In the United States, osteoporosis results in about 1.5 million annual fractures, costing approximately $15 billion. Calcitonin is safe and effective in slowing osteoporotic bone loss, but its effect is transient. The current studies were designed to explore the dose-dependent effects of salmon calcitonin on bone turnover in ovariectomized rats and to determine if the decrease in therapeutic effectiveness of calcitonin demonstrated over time with higher doses is due to oversuppression of bone turnover. Doses of 5, 15, & 50 IU/kg BW/day of calcitonin were compared to placebo in 12-week-old ovariectomized and sham-ovariectomized Sprague-Dawley rats for 24 weeks. The spinal …

Childhood Experiences Of Appalachian Women Who Have Experienced Intimate Partner Violence During Adulthood., Amy L. Reeves

Recent national surveys in the United States estimate one in five females will experience abuse by an intimate partner during her lifetime. Previous quantitative research linked childhood victimization to repeated victimization in adult relationships. This qualitative study explored the lived experiences of childhood in eight female victims of intimate partner violence who were born and reared in southern Appalachia. Interviews were analyzed using a descriptive-interpretative phenomenological method, as described by Van Manen. The three essential themes from childhood were identified as: living 'as if' an orphan; surviving in chaos; and, manifesting a devalued self. These themes were congruent with findings …

The estimated number of employees in the United Stated screened annually for illicit drugs is approximately 20 million, with marijuana being the most frequently abused drug. Urine adulterants provide an opportunity for illicit drug users to obtain a false negative result on commonly used primary drug screening methods such as the Fluorescence Polarized Immunoassay (FPIA) technique. Typical chemical adulterants such as nitrites are easily detected or render the urine specimen invalid as defined in the proposed federal guidelines for specimen validity testing based on creatinine, specific gravity and pH. Previous research has shown that parent implemented intervention is effective in increasing the speech and language development of children with cleft lip and palate. To further determine the efficacy of this intervention, this study compared the speech and language development of children with CLP, who received parent implemented intervention, with a group of younger, typically developing children, matched for vocabulary size. The human colonic cell line T84, a model for studying epithelial chloride secretion and cystic fibrosis chloride channel (CFTR) function, was used to investigate the regulatory role of estrogens in transepithelial ion transport. Estrogens and other steroid hormones do not stimulate chloride secretion by themselves. However, 17 β-estradiol (17β-E2) rapidly (within seconds to minutes) potentiates carbachol- and thapsigargin-stimulated chloride secretion measured as short circuit current in voltage-clamped T84 monolayer cultures. The effects of a multimodality treatment were investigated using a single subject experimental design across behaviors in two patients with different severity levels of Broca's aphasia. We hypothesized that Response Elaboration Training and an oral reading task would improve accuracy of sentence production, information content, and mean length of utterance. Results indicated that this treatment approach elicited significant improvement in the accuracy of sentence production and information content in the participant with very mild Broca's aphasia. Furthermore, the participant with severe Broca's aphasia demonstrated a very significant improvement in information content and mean length of utterance. The improvements support the …

Nurses believe that more liberal visitation policies will cause greater work overload and decrease the quality of patient care. The purpose of this study was to investigate the effect of a liberal visitation policy on work satisfaction, beliefs, and retention of nurses.

In this study, nurses were asked to complete a survey before and after implementation of a more liberal visitation policy. The nurses were surveyed using Stamps (1997) Index of Work Satisfaction and the Boswell Beliefs Inventory. This study investigated an enhanced dialogic reading (DR) approach in facilitating emergent literacy skills in typically developing preschool children. Eight children from a Title One preschool and their parents participated in five weekly 90-minute training sessions that focused on phonological awareness, print awareness, and alphabet knowledge. First order effects were examined in parent questioning and interaction behaviors on pre- and post-training videotapes. Second order effects were examined in the children's outcomes from pre- to posttesting of preliteracy, speech, and language skills. The purpose of this study was to investigate if there is an additive benefit in training Emergent Literacy (EL) skills with typically developing preschoolers using a combined intervention approach, Dialogic Reading (DR) plus classroom Phonological Awareness (CL), than a single intervention approach providing only DR training.

The study consisted of 8 preschoolers (5 DR+CL, 3 DR). The classroom PA training was conducted 3 times a week for 5 weeks for 20 minutes, using play-based activities. A subsample of 478 children who originally participated in a larger study (Tomblin et al., 1997) was examined for type and frequency of morphological noun and verb errors in oral and written narratives in 2nd and 4th grade. Each child represented one of four groups: Typical Language, Specific Language Impairment, Nonspecific Language Impairment, and Low Nonverbal IQ. The rate of graduation among students with histories of specific language impairment (SLI) was investigating by comparing the target population to matched, nondisabled peers. Data regarding the graduation outcomes for 176 matched pairs of students, over a five-year period, were analyzed to determine if a significant difference in achievement of a high school diploma was observable between the two groups of students. In addition, the study addressed the variables of socioeconomic status and attendance rate as related to graduation rate. Dietary flavonoids are ubiquitous and are marketed as supplements. Characterized as antioxidants, they offer protection against a number of degenerative diseases. Flavonoid mechanics involve free radical scavenging, metal chelation, and substrate association. The skeletal structure of flavonoids is a fused ring system modified by hydroxyl, sugar, and carbohydrate additions. Flavone is a structurally simple flavonoid. Quercetin and its glycosidic analog rutin are complex structures. Using a DNA oxidation/cleavage assay, flavone reduces DNA nicking by 91%. Depending on the solvent system used, quercetin can either increase or decrease DNA oxidation. Rutin exhibits neither pro- nor antioxidant activity. The molecular interactions responsible …

Protection Of Pifithrin-Α And Melatonin Against Doxorubicin-Induced Cardiotoxicity., Xuwan Liu

The current studies were designed to explore the protective effects of pifithrin-α and melatonin against doxorubicin-induced cardiotoxicity. Doxorubicin was injected at a dose of 22.5 mg/kg (i.p.) in mice to induce cardiotoxic effects. Meanwhile, doxorubicin caused a significant increase of cardiac cell apoptosis following injection (14.2 ± 1.1% for doxorubicin-5 d vs. 1.8 ± 0.12% for control, P < 0.01). Ribonuclease protection assays and Western blot analyses revealed that doxorubicin upregulated the p53-dependent genes Bax, BclxL, and MDM2 at least 2-fold. p53 was phosphorylated at Ser 15 in mouse hearts 1 h following doxorubicin injection, and p38 and ERK1/2 MAPKs mediated the phosphorylation of p53. In addition, caspases-3 and -9 were activated 24 h after doxorubicin injection. A p53 inhibitor, pifithrin-α, inhibited doxorubicin-induced apoptosis when administered at a dose of 2.2 mg/kg. Pifithrin-α abolished p53 transactivation activity, but did not influence doxorubicin-induced phosphorylation at Ser 15. By effectively inhibiting the expression of p53-dependent genes, pifithrin-α blocked doxorubicin-induced activation of caspases-3 and -9, thereby preventing cardiac apoptosis. In addition, pifithrin-α attenuated doxorubicin-induced structural and functional damages, without diminishing its anti-tumor efficacy on p53-null PC-3 cancer cells. The protective effects of melatonin and its metabolite 6-hydroxymelatonin on doxorubicin-induced cardiac dysfunction were evaluated in an isolated perfused mouse hearts and in vivo doxorubicin-treated mice. While perfusion of mouse hearts with 5 μM doxorubicin for 60 min resulted in a 50% suppression of HRxLVDP and a 50% reduction of coronary flow, pre-exposure of hearts to 1 μM melatonin or 6-hydroxymelatonin eased the cardiac …

Lithium continues to be widely prescribed for the management of bipolar disease, yet cognitive impairment-related side effects promote noncompliance of the treatment regimen. We have introduced a novel animal model, the black molly fish, to study dose-response effects of lithium on short-term (STM) and long-term (LTM) memories. We developed a method utilizing capillary ion analysis (CIA), to measure plasma and brain lithium levels employed in our behavioral studies. Hepatocyte Growth Factor/Scatter Factor (HGF/SF) stimulates migration of various cells and has been linked via Met tyrosine kinase signaling to transformation and the metastatic phenotype. HGF/SF-Met signaling also plays a role in malignancy. Migration of transformed MDCK-F cells depends on activation of a charybdotoxin (ChTX)-sensitive, volume-activated membrane K+ current. Patch-clamp electrophysiology and transwell migration assays were used to study the effects of HGF/SF on membrane K+ currents and cell migration in MDCK II cells. The purpose of this study is 1) to examine the presence of the antigen in serum by employing a newly developed ELISA immunoassay that quantifies the total antigen and bound antigen (antigen-antibody complex) using polyclonal chicken antibodies directed against an IgM-binding epitope of the new antigen, and 2) to determine the location of the antigen in carcinoma and normal cells. One mechanism for this pathology may involve endothelial activation either by HIV itself or by its secreted proteins, gp120 (a viral envelope protein) and tat (a protein that upregulates transcription of viral genes). We have studied the effects of gp120 and tat on signaling and production of inflammatory cytokines by Human Pulmonary Artery Endothelial Cells (HPAEC). Antibacterial agents specific for the 50S ribosomal subunit not only inhibit translation but also prevent assembly of that subunit. I examined the 30S ribosomal subunit in growing Escherichia coli cells to see if antibiotics specific for that subunit also had a second inhibitory effect. I used the aminoglycoside antibiotics paromomycin and neomycin, which bind specifically to the 30S ribosomal subunit. The purpose of this research effort was to examine whether the feature retention patterns and phonological knowledge of children with suspected apraxia of speech (AOSc) in comparison to those of children with phonological impairment (PI). A second purpose was to determine if a relationship exists between phonological knowledge and feature retention.

The study consisted of three groups of children: PI, AOSc, and typically developing (TD), ages four to seven. A 245-item speech sample was collected from each group. Feature retention percentages and phonological knowledge, represented by percent correct underlying representations (PCUR) were calculated for each child.

Studies of preschool children have shown early speech and language deficits in children with cleft lip and/or palate (CLP). For some children, the deficits during kindergarten diminish as they begin school while some children continue to show delays. The purpose of this study was to determine if a relationship exists between speech and language skills and early reading skills of phonological awareness, letter identification, and rapid naming in children with and without CLP.
