Pathological and immunohistochemical changes in livers were investigated in monocrotaline-treated rats. Eighty adult-male Wistar rats were distributed in 5 groups, each containing 16 animals according to monocrotaline dose (Group I: 50 mg/kg, Group II: 100 mg/kg, Group III: 150 mg/kg, and Group IV: 200 mg/kg). The control group was administered with physiological saline. Half of the animals in each group were euthanized at the 18th hour while the remaining half at the 6th week postinoculation. In this study, monensin (MON) was investigated for its apoptotic and genotoxic effects on chick liver tissue and cytotoxic and apoptotic effects on chick primary hepatocytes isolated from perfused liver tissue. Western blotting and real-time PCR (qPCR) were used to determine the apoptotic effect on primary hepatocytes and liver tissue, and the comet assay was used to determine the genotoxic effect on liver tissue. MON decreased cell viability in primary hepatocytes with increasing concentrations. This study was conducted to determine the ultrasonographic features of the liver, gallbladder, spleen, kidneys, and urinary bladder of 24 apparently healthy male and female long-tailed macaques (LTM) and the correlations of organ measurements with body weight and crown?rump length. The animals were grouped into 12 males and 12 females. Examination was performed in sedated LTM using an ultrasound machine with 6.0 MHz microconvex scanner. Ultrasound appearance, dimensions, and echotexture of the liver, gallbladder, kidneys, spleen, and urinary bladder were evaluated. Cannabinoid receptor 2 (CB2) has a role in the pathology of some liver diseases. Cypermethrin (CYP) is a synthetic pyrethroid insecticide used commonly for the control of pests and vectors, and for protection of foodstuffs. In this study, the involvement of CB2 in CYP-induced liver damage in rats was investigated. CYP was applied to rats orally at a dose of 125 mg/kg bw per day for 14 days. Evaluation Of Ultrasonography As A Diagnostic Tool For Hepatic Hydatid Cysts In Sheep, Hussein Awad Hussein, Mohammed Elrashidy

This retrospective study was designed to evaluate the use of ultrasonography for the diagnosis of hepatic hydatid cysts. This study was conducted on 22 sheep, which were classified into 2 groups according to their liver health group (healthy liver = 13 and cystic liver = 9). Biochemically, serum concentrations of gamma-glutamyl transferase, aspartate aminotransferase, total bilirubin, and globulins were significantly increased (P < 0.01) while albumin was lowered (P < 0.01) in sheep with cystic livers. Ultrasonographically, rounded, anechoic, and unilocular hydatid cysts with elliptical circumferences ranging from 6 to 10 cm were seen. The interiors of cysts contained echogenic particulate materials, septations, or fine echoes. At the 10th intercostal space (ICS), ventral margin, size, thickness, and angle of livers increased in sheep with hepatic cysts, while the diameter of portal vein decreased in sheep with liver cysts. At the 9th ICS, the circumference of the gall bladder was lowered in sheep with hepatic cysts (P < 0.01). The sensitivity, specificity, and positive and negative predictive values of ultrasonography for diagnosis of hepatic hydatid cysts were 80%, 100%, and 100% and 83%, respectively. In conclusion, ultrasonography alone or in combination with some biochemical parameters reflecting liver function could be helpful for the diagnosis of hepatic hydatid cysts in sheep.

Our focus has been to study and compare the anatomical helical computed tomography (CT) features of the normal rabbit liver with its native cross-sectional anatomy. Helical CT was used for scanning the cranial part of the abdominal cavity. The slice thickness was 5 mm. This study aimed to purify glutathione reductase (GR) from turkey liver and investigate some of its characteristic features. The purification procedure comprised 2 steps: homogenate preparation and 2',5'-ADP Sepharose 4B affinity gel chromatography. Thanks to the 2 consecutive procedures, the enzyme, having the specific activity of 606.67 EU mg protein^{-1}, was purified with a yield of 10.75% and 2476-fold, and, in order to control enzyme purity, SDS-PAGE was done. The present study was conducted to investigate the variations in blood serum and hepatic levels of vitamin A and b-carotene in Ghezel sheep throughout the year. From 1000 sheep aged 1-5 years, blood and liver samples were collected before and after slaughter. The levels of vitamin A and \beta-carotene in both samples were determined spectrophotometrically. The results showed significant differences between summer and winter values (P < 0.05), but no differences were found between the sexes (P > 0.05). There were significant correlations between the vitamin A levels in serum and the liver (P < 0.05). Due to low levels of vitamin A and \beta -carotene in Ghezel sheep during the winter, supplementary feeding of vitamin A is recommended during this season.

Glutathione reductase (GR) is important in maintaining high levels of reduced glutathione in cells. The enzyme is inhibited by some drugs used in antimicrobial chemotherapy. The effects of some antibiotics on GR activity were investigated in this study. At first, the enzyme, having a specific activity of 88.28 EU/mg proteins, was purified 1636-fold with a yield of 35%. Glutathione reductase (Glutathione: NADP^+ oxidoreductase, EC 1.8.1.7; GR) was purified from sheep liver. The purification included 4 steps: preparation of homogenate, ammonium sulfate fractionation, 2', 5'-ADP Sepharose-4B affinity chromatography, and gel filtration chromatography. GR was obtained with a yield of 14.1% having a specific activity of 47.27 EU/mg proteins. Optimal pH, stable pH, and optimal temperature were 8.0, 5.5, and 60 °C, respectively. K_M and V_max values for NADPH and GSSG substrates were 0.0258 and 0.0239 mM, and 0.266 and 0.255 EU/ml, respectively. The overall purification was about 1477-fold for liver GR. The effects of replacing fish oil with soybean oil, sunflower oil, corn oil and olive oil in starter feeds on the vicerosomatic index, hepatosomatic index and liver fatty acid composition of the sea bass were studied. Juvenile sea bass (initial weight = 7.58 ± 0.13 g) were fed experimental diets for 75 days. The fish were then killed and liver samples were collected. The lowest total crude fat value (17.61%) was found in the fish fed the fish oil diet. The highest total crude fat value (34.30%) was found in the fish fed the olive oil diet (P < 0.05). Similarly, the fish fed the fish oil diet had the lowest hepatosomatic index value (2.08) and all the fish fed vegetable oils had similarly high hepatosomatic index values (P < 0.05). Likewise, vicerosomatic index values were lowest (9.06%) in the fish fed fish oil and highest in the fish fed soybean and other vegetable oils (P < 0.05). Fatty acid analyses showed that EPA and DHA from n-3 HUFA's in the fish fed the fish oil diet, linolenic acid (18:3n-3) and linoleic acid (18:2n-6) in the fish fed the soybean oil diet and oleic acid (18:1n-9) in the fish fed the olive oil diet were higher than in the fish fed the other diets.

The liver damage and obstructions of the biliary tract were evaluated ultrasonographically in experimentally bile duct ligated dogs. Seven healthy dogs of different breeds were used. The common bile duct was double ligated at the duodenal region after clinical and ultrasonographic examinations. After that, clinical examinations, biochemical analyses and examinations of the liver, gallbladder and biliary tract were done ultrasonographically over 6 weeks. All dogs were necropsied and macroscopic and histologic alterations were determined at the end of the study. It was determined that the levels of serum ALT, ALP, GGT and T. Bilirubin were (p < 0.001-0.05) statistically significantly increased after the operation in all dogs. As a result of ultrasonographic examinations, it was determined that the bile ducts and gallbladder were distended, the neck of the gallbladder was larger and more tortuous than normal, hypoechoic sludge was prominent in the gallbladder, and perivascular, multifocal hyperechoic areas appeared in the liver parenchyma toward the end of the study. These alterations were confirmed macroscopically and microscopically at the end of the autopsy in all dogs. We consider that as well as blood serum parameters, ultrasonographic examinations are useful in the early diagnosis of biliary diseases and in differentiating them from hepatic diseases.

In this study, 3470 livers of beef cattle slaughtered by the Ankara Meat and Fish Company were inspected. In these animals, 82 livers had telangiectasis, and the incidence of lesions was 2.3%. They were characterized grossly by single or multiple red-brown foci, generally 1 to 5 mm in diameter, and occasionally larger. They were located on the parietal surface of the lobus dexter or rarely scattered throughout the organ. Microscopically, dilated sinusoids filled with erythrocytes were observed in initial lesions; the distortion, atrophy and necrosis of hepatocytic cordons were seen in advanced cases. In this study, the changes in pyruvate kinase enzyme activities in the liver and kidney tissues of rats with diabetes induced by streptozotocin were examined and the relations with the disease were investigated. Diabetes mellitus was induced in Wistar-Albino rats by the administration of a single intraperitoneal dose of streptozotocin (60 mg/kg body weight). After streptozotocin was injected, the pyruvate kinase activities in the livers and kidneys of the rats were analysed according to Beutler's method on days 3 and 12. The aim of the present study was to define the reference values for erythrocyte and liver tissue glucose-6-phosphate dehydrogenase (G6PDH), glutathione peroxidase (GPX), glutathione reductase (GR) and superoxide dismutase (SOD) activities and also reduced glutathione (GSH) levels in order to obtain basal data for subsequent toxicological investigations. In addition, the best storage conditions were determined for liver G6PDH enzyme. The effects of free radicals in the liver tissue of 20 mice were studied. The mice, separated into acute and chronic groups, were injected with phorbol myristate acetate (PMA) to trigger the release of free radicals. In this study, 80 carp specimens (Cyprinus carpio) (16 fish from each station) of ages between 1-2 were caught at five stations in the upper Sakarya river basin the four seasons between October 1995 and October 1996. The structural changes of gill, liver and spleen tissue of the carp were examined. The gills of fishes which live in aquatic habitat exhibit tissue lesions caused by water pollutants, because they are in direct contact with their environment. This paper reports an investigation of the effects of acute and chronic carbon tetrachloride intoxications on the histologic structure of the liver and hematological values and electrocardiograms in rabbits. The rabbits were divided into 3 groups of 7. The first group was the control group, the second group was acutely intoxicated and the third group was chronically intoxicated. In the acutely intoxicated group, caryorexsis, along with necrosis was observed in the hepatocyte of the centrolobular area of the liver.
