The Interaction Between Atp13a2 And Alpha-Synuclein In Mice, Emily Ribak Dirr

Abnormal accumulation of the presynaptic protein alpha-synuclein (aSyn) is associated with both familial and sporadic forms of Parkinson's disease. ATP13A2 is a lysosomal protein involved in protein degradation, including aSyn. Mice with a loss of ATP13A2 function develop age-related sensorimotor impairments and, in the brain, enhanced lipofuscin and insoluble aSyn accumulation. To further examine the potential interaction between aSyn and ATP13A2, aSyn overexpressing mice (aSyn) were crossbred with Atp13a2-defeicient (13a2) mice. Wildtype (WT), 13a2, aSyn, and double mutant aSyn/13a2 mice were generated and tested behaviorally at 3 ages. Sensorimotor function and cognition were examined using a battery of behavioral tests. …

In animals proprioceptive Ia afferents enable the animal to sense the location, orientation and movement of its limbs in space. These Ia afferents make connections with many different types of neurons within the spinal cord such as; motor neurons, Ia Inhibitory interneurons, and other interneurons. Ia Afferents connect with IaINs that project to antagonist muscle MNs to ensure that contraction of the antagonist muscle group is inhibited. For example the quadriceps Ia afferents contact IaINs that contact PBST motor neurons to inhibit excitation and thus contraction. Real-world expertise provides a valuable opportunity to understand how experience shapes human behavior and neural function. In the visual domain, the study of expert object recognition, such as in car enthusiasts or bird watchers, has produced a large, growing, and often-controversial literature. Here, we synthesize this literature, focusing primarily on results from functional brain imaging, and propose an interactive framework that incorporates the impact of high-level factors, such as attention and conceptual knowledge, in supporting expertise. This framework contrasts with the perceptual view of object expertise that has concentrated largely on stimulus-driven processing in visual cortex. One prominent version of …

This study explores the effect of citicoline on the permeability and expression of tight junction proteins (TJPs) in endothelial cells under hypoxia/aglycemia conditions. Hypoxia or oxygen and glucose deprivation (OGD) was utilized to induce endothelial barrier breakdown model on human umbilical vein endothelial cells (HUVECs) and mouse brain microvascular endothelial cells (bEnd.3s). The effect of citicoline on endothelial barrier breakdown models was determined at either low or high concentrations. FITC-Dextran flux was used to examine the endothelial permeability. Introduction: The purpose of this case study was to describe Dayton, Ohio’s process for implementing an active transportation initiative and suggests ways in which it might be improved. Methods: This paper compares the strategies used in Dayton, Ohio to the 5Ps of the Active Living by Design’s Community Action Model, and compares them with the strategies of three communities described in the literature. Results: The City of Dayton fully initiated two of the 5Ps but neglected the other Ps. Some interview respondents attribute this to a lack of staffing within the City of Dayton. We asked whether wolf re-colonization would facilitate increased growth and reproduction of three browse-sensitive plant species. We hypothesized plant size and the proportion of reproductive individuals would be lowest in areas with no wolves, intermediate where wolves had been present for 4-6 years, and highest where wolves had been present for 12-13 years. Two plant species exhibited significantly greater reproduction where wolves were present for 12-13 years. Mean leaf size of indicator plants was significantly greater in areas where wolves were present for 12-13 years, as compared with that in areas where wolves were not present or were present for …

Background/aims: The quaternary benzo-phenanthridine alkaloid (QBA) chelerythrine (CET) is a pro-apoptotic drug and Na(+)/K(+) pump (NKP) inhibitor in human lens epithelial cells (HLECs). In order to obtain further insight into the mechanism of NKP inhibition by CET, its sub-cellular distribution was quantified in cytosolic and membrane fractions of HLEC cultures by surface-enhanced Raman spectroscopy (SERS). Methods: Silver nanoparticles (AgNPs) prepared by the Creighton method were concentrated, and size-selected using a one-step tangential flow filtration approach. Cutaneous Manifestations Of Giant Cell Arteritis, Christina E. Borchers, Ronald E. Warwar

A 68 year old woman with advanced giant cell arteritis presented with progressive tongue, lip, and scalp necrosis. She was initially misdiagnosed and treated for oral thrush by her primary care physician and emergency department personnel. She was ultimately correctly diagnosed with giant cell arteritis after she developed a left central retinal artery occlusion, ischemic optic neuropathy, and third cranial nerve palsy. She was treated with corticosteroids and the vasculitis was arrested. She was left with permanent defects in her tongue and lip as well as blindness in the left eye.

Knowledge, Training And Experience Of Emergency Responders In The Context Of Functional Needs In General Shelters, Athena K. Fannin

Objective: Gathering and analyzing information on experience, training, and professional affiliation of disaster responders who will deal with functional needs clients in general shelters. Methods: The web-based survey administered in June, 2012 to 165 local Medical Reserve Corps (MRC) volunteers and 72 public health workers was part of a larger study designed to assess functional needs flow processes in general shelters. Descriptive statistics and simple logistic regression were used to analyze data.

Results: The response rate was 16.5%. The percent of survey respondents that had any disaster, medical, or functional needs training were approximately 80%, 70%, and 40% respectively. The …

National trends of c. difficile infections in hospitalized children and young adults with inflammatory bowel disease (IBD) are currently incomplete.

Patients with neurofibromatosis type 1 (NF1) and Costello syndrome Rasopathy have behavioral deficits. In NF1 patients, these may correlate with white matter enlargement and aberrant myelin. To model these features, we induced Nf1 loss or HRas hyperactivation in mouse oligodendrocytes. Enlarged brain white matter tracts correlated with myelin decompaction, downregulation of claudin-11, and mislocalization of connexin-32. Surprisingly, non-cell-autonomous defects in perivascular astrocytes and the blood-brain barrier (BBB) developed, implicating a soluble mediator. Nitric oxide (NO) can disrupt tight junctions and gap junctions, and NO and NO synthases (NOS1-NOS3) were upregulated in mutant white matter. Treating mice with the NOS inhibitor …

Objective:To systematically examine infant size and growth, according to the 2006 WHO infant growth standards, as risk factors for overweight status in young adulthood in a historical cohort. The db/db mouse is a widely used preclinical model in diabetes research. Recent studies have shown that these mice also display aspects of psychosis and depression-like behaviors as seen in some psychiatric disorders. Here, we have performed multiplex immunoassay and liquid chromatography mass spectrometry profiling of the plasma and brain samples from db/db and control mice to identify altered pathways, which could be related to these behavioral abnormalities. Alterations within the renal renin angiotensin system play a pivotal role in the development and progression of cardiovascular and renal disease. Angiotensin converting enzyme 2 (ACE2) is highly expressed in renal tubules and has been shown to be renoprotective in diabetes. The protease, a disintegrin and metalloprotease (ADAM) 17, is involved in the ectodomain shedding of several transmembrane proteins including ACE2. Renal ACE2 and ADAM17 were significantly increased in db/db mice compared to controls. We investigated the effect of the insulin sensitizer, rosiglitazone, on albuminuria, renal ADAM17 protein expression and ACE2 shedding in db/db diabetic mice. …
