5.2-7.3) at all sampling times. Ruminal NH_3-N concentrations were significantly greater (P < 0.05) in animals fed DMSBP as an energy source compared with animals fed barley as an energy source and were between 10.4 and 32.75 mg/100 ml in all groups. Formaldehyde treatments of protein sources reduced ruminal NH_3-N concentrations in animals fed DSBP as an energy source (P < 0.05). Acetic and propionic acid concentrations were affected by the energy source, although butyric acid concentrations were significantly higher in animals fed groups containing barley than in animals fed groups containing DMSBP. Formaldehyde treatment did not affect propionic acid concentrations, although propionic acid concentrations were generally significantly higher (P < 0.05) in animals fed barley + SBM combinations compared to treatments containing both SBM and CSM combinations (P < 0.05). Serum total protein concentrations were affected by the energy source and these were higher in groups containing DMSBP than in groups containing barley (P < 0.05). However, serum urea-N concentrations were similar among the different diets.

In this study, four male Kıvırcık yearlings with permanent rumen cannula were used. The animals were fed according to a 4 x 4 Latin square design over 20-day periods (15-day adaptation period, 5-day collection period). The animals were fed four different ration types. This study was carried out to investigate the growth of some testis characteristics of Norduz male kids reared at the Agricultural Farm of Yüzüncü Yıl University. The data were obtained from 22 Norduz male kids weaned at 3 months of age. The male kids were maintained in farm conditions. In male kids, least square means of testis diameter, testis length, scrotum circumference and scrotum length evaluated as testis measurements were 3.13 ± 0.08, 5.95 ± 0.14, 16.52 ± 0.39 and 10.60 ± 0.33 cm at 90 days, respectively, and 4.75 ± 0.08, 9.29 ± 0.14, 19.61 ± 0.40 and 12.38 …

Respiratory diseases due to infectious agents are an important problem in the broiler chicken industry. One of the most important infectious agents of chickens is Ornithobacterium rhinotracheale, which has recently been isolated in Turkey and around the world. In this report, O. rhinotracheale infection was detected in broiler chickens submitted to the poultry disease laboratory with respiratory problems, increased mortality and retarded growth.

Haemoglobin and transferrin types were investigated in Çine type sheep. Separation of transferrin (Tf) and haemoglobin (Hb) types was carried out using horizontal starch-gel and polyacrylamide gel electrophoresis, respectively. Three Hb A and 59 Hb B types, and 5 transferrin alleles (A, B, M, D, E) were isolated from Çine type sheep. Eight Tf phenotypes consisting of 3 homozygote types (Tf AA, Tf MM, Tf DD) and 5 hetorozygote types (Tf AM, Tf AD, Tf AE, Tf BD, Tf MD) were detected.

In this study, the isolation of Staphylococcus aureus from dairy desserts, and its thermonuclease activity and enterotoxigenic properties were investigated. A total of 175 samples were analyzed consisting of 100 pudding samples (sütlaç [rice pudding] 25, keşkül [milk pudding containing coconut] 30, supangile [chocolate pudding] 25, kazandibi [milk pudding slightly burned on the bottom] 20) and 75 cream pastries (butter-cream 25, chocolate-cream 25, fruit-cream 25). In this study, the thyroid hormone levels in sheep experimentally treated with selenium were investigated. Twelve Akkaraman sheep were used in the investigation. Selenium was given intramuscularly at doses of 0.1 (Group 1) and 0.4 mg/kg (Group 2). The blood samples were taken from the jugular vein at 1, 2, 4, 8, 24, 48, 96 and 144 hours after the administration. In this research, the colourfullness and the relationships between colourfullness and physical characteristics in the coarse and down fibres obtained from hair goats raised in the provinces of Antalya, Artvin, Diyarbakır, İzmir, Kars and Konya in Turkey were studied. This study was carried out to investigate the changes occurring in the levels of serum thyroid hormone in rams given indomethacin, furosemide and PGF2\alpha. In the study, 20 Akkaraman rams, approximately 2 years old were used. Indomethacin (3.0 mg/kg), furosemide (2.0 mg/kg) and PGF2\alpha (0.1 mg/kg) were intramuscularly given to the animals in one dose and repeated for four consecutive days. This study was carried out on cattle to detect the seroprevalence of babesiosis around Kayseri between July 1997 and January 1999. During this period a total of 191 cattle were examined for the Babesia species. Blood samples were collected from the cattle to obtain sera. During sampling, thick and thin blood smears were prepared from the punctured tail of each animal. Ticks were collected from the animals sampled. The applicability of the immunoperoxidase (IP) technique in unfixed-fresh tracheal organ cultures (TOCs) was investigated for rapid detection of infectious bronchitis virus (IBV). For this purpose, TOCs prepared from one-day-old SPF chicks were inoculated with IBV M41 strain in serial dilutions. IBV antigen detected by IP technique in TOCs 18 h postinoculation. IP staining was observed before siliostasis in all dilutions. The IP technique was also applied to detect IBV antigen in formalin-fixed, paraffin-embedded sections of TOCs inoculated with IBV M41 strain. Viral antigen was also detected in these sections. In this study we examined factors affecting the colonization of campylobacters in chick intestines, colonization times and elicited immune responses. In chicks, infected orally by minimal infective dose _{100} (MID_{100}), colonization in the intestines was observed to reach 100% during the 3-week period following C. jejuni and C. coli inoculation. Due to the recent increasing mortality rates in 1- to 4-month-old Thoroughbred foals with Rhodococcus equi infection in Turkey, serum samples from 454 foals between 30 and 60 days on studfarms of the Marmara Region, where the Thoroughbred foal population is concentrated, were investigated by ELISA for early diagnosis of the infection. The Tween 20-extracted antigen of the R. equi ATCC 6939 strain was used in the ELISA. Two hundred and nine (46%) of the 454 foals were found to be ELISA-positive. The purpose of this study was to isolate Campylobacter species from the intestines and livers of chicken and to identify Campylobacter coli and Campylobacter jejuni by both conventional methods and Polymerase Chain Reaction (PCR). Four specific primers derived from the ceuE gene present in the genomes of C. coli and C. jejuni were used for PCR identification. The aim of this study was to diagnose Theileria annulata infected cattle by microscopy, IFAT and nested PCR and to compare the sensitivity and specificity of these diagnostic methods. A total of 147 blood and sera samples were collected from healthy cattle in four localities of Polatlı district, Ankara, where tropical theileriosis is prevalent. Examination results revealed positivity rates of 31.3%, 44.9% and 61.2% for microscopy, IFAT and nested PCR, respectively. The liver damage and obstructions of the biliary tract were evaluated ultrasonographically in experimentally bile duct ligated dogs. Seven healthy dogs of different breeds were used. The common bile duct was double ligated at the duodenal region after clinical and ultrasonographic examinations. After that, clinical examinations, biochemical analyses and examinations of the liver, gallbladder and biliary tract were done ultrasonographically over 6 weeks. All dogs were necropsied and macroscopic and histologic alterations were determined at the end of the study. It was determined that the levels of serum ALT, ALP, GGT and T. Bilirubin were (p < 0.001-0.05) statistically significantly increased after the operation in all dogs. As a result of ultrasonographic examinations, it was determined that the bile ducts and gallbladder were distended, the neck of the gallbladder was larger and more tortuous than normal, hypoechoic sludge was prominent in the gallbladder, and perivascular, multifocal hyperechoic areas appeared in the liver parenchyma toward the end of the study. These alterations were confirmed macroscopically and microscopically at the end of the autopsy in all dogs. We consider that as well as blood serum parameters, ultrasonographic examinations are useful in the early diagnosis of biliary diseases and in differentiating them from hepatic diseases.

The objective of the present study was to transfer sheep embryos produced in vitro to recipient ewes. Ovaries were taken from slaughtered Kıvırcık ewes and transferred to the laboratory in phosphate buffered saline (PBS) at 30-35 ºC. The cumulus-oocyte complexes were obtained by slicing and washing 1-6 mm diameter follicles and matured for 24 h in medium 199 supplemented with sodium pyruvate, follicle stimulating hormone (FSH), luteinizing hormone (LH) and 10% fetal calf serum (FCS) at 38.5 ºC under 5% CO_2 in humidified atmosphere. In this study, 275 serum samples collected from cattle at various ages and of both sexes, slaughtered at state or private abattoirs in Elazığ, Malatya and Diyarbakır were examined by MAT and ELISA for the presence of antibodies against Leptospira species. In the serological analysis, using antigens prepared from reference Leptospira hardjo, L. pomona, L. grippotyphosa, L. hebdomadis, and L. icterohaemorrhagiae cultures, 44 (16%) and 64 (23.3%) of the samples were determined to be positive with MAT and ELISA, respectively.

The seasonal variation and meat composition of blue crabs (Callinectes sapidus Rathbun, 1896) were investigated. Twenty (10 male, 10 female) crabs were used each season. The total weight of the samples was estimated and the ratios of crude protein, lipid, dry matter and crude ash were obtained from meat of the body and claws. Comparatively higher values of dry matter, crude ash and lipid were determined for the claw meat of males in winter. Hydrocephalus associated lesions such as diverticula, clefts and tears observed in periventricular white matter were studied pathologically in three puppies with necropsy between 1993 and 1994. Enlargement of the cranial vault was present in the puppies. The animals were blind, and their morphological alterations were similar. Marked hydrocephalus caused by mesencephalic canal malformations and obstruction was detected in all cases. Compression atrophy was seen in the nucleus caudatus, hippocampus, putamen and cerebellum in all animals. Cerebral sulci malformations were also detected in the cranial parts of the brain. In this research 30 sucuk samples were obtained from 5 meat plants with high capacity, during 3 months (January 1998-March 1998) at 15 day intervals. At the results of determination of the 30 sucuk samples, 80% of them was not shown acceptable colour development and 93.33% had a soft texture. 83.33% of the samples cut from the surface was not indicated good colour development and 50% of them has not good marble form. In this study, leptospiral antibodies were investigated by Microscopic Agglutination Test (MAT) and Enzyme Linked Immunosorbent Assay (ELISA) in order to detect leptospirosis and serotypes which cause leptospira infection in cattle raised in Kars and Ardahan. Live and sonicated antigens prepared from Leptospira hardjo and Leptospira grippotyphosa serotypes were used. Blood samples were taken from 150 cattle under 2 years of age and 840 cattle 2 years of age or older (a total of 990) which were unvaccinated against leptospirosis in 30 rural settlements. This study was performed to determine the incidence and anatomo-pathologic aspects of reproductive organ tumors in laying chickens. For this research, 10,475 16-18-month-old, commercial laying hens of the Babcock genotype were obtained from four flocks in Afyon city. They were examined right after slaughtering, and reproductive organ tumors in 804 hens (7.67%) were observed. Out of these 804 hens, leiomyoma was found in 760 hens (7.25%), oviduct adenocarcinoma in 39 hens (0.37%) and leiomyoma together with oviduct adenocarcinoma in 5 hens (0.05%) were detected. The \alpha-amylase encoding gene from Bacillus subtilis ORBA97 strain was cloned and expressed in Escherichia coli with the plasmid vector called pUC18 as well as in the \alpha-amylase negative Bacillus subtilis YB886 strain by using the pUB110 vector. The specific activity of the enzyme was determined to be 5.052 µmol (mg protein min)^{-1}. Zymogram analysis revealed no sign of protease degradation in the YB886 strain. The optimum temperature and pH of the enzyme were found to be 50°C and 5 respectively. Relative enzyme activity decreased to 36% of the original activity when it was subjected to a temperature of 90°C …
