Leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5) is an established marker of normal and cancer stemlike cells. LGR5 has been implicated in promoting cancer cell plasticity that drives tumorigenesis, metastasis, and therapeutic resistance. LGR5 rapidly and constitutively internalizes and potentiates Wnt (Wingless/Int-1)/β-catenin and adhesion signaling pathways, though its precise mechanisms and interacting partners remain unresolved. An improved understanding of LGR5 signaling may provide invaluable insight into its intricate and important functions in cancer progression. Moreover, several LGR5-targeting therapies, including peptibody- and antibody-drug conjugates and bispecific antibodies, are showing promising efficacy and tolerability in colorectal cancer and other tumor types. Background: Autism spectrum disorder (ASD) is a developmental disorder that has increased in prevalence over the years. Diagnosis and subsequent intervention by the age of 3 is essential for development. However, recent estimates indicate that less than 50% of children receive an evaluation by age 3. Underserved populations are more likely to receive an evaluation at an older age. Several barriers in the diagnostic process have been identified; however, limited studies have aimed at understanding the specifics of these barriers among underserved populations and whether current strategies are beneficial for all children.
Objectives: The aim of this study was to …

Purpose: The Ki-67 proliferation index (Ki-67 PI) has been associated with meningioma recurrence, yet its clinical utility remains debated. Whether Ki-67 PI provides prognostic information across subgroups defined by both WHO grade and extent of resection remains to be investigated.
Methods: We analyzed 5,050 patients with intracranial meningiomas from the international PERNS cohort (42 centers, diagnosed between 1989-2019) who underwent surgical resection without postoperative radiotherapy. Ki-67 PI prognostic accuracy was assessed up to 10 years postoperatively by using ROC analyses and estimating its association with the risk of recurrence.
Results: Results demonstrated that the prognostic value of Ki-67 PI differed …

Mitochondrial Presequences Harbor Variable Strengths To Maintain Organellar Function, Youmian Yan, Baigalmaa Erdenepurev, Thiago N Menezes, Ian Collinson, Natalie M Niemi
2020-Current year OA Pubs
Hundreds of mitochondrial proteins rely on N-terminal presequences for organellar targeting and import. While generally described as positively charged amphiphilic helices, presequences lack a consensus motif and thus likely promote protein import into mitochondria with variable efficiencies. Indeed, the concept of presequence strength underlies biological models such as stress sensing, yet a quantitative analysis of what dictates strong versus weak presequences is lacking. Furthermore, the extent to which presequence strength affects mitochondrial function and cellular fitness remains unclear. Here, we capitalize on the MitoLuc protein import assay to define multiple aspects of presequence strength. We find that select presequences, including …

Amyotrophic lateral sclerosis (ALS) is a progressive disease that affects nerve cells in the brain and the spinal cord, leading to a broad range of neurological issues. Individuals with ALS experience progressive muscle weakness, dysphagia, slurred speech, and impaired movement and cognition. In addition to physical symptoms, around 15-20% of individuals with ALS also develop frontotemporal dementia (FTD). There are limited interventions to slow cognitive decline in this specific population. The Mediterranean-DASH Intervention for Neurodegenerative Delay (MIND) diet has been associated with slower cognitive decline and reduced risk in neurodegenerative disease among older adults; however, its effects have not been …

Background: A formal assessment of knowledge prior to the Ophthalmic Knowledge Assessment Program (OKAP) may help ophthalmology residents identify areas of weakness and organize their study plans.
Purpose: To assess the utility of an OKAP pre-test as a means of helping students prepare for the OKAP.
Methods: A 91-question OKAP pre-test was formulated from the American Academy of Ophthalmology self-assessment study bank and administered in January to 28 residents at two academic institutions. Residents were given the results of their January test scores with a breakdown by subtest before the OKAP. Overall and subtest scores from the …

The Effect Of Low And High-Carbohydrate Diets On Memory Performance In College Students, Samia Baugh, Zoe Zajko, Jamie Analuisa, Ivanna Jacome
Department of Nutrition Student Projects
Carbohydrates are the primary source of energy for the brain and play an important role in processes related to memory performance. The purpose of this study is to examine the impact of a low-carbohydrate diet compared to a high-carbohydrate diet on memory performance in college students aged 18–22 years. For the purposes of this study, a low-carbohydrate diet is defined as a diet providing fewer than 130 grams of carbohydrates per day, while a high-carbohydrate diet provides approximately 325 grams of carbohydrates. This population is particularly important to study because college students experience high academic demands that require strong memory …

Untangling Impacts Of Socioeconomic Position, Chronic Disease, And Low-Level Pm2.5 Exposure On Mortality Among Native American Medicare Beneficiaries, Judy Wendt Hess, Wenyaw Chan
Faculty, Staff and Student Publications
Ambient fine particulate matter (PM2.5) is associated with increased mortality at concentrations below current regulatory standards. Studies of low-level exposure often rely on large administrative cohorts whose geographic and demographic composition may influence observed associations. In a prior analysis, we observed an association between long-term PM2.5 and all-cause mortality among Native American Medicare beneficiaries living in zip codes within the lowest decile of PM2.5 exposure. The present study, a case-control analysis of 1,713,399 low-PM2.5-exposed beneficiaries enrolled in traditional Medicare during 2015-2016, evaluated whether this association could be explained by geographic context, socioeconomic position (SEP), or baseline health status. We used …

A 10-year-old female Pomeranian was referred after a hepatic cyst was incidentally identified during a routine health check. Ultrasound-guided simple drainage was initially performed, and cytological examination of the aspirated fluid confirmed a simple cyst. However, the cyst re-expanded to its original size within 2 weeks. Therefore, ultrasound-guided percutaneous sclerotherapy was elected using lower-concentration ethanol (83%), a readily available medical disinfectant, rather than conventional high-concentration ethanol. Approximately 50% of the aspirated volume was replaced with 83% ethanol and retained for 20 min to achieve effective epithelial ablation. Post-procedural follow-up revealed a transient increase in cyst size and serum alanine aminotransferase …

Autosomal dominant polycystic kidney disease (ADPKD), caused by the PKD1 c.10063C>A mutation, is a prevalent inherited feline disorder, but its allele frequency in Vietnam remains unidentified. This study aimed to determine the carrier frequency of this variant in British Shorthair, Maine Coon, and Ragdoll cats in Ho Chi Minh City and to correlate genotype with ultrasonographic phenotype. Ninety-seven client-owned cats were genotyped for the pkd1 mutation using PCR-RFLP. A subset of 47 cats also underwent abdominal ultrasonography to detect renal cysts. Genotypic results were correlated with sonographic findings, and risk factors were analyzed using chi-square tests and odds ratios …
