Drug development is a complicated, slow and expensive process with high failure rates. One strategy to mitigate these factors is to recycle existing drugs with viable safety profiles and have gained Food and Drug Administration approval following extensive clinical trials. Cardiovascular and neurodegenerative diseases are difficult to treat, and there exist few effective therapeutics, necessitating the development of new, more efficacious drugs. Recent scientific studies have led to a mechanistic understanding of heart and brain disease progression, which has led researchers to assess myriad drugs for their potential as pharmacological treatments for these ailments. The dopamine D3 receptor (D3R) is one of the most studied receptors involved in drug addiction. One of the most common strategies to treat substance use disorders is via D3R antagonism. The majority of the D3R antagonists synthesized so far have poor pharmacokinetic properties and/or lack selectivity toward D3R. In this thesis, the design, synthesis and biological evaluation of novel molecules that target the dopamine D1 receptor (D1R), D3R and the serendipitous discovery of molecules that target s receptors will be described.

Heat exposure studies over the last decade have shown little attention in assessing and reporting the psychometric properties of the various scales used to measure impacts of occupational heat stress on workers. A descriptive cross-sectional survey including 320 small- and large-scale mining workers was employed to assess the construct validity of the social impacts of occupational heat stress scale (SIOHSS) in the Western Region of Ghana in 2017. The primary focus of this manuscript comprises three sections. Initially, we introduce the concept of a simplified intuitionistic neutrosophic soft set. We impose an intuitionistic condition between the membership values of truth and falsity such that their sum does not exceed unity. Similarly, for indeterminacy, the membership value is a real number from the closed interval [0, 1]. Hence, the sum of membership values of truth, indeterminacy, and falsity does not exceed two. We present the notion of necessity, possibility, concentration, and dilation operators and establish some of its properties. Wildlife-aircraft collisions (wildlife strikes) pose a serious safety risk to aircraft. Raptors (i.e., hawks and owls) are one of the most frequently struck guilds of birds within North America. Although raptors (most notably red-tailed hawks [Buteo jamaicensis]) are commonly managed at most airports and military bases, there is no scientific information available regarding comparisons of the efficacy of raptor management programs for reducing raptor-aircraft collisions. Therefore, we conducted a study to examine the efficacy of 2 raptor hazard management programs implemented at Chicago's O'Hare International Airport (ORD). We investigated intramuscular administration of alfaxalone (5.3–10.0 mg/kg) as an immobilizing agent in free-ranging small Indian mongooses (Urva auropunctata) on the island of St. Kitts, West Indies. From 5–11 January 2020, we successfully immobilized 10 of 11 mongooses. Alfaxalone provided rapid onset (median = 3 min) of immobilization. Mean duration of immobilization was 16.8 ± 3 min. Mean recovery time was 5.6 ± 1 min. There was no effect of dose administered on induction, anesthesia, or recovery times at the dose range used. We concluded that alfaxalone represents an effective alternative to dissociative agents for the immobilization of free-ranging mongooses. …

More than 1.6 million Servere Acute Respiratory Syndrome Cronovirus 2(SARS-COV-2)tests were administered daily in the United States at the peak of the epidemic, with a significant focus on individual treatment. Here, we show that objective-driven, strategic sampling designs and analyses can maximize information gain at the population level, which is necessary to increase situational awareness and predict, prepare for, and respond to a pandemic, while also continuing to inform individual treatment. River otter (Lontra canadensis) populations in North America have been the focus of significant restoration efforts. Wildlife management agencies, concerned about the unintentional take of river otters incidental to beaver (Castor canadensis) trapping, may recommend techniques to avoid capturing river otters. River otter avoidance techniques that are ineffective or diminish trap performance for beavers are undesirable. We conducted a field evaluation in 2015 and 2016 in Wisconsin to assess how two trigger configurations (offset and center) on body-grip traps would affect the incidental capture rate of river otters during beaver trapping. While the coronavirus disease 2019 (COVID-19) pandemic wreaked havoc across the globe, we have witnessed substantial mis- and disinformation regarding various aspects of the disease. We conducted a cross-sectional study using a self-administered questionnaire for the general public (recruited via social media) and healthcare workers (recruited via email) from the State of Qatar, and the Middle East and North Africa region to understand the knowledge of and anxiety levels around COVID-19 (April–June 2020) during the early stage of the pandemic. An optimal experience termed flow can be experienced by users who are deeply involved in human-computer interaction (HCI). The electroencephalogram (EEG) provides an innovative way to assess in real-time if users are approaching or experiencing the flow state in various applications of HCI. However, the current literature remains unclear and inconsistent with regard to the relationship between EEG activity and flow experience. The objective of this research is to assess EEG activity corresponding to the state of flow. A laboratory experiment was conducted to collect EEG data of the flow and other states (including baseline) in the context of HCI.

Just-In-Time Adaptive Intervention (JITAI) is an emerging technique with great potential to support health behavior by providing the right type and amount of support at the right time. A crucial aspect of JITAIs is properly timing the delivery of interventions, to ensure that a user is receptive and ready to process and use the support provided. Some prior works have explored the association of context and some user-specific traits on receptivity, and have built post-study machine-learning models to detect receptivity. When attempting to mitigate the spread of an epidemic without the use of a vaccine, many measures may be made to dampen the spread of the disease such as physically distancing and wearing masks. The implementation of an effective test and quarantine strategy on a population has the potential to make a large impact on the spread of the disease as well. In this study, we used an integrative computational approach to examine molecular mechanisms underlying functional effects of the D614G mutation by exploring atomistic modeling of the SARS-CoV-2 spike proteins as allosteric regulatory machines. We combined coarse-grained simulations, protein stability and dynamic fluctuation communication analysis with network-based community analysis to examine structures of the native and mutant SARS-CoV-2 spike proteins in different functional states. Through distance fluctuations communication analysis, we probed stability and allosteric communication propensities of protein residues in the native and mutant SARS-CoV-2 spike proteins, providing evidence that the D614G mutation can enhance long-range signaling of the allosteric spike …

Smoking leads to disease and disability as well as harm nearly every organ of the body. Furthermore, smoking of tobacco is known to cause pulmonary dysfunction and lead to complications, pain, or even death. This study aimed to measure the risk factors for the respiration of carbon monoxide among smokers. A cross-sectional design was implemented by involving 156 smokers in Karyajaya Subdistrict, Palembang City. The Super Depo Sutorejo Surabaya (SDSS) Project was created to separate household waste into fractions, but its separation effect is unknown. This study assessed the significance of the project in sorting general waste into biodegradable, non-biodegradable, and assorted. The t-test compared the means of general and biodegradable waste (normally distributed). The non-biodegradable and assorted waste were not significantly distributed; therefore, the Wilcoxon Signed Ranks Test was used to compare their medians against general waste. This study pertains to stunting in children in Indonesian with z-scores of less than -2 Standard Deviation (SD) and less than -3 SD. Jakarta is one of the most air-polluted cities in the world, which can increase the health risk of commuting workers exposed to pollutants. This study aimed to determine the factors that directly and indirectly affect healthy behavior (wearing a mask) for commuters using the Bogor-Jakarta Commuter Line. Furthermore, a total of 155 respondents participated and the data collected were analyzed using descriptive and path analysis. Pre-lacteal feeding is widely known as a distraction to exclusive breastfeeding, and the malpractice continues to be prevalent in Indonesia. Therefore, this study aimed to explore the potential determinants of pre-lacteal feeding among mothers of infants below 24 months. A sample of 6,455 mother-infant pairs from the 2017 Indonesia Demographic and Health Survey (IDHS) was used. Also, multivariate logistic regression was employed to identify factors associated with pre-lacteal feeding practice. Socio-cultural concerns often restrict women's sexual rights and well-being living with HIV (WLHIV) in developing countries. These convergent parallel mixed methods study combined quantitative and qualitative approaches to explore factors influencing WLHIV sexual quality of life in Banten Province, Indonesia. A 2017 cross-sectional survey of 207 reproductive-aged WLHIV was analyzed using univariate and multivariate logistic regression statistics. Furthermore, in-depth interviews with a sub-sample of survey participants (n = 30) were thematically analyzed.
