This study evaluates the use of nanotubes (NTs) as a matrix for local drug delivery modified by a biodegradable polymeric coating on medical-grade nitinol (NiTi alloy) surfaces. For this purpose, NiTi was anodized within parameters that promote the formation of NTs, ultrasonicated, annealed and impregnated with vancomycin hydrochloride. To improve bioperformance, poly(lactic-co-glycolic acid) (PLGA) was also deposited on the drug-loaded NTs. The samples were characterized in terms of structure, wettability, drug delivery, corrosion and cytocompatibility. Scanning electron microscopy and water contact angle measurements signify the formation of open-top homogeneous NTs of 600– 700 nm in length and ~30 nm in …

The Influence Of Autonomic Function On Pain Modulation Before And After Exercise And Cognitive Task In Fibromyalgia, Abdulaziz Awali Oct 2020

The Influence Of Autonomic Function On Pain Modulation Before And After Exercise And Cognitive Task In Fibromyalgia, Abdulaziz Awali

Dissertations (1934 -)

Fibromyalgia (FM) is a chronic pain condition characterized by widespread pain. People with FM have alterations in autonomic function compared to healthy individuals. Exercise can decrease pain in people with FM, however some people with FM experience pain exacerbation when initiating exercise. Whether variability in pain at rest and following exercise is related to altered autonomic function, including the potential implications with other stressful events such as mental math, is not known. This dissertation aimed at investigating: 1) the relation between cardiovascular autonomic function and central pain facilitation and inhibition, measured by temporal summation of pain (TS) and conditioned pain …

Experimental & Simulation Approaches To Study Neuromuscular Control In People With Chronic Ankle Instability, Hoon Kim

Dissertations (1934 -)

Ankle sprains are among the most common musculoskeletal injuries, and up to 70% of people who sprain their ankles develop chronic ankle instability (CAI). Moreover, people who develop CAI have a significantly higher risk of developing ankle osteoarthritis. Recent research has identified neuromuscular deficits that may be responsible for the high recurrence rates of ankle sprains and for the progression towards ankle osteoarthritis in people with CAI. Unfortunately, current rehabilitation strategies are not completely successful because the mechanisms responsible for these deficits are not fully elucidated. Therefore, the purpose of this dissertation was to investigate individual muscle forces and force …

Carbosilane dendrimers are a particular type of dendrimer structure that has been used as delivery vehicles for drugs and nucleic acids. They have a defined structure according to their generation number, and their terminal groups can be rendered cationic or anionic. The cationic charges can address the limitation of electrostatic repulsion between the negatively charged phosphate groups of nucleic acids and negatively charged cell membranes. Specific drugs can be loaded into the central part of the dendrimer or attached at the exterior, and the overall positive charge may improve the efficacy of anti-inflammatory drugs. One promising feature of dendrimers is …

School of Dentistry Faculty Research and Publications

Most problems associated with chemotherapeutic agents involve non-specific cytotoxicity, low intratumoral accumulation and drug resistance. Targeted drug delivery systems (TDDS) based on nanoparticles (NPs) are a new strategy for better therapeutic efficiency, along with reduction of side effects commonly seen with cancer drugs. Poly (lactic-co-glycolic acid) (PLGA), as one of the furthest developed synthetic polymer, has gained significant attention because of excellent properties—including biodegradability and biocompatibility, controlled release of drug, protection of drug or gene from decomposition and ability to modify surface with targeting agents for both cancer diagnosis and therapy. Aptamers are single-stranded RNA or DNA that can fold …

Shedding Light On The Role Of Keratinocyte-Derived Extracellular Vesicles On Skin-Homing Cells, Golara Nasiri, Negar Azarpira, Aliakbar Alizadeh, Sanaz Goshtasbi, Lobat Tayebi

School of Dentistry Faculty Research and Publications

Extracellular vesicles (EVs) are secretory lipid membranes with the ability to regulate cellular functions by exchanging biological components between different cells. Resident skin cells such as keratinocytes, fibroblasts, melanocytes, and inflammatory cells can secrete different types of EVs depending on their biological state. These vesicles can influence the physiological properties and pathological processes of skin, such as pigmentation, cutaneous immunity, and wound healing. Since keratinocytes constitute the majority of skin cells, secreted EVs from these cells may alter the pathophysiological behavior of other skin cells. This paper reviews the contents of keratinocyte-derived EVs and their impact on fibroblasts, melanocytes, and …

Here, bismuth-based nanomaterials (Bi-based NMs) are introduced as promising theranostic agents to enhance image contrast as well as for the therapeutic gain for numerous diseases. However, understanding the interaction of such novel developed nanoparticles (NPs) within a biological environment is a requisite for the translation of any promising agent from the lab bench to the clinic. This interaction delineates the fate of NPs after circulation in the body. In an ideal setting, a nano-based therapeutic agent should be eliminated via the renal clearance pathway, meanwhile it should have specific targeting to a diseased organ to reach an effective dose and …

Neurotoxicity Of Isomers Of The Environmental Toxin L-Bmaa, Thomas Schneider, Catherine Simpson, Prachi Desai, Madeleine Tucker, Doug Lobner

Biomedical Sciences Faculty Research and Publications

There is evidence that the environmental toxin β-N-methylamino-L-alanine (L-BMAA) may be involved in neurodegenerative diseases. However, a number of controversies exist regarding L-BMAA, one of which is the possibility that when assaying for L-BMAA, its isomers are being detected instead. There are at least four isomers of BMAA that are known to occur: L-BMAA, β-N-methylamino-D-alanine (D-BMAA), 2,4-diaminobutyric acid (DAB), and N-(2-aminoethyl)glycine (AEG). The fact that isomers of BMAA exist in nature also leads to the possibility that they are involved in toxicity. We set out to determine both the potency and the mechanism of toxicity of L-BMAA, D-BMAA, DAB, and …
